Abstract
Description
角質層は、皮膚の最上位にあって層状をなし、種々の薬剤が皮膚から浸透するのを防ぐ、バリヤーの働きを行っている。一般的には、皮膚のバリヤー作用の約50~約90%は角質層で行われている。表皮層では角質層ほどのバリヤー作用を果たさないが、残りの約10%以上のバリヤー作用を果たしている。一方、真皮は、真皮層と表皮層の接合部付近に豊富な毛細血管網があり、そのため、薬剤が一度真皮の深さに到達すると、その毛細血管網を伝わって、より深部の組織(毛包、筋肉等)に素早く拡散する。そして、毛細血管から血液循環を経由して薬剤が全身に拡散される。
そこで、薬剤の皮膚透過性を上げるための方法の一つとして、特許文献1に示されるように、微小針(マイクニードルまたはマイクロシリンジ)を使用し、角質層を局所的に破壊して薬剤を真皮層に強制的に投与すると言うことが試みられてきた。
この目的で使用される微小針は、真皮層に微小針が到達すればよいことから、その針の長さは30μm以上であることが望ましく、その針を支持するために必要な基板があればよいとされている。そして、この微小針は、神経の末端が存在する真皮層に到達しないので痛くない。それ故、小児などに恐怖感を与えることなく薬剤の投与ができると言う長所が存在する。
しかし、このような衝撃力を利用したマイクロニードルの穿刺方法は、マイクロニードルの微小針が皮膚穿刺時に折れ易く、また過度の皮膚表面の損傷を招くことにもなる。そこで、より衝撃の少ない、ソフトな穿刺用のデバイスが求められていた。そこで、筒状の中にピストンを挿入し、ピストンを手で押し、ゆっくりと皮膚を穿刺する注射器形の剣山デバイスが報告されている(特許文献4)。
更には、当該アプリケータを用いて、皮膚にマイクロニードルを穿刺後、アプリケータを取り去る際には、剣山型マイクロニードルが皮膚にテープで貼付され留置されるような、テープ状チップ(マイクロニードルが収納された粘着テープ)が設置されたアプリケータを提供する。
更に、本発明者らは、皮膚がどのように盛り上がるのかを評価する器具(図16)を作製して、デバイスのサイズとデバイスにかかる力の位置によって皮膚表面がどう変化して盛り上がるのかを図17のごとく検討した。その結果、図18や表6~表7に示されるように、デバイスのサイズや、デバイスを押す力の位置に関わらず、盛り上がる皮膚表面はデバイスに対して垂直に上昇することが分かった。そのため、デバイスに設置された剣山型マイクロニードルは、皮膚表面に垂直に突き刺さることを見出した。
更に、皮膚固定部材を配置する位置は、設置されるマイクロニードルを中心にして点対称に配置する、またはマイクロニードルを中心にした正多角形の角に配置することがよいことが分かった。即ち、マイクロニードルを中心にして、上記の位置に皮膚固定部材が配置されていると、このようなアプリケータを押圧すると皮膚表面が垂直に上昇する。
その結果、支持体両面の粘着剤の粘着強度(粘着力)の差が10~15%以上になれば、アプリケータ等からテープ状のチップが剥離して、皮膚に固定付着したままテープ状のチップが留置されることを見出した。更に、アプリケータ等を斜めに引き剥がす場合には、より皮膚に留置され易くなることを見出した。また、弱い粘着剤の形状によりテープ状チップが、より皮膚に留置され易くなることも見出した。本発明者らは、これらの知見に基き、本発明を完成した。

なお、皮膚固定部材の表面に弱い粘着剤を塗布し、カバーフィルムと接着することによって、これをマイクロニードルの微小針の保護シートとして使用でき、微小針の損傷するのを防ぐことができる。本発明のチップ状器具を使用するためには、ます、このカバーシートを剥離させ、このチップ状器具を皮膚表面に設置する。
図2で示されるように、本発明のマイクロニードルデバイスを使用する場合、特に図1(1)の下駄型アプリケータの場合には、アプリケータの設置の方向性によって、図3と図4に示されるように、皮膚表面の盛り上がり距離が異なっている。しかし、図1(2)と(3)のデバイスにおいては、図5と図6に示されるようにデバイスの設置の方向性に影響を受けることはなかった。
下駄型アプリケータを用いる場合、例えば、図13に示されるように腕の方向に垂直設置し、設置した位置からアプリケータを約12mm押し下げる。そうすると、皮膚固定部材の間で形成される領域(距離aと距離bで形成される領域)内に、皮膚が盛り上がってくる。盛り上がった皮膚の高さ(皮膚固定部材で固定された皮膚と盛り上がった皮膚の頂点との間の距離)は,測定用にアプリケータ平板中央に開けた8mmφの穴を利用してノギスで測定する。
皮膚固定部材としては、材質の点で特に限定されないが、加工性の点で、ポリエチレン,ポリプロピレン,ポリ塩化ビニル,アクリル,ポリエチレンテレフタレート,ポリスチレン,アクリロニトル・ブタジエン・スチレン共重合体,ポリカーボネート,ポリアミド,フッ素樹脂,ポリブチレンテレフタレート等の樹脂が好適である。
