Abstract
本発明は、植物原料から、簡便且つ安価に、大量の高純度グルコシルセラミド含有物を製造する方法を提供することを課題としてなされた。グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分をアルコール沈殿処理して、上清を回収する工程を含む、グルコシルセラミド含有物の製造方法により、大量の高純度グルコシルセラミドを得ることができる。なお、当該植物原料由来画分は、アルコール又は含水アルコールを用いて植物原料から抽出して得ることができ、当該抽出後に脂質除去処理されていることが好ましい。
Description
本発明は、グルコシルセラミド含有物の製造方法に関し、より詳細には植物原料からグルコシルセラミド含有物を製造する方法に関する。
近年、植物由来のグルコシルセラミド並びにステロール及びステロール配糖体が機能性素材として注目されている。
グルコシルセラミドはグルコセレブロシドとも呼ばれ、セラミドの1位のヒドロキシ基にグルコースがβ-グリコシド結合した構造を有する。なお、セラミドは、スフィンゴイド塩基と脂肪酸が酸アミド結合した構造を有する。図1にグルコシルセラミドの構造を示す。グルコシルセラミドは、例えば皮膚保湿効果や抗アトピー効果、抗腫瘍効果がある素材として期待されている。
ステロール配糖体は、動物組織にはほとんど含まれていない一方、植物組織には様々な種類が含まれていることが知られている。ステロール配糖体の具体的な例を図2に示す。ステロール配糖体は、例えばコレステロール濃度を低下させることにより抗高脂血症効果及び動脈硬化予防効果等を得られる素材として期待されている。
しかし、グルコシルセラミドとステロール配糖体は、極性基が同じグルコースであることから分子全体の極性が類似している。また、脂溶部のステロールとセラミド、並びにβ-グリコシド結合は弱アルカリに対して共通して安定な性質であるため、これらを簡便且つ高純度で分離することは困難であった。
植物原料由来のグルコシルセラミドは既に複数の販社から市販されている(例を表1に示す)が、市販されているグルコシルセラミド素材を、ケイ酸薄層クロマトグラフィー(TLC)にて解析すると、グルコシルセラミドとステロール配糖体が含まれていることがわかる。さらにこれらのRf値(スポットの移動距離を溶媒の移動距離で割った値)があまり変わらないため、TLCではほとんど分離されないことがわかる(図3)。
従って、ステロール配糖体と分離した、高純度の植物原料由来のグルコシルセラミドを得るには、植物原料から溶媒にてグルコシルセラミド含有画分を抽出した後、高速液体クロマトグラフィー(HPLC)を用いる等して、少量ずつ分離を行う必要があった。このため、高純度の植物由来グルコシルセラミドは非常に高価であり、その機能性を動物実験にて確かめようにも、実験のための十分な量を容易には入手できない状況であった。
このような状況において、簡便且つ安価に高純度のグリコシルセラミドを大量に製造する方法も研究されてきている。例えば、特許文献1には、植物原料をアルカリ性エタノールで加水分解し、次いでヘキサン、アセトン及び水の混合溶媒による抽出工程に供することで、グリコシルセラミド含有抽出物を得る方法が記載されている。しかしながら、当該方法では、グリコシルセラミドを通常12~50重量%含有する抽出物が得られるに留まっており、さらにステロール配糖体は分離されずに元の原料濃度に依存してそのまま残存している。
本発明は、グルコシルセラミド及びステロール配糖体を含有する植物原料から、簡便且つ安価に、大量の高純度グルコシルセラミド含有物を製造する方法を提供することを目的とする。
本発明者らは、驚くべき事に、グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分をアルコール沈殿処理して、上清を回収することで、簡便且つ安価に、大量の高純度グルコシルセラミド含有物を製造することが可能であることを見出し、さらに改良を重ねて本発明を完成させるに至った。
すなわち、本発明は例えば以下の項1~6のグルコシルセラミド含有物の製造方法を包含する。

グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分をアルコール沈殿処理して、上清を回収する工程を含む、グルコシルセラミド含有物の製造方法。
項2.
植物原料由来画分が、アルコール又は含水アルコールを用いて植物原料から抽出されて得られたものである、項1に記載のグルコシルセラミド含有物の製造方法。
項3.
植物原料由来画分が、アルコール又は含水アルコールを用いて植物原料から抽出され、さらに脂質除去処理されて得られたものである、項2に記載のグルコシルセラミド含有物の製造方法。
項4.
