本発明において、グルコース誘導性不活化/分解耐性は、例えば次のようにして評価することができる。最初に、各トランスポータータンパクを発現する株が、0~2 mMの2-デオキシグルコースを含有するマルトース等の最少培地(Yeast Nitrogen Base w/o amino acids 6.7 g/L、 マルトースなど 20 g/L、ただし、栄養要求性のある形質転換株については、その栄養素を含む) あるいは、0~2 mMの2-デオキシグルコースを含有するマルトース等の完全合成培地(SCM) (Yeast Nitrogen Base w/o amino acids 6.7 g/L、 マルトース20 g/L、アデニン硫酸 20 mg/ml、ウラシル20 mg/ml、L-トリプトファン20 mg/ml、L-ヒスチジン塩酸塩20 mg/ml、L-アルギニン塩酸塩20 mg/ml、L-メチオニン20 mg/ml、L-チロシン30 mg/ml、L-ロイシン30 mg/ml、L-イソロイシン30 mg/ml、L-リジン塩酸塩30 mg/ml、L-フェニルアラニン50 mg/ml、L-グルタミン酸100 mg/ml、L-アスパラギン酸 100 mg/ml、L-バリン150 mg/ml、L-トレオニン200 mg/ml、L-セリン400 mg/ml)で生育することを確認し、グルコース誘導性の不活化/分解シグナルが生じている状態の酵母においても、そのトランスポーターがマルトース取り込み活性を持ち機能している株を選択する。次に、この株をYPD(イーストエキストラクト 10 g/L,ポリペプトン 20 g/L, グルコース 20 g/L)に植菌して、30 ℃にて一晩震盪培養する。培養液を、OD660=1.0になるようにYPM培地(イーストエキストラクト 10 g/L,ポリペプトン 20 g/L, マルトース 5 g/L)に植菌し、30 ℃にて2.5時間、震盪培養し、集菌する。60 OD660unitの細胞を測り取り、あらかじめ30 ℃にてインキュベートしておいた分解速度測定用培地(Yeast Nitrogen Base w/o amino acids and ammonia 1.6 g/L、 グルコース 20 g/L、シクロヘキシミド 25μg/L)30mlに懸濁し、30 ℃でインキュベートする。 細胞懸濁液は適当な時間(0、10、20、30および40 分あるいは、0、30、60、90および120 分)5mlサンプリングし、速やかに遠心して上澄みを捨て、細胞をエタノールドライアイスで凍結する。凍結した細胞から常法にしたがってトランスポータータンパク質を検出し、タンパクバンドのインテンシティーを測定し、その減少速度から半減期を求める。本発明において好ましいトランスポータータンパク質は、例えば、Agt1pと比較して、2倍以上、3倍以上、4倍以上、5倍以上、6倍以上または8倍以上の半減期を有する。
本発明において、上記(a)に記載される「配列番号:3の307~1659番目の塩基配列又は配列番号:5の283~1641番目の塩基配列」、及び(b)に記載される配列番号:4の103~553番目のアミノ酸配列又は配列番号:6の95~547番目のアミノ酸配列とは、酵母におけるAgt1p又はMtt1pの12回膜貫通ドメインとして予想される部分である。これらタンパク質の膜貫通ドメインは、Stockholm university Theoretical Chemistry Protein Prediction Servers のTopology prediction program であるToppred2 (http://bioweb.pasteur.fr/seqanal/interfaces/toppred.html)、あるいは、EMBnetのTransmembrane regions detection programであるTMPRED:Transmembrane regions detection (EMBnet http://www.ch.embnet.org/index.html)などで予測する事が可能である(アクセス日:2008年8月29日)。Mal61pの予測膜貫通ドメインについては、Cheng, Q., and Michels, C. A. (1989) Genetics 123(3), 477-484に書かれている。本明細書において、以下の予測TMD1~12のために、Stockholm university Theoretical Chemistry Protein Prediction Servers のTopology prediction program であるToppred2 (http://bioweb.pasteur.fr/seqanal/interfaces/toppred.html)を使用した。