押圧用に平板上に設置された突起物(構造)は、指で押圧するために適切なサイズであることが望ましく、形状は特に限定されるものではない。好ましくは、図23~図24に示される円柱状または半球状の突起物であり、更には図25~図26に示されるアーチ状の構造物を挙げることができる。これらの押圧用部材の材質は、平板部材の材質や皮膚固定部材の材質と同じものを使用することができる。更には、これらの部材(押圧用部材、平板部材、皮膚固定部材)を射出成型等で一度に形成することも出来る。
押圧用突起物(構造)の設置位置は、マイクロニードルの設置位置であるデバイスの中央部が好ましい。即ち、マイクロニードルの位置と同じ位置のデバイス平板の上部に設置することが好ましい。アーチ状の構造物の場合には、指で押圧する位置がデバイスの中央部に来るように設定することが好ましい。
なお、本発明のデバイスの平板上に、押圧用の突起物(構造)を設置しない場合には、平板上に押圧位置を明示するための印刷又はシールを設置することが好ましい。更には、平板の押圧位置を少し窪まして、押圧位置を明示することも好ましい。
一方、アプリケータデバイスのサイズが固定されれば、それに対応する形で、マイクロニードルの基盤のサイズが決まることになる。
また、本発明のアプリケータデバイスは、マイクロニードルの保護を皮膚固定部材間に貼付したカバーフィルムで行なうことができる。このカバーフィルムが存在することにより、マイクロニードルの微小針は、外部との不用意な接触から保護されており、本発明のアプリケータデバイスの携帯が容易となっている。そして、このカバーフィルムを剥離することにより、簡便に皮膚表面に本発明のアプリケータデバイスを設置し、マイクロニードルで皮膚を穿刺することができる。
また、図3~図6に示されるように、皮膚の盛り上がり高さは、皮膚固定部材の間で形成される領域(距離aと距離bで形成される領域)で決まってくる。それ故、皮膚固定部材の間隔としては5~70mmの間が好ましく、より好ましくは10~40mmを挙げることができる。更に、皮膚固定部材の間で形成される領域(距離aと距離bで形成される領域)としては、5×5mm~70×70mmの範囲が好ましく、より好ましくは10×10mm~40×40mmの範囲を挙げることができる。
本発明のアプリケータの平板の形状は、円形、楕円形、正方形や長方形等の四角形、五角形、六角形等の多角形が挙げられる。より好ましい形状としては、円形、正方形や長方形を挙げることができる。本発明のアプリケータの平板のサイズは、主に皮膚固定部材間の距離が5~70mmで規定され、皮膚固定部材の幅に影響される。例えば、図7のように皮膚固定部材の幅が5mmであれば、25~60mmの長さの辺を持つ正方形や長方形を挙げることができる。好ましくは、25~40mmの長さの辺を持つ正方形や長方形を挙げることができる。
皮膚固定部材のサイズとしては、例えば四角柱であれば5~7mmの長さの辺を持つ四角柱であり、5~7mmφの半球である。
本発明のマイクロニードル(剣山型)としては、種々の材質、用途の公知のマイクロニードルを使用できる。例えば、シリコンやセラミックス等のシリコン製、例えば、ステンレス鋼、タングステン鋼、ニッケル合金、モリブデン、クロム、コバルト、チタン及びその合金等の金属製のマイクロニードル;例えば、ポリ乳酸、ポリグリコール酸、乳酸・グリコール酸共重合体等の脂肪族ポリエステル樹脂に代表される生体内分解性樹脂製のマイクロニードル;例えば、マルトース,ラクトース,スクロース,マンニトール,ソルビトール等の多糖類製のマイクロニードル等を挙げることができる。なかでも、生体内分解性材料である脂肪族ポリエステル樹脂製のマイクロニードルや多糖類製のマイクロニードルは、比較的低強度であり、衝撃に弱く、折れ易い又は曲がり易い傾向にあるが、本発明のアプリケータを使用することで、マイクロニードルの針先の折損を起こすことなく、または、軽減して、皮膚表面にマイクロニードルを確実に穿刺することができる。それ故、これまで効果的に使用することが難しかった。ポリ乳酸,ポリグリコール酸,乳酸・グリコール酸共重合体等の脂肪族ポリエステル樹脂に代表される生体内分解性樹脂製のマイクロニードル、あるいは、マルトース,ラクトース,スクロース,マンニトール,ソルビトール等の多糖類製のマイクロニードルを製品として市販することが可能になった。
なお、マイクロニードル(剣山型)の全体形状は円形が望ましく、サイズは5~15mmφを挙げることができる。好ましくは、9~13mmφを挙げることができる。
また、本発明方法では、皮膚表面にマイクロニードルを穿刺後、粘着テープによりマイクロニードルを皮膚上に留置させることも簡単に行うことができる。従って、上記の生体内分解性材料のマイクロニードルを用いることにより、薬剤の吸収性向上や薬剤の徐放性向上などを図ることができる。
本発明の弱い粘着剤とは、上記粘着テープの支持体をアプリケータデバイスの平板に接着固定させるものである。粘着テープよりも粘着力が弱いものであるため、粘着テープが皮膚に貼付された際には、平板に固定された粘着テープが剥離することになる。このような弱い粘着テープ剤としては、市販のものを適宜選択して使用することができる。更には市販の両面テープ等を使用することができる。
