Abstract
Description
XSCID(X-linked Severe Combined Immuno Deficiency:X連鎖重症複合免疫不全症)の患者集団においては、JAK3タンパクレベルの低下、若しくは共通γ鎖の遺伝子欠損が見られる。このことは、免疫抑制がJAK3を介したシグナル経路を遮断することにより生じていることを示唆する(非特許文献3、4)。動物実験では、JAK3がB-およびT-リンパ球の成熟に重要な役割を果たすだけでなく、T細胞の機能維持にも重要であることが示されている。従って、このメカニズムを通じた免疫応答の制御により、臓器移植時の拒絶や自己免疫疾患のようなT細胞の増殖異常が関与する疾患の治療が見込まれる。
JAK1は、JAK1 KOマウスやJAK1欠損細胞の解析から、JAK1がインターフェロン(以下、IFNと言う)α、IFNβ、IFNγ、IL-2、IL-4、IL-6、IL-7、IL-15等の多様な受容体を介したシグナル伝達に関与していることが示されている(非特許文献5)。Stat3を活性化することにより、IL-6レセプターのシグナル伝達に深く関与すると考えられる。従って、このメカニズムを通じた炎症応答の制御により、自己免疫疾患や臓器移植時の急性及び慢性拒絶のようなマクロファージやリンパ球の活性化が関与する疾患の治療が見込まれる。
JAK2は、JAK2 KOマウスやJAK2欠損細胞の解析から、JAK2がエリスロポエチン(以下、EPOと言う)α、トロンボポエチン(以下、TPOと言う)、IFNγ、IL-3、GM-CSF等の多様な受容体を介したシグナル伝達に関与していることが示されている(非特許文献6、7、8)。これらはStat3を介し骨髄中の赤血球、血小板などの前駆細胞の分化に関与すると考えられる。なお、JAK2の617番塩基のフェニルアラニンがバリンに置換されている場合があり、骨髄増殖性疾患との関与が示唆されている(非特許文献6)。従って、これらのメカニズムを通じた骨髄前駆細胞の分化の制御により、骨髄増殖性疾患の治療が見込まれる。
当該化合物は、R21が、R3とともに一定の官能基を介して結合し、特定のヘテロ環を形成することを特徴とする。

更に、本発明は、式(I)の化合物又はその塩を含有する望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防用若しくは治療用医薬組成物、即ち、式(I)の化合物又はその塩を含有する望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防若しくは治療剤に関する。
また、本発明は、望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防用若しくは治療用医薬組成物の製造のための式(I)の化合物又はその塩の使用、望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防若しくは治療のための式(I)の化合物又はその塩、並びに、式(I)の化合物又はその塩の有効量を患者に投与することからなる望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防若しくは治療方法に関する。

「JAK1、JAK2及びJAK3がそれぞれ単独でもしくは互いに組み合わさって関与する疾患」のある態様としては、乾癬、器官又は組織(心臓、腎臓、肝臓、骨髄、皮膚、角質、肺、すい臓、小島、小腸、手足、筋肉、神経、椎間、気管、筋芽細胞、軟骨等)の移植に対する拒絶反応、骨髄移植後の対宿主性移植片反応、リウマチ、全身性エリテマトーデス(SLE)、骨髄増殖性疾患、喘息、I型糖尿病と糖尿病合併症等、多発性硬化症であり、別の態様としては、乾癬、器官又は組織(心臓、腎臓、肝臓、骨髄、皮膚、角質、肺、すい臓、小島、小腸、手足、筋肉、神経、椎間、気管、筋芽細胞、軟骨等)の移植に対する拒絶反応、リウマチ、骨髄増殖性疾患、喘息、I型糖尿病、多発性硬化症であり、更に別の態様としては、乾癬、器官又は組織(心臓、腎臓、肝臓、骨髄、皮膚、角質、肺、すい臓、小島、小腸、手足、筋肉、神経、椎間、気管、筋芽細胞、軟骨等)の移植に対する拒絶反応、リウマチである。

また、式(I)の化合物には、不斉炭素原子や軸不斉を有する場合があり、これに基づく光学異性体が存在しうる。本発明は、式(I)の化合物の光学異性体の分離されたもの、あるいはそれらの混合物も包含する。
式(I)の化合物及びその塩は、その基本構造あるいは置換基の種類に基づく特徴を利用し、種々の公知の合成法を適用して製造することができる。その際、官能基の種類によっては、当該官能基を原料から中間体へ至る段階で適当な保護基(容易に当該官能基に転化可能な基)に置き換えておくことが製造技術上効果的な場合がある。このような保護基としては、例えば、ウッツ(P. G. M. Wuts)及びグリーン(T. W. Greene)著、「Greene’s Protective Groups in Organic Synthesis(第4版、2006年)」に記載の保護基等を挙げることができ、これらの反応条件に応じて適宜選択して用いればよい。このような方法では、当該保護基を導入して反応を行なったあと、必要に応じて保護基を除去することにより、所望の化合物を得ることができる。
また、式(I)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行なうことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。
以下、式(I)の化合物の代表的な製造法を説明する。各製法は、当該説明に付した参考文献を参照して行うこともできる。なお、本発明の製造法は以下に示した例には限定されない。
本発明化合物(I)のうち化合物(I-1)は、化合物(6)を環化反応に付し、化合物(7)に変換後、脱保護反応を行うことにより得ることができる。ここで、置換基Qの例には、メトキシ基、エトキシ基、tert-ブトキシ基、ジメチルアミノ基、メチルチオ基等が、保護基RPr1の例には、tert-ブトキシカルボニル基、ベンジルオキシカルボニル基、2-(トリメチルシリル)エトキシメチル基等が含まれる。
まず、化合物(6)と等量若しくは過剰量のQ-Hとを、反応に不活性な溶媒中、又は無溶媒下、冷却下から加熱還流下、好ましくは0℃から80℃において、通常0.1時間~5日間撹拌し、アセタール類(6’)を発生させる。ここで用いられる溶媒の例としては、特に限定はされないが、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類、N,N-ジメチルホルムアミド、ジメチルスルホキシド、酢酸エチル、アセトニトリル及びこれらの混合物が挙げられる。酸性条件化で反応を行うのが、反応を円滑に進行させる上で有利な場合がある。ここで用いられる酸の例としては、塩酸、酢酸等が挙げられ、塩化アセチルを用いることにより反応系中でQ-Hと塩酸を発生させる方法も用いることができる。また、ここで用いられるQ-Hの例としては、アルコール類、アミン類、チオール類があげられ、特に限定されないが、より具体的には、メタノール、エタノール、tert-ブタノール、メチルアミン、エチルアミン、エタンチオール等が挙げられる。尚、本反応は、試薬として用いるQ-Hを溶媒として用い反応を進行させることもできる。
次に、発生させたアセタール類(6’)を、等量若しくは過剰量の水の存在下で、反応に不活性な溶媒中、又は無溶媒下、冷却下から加熱還流下、好ましくは0℃から80℃において、通常0.1時間~5日間撹拌することにより、化合物(7)を得ることができる。本反応は、アセタール類(6’)を単離し、あるいは単離することなく行うことができ、また、酸又は塩基の存在下に反応を行うのが好ましい場合がある。ここで、用いられる酸の例としては、特に限定されないが、p-トルエンスルホン酸、無水酢酸等の有機酸、塩酸、硫酸等の無機酸等が挙げられ、塩基の例としては、特に限定されないが、トリエチルアミン、N,N-ジイソプロピルエチルアミン若しくはN-メチルモルホリン等の有機塩基、又は炭酸カリウム、炭酸ナトリウム、水酸化カリウム若しくは水酸化ナトリウム等の無機塩基が挙げられる。
更に、化合物(I-1)は、化合物(7)の脱保護反応により得ることができる。
ここで、本脱保護反応は、例えばウッツ(P. G. M. Wuts)及びグリーン(T. W. Greene)著、「Greene’s Protective Groups in Organic Synthesis(第4版、2006年)」を参照して実施することができる。

本発明化合物(I)のうち化合物(Ia-1)は、化合物(13)とR1-Wとの置換反応により得ることができる。ここで、脱離基Wの例には、ハロゲン、メタンスルホニルオキシ、p-トルエンスルホニルオキシ基、メトキシ基、エトキシ基等が含まれる。