脂質除去処理が、ヘキサン沈殿処理、アセトン沈殿処理、ヘキサン及びアセトン混合溶媒沈殿処理、又はこれらの組み合わせを含む工程により行われる、項3に記載のグルコシルセラミド含有物の製造方法。

アルコール沈殿処理が、エタノール及び/又はイソプロパノール沈殿処理である、項1~4のいずれかに記載のグルコシルセラミド含有物の製造方法。

またさらに、本発明者らは、グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分をアルコール沈殿処理して、沈殿を回収することで、簡便且つ安価に、大量の高純度ステロール配糖体含有物を製造することも可能であることを見出した。よって、本発明は例えば以下の項A~Fに記載のステロール配糖体含有物の製造方法も包含する。

なお、これらの製造方法から明らかなように、当該グルコシルセラミド含有物はグルコシルセラミド含有抽出物といえる。また、当該ステロール配糖体含有物ステロール配糖体含有抽出物といえる。
本発明のグルコシルセラミド含有物の製造方法によれば、植物原料から、簡便且つ安価に、大量の高純度グルコシルセラミド含有物を製造することができる。また、製造工程において毒性の高い溶媒を用いることがないため、安心安全な高純度のグルコシルセラミドを提供することができる。このため、例えば、食品原料、化粧品原料等として試験や実施を行うために必要な量のグルコシルセラミドを、安価で提供することができる。
以下、本発明について、さらに詳細に説明する。
本発明は、グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分を、アルコール沈殿処理して上清を回収する工程を含む、グルコシルセラミド含有物の製造方法に係る。
グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分は、植物原料から得られ、グルコシルセラミド及びステロール配糖体を含有するものであれば特に制限されない。通常、植物原料からグルコシルセラミドを含有する画分を抽出等して得た場合は、当該画分にステロール配糖体も含まれる。これらを簡便に分離できないため、安価な高純度のグルコシルセラミドを大量に得ることが、より困難となっている。
植物原料由来画分を得るために用いる植物原料としては、グルコシルセラミド及びステロール配糖体を得られるものであれば特に制限されないが、例えば小麦(小麦胚芽)、大豆、米(米糠・米胚芽)、綿実、菜種、トウモロコシ(コーン胚芽)、サトウキビ、ゴマ、ピーナッツ、蒟蒻芋、柚子、蜜柑、甜菜等が挙げられる。主にこれらの食用部位を用いることができるが、これら植物原料から油を製造・精製する場合に、油の抽出残渣や油脂の精製過程で生じる脱ガム物、あるいは米糠等の低コスト資源がコストの面で特に好ましい。なお、植物原料は1種又は2種以上を組み合わせて用いることができる。
グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分として好ましいものとして、上記植物原料から有機溶媒又は含水有機溶媒、より好ましくはアルコール又は含水アルコール等により、植物原料から抽出されて得られる画分が挙げられる。
抽出に用いるアルコールは特に制限されず、例えば炭素数1~6の一価アルコール、好ましくは炭素数1~4の一価アルコールを用いることができ、具体的にはメタノール、エタノール、プロパノール、ブタノール、イソプロパノール、ペンタノール、ヘキサノール等を用いることができる。中でもエタノールが好ましい。なお、アルコールは、1種又は2種以上を組み合わせて用いることができる。
また、植物原料からの抽出には、アルコールに代えて又はアルコールに加えて、アルコール以外の有機溶媒や水を用いてもよい。アルコール以外の有機溶媒としては、グルコシルセラミドが抽出でき、毒性が低いものであれば特に制限されない。クロロホルム等の毒性を有する非食品系溶媒は好ましくない。これらの有機溶媒や水を、アルコールとは別に用いて各抽出工程を順々に行ってもよいし、アルコールと混合した上で抽出に用いてもよい。また、有機溶媒を水と混合した上で用いてもよい。アルコールと水を混合したものが含水アルコールである。含水アルコールの含水率は特に制限されないが、50%(v/v%)以下のものが好ましく、30%(v/v%)以下のものがより好ましい。なお、これらアルコール、有機溶媒及び水は1種又は2種以上を混合して用いることができる。