本発明は、上記(1)~(6)に記載のポリヌクレオチドのいずれかにコードされるタンパク質も提供する。本発明の好ましいタンパク質は、配列番号:14、16又は18のアミノ酸配列において、1~10個(好ましくは、1~9個、1~8個、1~7個、1~6個、1~5個、1~4個、1~3個、1~2個又は1個)のアミノ酸が欠失、置換、挿入および/または付加したアミノ酸配列からなり、かつグルコース誘導性不活化/分解耐性を有するトランスポータータンパク質である。このようなタンパク質としては、配列番号:14、16又は18のアミノ酸配列において、上記したような数のアミノ酸残基が欠失、置換、挿入および/または付加されたアミノ酸配列からなり、かつグルコース誘導性不活化/分解耐性を有するトランスポータータンパク質が挙げられる。また、このようなタンパク質としては、配列番号:14、16又は18のアミノ酸配列と上記したような相同性を有するアミノ酸配列を有し、かつグルコース誘導性不活化/分解耐性を有するトランスポータータンパク質が挙げられる。このようなタンパク質は、「モレキュラークローニング第3版」、「カレント・プロトコールズ・イン・モレキュラー・バイオロジー」、“Nuc. Acids. Res., 10, 6487 (1982)”、“Proc. Natl. Acad. Sci. USA, 79, 6409 (1982)”、“Gene, 34, 315 (1985)”、“Nuc. Acids. Res., 13, 4431 (1985)”、“Proc. Natl. Acad. Sci. USA, 82, 488 (1985)”等に記載の部位特異的変異導入法を用いて、取得することができる。
次に、本発明は、上記したポリヌクレオチドを含有するベクターを提供する。本発明のベクターは、好ましくは、上記(1)~(4)のいずれかに記載のポリヌクレオチド(DNA)、又は配列番号:13、15又は17の塩基配列からなるポリヌクレオチド、又は配列番号:14、16又は18のアミノ酸配列からなるタンパク質をコードするポリヌクレオチドを含有する。また、本発明のベクターは、通常、(x)酵母細胞内で転写可能なプロモーター;(y)該プロモーターにセンス方向またはアンチセンス方向で結合した、上記のいずれかに記載のポリヌクレオチド(DNA);及び(z)RNA分子の転写終結およびポリアデニル化に関し、酵母で機能するシグナルを構成要素として含む発現カセットを含むように構成される。なお、上記本発明のタンパク質を高発現させる場合は、上記(a)~(i)の何れかに記載のポリヌクレオチド(DNA)の発現を促進するようにこれらのポリヌクレオチドを該プロモーターに対してセンス方向に導入するのが好ましい。
上述した本発明のベクターを製造対象となる酒類の醸造に適した酵母に導入し、その酵母を用いることによってアミノ酸の組成に特徴のある酒類を製造することができる。対象となる酒類としては、これらに限定されないが、例えば、ビール、ワイン、ウイスキー、清酒などが挙げられる。
本発明において作製されたポリヌクレオチドを含有する発現ベクターを構築し、それを定法に基づいて酵母に導入し、最初に、2-デオキシグルコースを含むマルトース・マルトトリオース培地で培養して、その生育レベルを指標に用いてグルコースによる不活化を受けにくいトランスポータータンパク質を含む酵母を選択する。次に、その酵母をオリゴ糖培地(例えば、マルトース・マルトトリオース培地)で培養する。その培養の際、その酵母に含まれるトランスポーターのグルコース誘導性不活化/分解耐性、その酵母のオリゴ糖資化能、増殖速度、麦汁での醗酵速度などを測定することによって、その酵母の適性を評価することができる。グルコース誘導性不活化/分解耐性、オリゴ糖資化能、増殖速度、麦汁での醗酵速度などは、後述の実施例において用いられた方法によって評価することができる。
