Description
また、特許文献2ではアルコールまたはアルコールと水との混合溶媒中で、アミノ酸のアルカリ金属塩またはエステル化によりカルボキシル基を保護したアミノ酸に対して、炭素数4~30の1,2-エポキシアルカンを反応させることを特徴とするN-長鎖(炭素数4~30)アルキルアミノ酸誘導体が報告されている。
しかしながら、上記アルギニン誘導体やN-長鎖(炭素数4~30)アルキルアミノ酸誘導体は界面活性剤であり、つっぱり感やごわつき感を生じる場合があり、化粧料への配合量が制限されていた。
一方、特許文献3では、アルギニンとグリシドールまたは3-ハロ-1,2-プロパンジオールを反応させることで得られるアルギニンN-グリセリル誘導体(N-(2,3-ジヒドロキシプロピル)-L-アルギニン)が一例として報告されている。しかしながら、アルギニンN-グリセリル誘導体はアルギニンに比べ高い保湿性を有し、つっぱり感やごわつき感を生じないものの、肌や毛髪とのなじみ性、肌の柔軟化や整肌効果といった点から十分に満足できるものではなかった。
さらに、製造方法の観点から見ると、特許文献1の製造方法では、炭素数8~22のハロゲン化アシル化合物や炭素数12~22の高級アルコールを、また特許文献2の製造方法では炭素数4~30の1,2-エポキシアルカンを反応に際して相溶化させるため多量の有機溶媒を必要とすることから、環境負荷が大きく、化粧料用基剤として使用する場合は残留溶媒が懸念される。特許文献3の製造方法では、アミノ酸のα-アミノ基を活性化させるため、反応に際してアルカリ剤や酸剤でpHを8~11に調整している。しかしながら、アミノ酸とグリシドールまたは3-ハロ-1,2-プロパンジオールとをアルカリ金属等の存在下で反応させた場合、副反応に伴う不純物が発生してしまうことから、化粧料として使用する場合は不純物の影響による使用感の悪化が問題であった。
(1)下記一般式(1)で示されるアルギニン誘導体。
(2) 前記(1)項に記載のアルギニン誘導体を配合する化粧料。
(3) アルギニンを水に溶解し、炭素数2~3のアルキレンオキシドを付加する際に無触媒で反応を行う下記一般式(1)で示されるアルギニン誘導体の製造方法。
式(1)で示されるアルギニン誘導体において、R1およびR2は、水素原子、もしくは、炭素数2~3の直鎖または分岐のモノヒドロキシアルキル基であり、具体的にはモノヒドロキシエチル基、モノヒドロキシプロピル基が挙げられる。肌や毛髪へのなじみ性、肌の柔軟化効果についてはモノヒドロキシプロピル基がより好ましく、整肌効果についてはモノヒドロキシエチル基が好ましい。モノヒドロキシアルキル基のアルキル鎖長が3を超える場合、界面活性能が発現されつっぱり感の原因となる場合がある。
出発原料としてのアルギニンは、D体、L体およびDL体のいずれも使用できるが、汎用的に入手できる点でL体が好ましい。
水素原子以外のR3は、アルカリ金属水酸化物、アンモニア、有機アミンなどの塩基をアルギニンおよび式(1)で表されるアルギニン誘導体に作用させることで導入できる。
しかしながら、アルギニンとアルキレンオキシドとの反応前に上記塩基を作用させると、アルキレンオキシドの副反応が促進されて不純物が生成しやすくなる。このためR3を導入する場合、式(1)で表されるアルギニン誘導体に上記塩基を作用させることが好ましい。
式(1)で表されるアルギニン誘導体を得るためのアルギニンとエチレンオキシドおよびプロピレンオキシドとの反応は、水溶媒中、無触媒で行うことができる。具体的には、所定濃度のアルギニン水溶液を調製し、pH無調整で、加温しながら、撹拌下、エチレンオキシドやプロピレンオキシドを滴下しながら反応を行うことで、式(1)で表されるアルギニン誘導体が得られ、酸剤でpH調製をすることでアルギニン誘導体の酸付加塩が得られる。
この反応に際して、アルコールなどの水溶性有機溶媒を使用することもできるが、エチレンオキシドやプロピレンオキシドは任意に水と相溶するため、水溶性有機溶媒を使用しなくても全く問題なく反応は進行する。また、アルコールとアルキレンオキシドとの副反応が生じる可能性、残留溶媒の危険性、環境への負荷等を考慮すると、水溶性有機溶媒を使用せず、水を溶媒とすることが好ましい。
反応温度はこの好ましくは30~90℃、より好ましくは40~80℃、さらに好ましくは45~70℃である。反応温度が30℃未満では、反応速度が遅くなり、反応温度が90℃より高いと臭いの発生や着色が起こりやすくなる。
反応時のエチレンオキシドやプロピレンオキシドの滴下時間は反応量によって異なるが、滴下終了後は反応を完結させるために、加温しながら3~10時間攪拌を続けるのが好ましい。
1モル付加体を合成する場合、反応はほぼ定量的に進行するため過剰のエチレンオキシドまたはプロピレンオキシドは不要であるが、2モル付加体を合成する場合は、2級化されたアミノ基の反応性が1級アミノ基に較べて低下するため、過剰のエチレンオキシドまたはプロピレンオキシドが必要になる。過剰量としては、必要付加モル数に対して、1.1~1.4倍であり、より好ましくは1.2~1.3倍である。
反応終了後は、窒素によるバブリング等で未反応のエチレンオキシドやプロピレンオキシドを容易に除去できることも本発明のアルギニン誘導体の製造方法の特徴である。得られたアルギニン誘導体は、水溶液として用いることも、スプレードライ等の処理を行って粉体化して用いることも可能である。
本発明の化粧料の剤型は任意であり、溶液系、可溶化系、乳化系、ゲル系、粉末分散系、水-油二層系等いずれも可能であり、目的とする製品に応じて上記式(1)で表されるアルギニン誘導体と上記任意配合成分とを配合して製造することができる。
<分析条件>
○1H-NMR 核磁気共鳴分光法(NMR)は、日本電子(株)製 ECP-400(400MHz)を使用した。1.5~1.7ppmに出現するアルギニンのβ-炭素、γ-炭素に結合している水素のピーク面積と3.8ppm付近に出現する2-ヒドロキシアルキル基の2位の炭素に結合している水素のピーク面積との比率から、α-アミノ基に導入された2-ヒドロキシアルキル基の数を算出した。
キャピラリー電気泳動(CP) キャピラリー電気泳動装置は、大塚電子(株)製 CAPI3330を使用し、下記の条件で各試料について測定を行った。