上記粘着テープは、剣山型マイクロニードルと共に、盛り上がってきた皮膚に密着して、アプリケータから脱離する。このように、本発明のアプリケータデバイスを皮膚に押し付けるだけで、他に特別な操作をすることなく、剣山型マイクロニードルを皮膚に貼付することができる。
本発明のテープ状チップが、アプリケータから剥離して皮膚に留置するためには、アプリケータ側の粘着剤の粘着力が、皮膚側の粘着剤の粘着力の85~90%以下である必要がある。また、テープ状チップの両面に使用される粘着剤が同じ材質である場合、あるいは粘着剤の粘着強度(N/cm2)が同じである場合には、粘着力は粘着剤の接着面積に比例する。そこで、テープ状チップを皮膚に留置させ、アプリケータから脱離させるためには、アプリケータ用の粘着剤の面積は、皮膚用の粘着剤の面積の85~90%以下である必要がある。
本発明のテープ状チップの形状は、円形、方形あるいは長方形の形が使用され、好ましくは円形、長方形のものを挙げることができる。
本発明の粘着剤とは、公知の粘着剤のことであり、例えばアクリル系重合体からなるアクリル系粘着剤、例えばスチレン-イソプレン-スチレンブロック共重合体,スチレン-ブタジエン-スチレンブロック共重合体等のスチレンブロック共重合体、例えばポリイソプレン,ポリイソブチレン,ポリブタジエン等のゴム系粘着剤、例えばシリコンゴム,ジメチルシロキサンベース,ジフェニルシロキサンベース等のシリコン系粘着剤、例えばポリビニルメチルエーテル,ポリビニルエチルエーテル,ポリビニルイソブチルエーテル等のビニルエーテル系粘着剤、例えば酢酸ビニル-エチレン共重合体等のビニルエステル系粘着剤、例えばジメチルテレフタレート,ジメチルイソフタレート,ジメチルフタレート等のカルボン酸成分とエチレングリコール等の多価アルコール成分からなるポリエステル系粘着剤を挙げることができる。これらの粘着剤は、いずれか1種を使用しても2種以上を配合して用いてもよい。
剥離されやすい形状とは,アプリケータから支持体が剥離されるときに最初に負荷がかかる部分の負荷が少なくなる形状であり,例えば,図34に示される鋭角な形状などが剥離される一つの切っ掛けを持つ形状である。更に、粘着剤が塗布される位置においても調整が可能である。例えば、図35に示されるように、同じ粘着剤の塗布面積であっても塗布の位置を変えることによって、剥離されやすさを調整することができる。また、図40(右図)に示されるテープ状チップの外周より内部の位置に粘着剤を塗布する場合と、図40(左図)に示される外周より突出させた位置に粘着剤を塗布する場合とを比較すると、図40(右図)に示される場合の方が、剥離が容易であることが見出された。このことから、好ましい粘着剤の塗布形状と位置として、例えば図36~図39、図41を挙げることができる。なお、好ましい粘着剤の設置位置としては、テープ状チップの外周から、距離として0.1mm以上離れたところに位置するように、粘着剤を設置することが望ましい。
なお、これらに使用する粘着剤としては、両面テープを適宜必要な形状にカットして使用することができる。例えば、図32に示されるように、三角形の両面テープを4箇所に設置して、支持体を接着することができる。即ち、マイクロニードルを皮膚用の粘着テープ上に固定し、上記粘着テープの支持体を両面テープ等の弱い接着部材でアプリケータの平板に固定した器具(下駄型アプリケータ)である。
また、平板部材上に最初に剣山型マイクロニードルを設置する操作を行ない、その後に、平板部材の両端に四角柱を接着するように、アプリケータデバイスの製造順序を変えてもよい。
図16(1)に示されるように、ポリブロピレン樹脂製の直径15mmの円盤の中央に3.5mmの円柱が垂直に設置された平行性測定治具を作製する。この治具を腕の皮膚上に載せる。次いで、例えば図16(2)に示されるように、中央部に治具の円柱を通すための直径8mmの開口部を有する下駄型アプリケータ、あるいは、図19に示されるようなワッペンアプリケータを作製する。図17(1)に示されるように、アプリケータの開口部に治具の円柱を通し、アプリケータを腕の皮膚上に設置する。次いで、島津製作所社製レオメーターを用いて一定の距離(12mm)だけ、アプリケータを押し下げる。アプリケータを押し下げるために、直径5mmの円柱状の棒が使用される。上記棒でアプリケータの平板上の色々な位置(アプリケータの中心部から周辺部)を押し下げ、棒が当たる平板の位置によって、平行性測定治具とアプリケータの平板との角度がどう変化するかを図18で示されるように測定した。また、上記の一定距離(12mm)押し下げると皮膚がどれだけ上に上昇するかを8mmφの開口部を利用してノギスで測定した。
また、皮膚の上昇する高さ(距離)をノギスで測定した。その結果、本発明のアプリケータは、図3~図6に示されるように、アプリケータの平板部の形状が、正方形であっても長方形であっても、皮膚の盛り上がりの高さにはあまり相違がないことが示された。このように、皮膚の盛り上がる高さは、平板部の形状の影響をあまり受けないが、皮膚固定部材の種類(形状)の影響を大きく受けることが分かった。即ち、下駄型アプリケータ、四隅アプリケータ、3点アプリケータの順に皮膚が盛り上がりやすいことが分かった。