この反応では、化合物(13)と等量若しくは過剰量のR1-Wを用い、これらの混合物を、反応に不活性な溶媒中、又は無溶媒下、冷却下から加熱還流、好ましくは0℃から200℃、更に好ましくは20℃から120℃において、通常0.1時間~5日間撹拌する。マイクロウェーブの照射下で反応を行うことが、反応を円滑に進行させる上で有利な場合がある。ここで用いられる溶媒の例としては、特に限定はされないが、メタノール、エタノール、tert-ブタノール等のアルコール類、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類、N,N-ジメチルホルムアミド、ジメチルスルホキシド、酢酸エチル、アセトニトリル及びこれらの混合物が挙げられる。トリエチルアミン、N,N-ジイソプロピルエチルアミン若しくはN-メチルモルホリン等の有機塩基、又はナトリウム tert-ブトキシド、炭酸カリウム、ビス(メチルシリル)ナトリウムアミド、炭酸ナトリウム若しくは水酸化カリウム等の無機塩基の存在下で反応を行うのが、反応を円滑に進行させる上で有利な場合がある。
また、上記反応は、特に限定はされないが、例えばウルマン反応やブッフバルト・ハートウィグ反応等に使用されるような触媒を用いて行うこともできる。ここで用いられる触媒は、特に限定はされないが、トリス(ジベンジリデンアセトン)パラジウム、テトラキス(トリフェニルホスフィン)パラジウム等と4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン(Xantphos)、2-ジシクロヘキシルホスフィノ-2’,6’-ジメトキシビフェニル(SPhos)、2-ジシクロヘキシルホスフィノ-2’,4’,6’-トリイソプロピルビフェニル(XPhos)等を適宜組み合わせて用いることができる。
更に、上記反応は、縮合剤の存在下で行うこともできる。ここで用いられる縮合剤の例としては、特に限定されないが、ジシクロヘキシルカルボジイミド、ジイソプロピルカルボジイミド、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩等を用いることができる。

本発明化合物(I)のうち化合物(I-2)は、化合物(7)のブロモ化反応及びその後の脱保護反応により得ることができる。
ブロモ化反応は、化合物(7)と等量若しくは過剰量のブロモ化試薬を用い、これらの混合物を、反応に不活性な溶媒中、又は無溶媒下、冷却下から加熱還流、好ましくは-20℃から200℃、更に好ましくは-10℃から150℃において、通常0.1時間~5日間撹拌する。ここで用いられる溶媒の例としては、特に限定はされないが、メタノール、エタノール、tert-ブタノール等のアルコール類、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類、N,N-ジメチルホルムアミド、ジメチルスルホキシド及びこれらの混合物が挙げられる。塩化アルミニウム(AlCl3)や三フッ化ホウ素(BF3)等のルイス酸の存在下で反応を行うのが、反応を円滑に進行させる上で有利な場合がある。ブロモ化試薬の例としては、臭素(Br2)の他、N-ブロモスクシンイミド等が挙げられる。
脱保護反応は、前述の製造法(第1製法)と同様の方法を用いて行うことができる。
一方、化合物(I-3)は、化合物(8)の置換反応により、化合物(9)に変換後、脱保護反応を行うにより得ることができる。化合物(8)の置換反応は、前述の製造法(第2製法)、続く脱保護反応は、前述の製造法(第1製法)と同様の方法を用いて行うことができる。
本発明化合物(I)のうち化合物(I-4)は、化合物(10)のブロモ化反応及びその後の脱保護反応により得ることができる。又、化合物(I-5)は、化合物(11)の置換反応により、化合物(12)に変換後、脱保護反応を行うにより得ることができる。
ブロモ化反応は、前述の製造法(第3製法)、置換反応は、前述の製造法(第2製法)、脱保護反応は、前述の製造法(第1製法)と同様の方法を用いてそれぞれ行うことができる。
化合物(6)は、化合物(1)から4段階で合成できる。
まず、化合物(3)は、化合物(1)とアミン類(2)との反応により得ることができる。この反応は、前述の製造法(第2製法)と同様の方法を用いて行うことができる。
次に、化合物(4)は、化合物(3)の還元反応により得ることができる。
この反応では、反応に不活性な溶媒中、冷却下から加熱下、好ましくは-20℃から80℃で、化合物(3)を等量若しくは過剰量の還元剤で、通常0.1時間~3日間処理する。ここで用いられる溶媒の例としては、特に限定されないが、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、ベンゼン、トルエン、キシレン等の芳香族炭化水素類及びこれらの混合物が挙げられる。還元剤としては、水素化アルミニウムリチウム若しくはジイソブチルアルミウムヒドリド等のヒドリド還元剤、その他、下記文献中の還元剤が好適に用いられる。
又、化合物(5)は、化合物(4)の酸化反応により得ることができる。
この反応では、反応に不活性な溶媒中、冷却下から加熱下、好ましくは-20℃から80℃で、化合物(4)を等量若しくは過剰量の酸化剤で、通常0.1時間~3日間処理する。ここで用いられる溶媒の例としては、特に限定されないが、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、ジクロロメタン、1,2-ジクロロエタン若しくはクロロホルム等のハロゲン化炭化水素類、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、N,N-ジメチルホルムアミド、ジメチルスルホキシド、酢酸エチル、水或いはこれらの混合物が挙げられる。酸化剤としては、例えば、活性二酸化マンガンが好適に用いられる。
本反応においては、スワン(Swern)酸化等のDMSO酸化又はデスマーチン(Dess-Martin)試薬を用いた酸化剤が好適に用いられる。
最後に、化合物(6)は、化合物(5)を付加・脱離反応に付することにより合成できる。本反応は、特に限定はされないが、例えばウィッティヒ反応に用いるリン化合物により行うことができる。
この反応では、反応に不活性な溶媒中、冷却下から加熱下、好ましくは-20℃から80℃で、化合物(5)を等量若しくは過剰量のリン化合物の存在下で、通常0.1時間~3日間処理する。ここで用いられる溶媒の例としては、特に限定されないが、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、N,N-ジメチルホルムアミド、ジメチルスルホキシド或いはこれらの混合物が挙げられる。リン化合物としては、例えば、アルキルトリフェニルホスホニウム塩が好適に用いられ、より具体的には、(メトキシメチル)トリフェニルホスホニウム クロリド、(メチルチオメチル)トリフェニルホスホニウム クロリド等があげられる。ナトリウムビス(トリメチルシリル)アミド、n-ブチルリチウム、tert-ブトキシカリウム、ナトリウムエトキシド、ナトリウムメトキシド等の塩基の存在下で反応を行うのが、反応を円滑に進行させる上で有利な場合がある。

次に、化合物(5)は、化合物(15)の還元反応により得ることができる。この反応は、前述の(原料合成1)記載の還元反応と同様の方法を用いて行うことができる。
単離、精製は、抽出、分別結晶化、各種分画クロマトグラフィー等、通常の化学操作を適用して行なわれる。
各種の異性体は、適当な原料化合物を選択することにより製造でき、あるいは異性体間の物理化学的性質の差を利用して分離することができる。例えば、光学異性体は、ラセミ体の一般的な光学分割法(例えば、光学活性な塩基又は酸とのジアステレオマー塩に導く分別結晶化や、キラルカラム等を用いたクロマトグラフィー等)により得られ、また、適当な光学活性な原料化合物から製造することもできる。
試験例1:JAK阻害試験
精製ヒトJAK1キナーゼドメインはカルナバイオサイエンス社(神戸)から購入した。これは、ヒトJAK1タンパク(accession number #BAD92294.1)の850~1154(C末端)番の断片のN末端にGSTタグ(62 kDa)を結合させたものをバキュロウイルスの発現系を用いて発現させ、グルタチオンセファロースクロマトグラフィーを用いて精製したものである。
(2)JAK1活性の測定
基質はBiotin-Lyn-Substrate-2(Biotin-XEQED EPEGF YFEWL EPE、 X = ε-Acp(ペプチド研、大阪)とATPを用いた。アッセイバッファーは0.01 % Tween 20と2 mM DTTを含む15 mM Tris-HCl pH 7.5を用いた。標準的には、20μLの基質溶液(627 nM Biotin-Lyn-Substrate-2、500μM ATPおよび25 mM MgCl2を含むアッセイバッファー)、10μLの被検化合物を含むアッセイバッファーおよび20μLの酵素溶液をマイクロプレートに添加し、よく攪拌した。
室温で1時間インキュベートした後、プレートを洗浄用バッファー(50 mM Tris-HCl pH=7.5、 150 mM NaCl、 0.02 % Tween 20)で洗浄し、ブロッキングバッファー(0.1 % Bovineserum albuminを含む洗浄用バッファー)をプレートに添加した。室温で30分インキュベートした後、ブロッキングバッファーを除去して、HRP-PY-20溶液(HRP-PY-20溶液をブロッキングバッファーで500倍希釈したもの)を添加した。室温で30分インキュベートした後、プレートを4回洗浄して、TMB substrate solution(Sigma)をプレートに添加した。室温で4分間インキュベートした後、1Mの硫酸を添加して反応を停止した。酵素活性は450 nmの吸光度として測定した。被験化合物のJAK1阻害活性は、JAK1活性を50 %抑制する被験化合物の濃度をIC50値として算出した。