なお、各アルギニン誘導体は凍結乾燥したものを測定試料とした。

反応組成物の一部を取り出し、凍結乾燥にて水分を除去したものを分析用のサンプルとした。1H-NMRスペクトルから、α-アミノ基へ導入された2-ヒドロキシエチル基は1.02モルであった。また、キャピラリー電気泳動(CP)による分析から、保持時間8分に主ピークが確認され、その他にごく少量の他成分が含まれていたが、1H-NMRとCPによる分析結果から、主成分はN-(2-ヒドロキシエチル)-L-アルギニンであると判断した。
オートクレーブから取り出した反応生成物に対し、固形分濃度が30質量%になるようにイオン交換水を加え、N-(2-ヒドロキシエチル)-L-アルギニン30質量%水溶液を調製し、以下の検討に用いた。

オートクレーブから取り出した反応生成物に対し、固形分濃度が30質量%になるようにイオン交換水を加え、N,N-ビス(2-ヒドロキシプロピル)-L-アルギニン30質量%水溶液を調製し、以下の検討に用いた。

オートクレーブから取り出した反応生成物に対し、固形分濃度が30質量%になるようにイオン交換水を加え、N-(2,3-ジヒドロキシプロピル)-L-アルギニン30質量%水溶液を調製し、以下の検討に用いた。
L-アルギニン174g(1モル)をイオン交換水203gおよび2-プロパノール203gに溶解し、オートクレーブに仕込み、オートクレーブ内の空気を乾燥窒素で置換した。次に80℃にオートクレーブを加温し、滴下装置によりブチレンオキシド72g(1モル)を1時間かけて徐々に滴下し、滴下終了後、さらに6時間攪拌した。反応終了後、オートクレーブから反応組成物を取り出した。これをロータリーエバポレータにより、2-プロパノールおよび水を留去し、さらに減圧乾燥して白色固体の反応生成物を得た。
1H-NMRスペクトルから、α-アミノ基へ導入された2-ヒドロキシブチル基は0.97モルであった。また、キャピラリー電気泳動(CP)による分析から、保持時間9分10秒に主ピークが確認され、その他にごく少量の他成分が含まれていたが、1H-NMRとCPによる分析結果から、主成分はN-(2-ヒドロキシブチル)-L-アルギニンであると判断した。
減圧乾燥した反応生成物に対し、固形分濃度が30質量%になるようにイオン交換水を加え、N-(2-ヒドロキシブチル)-L-アルギニン30質量%水溶液を調製し、以下の検討に用いた。
L-アルギニン174g(1モル)をイオン交換水348gおよび2-プロパノール348gに溶解し、オートクレーブに仕込み、オートクレーブ内の空気を乾燥窒素で置換した。次に80℃にオートクレーブを加温し、滴下装置によりドデシレンオキシド(1,2-エポキシドデカン)184g(1モル)を1時間かけて徐々に滴下し、滴下終了後、さらに6時間攪拌した。反応終了後、オートクレーブから反応組成物を取り出した。これをロータリーエバポレータにより、2-プロパノールおよび水を留去し、さらに減圧乾燥して白色固体の反応生成物を得た。