なお、下駄型アプリケータにおいて、アプリケータの設置の仕方で、皮膚の盛り上がる高さが異なることが想定されたが、表1と表2に示されるように、アプリケータの設置の仕方を変化させても(腕の方向性に対して垂直設置と平行設置)、実際上の皮膚の盛り上がり距離には差が見られなかった。
なお、本発明で使用する剣山型マイクロニードルには、薬剤が塗布されている。塗布される薬剤は、治療のためにこれまで使用されてきたものであれば特に限定されることはない。薬剤が蛋白質や抗原、抗体等の生体高分子である場合には水に対する溶解性が高いため、水溶液としてマイクロニードルに塗布することができる。抗生物質や抗精神病薬のような低分子化合物である場合には、溶解性に応じて水溶液でもよく、有機溶媒溶液を使用して塗布することができる。
なお、効果を示すために必要な量の薬剤が必要な場合、薬剤の水溶液等にマイクロニードルの微小針を浸漬し、乾燥することを繰り返して必要量を担持することが出来る。
(1)下駄型アプリケータの製造
アクリル製の長方形平板(30×40mm、厚さ2mm)の外周部に、図7に示すように、2本のアクリル製の四角柱(3×5×30mm)の側面(5×30mm)を接着し、皮膚固定部材とした。その結果、高さ3mmの下駄型アプリケータを作製した。得られた下駄型アプリケータの斜視図を図8に示す。2つの四角柱(皮膚固定部材)で囲まれる面積(皮膚が盛上がる部分)は30×30mmである。
アクリル製の正方形平板(30×30mm、厚さ2mm)の四隅に、図9に示すように、4個のアクリル製の半球(半径3mm)の上部を接着し、皮膚固定部材とした。その結果、高さ3mmの半球を四隅に持つ四隅アプリケータを作製した。得られた四隅アプリケータの斜視図を図10に示す。4個の半球で囲まれる面積(皮膚が盛上がる部分)は24×24mmである。
(3)四隅アプリケータの製造(b)
ポリプロピレン製の正方形平板(20×20mm、厚さ2mm)の四隅に、図1(2)に示すように、4個のポリプロピレン製の四角柱(5×5×10mm)の上部を溶着し、皮膚固定部材とした。その結果、高さ10mmの四角柱を四隅に持つ四隅アプリケータを作製した。得られた四隅アプリケータの斜視図を図11に示す。4個の四角柱で囲まれる面積(皮膚が盛上がる部分)は10×10mmである。
(4)3点アプリケータの製造
ポリプロピレン製の正方形平板(20×20mm、厚さ2mm)の外周部の中央に、図1(3)に示すように、1本のポリプロピレン製の四角柱(5×5×10mm)の上部を溶着する。その四角柱から10mm離れた所の両端部分に、図1(3)のように四角柱(5×5×10mm)を接着し、皮膚固定部材とした。その結果、高さ10mmの四角柱を3箇所に有する3点アプリケータを作製した。得られた3点アプリケータの斜視図を図12に示す。3個の四角柱で囲まれる面積(皮膚が盛上がる部分)は1×0.9mmである。
図1に示された各種のアプリケータを実施例1に準じて作製した。これらのアプリケータを使用して、腕の皮膚に設置し、図13の写真に示すようにアプリケータを設置した位置から約12mm押し下げ、皮膚に圧着させて、皮膚の盛り上がりの高さ(皮膚の盛り上がり距離)をノギス測定した。
以下の表1~表5に示すように、アプリケータの種類と、皮膚固定部材で囲まれる皮膚の面積の大きさを変化させた各種デバイスを作製した。なお、皮膚固定部材の高さは、皮膚固定部材で囲まれる面積の大きさによって適宜選択した。
実施例1の(1)に準じて作製された各種の下駄型アプリケータを用いて、皮膚に圧着させ、皮膚の盛り上がりの高さ(皮膚の盛り上がり距離)を評価した。皮膚に圧着させる場合には、図2左図と図13に示されるように、腕の方向に対して皮膚固定部材を直交するように設置し、押圧する場合(垂直設置)と、図2右図に示されるように、腕の方向に対して皮膚固定部材を平行に設置し、押圧する場合(平行設置)の2つの場合がある。この垂直設置と平行設置の相違で、アプリケータを皮膚に設置した際の皮膚表面の初期の高さが異なる。そこで、アプリケータの押圧時に皮膚が盛り上がる、実際の皮膚の盛り上がり高さ(距離)を測定した。その結果を、それぞれ区別して以下の表1(垂直設置)と表2(平行設置)、図3(垂直設置)と図4(平行設置)に示した。
実施例1の(3)に準じて作製された各種の四隅アプリケータを用いて、皮膚に圧着させ、皮膚の盛り上がりの高さ(皮膚の盛り上がり距離)を評価した。正方形の四隅アプリケータを使用した場合、皮膚の盛り上がりの高さ(皮膚の盛り上がり距離)は表3のようになった。
一方、長方形の四隅アプリケータを使用する場合には、図2左図や図2右図に示されるように腕の方向に対して長辺を直交するように設置・押圧する場合(垂直設置)と、腕の方向に対して長辺を平行に設置・押圧する場合(平行設置)の2つの場合がある。長方形の四隅アプリケータの垂直設置と平行設置を区別して得られた結果を表4にまとめて示し、また図5に示した。
なお、この盛り上がり距離は、下駄型アプリケータの場合と比較し、より高くなる(距離が出る)ことが分かった。
更に、皮膚固定部材が4つの四角柱であるので、皮膚に押圧する場合、四角柱の角が皮膚に当たり、痛みを招くことになった。そこで、痛みを軽減するため、四角柱の角を丸めることを行なった。