本実施例で用いた試験項目および試験方法を以下に示す。特に断りのない限り、本実施例における試験方法はこれに準じた。
酵母サッカロミセス・セレビジエーのAGT1遺伝子は既にクローニングされており、その塩基配列が報告されている。本明細書中に記載されるAGT1のヌクレオチド配列(配列番号:3)はSaccharomyces Genome Database(登録番号:YGR289C)から入手した。AGT1遺伝子は、その塩基配列情報をもとに、AGT1遺伝子を持つ酵母サッカロミセス・セレビジエーから調製した染色体DNAをPCRの鋳型に使用して、PCRによって増幅、単離することにより取得した。
また、MAL21については、染色体III番にコードされていることが知られているが、DNA配列は知られていなかった。しかし、染色体II番にコードされるMAL31, 染色体VIII番にコードされるMAL61遺伝子が99 %以上のアイデンティティーを持つことから考えて、MAL21もかなり高いアイデンティティーを持つことが予想された。
実際に我々はGenBank(登録番号:X17391)から入手したMAL61のDNA配列からプライマー(5’AGAGCTCAGCATATAAAGAGACA 3’ (配列番号:19)と5’TGGATCCGTATCTACCTACTGG 3’(配列番号:20)を設計した。そしてMAL21遺伝子を持つが、他のα-グルコシドトランスポーター遺伝子を持たない酵母の染色体DNAを鋳型として、PCRでMAL21を取得することができた(そのヌクレオチド配列は配列番号:1、アミノ酸配列は配列番号:2に示される)。AGT1はプライマー(5’TGAGCTCACATAGAAGAACATCAAA 3’ (配列番号:21)と5’ATGGATCCATATGAAAAATATCATT 3’ (配列番号:22))を用いて取得した。具体的にはAGT1はサッカロミセス・セレビジエー S288C (ATCC204508(Rose, M.D., Winston, F. and Hieter, P. (1990): Methods in Yeast Genetics: A Laboratory Course Manual, Cold Spring Harbor Laboratory Press, Cold Spring Harbor, NY))、MAL21はサッカロミセス・セレビジエー ATCC20598からPCRによって取得した。取得したDNA断片は、Invitrogen社のTOPO TA cloning kitを用いて、ベクター、pCR(登録商標)2.1-TOPOに挿入した後、DNAシークエンスにより挿入した遺伝子配列を確認した。
AGT1については、データバンクに登録されている登録番号:YGR289Cの塩基配列と同一であることを確認した。MAL21については、独立に10クローン以上をシークエンスして、塩基配列を確定した(配列番号:1)。
なお、使用したプライマーには開始コドンの上流にXbaIあるいはSacIサイト、終始コドンの下流にBamHIサイトがあり、発現ベクターに組み込めるようになっている。染色体DNAを用いたPCRによる目的遺伝子の増幅およびその後の単離は、PCRプライマーの調製を含め、当業者に周知な方法で行うことができる。MAL21の塩基配列を図1にアミノ酸配列を図2に示す。
本発明においては(1)~(3)の3種類の発現ベクターを用いた。