具体的な抽出操作は、特に限定されず、常法にて行うことができる。例えば、冷浸・温浸等の浸漬法、パーコレーション法等により行うことができる。好適な一例として、例えば、植物原料にアルコール又は含水アルコールを加え常温で1分~5時間程度静置又は撹拌を行う方法が例示できる。
このようにして得られる画分、及び当該画分を濃縮乾固するなどして固体としたものを、そのまま“グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分”として用いてもよいし、これをさらに脂質除去処理して、“グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分”として用いてもよい。脂質除去により、得られるグルコシルセラミド含有物の純度がさらに高まり得るため、好ましい。
好ましい脂質除去処理方法としては、特に限定するものではないが、例えばヘキサン及び/又はアセトン沈殿処理が挙げられる。当該処理は、常法により行うことができる。例えば、ヘキサン沈殿処理及びアセトン沈殿処理とも、試料に対して適量(例:試料1gに対し5~10mL)のヘキサン又はアセトンを加え、撹拌後遠心処理又は減圧ろ過して沈殿又は不溶部を回収する、という操作を行うことで実施できる。さらに、ヘキサン及びアセトンの混合溶媒を用いた沈殿処理(ヘキサン/アセトン沈殿処理)を行ってもよい。この場合ヘキサンとアセトンの混合比率は適宜設定できる(例えば容量比がヘキサン:アセトン=1:1)。ヘキサン沈殿処理、アセトン沈殿処理、並びにヘキサン/アセトン沈殿処理は繰り返し行ってもよく、また、単独又は任意の順番で組み合わせて複数回行なってもよい。組み合わせる場合は、例えば各処理をそれぞれ1~5回、好ましくは1~3回ずつ組み合わせるのが好ましい。なお、アセトン沈殿のみでは除去しきれない色素が、ヘキサンを用いることによって除去可能となることがあるため、ヘキサン沈殿処理、あるいはヘキサン/アセトン沈殿処理を用いることが好ましい。好ましい脂質除去処理の一態様として、アセトン沈殿処理を1~5回繰り返した後、ヘキサン/アセトン沈殿処理を1~5回繰り返すという処理を挙げることができる。また、試料としては、上述の植物原料から有機溶媒又は含水有機溶媒等により抽出して得られる画分を、減圧乾燥する等して、濃縮乾固したものを用いるのが好ましい。なお、限定的な解釈を望むものではないが、ヘキサン沈殿処理により主にリン脂質及びグリセロ糖脂質を除去でき、また、アセトン沈殿処理により主に中性脂質を除去できると考えられる。
また、脂質除去処理として、上述のヘキサン沈殿処理及び/又はアセトン沈殿処理に代えて又は加えて、アルカリ加水分解処理を行ってもよい。当該処理に用いることができるアルカリ溶液としては、例えば水酸化ナトリウム溶液等を挙げることができ、当該溶液の溶媒としては水、エタノール等を用いることができる。具体的な処理方法としては、特に限定されず常法にて行うことができる。
アルカリ加水分解処理は、ヘキサン沈殿処理及び/又はアセトン沈殿処理の前後どちらに行ってもよいが、前に行うことが好ましい。なお、アルカリ加水分解処理により、主にグリセロリン脂質とグリセロ糖脂質などのエステル脂質が分解され、極性脂質画分から除去されると考えられる。
このようにして得られる脂質除去画分も、“グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分”として好ましく用いることができる。なお、当該脂質除去処理を経て得られる画分を濃縮乾固するなどして固体としたものは、植物原料由来画分のほとんど(90質量%以上、好ましくは95質量%以上、より好ましくは97質量%以上)がグルコシルセラミド及びステロール配糖体からなる。
上述のようにして得られる“グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分”を、アルコール沈殿処理して上清を回収することにより、グルコシルセラミドが分離され、高純度のグルコシルセラミド含有物を得ることができる。なお、上述した方法は植物原料由来画分を得るための例であって、これに限定される訳ではない。公知の方法(例えば上述の特許文献1に記載の方法)によって得られる“グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分”であっても、これをアルコール沈殿処理して上清を回収することにより、グルコシルセラミドが分離され、高純度のグルコシルセラミド含有物を得ることができる。