実施例1の(4)で作製された3点アプリケータを用いて、皮膚に圧着させ、皮膚の盛り上がりの高さ(皮膚の盛り上がり距離)を評価した。その結果を表5と図6に示す。
以上のように、皮膚固定部材で圧接する皮膚面積が少ないほど、皮膚の盛り上がり距離が大きいことが示された。また、四隅アプリケータと3点アプリケータは、下駄型アプリケータと比較して、腕に対する設置方向(垂直設置、平行設置)の影響を受けないことが示された。
(1)下駄型マイクロニードルデバイス
実施例1の(1)で作製された図8の下駄型アプリケータに対して、図20に示されるように4個の三角形の両面テープで円形のテープを設置し、23mmの円形テープの中央に円形の直径10mmの剣山型マイクロニードルを設置した。その結果、図14で示される下駄型マイクロニードルデバイスが得られた。
(2)四隅マイクロニードルデバイス
実施例1の(2)で作製された図10の下駄型アプリケータに対して、上記(1)と同様にして4個の三角形の両面テープで円形のテープを設置し、23mmの円形テープの中央に円形の直径10mmの剣山型マイクロニードルを設置した。その結果、図15で示される四隅マイクロニードルデバイスが得られた。
試験例1の結果から、皮膚固定部材の存在しない位置に近い、アプリケータの天板部分を押圧することは、穿刺性の観点で好ましくないことが分かった。そこで、天板部分の中央部を押圧できるように、または皮膚固定部材に力が均等に掛けられるように、図23~図26に示される押圧用の突起物(構造)をアプリケータの天板に設置した。
(1)下駄型アプリケータの天板中央部に押圧用の突起物を設置したマイクロニードルデバイス
図23と図24に示されるように、下駄型アプリケータの天板の中央部分に、円柱状または半球状の突起物を設置したアプリケータを作製した。その結果、押圧がし易くなり、安定した穿刺性が得られるようになった。
(2)下駄型アプリケータの皮膚固定部材に均等に力を掛けるためのアーチ型構造物を天板に設置したマイクロニードルデバイス
アーチ状構造物を下駄型アプリケータの天板に設置し、図25に示されるようなマイクロニードルデバイスを作成することができる。アーチ状構造物として補強のため、補助板をアーチの中央部分に付加することができる。そのような形状のものとして、図26に示されるような、下駄型アプリケータの天板にアーチ状構造物が設置された、マイクロニードルデバイスを作製した。その結果、押圧がし易くなり、安定した穿刺性が得られるようになった。
(1)両面に粘着剤が貼付されたテープ状チップの製造
テープ状チップとしてニチバン製カテリープFSロール(23φ)を使用し、紙両面テープ(ナイスタックNW-40)を5×5mmの直角2等辺三角形に切り、それを4個、図30のように、カテリープFSロールの上部に設置した。
これにより、両面に粘着剤が貼付されたテープ状チップを製造した。
(2)テープ状チップを収納固定したアプリケータの製造
アプリケータとして、図7~図8に示されるアクリル製のものを使用する。このアプリケータの中央部に上記(1)のテープ状チップを設置し、上記紙両面テープでテープ状チップをアプリケータに固定した。
これにより、テープ状チップを設置したアプリケータを製造した。
(3)マイクロニードルが設置されたテープ状チップを固定したアプリケータの製造
(2)で得られたアプリケータ中のカテリープFSロールの粘着剤の保護紙を剥がし、その中央部にマイクロニードル(10mmφ)を設置する。
これにより、図32に示されるような、マイクロニードルが設置されたテープ状チップを固定したアプリケータを製造した。
アプリケータとテープ状チップを接着するための両面テープ(粘着剤A)が、試験例6の結果から、皮膚に接着している粘着剤の面積の約90%以下の面積であるならば、粘着剤Aの部分でアプリケータとテープ状チップが剥離する。また、試験例7の結果から、粘着剤Aの形状、数、配置が剥離し易さに寄与していることが示された。そこで、粘着剤Aの面積を皮膚に接着している粘着剤の面積の約90%以下の面積になるように設定し、粘着剤Aの形状を変化させ、各種のテープ状チップを作製する。
粘着剤Aの形状としては、図34~図41に記載の各種の形状のものを採用することができる。
実施例3で作製された各種マイクロニードルデバイスを使用すれば、マイクロニードルが皮膚を垂直に穿刺することになるか否かの確認を行なった。更に、当該デバイスの平板のどの位置に力を架けると、マイクロニードルが皮膚を垂直に穿刺することになるかの検討を行なった。
(1)デバイスの中心から皮膚固定部材の方向(垂直)にズレた位置を押圧する場合に生じる、マイクロニードルの皮膚穿刺の角度
この目的のために、図16に示されるように、実施例1の(2)に準じて作製された下駄型アプリケータ(皮膚固定部材で囲まれる面積は30×30mm、高さ7mm)を作製した。その平板の中央に直径8mmの穴を開設する。直径15mmの円盤の中央に、直径3.5mmの円柱を直角に立て、皮膚の上に図17のように設置する。更に上記下駄型アプリケータの穴にその円柱を通して重ね、アプリケータの平板の上を直径5mmの棒で押圧する。アプリケータ押圧時に見出される、アプリケータの平板と皮膚上に設置した円柱との角度の変化を図18のようにして評価した。