制限酵素認識サイトの配列を持つプライマーでPCRを行なうことにより、制限酵素認識サイトの導入を行なった。プライマーのDNA配列は表3にあるとおりである。具体的には、AGT1あるいはMAL21遺伝子が挿入されたプラスミドpCR(登録商標)2.1-TOPOを鋳型として、表4に示したプライマーの組み合わせで、PCRを行うことにより、AGT1遺伝子については、AGT1-SaS, AGT1-SaK AGT1-SK, AGT1-SB及びAGT1-KBの5つのDNA断片、MAL21遺伝子については、MAL21-SaS, MAL21-SaK, MAL21-SK, MAL21-SB及びMAL21-KBの5つのDNA断片を得た。各断片の模式図は、下記の表2および3に示される。得られたDNA断片はプラスミドpCR(登録商標)2.1-TOPOに挿入して、シークエンスを行い、導入した制限酵素サイト以外に元の遺伝子から置換された塩基がないことを確認した。各々のDNA断片はプラスミドより制限酵素、SacI, SalI, KpnI, BamHIなどを用いて切り出し、下記ハイブリッドトランスポーター遺伝子の構築に用いた。
図7に示した6種類のハイブリッドトランスポーター遺伝子、AAM, AMA, AMM, MAA, MAM及びMMAを構築した。各々のハイブリッドトランスポーターは上記項で得られたそれぞれの断片を、図7に示した組み合わせで、発現ベクターpYCGPYのSacI-BamHIサイトに組み込むことにより取得した。詳しくは、pYCGPYをSacIとBamHIで切断し、Bacterial alkali phosphataseで脱リン酸化処理したものを用意して、その直線化ベクターと、上記DNA断片をライゲーション後、E.coli DH5αに形質転換した。アンピシリン耐性となったコロニー数個よりプラスミドを調製し、MAL21あるいはAGT1遺伝子にしか存在しない制限酵素の組み合わせで切断後、アガロースゲル電気泳動を行なうことにより、検出された断片の大きさから、目的のハイブリッドトランスポーター遺伝子がベクターpYCGPYに組み込まれたことを確認した。pYCGPYにおいて、トランスポーター遺伝子はPYK1プロモーターより転写される。また、pYCGPYからSacI-BamHIで各々のハイブリッドトランスポーターを切り出し、発現ベクターpJHXSBのSacI-BamHIサイトに組み込んだ。pJHXSBにおいて、トランスポーター遺伝子はTPI1プロモーターより転写される。
前記方法により得られた天然型、あるいはハイブリッドトランスポーター遺伝子を発現することのできるプラスミドで酵母を形質転換した。形質転換株(アルファーグルコシドトランスポーター遺伝子を持たない株を宿主とする)は、使用したプラスミドのマーカーに従って、pYCGPYについては、G418を300 μg/ml含むYPD培地で選択した。また、pJHXSBについては、宿主にHH1001などのura-株を用いた場合、ウラシルを含まない合成培地、例えば、最少培地(Yeast Nitrogen Base w/o amino acids 6.7 g/L、グルコース 20 g/L)などで選択した。
天然型トランスポーター、あるいは、構築したハイブリッドトランスポーターの形質転換株(アルファーグルコシドトランスポーター遺伝子を持たない株を宿主とする)における、導入したトランスポーター遺伝子の発現は、0.5 %のマルトースあるいはマルトトリオースを唯一の炭素源とする最少培地(ただし、栄養要求性のある形質転換株については、その栄養素を含む)での生育の有無により調べることができる。例えば、供試菌株をYPDプレート(イーストエキストラクト 10 g/L,ポリペプトン 20 g/L, グルコース 20 g/L)から一白金耳とって、1 mlの滅菌水で一度洗浄した後、OD660=0.2になるように滅菌水に再懸濁した。これを集菌して、もう一度1 mlの滅菌水に懸濁した後、細胞懸濁液をマルトース又はマルトトリオースを唯一の炭素源とする試験培地に直接ストリークして生育を確認することにより、マルトース又はマルトトリオースの取り込み活性を持つことを確認した。