脂質除去処理を経て得られた植物原料由来画分を用いる場合は、当該画分のほとんどがグルコシルセラミド及びステロール配糖体であるので、アルコール沈殿処理を行うことにより、特に高純度(乾燥質量換算で、通常90質量%以上、好ましくは95質量%以上、より好ましくは97質量%以上)のステロール配糖体含有物(沈殿)及び高純度(乾燥質量換算で、通常90質量%以上、好ましくは95質量%以上、より好ましくは97質量%以上)のグルコシルセラミド含有物(上清)を得ることができる。このように、本発明は、グルコシルセラミド含有物の製造方法のみならず、上述した工程を経てステロール配糖体含有物を製造する方法にも係る。
アルコール沈殿処理に用いるアルコールとしては、特に制限されないが、例えば炭素数1~6の一価アルコール、好ましくは炭素数1~4の一価アルコールを用いることができ、具体的にはエタノール及びイソプロパノールを好ましく用いることができる。アルコールは1種又は2種以上を混合して用いることができる。
また、アルコール沈殿処理は、常法により行うことができる。例えば、植物原料由来画分に対して適量(例:濃縮乾固画分1gに対し約20mL)のアルコールを加え、撹拌後遠心処理又は減圧ろ過する、という操作により行うことができる。アルコール沈殿処理は繰り返し行ってもよい。グルコシルセラミドはアルコール沈殿処理後の上清に含まれるため、アルコール沈殿処理を繰り返し行う場合は、各処理毎に上清を回収しておき、合わせてもよい。なお、多回数繰り返すと上清へのステロール配糖体の混入が起こるおそれがあるため、アルコール沈殿処理は、通常1~5回、好ましくは1~3回、より好ましくは1~2回行う。
当該処理により得られる上清をグルコシルセラミド含有物として利用してもよいし、当該上清を濃縮乾固して固形物(粉末)とし、これをグルコシルセラミド含有物として利用してもよい。上清は常法に従って濃縮乾固することができる。例えば、当該上清をロータリーエバポレーター等で濃縮乾固し、必要に応じて洗浄すればよい。
このようにして得られるグルコシルセラミド含有物は、例えば、経口摂取又は経皮摂取により、肌の保湿、コラーゲンの分解抑制、メラニン生成抑制、アトピー症状緩解等のために好ましく用いることができる。また、グルコシルセラミドの大腸腺腫(ポリープ)発症抑制作用も知られており、このためにも用いることができる。さらに、未だ解明されていないグルコシルセラミドの消化吸収などの体内動態、急性・慢性毒性などの安全性試験や、グルコシルセラミドの摂取による遺伝子発現変動解析のためにも用いることができる。さらにまた、当該グルコシルセラミド含有物は、特に優れた脂質代謝改善作用(LDL-コレステロール低下作用、アディポネクチン分泌促進作用等)、抗高血糖作用を有すると考えられる。
よって、本発明は、当該グルコシルセラミド含有物を有効成分として含む保湿剤、コラーゲン分解抑制剤、メラニン生成抑制剤、抗アトピー剤、抗高脂血症剤、抗高血糖剤をも提供する。さらに、本発明は、当該グルコシルセラミド含有物を含みこれらの効果を奏する化粧品や食品添加物も提供する。またさらに、本発明は、当該グルコシルセラミド含有物を含む食品も提供し、当該食品からなる保湿剤、コラーゲン分解抑制剤、メラニン生成抑制剤、抗アトピー剤、抗高脂血症剤、抗高血糖剤をも提供する。
以下、本発明を具体的に説明するが、本発明は下記の例に限定されるものではない。なお、本発明のグルコシルセラミド含有物の製造方法の工程の概要を図4に示す。
グルコシルセラミド及びステロール配糖体含有画分の抽出
コーン胚芽4kgにエタノール8Lを加え、1時間撹拌した後、ろ別して可溶部を回収した。そして、これをロータリーエバポレーターにより濃縮乾固した結果、抽出物が約480g得られた。これに常法に従い脱ガム処理を行い、80gのコーン由来粗グルコシルセラミド画分(脱ガム物)を得た。
コーン胚芽4kgにエタノール8Lを加え、1時間撹拌した後、ろ別して可溶部を回収した。そして、これをロータリーエバポレーターにより濃縮乾固した結果、抽出物が約480g得られた。これに常法に従い脱ガム処理を行い、80gのコーン由来粗グルコシルセラミド画分(脱ガム物)を得た。
脂質除去処理
コーン由来粗グルコシルセラミド画分72g(脱ガム物)に対し、アセトンを700mL加えて激しく撹拌し、遠心分離して沈殿を回収した。当該アセトン沈殿処理を5回繰り返した。さらに、アセトン沈殿処理にて得られた沈殿に対し、ヘキサン/アセトン(容量比1:1)混合溶媒を350mL加えて激しく撹拌し、遠心分離して沈殿を回収した。当該ヘキサン/アセトン沈殿処理を3回繰り返した。