(3)ヒトJAK2の調製
精製ヒトJAK2キナーゼドメインはカルナバイオサイエンス社(神戸)から購入した。これは、ヒトJAK2タンパク(accession number #NP_004963.1)の826~1132(C末端)番の断片のN末端にHisタグ(39 kDa)を結合させたものをバキュロウイルスの発現系を用いて発現させ、Ni-NTAアフィニティカラムクロマトグラフィーを用いて精製したものである。
(4)JAK2活性の測定
基質はBiotin-Lyn-Substrate-2(Biotin-XEQED EPEGF YFEWL EPE、 X = ε-Acp(ペプチド研、大阪)とATPを用いた。アッセイバッファーは0.01 % Tween 20と2 mM DTTを含む15 mM Tris-HCl pH 7.5を用いた。標準的には、20μLの基質溶液(627 nM Biotin-Lyn-Substrate-2、25μM ATPおよび25 mM MgCl2を含むアッセイバッファー)、10μLの被検化合物を含むアッセイバッファーおよび20μLの酵素溶液をマイクロプレートに添加し、よく攪拌した。

(6)JAK3活性の測定

本発明化合物の代表化合物のJAK1、JAK2及びJAK3阻害活性測定試験の結果を表1に示す。尚、「Ex」は、後記実施例化合物を示す。
PD活性の測定は血中CD3+リンパ球のstat5リン酸化の程度を評価することでおこなった。
正常ラットに化合物(20 mg/kg)を経口投与した後、4時間後に200μL採血をし、そこに刺激サイトカイン(IL-2, 7, 15等、Peprotec)並び蛍光標識されたCD3抗体(BD pharmingen)を1/20用量加えて37℃でインキュベートすることでstat5のリン酸化を惹起した。Lysing buffer(BD pharmingen)を5 mL加えて反応を停止させた後、PBS (-)(Phosphate Buffered Saline (-)、伸陽化学工業製)を加えて洗い、permeabilization buffer(BD pharmingen)を300μL加えて氷上で固定した。facs bufferで洗った後、蛍光標識リン酸化stat5抗体(BD pharmingen)を加えて室温でさらにインキュベートした。インキュベーションの後、facs bufferで細胞を洗い、フィルターを通して不溶物を除いた。蛍光の測定はFACScaliburを用いておこなった。ラットそれぞれの個体に対し、サイトカインを添加しているサンプルと添加していないサンプルを作り、それぞれの平均蛍光強度の値の差(ΔMFI)を求めた。コントロールには化合物を投与していないラットの血液を用い、このコントロールサンプルの平均蛍光強度に対する各化合物投与群のサンプルの平均蛍光強度から抑制率を算出した(抑制率(%)=100*(1-(サンプルのΔMFI)/(コントロールのΔMFI))。
以下に代表的な化合物の結果を示す。
試験例3:ラット心移植
ラット腹部への異所性心臓移植は参考資料に準じて行った。動物はACI系雄性ラットをDonorとし、Lewis系雄性ラットをRecipientとして使用した。尚、Lewisラットは移植の前日に絶食した。被検薬剤は移植の当日から1日1回14日間、20mg/kgの投与量で、経口投与した。移植心の拒絶判定は、移植後29日間毎日移植心を触診により観察し、拍動の停止を拒絶とした。生着日数は拒絶の前日までとした。本移植試験を複数回行い、生着日数より、MST(Median Survival Time、生着期間の中央値)の値を算出した。本発明化合物の代表化合物の結果を表に示す。

本試験の結果、in vivo動物モデルにおいて、本発明化合物は、低用量の経口投与において、臓器移植による拒絶反応を抑制することが確認された。
試験例4:ラットアジュバンド関節炎(AIA)モデル試験
本試験は、右後肢足蹠皮下に流動パラフィンに懸濁した結核死菌を注射してアジュバント関節炎を誘発したラットを用いて行った。関節炎誘導前後、左後肢の用量をラット足容積測定装置(室町機械)を用いて、水置換法にて測定した。関節炎の指標とする足腫脹は、アジュバント感作日からの足容積の変化として表示した。プロピレングリコール、HCO40(NIKKOL社製)、Tween80(林純薬工業株式会社製)及び水に溶解した薬剤を1日1回、アジュバント感作当日から24日目までの25日間経口投与し、当該炎症に対する効果を確認した。

製造例1及び製造例3と同様の方法を連続的に行うことにより、後記表に示す製造例2及び製造例2-1の化合物を製造した。
rac-N-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-5-(2-メトキシビニル)-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-4-アミン(1.34 g)にMeOH (9.0 mL)、塩化アセチル(0.56 mL)を加え、80℃で1時間攪拌した。反応混合物に水(1.0 mL)を加え、80℃にて4時間攪拌した。反応終了後、反応混合物に飽和炭酸水素ナトリウム水溶液を加えた後、混合物をEtOAcで抽出し、brineで洗浄した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から30/70)で精製し、rac-1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン (720 mg)を得た。
rac-エチル 5-アミノ-4-{[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]アミノ}-6-[(3,4-ジメトキシベンジル)アミノ]ニコチナート(1.4 g)のオルトギ酸トリエチル(26 mL)混合液に氷冷下で濃塩酸(0.44 mL)を滴下し、室温にて16時間攪拌した。反応混合物に飽和炭酸水素ナトリウム水溶液を加えた後、混合物をCHCl3にて抽出し、水で洗浄した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH = 100/0から95/5)で精製し、rac-エチル 7-{[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]アミノ}-3-(3,4-ジメトキシベンジル)-3H-イミダゾ[4,5-b]ピリジン-6-カルボキシラート(802 mg)を白色アモルファスとして得た。
エチル 4-クロロ-1H-ピロロ[2,3-b]ピリジン-5-カルボキシラート(2.00 g)にDMF (20 ml)を加え、混合物を氷冷した。そこに水素化ナトリウム(鉱油中60%分散)(427 mg)を加え、氷冷下のままで1時間攪拌した。その後、[2-(クロロメトキシ)エチル](トリメチル)シラン(1.71 ml)を滴下し、室温に昇温させ30分間攪拌した。反応終了後、反応混合物に飽和炭酸水素ナトリウム水溶液を加え、混合物をEtOAcで抽出し、brineで洗浄した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 0/100から10/90)で精製し、エチル 4-クロロ-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボキシラート(2.50 g)を無色透明のオイル状物質として得た。
rac-1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(720 mg)にCH2Cl2 (3.0 mL)、TFA (3.0 mL)を加え、室温で1.5時間攪拌した。反応混合物に飽和炭酸水素ナトリウム水溶液を加えた後、CHCl3にて抽出した。抽出液を減圧下濃縮した。残渣にCH2Cl2 (2.0 mL)、MeOH (1.0 mL)、エチレンジアミン(2.0 mL)、1 M水酸化ナトリウム水溶液(3.0 mL)を加え、室温で終夜攪拌した。反応混合物に水を加えた後、CHCl3で抽出した。抽出液をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(MeOH/CHCl3 = 0/100から10/90)で精製し、rac-1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン (522 mg)を得た。
rac-1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(310 mg)(製造例6)をHPLC (ダイセル社製CHIRALPAK IA (5μm 20mmφX250mm))による光学分取を行い(移動層:Hx/EtOH = 95/5、流速8 mL/min、室温)、第一ピークとして1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(112 mg)(製造例6a)、第二ピークとして1-[(3S,4S)-1-ベンジル-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(119 mg)(製造例6b)をそれぞれ白色固体として得た。