減圧乾燥した反応生成物に対し、固形分濃度が30質量%になるようにイオン交換水を加え、N-(2-ヒドロキシドデシル)-L-アルギニン30質量%水溶液を調製し、以下の検討に用いた。
表1に示すように本発明のアルギニン誘導体として化合物1~4、比較成分として化合物5~7およびL-アルギニン、共通添加成分として表2に示す成分を選定し、スキンローションを調製した。尚、L-グルタミン酸およびクエン酸によってpHを6.0~7.0に調整し、以下の使用感テストに用いた。
<評価方法>
A.「肌へのなじみ性」について
20名の専門パネラーによる使用感テストを行い、スキンローション0.5mLを手の甲に塗布した際の肌へのなじみ性について、パネラー各人が下記絶対評価にて4段階に評価し評点を付けた。そして、試料ごとにパネラー全員の評点を合計し、30点以上を合格とした。ただし、3名以上のパネラーが0点の評点を付けた場合は不合格とした。

3 : 肌へのなじみ性が良好
2 : 肌へのなじみ性がやや良好
1 : 肌へのなじみ性がやや悪い
0 : 肌へのなじみ性が非常に悪い
20名の専門パネラーによる使用感テストを行い、スキンローション0.5mLを手の甲に塗布し、塗布後2時間後の肌の柔軟化効果について、パネラー各人が下記絶対評価にて4段階に評価し評点を付けた。そして、試料ごとにパネラー全員の評点を合計し、30点以上を合格とした。ただし、3名以上のパネラーが0点の評点を付けた場合は不合格とした。

3 : 塗布後の肌にふっくらとした柔軟性が感じられる
2 : 塗布後の肌にふっくらとした柔軟性がやや感じられる
1 : 塗布後の肌にふっくらとした柔軟性があまり感じられない
0 : 塗布後の肌にふっくらとした柔軟性が全く感じられない
20名の専門パネラーによる使用感テストを行い、スキンローション0.5mLを手の甲に塗布した際のべたつき感について、パネラー各人が下記絶対評価にて4段階に評価し評点を付けた。そして、試料ごとにパネラー全員の評点を合計し、30点以上を合格とした。ただし、3名以上のパネラーが0点の評点を付けた場合は不合格とした。

3 : 塗布後の肌にべたつきをほとんど感じない
2 : 塗布後の肌にべたつきをあまり感じない
1 : 塗布後の肌にべたつきをやや感じる

20名の男性パネラーによる長期連用テストを行った。スキンローション0.5mLを朝夕一日2回、2週間にわたって前腕内側部に塗布し、2週間後の肌のキメの状態についてマイクロスコープで観察し、専門評価者が下記絶対評価にて4段階に評価し評点を付けた。そして、試料ごとに評点を合計し、30点以上を合格とした。
<絶対評価基準>

3 : 皮溝が鮮明かつ均質で、試験前に比べて改善がみられる
2 : 皮溝の鮮明度および均質性に関し、試験前に比べてやや改善がみられる
1 : 皮溝の鮮明度および均質性に関し、試験前に比べてやや悪化がみられる
0 : 皮溝の不鮮明かつ不均質で(異方性が強まり)、試験前に比べて悪化がみられる
20名の専門パネラーによる使用感テストを行い、洗顔後、スキンローションを顔面に塗布した後、塗布後2時間以内に感じた肌のツッパリ感について、パネラー各人が下記絶対評価にて4段階に評価し評点を付けた。そして、試料ごとにパネラー全員の評点を合計し、30点以上を合格とした。ただし、3名以上のパネラーが0点の評点を付けた場合は不合格とした。

3 : 塗布後、2時間以内に全くつっぱり感を感じなかった
2 : 塗布後、2時間以内にややつっぱり感を感じた
1 : 塗布後、2時間以内につっぱり感を感じた
0 : 塗布後、2時間以内に強いつっぱり感を感じた
一方、比較例1~4では十分な効果が得られていない。比較例1では、R1またはR2がアルギニンとグリシドールとの反応から誘導された2,3-ジヒドロキシプロピル基であるため、肌へのなじみ性や肌の柔軟化効果および塗布後のべたつき感において不十分であった。比較例2では、モノヒドロキシアルキル基の炭素数が4であるため、整肌効果が不十分であり、塗布後につっぱり感が感じられた。比較例3では、モノヒドロキシアルキル基の炭素数が12であるため、整肌効果、柔軟化効果が不十分であり、塗布後につっぱり感が感じられた。比較例4では、未修飾のL-アルギニンであるため、肌へのなじみ性、柔軟化効果、整肌効果において不十分であった。