2-デオキシグルコース(2-DOG)は 2-DOG-6リン酸以上に代謝されないため、炭素源にはなり得ない糖アナログであるが、グルコースと同じレベルにグルコースリプレッション、あるいはグルコースによる不活化を起こすことが知られている。従って、このプレートで生育する株は、グルコースによる不活化を受けにくいアルファーグルコシドトランスポーターを持つ可能性が高い。2-DOGに対する耐性を調べるため、0~2 mMの2-デオキシグルコースを含有するマルトース等の最少培地(Yeast Nitrogen Base w/o amino acids 6.7 g/L、 マルトースなど 20 g/L、ただし、栄養要求性のある形質転換株については、その栄養素を含む)、あるいは、0~2 mMの2-デオキシグルコースを含有するマルトース完全合成培地(SCM)(Yeast Nitrogen Base w/o amino acids 6.7 g/L、 マルトース20 g/L、アデニン硫酸 20 mg/ml、ウラシル20 mg/ml、L-トリプトファン20 mg/ml、L-ヒスチジン塩酸塩 20 mg/ml、L-アルギニン塩酸塩20 mg/ml、L-メチオニン20 mg/ml、L-チロシン30 mg/ml、L-ロイシン30 mg/ml、L-イソロイシン30 mg/ml、L-リジン塩酸塩30 mg/ml、L-フェニルアラニン50 mg/ml、L-グルタミン酸 100 mg/ml、L-アスパラギン酸 100 mg/ml、L-バリン150 mg/ml、L-トレオニン200 mg/ml、L-セリン400 mg/ml)を作製した。各トランスポーター発現株のOD660=0.2の細胞懸濁液の希釈系列を試験培地に各々3μlスポットし、30 ℃にて2~3日培養することによって調べた。
トランスポータータンパクの菌体内蓄積量は、例えばウエスタンブロッティングにより調べることができる。例えば、供試菌株を10 mlの培養液から対数増殖期で回収し、溶解バッファー(8 M 尿素、5 % (w/v) SDS、40 mM Tris-HCl (pH6.8)、0.1 mM EDTA、1 % β-メルカプトエタノール)中でグラスビーズによる撹拌によって破壊し、細胞抽出液を得る。総タンパク60 μgのサンプルをSDS-ゲル電気泳動で展開し、ニトロセルロース膜に転写した後にウサギポリクローナル抗Mal61p抗体を用いてウエスタンブロッティングを行う。
ウサギポリクローナル抗Mal61p抗体は以下のようにして取得した。Mal61pのN末領域 (Met1-Leu181) をコードするDNAをpET Expression vector(Novagen社)のGST tagの下流につなぎ、大腸菌BL21(DE3)に形質転換し、その形質転換体の細胞破砕液をGST bind resin columnにアプライして、カラムに結合したタンパクを溶出する事によって調製した。詳しくは、Novagen 社のpET Expression System, GST・BindTM Affinity Resins(Novagen社)に添付されたマニュアルのとおりである。調製した融合タンパクは、SDS-PAGEにて純度を確認した後、これを抗原としてウサギを免疫し、ポリクローナル抗体を得た。抗体の有効性については、α-グルコシドトランスポーター遺伝子を発現させた酵母株と、当遺伝子を持たないその宿主株とをYPM培地(イーストエキストラクト 10 g/L,ポリペプトン 20 g/L, マルトース 5.0 g/L)で培養し、その細胞破砕液を、本抗体を用いて上記方法にてウエスターンブロッティングを行なうことにより確認した。本抗体により、α-グルコシドトランスポーター遺伝子を発現させた酵母株破砕液だけに、68 kDaのα-グルコシドトランスポーターの分子量に一致するポジティブバンドを検出した。なお、ハイブリッドトランスポーターについては、N末部がMal21p型であるトランスポーター、MMAp, MAMp及びMAApのみ、本抗体で検出可能である。
Agt1pの菌体内蓄積量は、Agt1pのC末端部に、ヘマトアグルチニンタグをタンデムに2個つないだ融合タンパクをコードする遺伝子を構築し、上述の方法にてそれを発現する株を取得して用いた。抗体は、マウスモノクローナル抗ヘマトアグルチニン抗体(Covance, Research, Products, Inc.社)を用いた。