rac-1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(570 mg)と20 %水酸化パラジウムカーボン粉末(84 mg)の混合物にMeOH (10 mL)を加えた。さらにDIBOC (314 mg)を加え、反応容器内を水素置換し、室温にて4時間30分攪拌した。反応終了後、反応混合物をセライトろ過し、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 15/85から20/80)で精製し、rac-tert-ブチル (3R,4R)-4-メチル-3-[6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]ピペリジン-1-カルボキシラート(582 mg)を得た。
rac-1-[(3R,4R)-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(103 mg)とCH2Cl2 (2.0 mL)の混合物にDIBOC (220 mg)とN,N-ジメチルピリジン-4-アミン(4.9 mg)を加え、室温で30分間攪拌した。反応終了後、反応混合物に飽和塩化アンモニウム水溶液を加え、混合物をCH2Cl2で抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(AcOEt/Hx = 30/70から50/50)で精製し、tert-ブチル rac-1-[(3R,4R)-1-(tert-ブトキシカルボニル)-4-メチルピペリジン-3-イル]ジピロロ[2,3-b:2',3'-d]ピリジン-6(1H)-カルボキシラート(170 mg)を得た。
tert-ブチル rac-(3R,4R)-3-[8-ブロモ-6-{[2-(トリメチルジリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]-4-メチルピペリジン-1-カルボキシラート(60 mg)にCH2Cl2 (1.0 ml)とTFA (1.0 ml)を加え、室温で2時間攪拌した。反応混合物を減圧下濃縮し、残渣にCH2Cl2 (1.0 mL)、MeOH (0.5 mL)、エチレンジアミン(7.0 mL)及び1M水酸化ナトリウム水溶液(0.1 mL)を加え、室温で1時間攪拌した。反応混合物に水を加え、混合物をCH2Cl2で抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣を塩基性シリカゲルカラムクロマトグラフィー(MeOH/CHCl3 = 0/100から5/95)で精製し、rac-8-ブロモ-1-[(3R,4R)-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(33 mg)を得た。
製造例9の方法と同様にして後記表に示す製造例10及び10-1~10-2の化合物を製造した。
1-(1-ベンジルピロリジン-3-イル)-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(126 mg)と20%パラジウムカーボン粉末水酸化パラジウムタイプ(含水品、エヌ・イーケムキャット社製)(30.0 mg)をMeOH (1.9 mL)及びTHF (1.9 mL)に加え、更にギ酸アンモニウム(100 mg)を加えて、80 ℃で2時間30分攪拌した。反応混合物にギ酸アンモニウム(100 mg)を加え、80 ℃で3時間30分攪拌した後、さらにギ酸アンモニウム(200 mg)を加えて80 ℃で1時間攪拌した。反応混合物をセライトろ過し、ろ液を減圧下濃縮した。残渣に飽和炭酸水素ナトリウム水溶液を加え、90/10の混合溶媒(MeOH/CHCl3)で抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣を塩基性シリカゲルカラムクロマトグラフィー(MeOH/CHCl3 = 0/100から5/95)により精製し、1-ピロリジン-3-イル-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(71.7 mg)を得た。
rac-8-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-3-(3,4-ジメトキシベンジル)-3,8-ジヒドロイミダゾ[4,5-b]ピロロ[2,3-d]ピリジン(490 mg)をTFA (10 mL)に溶解し、室温で2日間攪拌した。反応混合物を減圧下濃縮し、残渣にジイソプロピルエーテルを加え、沈殿物をろ取して、rac-8-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-3,8-ジヒドロイミダゾ[4,5-b]ピロロ[2,3-d]ピリジン トリフルオロ酢酸塩(454 mg)を白色固体として得た。
rac-2-[(1R,4S,6S)-2-ベンジル-2-アザビシクロ[2.2.1]ヘプト-6-イル]-1H-イソインドール-1,3(2H)-ジオン(700 mg)にMeOH (11 mL)とTHF (11 mL)を加えた後、更にヒドラジン・一水和物(0.409 mL)を加え、加熱還流下 2 時間攪拌した。析出物をTHFでろ取し、そのろ液を減圧下濃縮した。残渣に、1M水酸化ナトリウム水溶液を加えて塩基性とした後に、CHCl3で抽出した。有機層をMgSO4で乾燥後、ろ過し、ろ液を減圧濃縮し、rac-(1R,4R,6S)-2-ベンジル-2-アザビシクロ[2.2.1]ヘプタン-6-アミン(436 mg)を得た。
エチル4-{[(1R,2R,3S,5s)-5-ヒドロキシアダマンタン-2-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボキシラート(2.07 g)とCH2Cl2 (8.0 ml)の混合物に2,6-ジメチルピリジン(0.99 mL)を加え、氷冷した。氷冷下でtert-ブチル(ジメチル)シリル トリフルオロメタンスルホナート(1.46 mL)を滴下した後、室温に昇温させ20分間攪拌した。反応終了後、反応混合物に飽和炭酸水素ナトリウムを加え、混合物をCH2Cl2で抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から20/80)で精製し、エチル4-{[(1R,2R,3S,5s)-5-{[tert-ブチル(ジメチル)シリル]オキシ}アダマンタン-2-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボキシラート(2.55 g)を得た。
4-クロロ-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボニトリル(400 mg)、1-ベンジルピロリジン-3-アミン(344 mg)及びN,N-ジイソプロピルエチルアミン(0.679 mL)の混合物をマイクロウェーブ照射下180℃で1時間撹拌した。反応混合液をEtOAcで希釈した後、brineで2回洗浄した。有機層をMgSO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣を塩基性シリカゲルカラムクロマトグラフィー(EtOAc/Hx = 0/100から20/80)により精製し、4-[(1-ベンジルピロリジン-3-イル)アミノ]-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボニトリル(552 mg)を得た。
4,6-ジクロロ-5-ニトロニコチン酸エチルエステル(5.7 g)のDMF (40 mL)の混合液にrac-(3R,4R)-1-ベンジル-4-メチルピペリジン-3-アミン(4.39 g)とN,N-ジイソプロピルエチルアミン(3.7 mL)を加えた後、室温下で1時間攪拌した。反応混合物に1-(3,4-ジメトキシフェニル)メタンアミン(4.8 mL)とN,N-ジイソプロピルエチルアミン(9.4 mL)を加え、110℃で2時間攪拌した。反応混合物を水でクエンチした後、混合物をEtOAcで抽出し、水で洗浄した。有機層をMgSO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 1/4から1/2)で精製し、エチル rac-4-{[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]アミノ}-6-[(3,4-ジメトキシベンジル)アミノ]-5-ニトロニコチナート(9.9 g)を黄色アモルファスとして得た。
tert-ブチル rac-(3R,4R)-4-メチル-3-[6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]ピペリジン-1-カルボキシラート(500 mg)をCH2Cl2(5.0 ml)に溶解させ、氷冷した。氷冷下でN-ブロモスクシンイミド(183 mg)を加え、10分後、さらに飽和炭酸水素ナトリウム水溶液を加え、室温に昇温させ、CH2Cl2にて混合物を抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から30/70)で精製し、rac-tert-ブチル (3R,4R)-4-メチル-3-[8-ブロモ-6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]ピペリジン-1-カルボキシラート(202 mg)を得た。