具体的には直径5mmの棒で押圧する平板上の位置と上記棒の押し込み距離を変化させ、角度の変化を評価することにより、皮膚表面がどのように盛り上がってくるかを評価した。
その結果を表6に示す。
固定部材を平行方向に:図2(b)に示すように、腕に対して固定部材を平行方向に設置する(アプリケータ平行置き)。
固定部材を垂直方向に:図2(a)に示すように、腕に対して固定部材を垂直方向に設置する(アプリケータ垂直置き)。
押圧位置(中心からのズレ):アプリケータの中心から固定部材の方向(垂直)に、6mm、9mm、12mmの距離だけ離れたところを押し込む位置とする。
即ち、剣山型マイクロニードルに対して、皮膚は垂直の位置を保ちながら、押圧に伴い、盛り上がってくることが示された。
(2)デバイスの中心から皮膚固定部材と平行方向にズレた位置を押圧する場合に生じる、マイクロニードルの皮膚穿刺の角度
この目的のために、図22に示されるような、実施例1の(2)に準じて作製された下駄型アプリケータ(皮膚固定部材で囲まれる面積は30×30mm、高さ7mm)を作製した。図22に示されるように、平板の中央から右側に7×18mmの長方形の穴(空洞)を開設する。直径15mmの円盤の中央に、直径3.5mmの円柱を直角に立て、それを平行性測定用冶具として使用し、皮膚の上に設置する。更に上記下駄型アプリケータの穴にその円柱を通して重ね、アプリケータの平板の上を直径5mmの棒で押圧する。アプリケータ押圧時に見出される、アプリケータの平板と皮膚上に設置した円柱との角度の変化を図18のようにして評価した。
上記と同様にして、押圧する平板上の位置と上記棒の押し込み距離を変化させ、平行性測定用冶具で示される角度の変化を評価することにより、皮膚表面がどのように盛り上がってくるかを評価した。
その結果を表7に示す。
この結果は、上記表6の結果とは大きく異なり、下駄型のアプリケータを使用する場合には、押圧の位置に配慮することが必要になる。そのため、下駄型アプリケータを使用する場合には、押圧のための突起(構造物)をアプリケータの平板(中央部)に設置することが必要と考えられた。
一方、試験例2に示されるように、ワッペン型のアプリケータの場合であれば、アプリケータが接触する皮膚は、例えば円形状で周囲が囲まれているため、アプリケータのどの位置を押しても、アプリケータの平板と皮膚は平行を保っている。即ち、下駄型アプリケータの場合には、2箇所の平行する板で皮膚を固定するため、皮膚を固定する部材のない位置で押圧すると、上述するように、皮膚とアプリケータ平板が平行を保つのは難しい(マイクロニードルが皮膚を垂直に穿刺することは難しい)と考えられる。
試験例1と同じ方法で、図19に示されるワッペン型アプリケータを作製して、皮膚表面とアプリケータ平板の平行性を評価した。ワッペン型アプリケータの中央に直径8mmの穴を開設し、試験例1と同様に、中心から離れた位置を直径5mmの棒で押圧して、皮膚表面に設置した円柱とアプリケータ平板との角度変化を評価した。即ち、上記棒で押圧する平板上の位置と上記棒の押し込み距離を変化させ、角度の変化を評価して、皮膚表面がどのように盛り上がってくるかを判断した。その結果を表8に示す。
実施例3(1)で作製された下駄型アプリケータデバイスを用いて、皮膚への穿刺実験を行った。評価方法は、図21のヒト皮膚モデルを採用した。まず、スチレン・イソプレン・スチレンブロック共重合体(SIS)のゴム状平板上に、ラット皮膚(雄性5週齢腹部)を置き、その上に実施例1で作製されたマイクロニードルデバイスを設置した。該デバイスを島津製作所製レオメーターで12mm押した。デバイスとマイクロニードルを除去した後、ラット皮膚をゲンチアナバイオレット溶液で染色し、ラットの皮膚表面を観察した。皮膚が穿刺された部分は、青く染まるので、穿刺された個所が読み取れることになる。
穿刺の結果によれば、ラットの皮膚には、微小針で穿刺された箇所が規則正しい青色のスポットとして表われている。
また、穿刺後のマイクロニードルの微小針の折損等の変化を評価したが、微小針の形状に大きな変化はなく、折損や横曲がり等がみられない。
このように、本発明のアプリケータデバイスは、皮膚を適切に穿刺できるものである。
(1)ポリプロピレン製平板に対する粘着剤の引っ張り応力
粘着剤の粘着力(引っ張り応力)を応力測定機器(島津製作所製レオメータEZ-Test)を用いて、粘着力の最大応力を測定した。
3×3cm×2.5mmのポリプロピレン製平板を重ね、その中に幅3cm、長さ25cm、厚さ0.1mmのPET製シートを挟んで接着し一体とする。図27に示されるように、PET製シートを上記レオメーターに設置された引っ張り試験治具に固定する。上記ポリプロピレン製平板(A)の直下に、同じサイズのポリプロピレン製平板(B)を水平に設置する。上下の2つのポリプロピレン製平板を粘着剤(ニチバン製ナイスタックNW-40)で接着した。接着した平板の下部のもの(B)を固定し、図27に示されるように引っ張り治具を上昇させ、上部のポリプロピレン製平板(A)を垂直に引き上げる。そして、上下のポリプロピレン製平板が剥離する時の最大応力を測定した。