各トランスポータータンパクを発現する株をYPDに植菌して、30 ℃にて一晩震盪培養した。培養液を、OD660=1.0になるようにYPM培地に植菌し、30 ℃にて2.5時間、震盪培養し、集菌した。60 OD660unitの細胞を測り取り、あらかじめ30 ℃にてインキュベートしておいた分解速度測定用培地(Yeast Nitrogen Base w/o amino acids and ammonia 1.7 g/L、 グルコース 20 g/L、シクロヘキシミド 25 mg/L)30mlに懸濁し、30 ℃でインキュベートした。 細胞懸濁液は適当な時間(0、10、20、30及び40分あるいは0、30、60、90及び120 分)5mlサンプリングし、速やかに遠心して上澄みを捨て、細胞をエタノールドライアイスで凍結した。凍結した細胞は、上記の方法にてトランスポータータンパクを検出し、タンパクバンドのインテンシティーを測定し、その減少速度から半減期を求めた。
トランスポータータンパク質を構成的に発現している酵母によるマルトースの資化について、当該酵母に適した条件下にて酵母を好気培養あるいは醗酵させ、培地中のマルトース量を測定することにより評価した。糖の測定は、当業者に周知の方法、例えばIR検出器を用いた液体クロマトグラフィーにより測定することができる。
PCRにより取得した、MAL21遺伝子とAGT1遺伝子を発現ベクターpYCGPYに挿入し、酵母Δ152に形質転換した。0.5 %のマルトースあるいは、マルトトリオースを唯一の炭素源として含み、20mg/Lのウラシル、20mg/Lのヒスチジン、30mg/Lのロイシン、300μg/mlのジェネティシンを含む最少培地(MM)に形質転換株をストリークした(図5)。MAL21を持つ株はマルトース培地でしか生育できなかったが、AGT1を持つ株はマルトース、マルトトリオースどちらの培地でも生育し、AGT1だけが両糖を取り込むことができることを確認した。次にAgt1pのC末端部にヘマトアグルチニンタグをタンデムに2個つないだ融合タンパクをコードする遺伝子、AGT1-2HAを構築し、発現ベクターpJHXSBに挿入した。MAL61とMAL21もpJHXSBに挿入した。これらを酵母HH1001に形質転換した。両形質転換株を用いて、試験方法の項に示した方法で、グルコース存在下での分解速度をウエスターンブロッティングにて調べた。その結果を図6に示す。Agt1-2HApはMal61pやMal21pに比べ、半減期が短い(図6を参照して、Agt1-2HAp:14分、 Mal61p:25分、Mal21p:118分)事がわかった。また、同じα-グルコシドトランスポーターでもMal21pはMal61pに比べ、はるかに長い半減期を持つことがわかった(Mal61p:25分, Mal21p:118分)。
Agt1pとMal21pはMFS(Major facilitated sugar transporter)の仲間に属し、そのアミノ酸配列の類似性から、12の膜貫通ドメインを持ち、N末端とC末端は細胞質ドメインだと考えられている。Agt1pとMal21pの基質特異性や活性に関わる部位については知られていない。両者のアミノ酸配列のアラインメントを見ると、N末端とC末端は細胞質ドメインと考えられる部分は類似性が低く、中央の12の膜貫通ドメイン部分は類似性が高い(図3を参照のこと)。そこで、我々は中央の12の膜貫通ドメイン部分は取り込み活性を持ち、あるいは基質特異性を左右する部分である可能性があると考えた。AGT1とMAL21遺伝子をそれぞれ、N末あるいはC末の細胞質ドメインをコードする部分と、中央部の12回膜貫通ドメインをコードする部分とを両端に制限酵素サイトを持つ形で、PCRにより取得した。使用したプライマーを表3に、取得した断片と使用したプライマー対との関係を表4に示した。これらを組み合わせることにより数種のハイブリッドトランスポーター遺伝子を構築した。使用した組換え位置を図3中の十字印として表し、各ハイブリッドの名称および模式図を図7に示す。詳しくは試験方法に述べた通りである。図7に示した、各ハイブリッドトランスポーター遺伝子を挿入した発現ベクター、pYCGPYを酵母Δ152に形質転換した。0.5 %のマルトースあるいは、マルトトリオースを唯一の炭素源とし、20mg/Lのウラシル、20mg/Lのヒスチジン、30mg/Lのロイシン、300μg/mlのジェネティシンを含む最少培地(MM) (MM:Yeast Nitrogen Base w/o amino acids 6.7 g/L、5 g/L マルトースあるいはマルトトリオース、20 mg/L ヒスチジン、30 mg/L ロイシン、20 mg/L ウラシル300μg/ml ジェネティシン)に形質転換株をストリークした(図8)。その結果、中央部の12回膜貫通ドメインがAgt1p由来である3つのハイブリッドトランスポーター、AAMp、MAMp、MAApはマルトトリオースを取り込む活性を持つことがわかった。
