Description
生のタモギタケ50kgを300Lの沸騰水に入れ、5分間ボイルを行った。ボイル後水道水で25℃に冷却し、65℃の熱風式乾燥機で8時間乾燥した。乾燥後粉砕機で0.2mm以下に粉砕し、タモギタケ乾燥粉末3.5kgを得た。繰り返し操作で得られたタモギタケ乾燥粉末20kgをΦ300mm、高さ1,000mmのステンレス製カラムに入れ、エタノールを入れてフタをした。加圧エアをカラム内圧が0.4MPaになるよう上から入れ、カラム下から抽出液を回収した。エタノールは合計100L使用して、カラム下から80L抽出液を回収した。繰り返し操作で得られた抽出液400Lを減圧濃縮機で50℃、-0.95MPa、毎時60L蒸発で濃縮乾固して、4.5kgのペースト状の抽出物を得た。この抽出物のグルコシルセラミド含有量は4.6%であった。
製造例1で得られた抽出物500gにエタノール5Lを加え、ホットスターラー上で加熱攪拌しながら完全に溶解した。次に4℃で2時間以上冷却して沈殿を生じさせた後、5000rpm、15℃にて30分間遠心した。上清を回収後、等量の蒸留水を加え、室温にて一晩攪拌した。5000rpm、15℃にて30分間遠心後、上清と沈殿下層(茶色の沈殿)を吸引により除去し、沈殿上層を回収し、エバポレーターによりエタノールと水を留去した。回収物は227g、グルコシルセラミド含有量は6.2%であった。
製造例1で得られた抽出物1000gにエタノール5Lを加え、ホットスターラー上で加熱攪拌しながら完全に溶解した。次に4℃で2時間以上冷却して沈殿を生じさせた後、5000rpm、15℃にて30分間遠心した。上清を回収後、等量の蒸留水を加え、室温にて一晩攪拌した。5000rpm、15℃にて30分間遠心後、上清と沈殿下層(茶色の沈殿)を吸引により除去し、沈殿上層を回収し、エバポレーターによりエタノールと水を留去した。回収物は500g、グルコシルセラミド含有量は5.5%であった。
Claims (3)
- 植物および菌類から抽出したグルコシルセラミドを精製する方法であって、
(1)植物および菌類から有機溶媒抽出を行った抽出液を濃縮し、抽出濃縮物を得る工程、
(2)前記抽出濃縮物をエタノールに溶解する工程、
(3)前記エタノール溶液1重量部に対して水を0.5〜2重量部加えた後、遠心分離する工程、
(4)遠心分離の後、上清および沈殿下層を除去して、グルコシルセラミドを含有する沈殿上層を回収する工程、
を含むことを特徴とする、方法。 - (5)前記回収した沈殿上層から、エタノールと水を留去する工程、
をさらに含む、請求項1に記載の方法。 - 植物および菌類が米、小麦、大豆、トウモロコシ、こんにゃく芋、タモギタケ、シイタケ、なめこ、マイタケ、エノキタケ、ひらたけ、ブナシメジ、エリンギ、アガリクス、マッシュルームからなる群より選択される,請求項1−2のいずれかに記載の方法。