その際、粘着剤のサイズ(20、15、10、5mmφ、14×14mm、0.3×0.3mm)を換えて、それぞれの剥離時の最大応力を測定した。
その結果を表9にまとめて示した。
上記(1)と同様にして粘着剤の粘着力(引っ張り応力)を測定した。なお、ポリプロピレン製平板(B)の代わりに、ヒト上腕を使用し、図28に示すように引っ張り応力の評価試験を行なった。
その結果を表10にまとめて示した。
そこで、ヒトの皮膚に対する粘着剤の粘着力(応力/cm2)とポリプロピレン(PP)製平板に対する粘着剤の粘着力(応力/cm2)を対比すると、次の表11のようになった。
テープ状チップの両面に塗布された接着剤の粘着力に強弱の相違があると、粘着力の弱い部分の方が剥離することになる。そこで、粘着力にどの程度の強弱の差があると弱い粘着力の方が剥離することになるのかを検討評価した。
(1)ポリプロピレン(PP)製平板における両面の粘着力の差と剥離部位の評価
モデル実験として、図27に示されるように、PP製平板(3×3cm×5mm)のチップの両面に粘着剤Aと粘着剤Bを塗布した。試験例4と同様に、上記チップをPP製平板AとBで挟み接着させ、引っ張り冶具にPP製平板Aを固定する。冶具を上昇させ、粘着剤Aの部分で剥離するか、粘着剤Bの部分で剥離するかを確認した。
粘着剤Aとして、ニチバン製ナイスタックNW-40を使用し、貼付する粘着剤の面積を以下の表11のように変化させた。一方、粘着剤Bは、同じニチバン製ナイスタックNW-40を使用するものの、貼付する粘着剤の面積は20mmφで固定した。
その結果を以下の表12で示す。
図3に示されるように、PP製平板Cの代わりにヒトの前腕を用いた。そして、前項(1)と同様にして剥離試験を行なった。
その結果を以下の表13に示す。
一方、表11の結果から、皮膚に貼付した粘着剤の粘着力は、PP製平板と比較し約6%前後に低減することが示されている。即ち、同じ材質の粘着剤を使用し、PP製平板と比較するためには、皮膚に対して粘着する粘着剤Bの面積の約6%と対比することになる。
そこで、図27のようなPP製平板同士の比較に変換すると、以下の表14のように表わすことができる。
「粘着剤Aの粘着力」<「粘着剤Bの粘着力」×(85~90%)
であることが分かる。
また、同じ材質の粘着剤を使用する場合には、粘着力は粘着剤の貼付面積で置き換えることができる。更に、皮膚に対する粘着力は、表11の結果から、PP製平板間の粘着力の約6%であることが示されているので、上記の式は、次のように書換えることができる。
「粘着剤Aの貼付面積」<(「貼付剤Bの皮膚付着面積」×約6%)×(85~90%)
の関係があることが分かる。
試験例5では、応力測定機器(島津製作所製レオメータEZ-Test)を用いて、図27、図28に示されるように垂直にPP板Aを引上げ、その両面に塗布された接着剤の剥離試験を行ない、粘着力にどの程度の強弱の差があると弱い粘着力の方が剥離することになるのかを検討した。
試験例6では、図30に示されるように、PP板Aを斜めに引き上げることで、どの程度、引っ張り応力が軽減されるかを評価した。
(1)粘着剤を貼付したテープ状チップの作製
図30に示されるように、テープ状チップとしてニチバン製カテリープFSロール(23φ)を使用し、その上部(PP板A側)に接着剤A(ナイスタックNW-40)を4ヶ所貼付する(5×5mmの直角2等辺三角形を4個)。
皮膚への付着面積は約415mm2であり、PP板Aへの付着面積は50mm2となる。
従って、試験例5(垂直引上げ)の結果から、以下の式に数字を記載すると、
「粘着剤Aの貼付面積」<(「貼付剤Bの皮膚付着面積」×約6%)×(85~90%)
(50mm2) > (25mm2)×(85~90%)
となり、試験例5のように垂直引き上げの場合であれば、皮膚側部分が剥離すると考えられた。
図30に示されるように、皮膚に上記(1)のテープ状チップを貼付し、PP板Aを設置して、応力測定機器(島津製作所製レオメータEZ-Test)に接続する。応力測定器の冶具を引上げて、PP板Aを斜めに引上げ、PP板に付着している接着剤Aが剥離する際の引張り応力を評価した。その結果を以下の表15に示す。
従って、斜めに引っ張り上げる場合に、皮膚側の接着剤Bが剥離せず、接着剤Aが剥離するためには、以下のような式が成り立つことが必要である。
「粘着剤Aの粘着力」×約5% <「粘着剤Bの粘着力」×(85~90%)
また、同じ材質の粘着剤を使用する場合には、粘着力は粘着剤の貼付面積で置き換えることができ、皮膚に対する粘着力は、表11の結果から、PP製平板間の粘着力の約6%であることが示されているので、上記の式は、次のように書換えることができる。
「粘着剤Aの貼付面積」×約5% <(「貼付剤Bの皮膚付着面積」×約6%)×(85~90%)
更に、これを簡略化すると、次のような近似式になる。
「粘着剤Aの貼付面積」<「貼付剤Bの皮膚付着面積」×約90%