実施例2に記載したハイブリッドトランスポーター遺伝子をpYCGPYからSacI-BamHIで切り出し、pJHXSBのSacI-BamHIサイトに組み込んだ。構築した各々の発現ベクターをHH1001に形質転換した。HH1001はmal-株であるX2180-1Aのura3-のシブリングであり、TPI1p::MAL32(マルターゼ遺伝子をコードしている)が組み込まれているので、マルターゼが構成的に発現している。2 %のマルトースを含む完全合成培地(SCM)に2-デオキシグルコース(2-DOG)を0 mM~2 mM加えたプレートを作製し、試験方法に記載したように、各形質転換株の2-DOG 耐性を調べた。2-DOGは2-DOG-6リン酸以上に代謝されないため、炭素源にはなり得ない糖アナログであるが、グルコースと同じレベルにグルコースリプレッション、あるいはグルコースによる不活化を起こすことが知られている。従って、このプレートで生育する株は、グルコースによる不活化を受けにくいα-グルコシドトランスポーターである可能性が高い。その結果を図9に示す。MAL21由来のN末の細胞質ドメインを持つハイブリッドトランスポーター MMAp、MAApは1 mMの2-DOGの培地でも生育した。また、MAMpは2 mMの2-DOGの培地でも生育し、天然型のMal21pと同じ2-DOGに対する耐性を持っており、グルコースによる分解誘導を受けにくいことがわかった。
実施例3で示したとおり、MAL21由来のN末の細胞質ドメインを持つハイブリッドトランスポーター MMAp、MAAp及びMAMpはグルコースによる分解誘導を受けにくいと考えられる。そこでこれを確かめるため、試験方法に示した方法で、グルコース存在下でのMMAp、MAAp及びMAMpの分解速度を測定した(図10)。いずれのハイブリッドトランスポーターもAgt1-2HApよりも長い半減期を持っていた。特にMAMpは半減期が119分で、Mal21p(118分)とほぼ同じ半減期を持っており、グルコースにより速やかに分解されるアルファーグルコシドトランスポーターであるAgt1pを改良し、分解されにくいトランスポーターを構築することが出来た。
グルコース誘導性分解に対する耐性を持つトランスポーター、MAL21をプラスミドpUP3GLPのXbaI(あるいはSacI)-BamHIサイトに組み込んだ。pUP3GLPを図14に示す。pUP3GLP はYIp型プラスミドであり、トランスポーター遺伝子はグリセルアルデヒド三リン酸デヒドロゲナーゼプロモーター(TDH3p)によって発現する。各プラスミドをURA3の中にあるEcoRVサイトで切断後、下面ビール酵母(Weihenstephan 194)に形質転換し、0.3μg/mlのシクロヘキシミドを含むYPGプレート(イーストエキストラクト 10 g/L,ポリペプトン 20 g/L, ガラクトース 20 g/L)に塗布した。目的の発現カセットがWeihenstephan 194の染色体上のURA3遺伝子に挿入されたことはPCRによって確認した。
Weihenstephan 194 (URA3::TDH3p::MAL21)と親株Weihenstephan 194 を2種類の発泡酒麦汁に植菌した。発泡酒麦汁は水を除く原料中の麦芽の使用比率が25%未満で、糖化スターチ、ホップなどを用いた麦汁である。発泡酒用麦汁のうちの1つは初期エキス濃度が14.0 %で、グルコース 1.2 %、マルトース 6.6 %、マルトトリオース 2.2 %の組成比で糖が含まれている。もう一つのグルコースリッチ発泡酒麦汁は、初期エキス濃度が15.6 %で、グルコース 4.7 %、マルトース 5.4 %、マルトトリオース 1.7 %の組成比で糖が含まれている。それぞれ、同じ量の麦汁(終濃度、麦芽比率25 %未満)に対して、糖組成の異なる糖化スターチを加えることにより調製した。各発泡酒麦汁に湿菌体を7.5 g/Lになるようにピッチングし、15 ℃にて醗酵を行い、醗酵中のもろみのマルトース濃度を測定した。その結果を図11に示す。