rac-tert-ブチル (3R,4R)-4-メチル-3-[8-ブロモ-6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]ピペリジン-1-カルボキシラート(21.2 mg)、ジシアノ亜鉛(6.6 mg)、テトラキストリフェニルホスフィンパラジウム(4.3 mg)、DMF (0.3 mL)を加え、マイクロウェーブ反応装置にて100℃で、1時間反応を行った。反応終了後、反応混合物に飽和炭酸水素ナトリウム水溶液を加え、EtOAcにて混合物を抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から20/80)で精製し、rac-tert-ブチル (3R,4R)-3-[8-シアノ-6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]-4-メチルピペリジン-1-カルボキシラート(15.4 mg)を得た。
(4-{[(1R,2R,3S,5s)-5-{[tert-ブチル(ジメチル)シリル]オキシアダマンタン-2-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-イル)メタノール(536 mg)と二酸化マンガン(835 mg)の混合物にCH2Cl2 (10 ml)を加え、2時間加熱還流した。反応終了後、反応混合物をセライトろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から20/80)で精製し、4-{[(1R,2R,3S,5s)-5-{[tert-ブチル(ジメチル)シリル]オキシ}アダマンタン-2-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルバルデヒド(465 mg)を得た。
rac-エチル 4-{[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]アミノ}-6-[(3,4-ジメトキシベンジル)アミノ]-5-ニトロニコチナート(2.65 g)をEtOH (88 mL)と水(11 mL)に溶解した後、混合液に、還元鉄(788 mg)と塩化アンモニウム(251 mg)を加え、120℃で4時間攪拌した。反応混合物を室温まで冷却し、飽和炭酸水素ナトリウム水溶液を加えて30分攪拌した。不溶物をセライトろ過し、ろ液を減圧下濃縮した。残渣をCHCl3にて抽出し、水で洗浄した。有機層をMgSO4で乾燥後、ろ過し、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH = 100/0から95/5)で精製し、rac-エチル 5-アミノ-4-{[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]アミノ}-6-[(3,4-ジメトキシベンジル)アミノ]ニコチナート(1.4 g)を黄色アモルファスとして得た。
4-[(1-ベンジルピロリジン-3-イル)アミノ]-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボニトリル(548 mg)とTHF (5.5 mL)の混合物を-70℃に冷却し、水素化ジイソブチルアルミニウム(1.0M Hx溶液、3.06 mL)を加え、氷冷下で1時間撹拌した。反応混合物にMeOHをゆっくり加えた後に、0.5M塩酸水溶液(8 mL)とEtOAc (4 mL)を加えて1時間攪拌した。反応混合物に1M水酸化ナトリウム水溶液を加えて中和し、混合物をEtOAcで抽出した。有機層をMgSO4で乾燥後、ろ過し、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(MeOH/ CHCl3 = 0/100から2/98)により精製し、4-[(1-ベンジルピロリジン-3-イル)アミノ]-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルバルデヒド(236 mg)を得た。
製造例22の方法と同様にして後記表に示す製造例23及び23-1~23-2の化合物を製造した。
THF (75 mL)を氷冷し、水素化アルミニウム(316 mg)の粉末を加えた。続いて、エチル 4-{[(1R,2R,3S,5s)-5-{[tert-ブチル(ジメチル)シリル]オキシ}アダマンタン-2-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボキシラート(2.50 g)のTHF溶液(75 mL)を30分かけて滴下し、室温に昇温させ、4時間攪拌した。その後、室温下で15時間静置した後、35 ℃で二時間攪拌した。再び氷冷し、水(0.32 mL)、15 %水酸化ナトリウム水溶液(0.32 mL)、水(0.32 mL)を順次加え、室温に戻し、1時間攪拌した。反応混合物をセライトろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から30/70)で精製し、(4-{[(1R,2R,3S,5s)-5-{[tert-ブチル(ジメチル)シリル]オキシ}アダマンタン-2-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-イル)メタノール(1.10 g)を得た。
rac-(1R,4S,6S)-2-ベンジル-2-アザビシクロ[2.2.1]ヘプタン-6-オール(1.00 g)に1H-イソインドール-1,3(2H)-ジオン(796 mg)、トリブチルホスフィン(1.46 mL)及びトルエン(30 mL)の混合物に、1,1'-(アゾジカルボニル)ジピペリジン(1.49 g)を氷冷下で加え、室温で終夜撹拌した。次に、反応混合物に1H-イソインドール-1,3(2H)-ジオン(362 mg)、トリブチルホスフィン(0.728 mL)及び1,1'-(アゾジカルボニル)ジピペリジン(745 mg)を加えて、室温で4時間攪拌した。反応混合物にジイソプロピルエーテル(30 mL)を加えて、30分攪拌後、反応混合物をろ過し、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 0/100から34/66)により精製し、rac-2-[(1R,4S,6S)-2-ベンジル-2-アザビシクロ[2.2.1]ヘプト-6-イル]-1H-イソインドール-1,3(2H)-ジオン(1.21 g)を得た。
tert-ブチル (4-メチルピリジン-3-イル)カルバマート(12 g)のアセトン(480 mL)溶液にベンジルブロミド(6.98 mL)を加え、75℃で3時間撹拌した。室温に戻した後、生じた固体を濾取し、アセトン(120 mL)で洗浄し、1-ベンジル-3-[(tert-ブトキシカルボニル)アミノ]-4-メチルピリジニウム ブロミド(20.96 g)を淡黄色結晶として得た。
rac-1-ベンジル-3-[(tert-ブトキシカルボニル)アミノ]-4-メチルピリジニウム ブロミド(1.2 g)のEtOH (48 mL)溶液に、酸化白金(Adam's Catalyst)(36 mg)を加え、40℃、3気圧で4時間接触還元を行った。反応混合物をセライトで濾別し、ろ液を減圧下濃縮した。得られた残渣を飽和炭酸水素ナトリウム水溶液(50 mL)で中和後、混合物をCHCl3 (200 mL)で抽出した。有機層をMgSO4 (20 g)で乾燥後、ろ過し、ろ液を減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から13/87)により精製し、tert-ブチル rac-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]カルバマート(664 mg)を無色油状物として得た。
製造例1、製造例3及び製造例6と同様の方法を連続的に行うことにより、後記表に示す製造例28及び28-1~28-2の化合物を製造した。
rac-tert-ブチル (3R,4R)-3-[3-ブロモ-8-シアノ-6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]-4-メチルピペリジン-1-カルボキシラート(125 mg)、ピリジン-4-イルボロン酸(31.3 mg)、テトラキストリフェニルホスフィンパラジウム(24.5 mg)、ジオキサン(1.6 mL)、2M 炭酸ナトリウム水溶液(0.8 mL)を加え、100 ℃で1時間反応を行った。反応終了後、反応混合物に水を加え、CH2Cl2にて抽出した。得られた抽出液をbrineで洗浄した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残留をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 30/70から95/5)で精製した。さらに、シリカゲルカラムクロマトグラフィー(MeOH/CHCl3 = 0/100から5/95)で精製し、rac-tert-ブチル (3R,4R)-3-[8-シアノ-3-ピリジン-4-イル-6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]-4-メチルピペリジン-1-カルボキシラート(120 mg)を得た。