コーン由来粗グルコシルセラミド画分72g(脱ガム物)に対し、アセトンを700mL加えて激しく撹拌し、遠心分離して沈殿を回収した。当該アセトン沈殿処理を5回繰り返した。さらに、アセトン沈殿処理にて得られた沈殿に対し、ヘキサン/アセトン(容量比1:1)混合溶媒を350mL加えて激しく撹拌し、遠心分離して沈殿を回収した。当該ヘキサン/アセトン沈殿処理を3回繰り返した。
以上の処理により得られた沈殿30gをコーン由来グルコシルセラミド画分とした。
エタノール沈殿処理
コーン由来グルコシルセラミド画分30gにエタノール600mL(1g/20mL)を加え、撹拌し、遠心分離して上清を回収した。当該エタノール沈殿処理を2回繰り返し、得られた上清を全て合わせてグルコシルセラミド含有画分とした。なお、当該エタノール沈殿処理で得られた沈殿(ステロール配糖体含有物)も回収した。
コーン由来グルコシルセラミド画分30gにエタノール600mL(1g/20mL)を加え、撹拌し、遠心分離して上清を回収した。当該エタノール沈殿処理を2回繰り返し、得られた上清を全て合わせてグルコシルセラミド含有画分とした。なお、当該エタノール沈殿処理で得られた沈殿(ステロール配糖体含有物)も回収した。
当該グルコシルセラミド含有画分をロータリーエバポレーターで濃縮乾固し、得られた固形物をヘキサンに溶解してチューブへ移し、さらに4倍容量のアセトンでアセトン沈殿して、沈殿を18.4g得た。当該沈殿を、以下グルコシルセラミド含有物として用いた。
蒸発光散乱検出-高速液体クロマトグラフィー(HPLC-ELSD)による純度の検定
上述のようにして得られたグルコシルセラミド含有物10mgを、クロロホルム/メタノール(2:1)液100mlに溶解させ、そこから15μlを注入してHPLC-ELSDにより解析してクロマトグラムを得た。得られたクロマトグラムを図5に示す。上図にグルコシルセラミド含有物が1.5μg含まれる溶液を解析した結果を、下図にグルコシルセラミド含有物が10μg含まれる溶液を解析した結果を示す。なお、当該HPLC-ELSD解析の条件は以下の通りである。
上述のようにして得られたグルコシルセラミド含有物10mgを、クロロホルム/メタノール(2:1)液100mlに溶解させ、そこから15μlを注入してHPLC-ELSDにより解析してクロマトグラムを得た。得られたクロマトグラムを図5に示す。上図にグルコシルセラミド含有物が1.5μg含まれる溶液を解析した結果を、下図にグルコシルセラミド含有物が10μg含まれる溶液を解析した結果を示す。なお、当該HPLC-ELSD解析の条件は以下の通りである。

当該クロマトグラムの全ピーク面積に対するグルコシルセラミドのピーク面積はいずれも98%であった。従って、得られたグルコシルセラミド含有物のグルコシルセラミド純度は98%であることが確認できた。
なお、同様にして、エタノール沈殿処理で得られた沈殿(ステロール配糖体含有物)を解析したところ、得られたクロマトグラム(図6)のピーク面積から、当該ステロール配糖体含有物のステロール配糖体純度は98%であることが確認できた。
Claims (6)
- グルコシルセラミド及びステロール配糖体を含有する植物原料由来画分をアルコール沈殿処理して、上清を回収する工程を含む、グルコシルセラミド含有物の製造方法。
- 植物原料由来画分が、アルコール又は含水アルコールを用いて植物原料から抽出されて得られたものである、請求項1に記載のグルコシルセラミド含有物の製造方法。
- 植物原料由来画分が、アルコール又は含水アルコールを用いて植物原料から抽出され、さらに脂質除去処理されて得られたものである、請求項2に記載のグルコシルセラミド含有物の製造方法。
- 脂質除去処理が、ヘキサン沈殿処理、アセトン沈殿処理、ヘキサン及びアセトン混合溶媒沈殿処理、又はこれらの組み合わせを含む工程により行われる、請求項3に記載のグルコシルセラミド含有物の製造方法。
- アルコール沈殿処理が、エタノール及び/又はイソプロパノール沈殿処理である、請求項1~4のいずれかに記載のグルコシルセラミド含有物の製造方法。
- 植物原料が、小麦、大豆、米、綿実、菜種、トウモロコシ、サトウキビ、ゴマ、ピーナッツ、蒟蒻芋、柚子、蜜柑及び甜菜からなる群より選択される少なくとも1種である、請求項1~5のいずれかに記載のグルコシルセラミド含有物の製造方法。