図34で示される粘着剤の形状でアプリケータにテープ状チップを固定したタグ付きアプリケータ(図33の(c))を作成した。このアプリケータを用いて,図30に示される斜めの引上げ方法により、ヒト前腕部に使用した場合、皮膚へのテープ状チップの移行のしやすさを5段階で評価した。
その結果を表16に示す。
(1)テープ状チップの外周より粘着剤(両面テープ)が突出する場合と内部に設置される場合の比較評価
図40で示されるように、同じ形状の粘着剤(ナイスタックNW)を使用し、テープ状チップ(トランスポアSP1527SP-1)の外周より粘着剤が突出する場合(左図)と、内部に設置される場合(右図)の下駄型アプリケータ(デバイス)を作製した。図40a)で示される長方形のテープ状チップを使用する場合、左図のように粘着剤が外周より0.5mm突出しているものを作製した。また、右図のように外周より2mm内部に粘着剤が設置されたものを作製した。また、マイクロニードルのダミーとして、PP板(0.8mm厚、10mmφ)を設置した。
このテープ状チップが設置された下駄型アプリケータを使用し、ヒトの前腕に手で押し付け、テープ状チップの皮膚への移行性を評価した。その結果、外周より内部に粘着剤が設置された、右図に示されるものの方が皮膚への移行性が良好であった。
同様に、図40b)で示される円形状のテープ状チップを使用し、左図のように粘着剤が外周より1.0mm突出しているもの、また、右図のように外周より1mm内部に粘着剤が設置されたもの、を作製した。また、マイクロニードルのダミーとして、PP板(0.8mm厚、10mmφ)を設置した。
上記と同様に皮膚への移行性を評価したが、外周より内部に粘着剤が設置された、右図に示されるものの方が皮膚への移行性が良好であった。
粘着剤(ナイスタックNW)が長方形のテープ状チップ(トランスポアSP1527SP-1)に設置された、図42に示されるものを作製し、下駄型アプリケータに設置した。外周からの距離aとbの相違により、どのように皮膚移行性が変化するかを確認した。また、マイクロニードルのダミーとして、PP板(0.8mm厚、10mmφ)を設置した。
ヒトの前腕に手で押し付け、テープ状チップの皮膚への移行性を評価した。その結果を以下の表17に示す。
これまでの試験例の結果から、テープ状チップをアプリケータ(デバイス)に固定し、かつ、皮膚に移行しやすくするために、粘着剤(両面テープ等)の粘着力の制御、あるいは粘着剤の形状、設置位置を選択することができる。特に、粘着剤の設置位置は、テープ状チップの外周の内部(外周からの距離が0.1mm以上)にあることが好ましいことが分かった。
Claims (14)
- マイクロニードルを皮膚に穿刺するためのマイクロニードルデバイスであって、
a)平板の片面に2本以上の皮膚固定部材が設置され、
b)上記皮膚固定部材の間隔が10~70mmであり、
c)上記皮膚固定部材の形状が、角柱状、円柱状、半円柱状(かまぼこ型)または半球状であり、
d)上記皮膚固定部材の間の中央部にマイクロニードルを設置し、
e)マイクロニードルの微小針の先端部が、上記皮膚固定部材の先端面よりデバイス内に留まっている、
ことを特徴とする、マイクロニードルデバイス。 - 上記皮膚固定部材の間隔が10~40mmである、請求項1記載のマイクロニードルデバイス。
- 上記皮膚固定部材がデバイス平板の両端に設置され、下駄状の形状であることを特徴とする、請求項1又は2に記載のマイクロニードルデバイス。
- 上記皮膚固定部材がデバイス平板の四隅に設置されていることを特徴とする、請求項1又は2に記載のマイクロニードルデバイス。
- 上記皮膚固定部材がデバイス平板の3箇所に設置されることを特徴とする、請求項1又は2に記載のマイクロニードルデバイス。
- 上記皮膚固定部材が角柱、円柱又は半球であることを特徴とする、請求項4又は5に記載のマイクロニードルデバイス。
- 上記角柱が四角柱であることを特徴とする、請求項1~7のいずれかに記載のマイクロニードルデバイス。
- 上記皮膚固定部材が半球であることを特徴とする、請求項4又は5に記載のマイクロニードルデバイス。
- 押圧用の突起物(構造)が、上記平板の中央部に設置されていることを特徴とする、請求項1~8のいずれかに記載のマイクロニードルデバイス。
- 上記突起物が、円柱状または半球状である、請求項9記載のマイクロニードルデバイス。
- 押圧用の突起物(構造)として、アーチ状の構造物が上記平板の上に設置されていることを特徴とする、請求項1~8のいずれかに記載のマイクロニードルデバイス。
- 皮膚貼付用のテープ状チップが設置されていることを特徴とする、請求項1~11のいすれかに記載のマイクロニードルデバイス。
- 上記テープ状チップが、
a)1~50cm2のテープ状チップであって、
b)テープ状チップの支持体とデバイスを粘着剤で接着させ、
c)中央部に5~15mmφのマイクロニードルが設置されている、
ことを特徴とする、請求項12記載のマイクロニードルデバイス。 - 上記粘着剤の粘着力が、皮膚に接着させる粘着剤の粘着力の85~90%以下である、請求項12または13に記載のマイクロニードルデバイス。