rac-4-{[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ [2,3-b]ピリジン-5-カルバルデヒド(1.75g)、トリフェニルホスフィン(5.75g)、CH2Cl2 (17.5mL)の混合物に氷冷下で四臭化炭素(3.64g)を加え、室温で1時間攪拌した。反応混合物に氷冷下で飽和炭酸水素ナトリウム水溶液を加えた後に、CHCl3で抽出し有機層を飽和塩化ナトリウム水溶液で洗浄した。有機層をMgSO4で乾燥後、ろ過し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィーにより精製し(EtOAc/Hx = 0-10%)、rac-N-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-5-(2,2-ジブロモビニル)-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-4-アミン(2.07g)を得た。
ベンジル rac-(3R,4R)-3-[3-(1-アセチル-1,2,3,6-テトラヒドロピリジン-4-イル)ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]-4-メチルピペリジン-1-カルボキシラート(27 mg)、10%パラジウムカーボン粉末(含水品、エヌ・イーケムキャット社製)(5.6 mg)、MeOH (0.6 ml)、ギ酸アンモニウム(33 mg)を加え、80℃で15分間攪拌した。反応混合物をセライトでろ過し、ろ液を減圧下濃縮した。残渣に飽和炭酸水素ナトリウム水溶液を加え、MeOH/CHCl3 (9/1)の混合溶液で抽出し、得られた有機層をNa2SO4で乾燥後、ろ過し、減圧下濃縮した。得られた残渣をジイソプロピルエーテルにて洗浄し、白色粉末のrac-1-(4-{1-[(3R,4R)-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン-3-イル}ピペリジン-1-イル)エタノン(10 mg)を得た。
rac-1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(160 mg)のDMF (3.2 mL)溶液にN,N-ジメチレンアンモニウム ヨージド(81 mg)加え、反応混合物を60℃にて1.5時間攪拌した。室温に冷却後、反応混合物に飽和炭酸水素ナトリウム水溶液を加え、攪拌した。反応混合物をCHCl3にて抽出し、水で洗浄した。有機層をMgSO4で乾燥後、ろ過し、ろ液を減圧濃縮した。得られた残留をシリカゲルカラムクロマトグラフィー(溶出液;CHCl3:MeOH =100 : 0~92 : 8)で精製し、rac-1-(1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン-8-イル)-N,N-ジメチルメタンアミン(172 mg)を淡黄色油状物として得た。
1-(ピペリジン-4-イル)-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン (200 mg)、K2CO3 (224 mg)及びKI (448 mg)をアセトニトリル(4 mL)に加え、更に室温でブロモアセトニトリル(324 mg)を加えて4時間攪拌した。反応混合物に純水を加え、EtOAcで抽出した。有機層を飽和塩化ナトリウム水溶液で一回洗浄し、有機層を分離後、MgSO4で乾燥した。有機層をろ過し、ろ液を濃縮して、褐色油状物を得た。これをシリカゲルカラムクロマトグラフィー(CHCl3/MeOH (1 : 0~20 : 1)にて精製し、薄褐色油状物の{4-[6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1-(6H)-イル]ピペリジン-1-イル}アセトニトリル(205 mg)を得た。
rac-N-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-5-(2,2-ジブロモビニル)-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-4-アミン(2.06 g)とDMSO (20 mL)の混合物に1,8-ジアザビシクロ[5.4.0]-7-ウンデセン(1.46 mL)を水冷下で加え、室温で1時間攪拌した。水冷後、冷却した1N塩酸で反応混合物を中和し、EtOAcで抽出した。有機層を水、飽和塩化ナトリウム水溶液で順次洗浄後、有機層を分離し、有機層をMgSO4で乾燥した。この有機層をろ過し、減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィーで精製し(EtOAc/Hx = 0-10%)、褐色油状物のrac-N-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-5-(ブロモエチニル)-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-4-アミン(1.80 g)を得た。
rac-N-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-5-(ブロモエチニル)-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ [2,3-b]ピリジン-4-アミン(710 mg)とTHF (7.1mL)の混合物に-50℃でn-ブチルリチウム(1.65M Hx溶液, 1.71 mL)を滴下し、30分攪拌した。その後、反応混合物にヨウ化メチル (0.958 mL)を加え、室温で1.75時間攪拌した。反応混合物に飽和塩化アンモニウム水溶液を加え、EtOAcで抽出し、有機層を飽和塩化ナトリウム水溶液で洗浄した。有機層を分離後、MgSO4で乾燥した。有機層をろ過し、減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 0/100から7/93)で精製し、黄色油状物のrac-N-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-5-プロプ-1-イン-1-イル-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-4-アミン (316 mg)を得た。
rac-N-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-5-プロプ-1-イン-1-イル-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-4-アミン(440 mg)とTHF (4.4 mL)の混合物に室温でカリウム tert-ブトキシド(253mg)を加え、50℃に昇温して1時間攪拌した。その後、室温に戻し、カリウム t-ブトキシド(253 mg)を加えて、50℃で更に1.5時間攪拌した。反応混合物に飽和塩化アンモニウム水溶液を加え、CHCl3で抽出した。有機層を飽和塩化ナトリウム水溶液で洗浄後、有機層を分離した。この有機層をMgSO4で乾燥後、ろ過し、減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 0-15%)で精製し、黄色油状物のrac-1-[(3R,4R)-1-ベンジル-4-メチルピペリジン-3-イル]-2-メチル-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロイジピロロ [2,3-b:2',3'-d]ピリジン(294 mg)を得た。
cis-4-[6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d] ピリジンー1(6H)-イル]シクロヘキシル ベンゾアート (214 mg)をMeOH (4.3 mL)に加え、更に室温で1 M NaOH (0.9 mL)を加えて、60℃で1時間攪拌した。その後、MeOH (4.3 mL)及び1 M NaOH (0.9 mL)を室温で順次加え、60℃で1時間攪拌した。反応混合物に純水を加え、EtOAcで一回抽出した。有機層を飽和塩化ナトリウム水溶液で一回洗浄し、有機層を分離後、MgSO4で乾燥した。有機層を濾過し、濃縮して、無色油状物のcis-4-[6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d] ピリジンー1(6H)-イル]シクロヘキサノール (168 mg)を得た。