どちらの発泡酒麦汁においても、MAL21高発現株のマルトースの資化速度は、親株のWeihenstephan 194よりも著しく早くなった。特にグルコースリッチ発泡酒麦汁の場合、その効果は顕著であった。初期エキス濃度が高いということは、グルコース濃度が高いことを意味しており、グルコース誘導性分解に対する耐性を持つトランスポーターの効果が十分見られた。したがって、ハイブリッドトランスポーターMAAp又はMAMpを導入した酵母の場合は、例えば図10および図11の結果から、例えばグルコースなど単糖が高濃度の場合におけるマルトトリオースの資化促進が十分期待される。
Claims (16)
- グルコース誘導性不活化/分解耐性を有するトランスポータータンパク質をコードするポリヌクレオチドであって、以下の(a)~(d)のいずれかに記載のポリヌクレオチドからなる12回膜貫通ドメインコード領域を含み、5'側配列及び/又は3'側配列を異種ポリヌクレオチドと組み換えてなるポリヌクレオチド:
(a)配列番号:3の307~1659番目の塩基配列又は配列番号:5の283~1641番目の塩基配列からなるポリヌクレオチド;
(b)配列番号:4の103~553番目のアミノ酸配列又は配列番号:6の95~547番目のアミノ酸配列をコードするポリヌクレオチド;
(c)配列番号:4の103~553番目のアミノ酸配列又は配列番号:6の95~547番目のアミノ酸配列において、1~10個のアミノ酸が欠失、置換、挿入および/または付加したアミノ酸配列をコードするポリヌクレオチド;及び
(d)配列番号:4の103~553番目のアミノ酸配列又は配列番号:6の95~547番目のアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列をコードするポリヌクレオチド。 - 異種ポリヌクレオチド配列が配列番号:1の12回膜貫通ドメインの5'側配列及び/又は3'側配列である、請求項1に記載のポリヌクレオチド。
- 12回膜貫通ドメインコード領域と5'側配列との組換え位置が12回膜貫通ドメインの予測TMD1コード領域内であり、12回膜貫通ドメインと3'側配列との組換え位置が12回膜貫通ドメインの予測TMD12コード領域開始位置から予測TMD12コード領域終止位置より3'側に96塩基以内の領域内であって、かつ配列番号:2と配列番号:4との対応するアミノ酸配列において同一性が80%以上の領域内である、請求項1に記載のポリヌクレオチド。
- 12回膜貫通ドメインコード領域と5’側配列との組換え位置が配列番号:1又は配列番号:5の175番目の塩基より予測TMD1コード領域終止位置までの領域内であり、12回膜貫通ドメインコード領域と3'側配列との組換え位置が予測TMD12コード領域開始位置から予測TMD12コード領域終止位置より3'側に180塩基以内の領域内である、請求項1に記載のポリヌクレオチド。
- 配列番号:13、15又は17の塩基配列からなるポリヌクレオチドを含有する請求項1~4のいずれかに記載のポリヌクレオチド。
- 配列番号:14、16又は18のアミノ酸配列からなるタンパク質をコードするポリヌクレオチドを含有する請求項1~4のいずれかに記載のポリヌクレオチド。
- DNAである、請求項1~6のいずれかに記載のポリヌクレオチド。
- 請求項1~7のいずれかに記載のポリヌクレオチドにコードされるタンパク質。
- 請求項1~7のいずれかに記載のポリヌクレオチドを含有するベクター。
- 請求項9に記載のベクターが導入された形質転換酵母。
- 請求項9に記載のベクターを導入することによって、オリゴ糖資化能が向上した請求項10に記載の醸造用酵母。
- 請求項8に記載のタンパク質の発現量を増加させることによってオリゴ糖資化能が向上した請求項11に記載の醸造用酵母。
- 請求項10~12のいずれかに記載の酵母を用いた酒類の製造方法。