窒素気流下、スルフリルジクロリド (1.80 g)とCH2Cl2 (30 ml)の混合物を-60℃に冷却した。混合物に(メチルアミノ)アセトニトリル(920 mg)、4-ジメチルアミノピリジン (1.48 g)及びCH2Cl2 (20 ml)の混合物を10分かけて滴下し、室温に昇温して終夜攪拌した。反応混合物にシリカゲルを加えて、溶媒を減圧留去した。得られた残渣をシリカゲルでろ過(hexane/EtOAc = 1/1)し、(シアノメチル)メチルスルファミルクロリド(1.09 g)を得た。
6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(200 mg)にジオキサン(2 ml)、3-ヨードピリジン(171 mg)、リン酸カリウム(310 mg)、rac-(1R,2R)-シクロヘキサン-1,2-ジアミン (64 mg)及びヨウ化銅(I)(53 mg)を加えて110℃で終夜攪拌した。反応混合物にEtOAcを加え、不溶物をろ去した。ろ液を減圧下濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(Hx/EtOAc = 100/0~50/50)で精製し、淡黄色油状物の1-(ピリジン-3-イル)-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(192 mg)を得た。
4-[(4-メトキシベンジル)アミノ]-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボニトリル(120 mg)にトルエン(4 ml)、水(1 ml)、2,3-ジクロロ-5,6-ジシアノ-1,4-ベンゾキノン(200 mg)を加え、80℃で2時間攪拌した。反応混合物に飽和炭酸水素ナトリウム水溶液を加え、CHCl3にて抽出した。抽出液をMgSO4で乾燥後、ろ過し、ろ液を減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(Hx/EtOAc = 100/0~70/30)で精製し、白色固体の4-アミノ-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルボニトリル(81 mg)を得た。
rac-tert-ブチル (3R,4R)-3-[3-ブロモ-6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]-4-メチルピペリジン-1-カルボキシラート(370 mg)をMeOH (3.0 ml)に溶解させた。混合物に4M 塩化水素ージオキサン溶液 (3.0 ml)を加えて、室温で30分間攪拌した。反応混合物を減圧下濃縮し、飽和炭酸水素ナトリウム水溶液を加えた後にCH2Cl2にて抽出した。有機層をNa2SO4で乾燥後、ろ過し、減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(MeOH:CHCl3=0:100~10:90)で精製し、rac-3-ブロモ-1-[(3R,4R)-4-メチルピペリジン-3-イル]-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(283 mg)を得た。
前述の実施例7の方法と同様にして製造例42の化合物を製造した。
rac-3-ブロモ-1-[(3R,4R)-4-メチルピペリジン-3-イル]-6-{[2-(トリメチルシリル)エトキシ]メチル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(280 mg)、CH2Cl2 (3.0 ml)及び飽和炭酸水素ナトリウム水溶液(3.0 ml)の混合物にクロロギ酸ベンジル(104μl)を加え、室温にて15分間攪拌した。反応終了後、飽和炭酸水素ナトリウム水溶液を加え、CH2Cl2にて抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc:Hx=10:90~30:70)で精製し、ベンジル rac-(3R,4R)-3-[3-ブロモ-6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピルジン-1(6H)-イル]-4-メチルピペリジン-1-カルボキシラート(328 mg)を得た。
製造例25の方法と同様にして、trans-4-[6-{[2-(トリメチルシリル)エトキシ]メチル}ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル]シクロヘキサノールを原料として後記表に示す製造例44の化合物及び製造例44-1の化合物を製造した。
(メトキシメチル)トリフェニルホスホニウム クロリド(315 mg)にTHF (1.0 mL)を加え、氷冷下でナトリウムビス(トリメチルシリル)アミド(1.07 M THF溶液、1.01 mL)を滴下し、40分間攪拌した。この反応混合物に4-{[(1R,2R,3S,5s)-5-{[tert-ブチル(ジメチル)シリル]オキシ}アダマンタン-2-イル]アミノ}-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-5-カルバルデヒド(465 mg)のTHF (5.0 mL)溶液を滴下した後、室温で2時間攪拌した。反応混合物に飽和塩化アンモニウム水溶液を加えた後、混合物をEtOAcで抽出し、brineで洗浄した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(EtOAc/Hx = 10/90から20/80)で精製し、N-[(1R,2R,3S,5s)-5-{[tert-ブチル(ジメチル)シリル]オキシ}アダマンタン-2-イル]-5-(2-メトキシビニル)-1-{[2-(トリメチルシリル)エトキシ]メチル}-1H-ピロロ[2,3-b]ピリジン-4-アミンの粗生成物を得た。
この粗生成物(445 mg)にMeOH (5.0 mL)と塩化アセチル(0.16 mL)を加え、80 ℃で1時間攪拌した。反応混合物に水(0.5 mL)を加え、80 ℃で30分間攪拌した。反応終了後、反応混合物に飽和炭酸水素ナトリウム水溶液を加えた後、混合物をCHCl3で抽出し、brineで洗浄した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣にCH2Cl2 (3.0 mL)とTFA (3.0 mL)を加え、室温で2時間攪拌した。反応混合物を減圧下濃縮し、残渣にCH2Cl2 (2.0 mL)、MeOH (1.0 mL)、エチレンジアミン(0.2 mL)、1M 水酸化ナトリウム水溶液(3.0 mL)を加え、室温で1時間攪拌した。反応混合物に水を加えた後、混合物をCHCl3で抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をMeOH及びジイソプロピルエーテルにより洗浄し(1s,3R,4R,5S)-4-(ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル)アダマンタン-1-オール(145 mg)を白色固体として得た。
1-ピロリジン-3-イル-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(69.2 mg)、シアノ酢酸(52.0 mg)、1-ヒドロキシベンゾトリアゾール(62.0 mg)、N-(3-ジメチルアミノプロピル)-N'-エチルカルボジイミド塩酸塩(87.9 mg)、Et3N (0.0853 mL)、DMF (2.5 mL)を加え50℃で30分間攪拌した。反応終了後、反応混合物を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(MeOH/CHCl3 = 0/100から10/90)により精製した。さらに、得られた混合物を塩基性シリカゲルカラムクロマトグラフィー(MeOH/CHCl3 = 0/100から4/96)により精製した。得られた固体をジイソプロピルエーテルで洗浄し、減圧下で乾燥することで3-(3-ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イルピロリジン-1-イル)-3-オキソプロパンニトリル(65.4 mg)を得た。
前述の製造例19の方法と同様にして、後記表に示す実施例3の化合物を製造した。
rac-1-[(3R,4R)-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(50 mg)と4-ニトロフェニル (シアノメチル)カルバマート(87 mg)のDMF(1.0 mL)溶液に、Et3N (0.082 mL)を加え、マイクロウェーブ照射下、120℃で30分間攪拌した。反応終了後、反応混合物に水を加え、混合物をCHCl3で抽出し、1N水酸化ナトリウム水溶液及びbrineで洗浄した。有機層をMgSO4で乾燥後、ろ過し、ろ液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(MeOH/CHCl3 = 5/95)で精製し、得られた固体をイソプロパノール-ジイソプロピルエーテルで洗浄後、濾取し、rac-(3R,4R)-N-(シアノメチル)-3-ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル-4-メチルピペリジン-1-カルボキサミド(25 mg)を白色粉末として得た。
rac-1-[(3R,4R)-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(130 mg)のDMF (20 mL)溶液1.00 mLとフェニルメタンスルホニル クロリド (0.040 mmol)との混合物にピリジン(0.5 mL)を加え、室温で終夜攪拌した。反応混合物にMP-Carbonate (Biotage 800269) (50 mg)とPS-NCO (Biotage 800262) (50 mg)を加え、2時間攪拌した。反応混合物をろ過し、ろ液を濃縮した。この濃縮液をMSトリガーを備える液体クロマトグラフィー(LC)(aq. HCOOH/MeOH)にて分取(LC分取条件:SunFireカラム 5μm 19*100mm、MeOH/0.1 % aq. HCOOH = 10/90 (0 min) - 10/90 (1 min) - 95/5 (8 min) - 95/5 (12 min) - 10/90 (13 min))精製し、rac-1-[(3R,4R)-1-(ベンジルスルホニル)-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ [2,3-b:2',3'-d]ピリジン (3.2 mg)を得た。
rac-1-[(3R,4R)-4-メチルピペリジン-3-イル]-1,6-ジヒドロピロロ[2,3-b:2',3'-d]ピリジン(67 mg)に室温でジクロロエタン(2 mL)を加えた後、氷冷下でジイソプロピルエチルアミン(95μL)及びトリフルオロ酢酸無水物(56μL)を加えた。室温で1時間攪拌した後、反応混合物に飽和塩化アンモニウム水溶液を加え、混合物をCH2Cl2で抽出した。有機層をNa2SO4で乾燥後、ろ過し、ろ液を減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH = 100/0~92/8)で精製し、白色固体のrac-1-[(3R,4R)-4-メチル-1-(トリフルオロアセチル)ピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(56 mg)を得た。
前述の製造例6の方法と同様にして、後記表に示す実施例8及び実施例8-1~8-8の化合物を製造した。
rac-1-[(3R,4R)-4-メチルピペリジン-3-イル]-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(100 mg)のDMF (4 ml)溶液にEt3N (200μl)、2,6-ジメチルモルホリン-4-スルホニルクロリド(100 mg)を加え、室温で2時間攪拌した。反応混合物に水を加え、EtOAcで抽出した。抽出液をbrineで洗浄し、MgSO4で乾燥後、ろ過し、ろ液を減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH = 100/0~95/5)で精製し、白色固体のrac-1-{(3R,4R)-1-[(2,6-ジメチルモルホリン-4-イル)スルホニル]-4-メチルピペリジン-3-イル}-1,6-ジヒドロジピロロ[2,3-b:2',3'-d]ピリジン(131 mg)を得た。
Claims (16)
- 式(I)の化合物又はその塩。
Aは、置換されていてもよいシクロアルキル、置換されていてもよいシクロアルケニル、又は、置換されていてもよい含窒素ヘテロ環基であり、
Xは、CRX、又は、Nであり、
RXは、H、ORXY1、NRXY2RXY3、SRXY4、ハロゲン、シアノ、又は、低級アルキル、アリール若しくはヘテロ環基であり、
Yは、H、ORXY1、NRXY2RXY3、SRXY4、ハロゲン、シアノ、置換されていてもよい低級アルキル、置換されていてもよいアリール、又は、置換されていてもよいヘテロ環基であり、
Zは、H、又は、低級アルキルであり、
RXY1、RXY2、RXY3及びRXY4は、互いに同一又は異なって、H、又は、低級アルキルであり、
R1は、H、OH、-(CR11R12)m-R13、-SO2-R14、又は、置換されていてもよいヘテロ環基であり、
R11及びR12は、互いに同一又は異なって、H、ハロゲン、OH、、置換されていてもよい低級アルキル、置換されていてもよいアリール、又は、置換されていてもよいヘテロ環基、
R11とR12が一体となってオキソ(=O)、若しくは、
R11とR12がそれらが結合する炭素原子と一体となって置換されていてもよいシクロアルキルを形成してもよく、
R13は、H、ハロゲン、シアノ、-NRN1RN2、又は、置換されていてもよいアリール、置換されていてもよいシクロアルキル、又は、置換されていてもよいヘテロ環基であり、
RN1及びRN2は、互いに同一又は異なって、H、置換されていてもよい低級アルキル、又は、置換されていてもよいアリールであり、
R14は、NRN3RN4、又は、置換されていてもよい低級アルキル、又は、置換されていてもよいヘテロ環基であり、
RN3及びRN4は、互いに同一又は異なって、H、置換されていてもよい低級アルキル、又は、置換されていてもよいアリールであり、
mは、1、2、3若しくは4である。) - Aが、置換されていてもよいシクロアルキル、又は、置換されていてもよい含窒素ヘテロ環基であり、
Xが、CRX、又は、Nであり、
RXが、H、ORXY1、NRXY2RXY3、SRXY4、ハロゲン、シアノ、又は、低級アルキル、アリール若しくはヘテロ環基であり、
Yが、H、ORXY1、NRXY2RXY3、SRXY4、ハロゲン、シアノ、又は、低級アルキル、アリール若しくはヘテロ環基であり、
RXY1、RXY2、RXY3及びRXY4が、互いに同一又は異なって、H、又は、低級アルキルであり、
R1が、H、OH、-(CR11R12)m-R13、-SO2-(置換されていてもよい低級アルキル)、又は、置換されていてもよいヘテロ環基であり、
R11及びR12が、互いに同一又は異なって、H、ハロゲン、OH、置換されていてもよい低級アルキル、置換されていてもよいアリール、又は、置換されていてもよいヘテロ環基、
R11とR12が一体となってオキソ(=O)
R11とR12がそれらが結合する炭素原子と一体となってシクロアルキルを形成してもよく、
R13が、H、ハロゲン、シアノ、-NRN1RN2、置換されていてもよいアリール、置換されていてもよいシクロアルキル、又は、置換されていてもよいヘテロ環基であり、
RN1及びRN2が、互いに同一又は異なって、H、置換されていてもよい低級アルキル又は、置換されていてもよいアリールであり、
mは、1、2、3若しくは4である請求項1記載の化合物又はその塩。 - RA1は、H、又は、メチルである請求項4記載の化合物又はその塩。
- R1が、-(CR11R12)m-R13であり、R13が、シアノ、-NRN1RN2、又は、低級アルキルで置換されていてもよいヘテロ環基である請求項5記載の化合物又はその塩。
- R1が、-C(=O)-R13、又は、-C(=O)-CH2-R13であり、R13が、シアノ、-NRN1RN2、又は、低級アルキルで置換されていてもよい1H-テトラゾール-1-イルであり、RN1及びRN2が、互いに同一又は異なって、H、又は、シアノで置換されていてもよい低級アルキルである請求項6記載の化合物又はその塩。
- R1が、5-シアノピリジン-2-イル若しくは5-シアノピラジン-2-イルである請求項5記載の化合物又はその塩。
- Xが、CRXであり、RXが、H、ブロモ、又は、シアノであり、Yが、H、ハロゲン、シアノ、又は、ヘテロ環基である請求項7及び請求項8記載の化合物
- rac-3-[(3R,4R)-3-(ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル)-4-メチルピペリジン-1-イル]-3-オキソプロパンニトリル、
rac-(1S,3R,4R,5S)-4-(ジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル)-アダマンタン-1-オール、
rac-3-[(3R,4R)-3-(3-ブロモジピロロ[2,3-b:2',3'-d]ピリジン-1(6H)-イル)-4-メチルピペリジン-1-イル]-3-オキソプロパンニトリル、

- 請求項1に記載の化合物又はその塩を含有する望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防用若しくは治療用医薬組成物。
- 望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防用若しくは治療用医薬組成物の製造のための請求項1に記載の化合物又はその塩の使用。
- 望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防若しくは治療のための請求項1に記載の化合物又はその塩の使用。
- 請求項1に記載の化合物又はその塩の有効量を患者に投与することからなる望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防若しくは治療方法。
- 望ましくないサイトカインシグナル伝達によって引き起こされる疾患、あるいは異常なサイトカインシグナル伝達によって引き起こされる疾患の予防若しくは治療のための請求項1に記載の化合物又はその塩。
