トリコデルマ・リーセイQM9414(NBRC 31329)をポテトデキストロース寒天培地上で28℃、7日間培養して胞子を充分形成させた。その1白金耳を、結晶セルロース(旭化成ケミカルズ製、商品名セオラス):1g、KH2PO4:0.2g、(NH4)2SO4:0.14g、CaCl2・2H2O:0.03g、MgSO4・7H2O:0.03g、ポリペプトン0.2g、酵母エキス0.05g、ツイーン80:0.1ml、微量元素液(H3BO4 6mg、(NH4)6Mo7O24・4H2O 26mg、FeCl3・6H2O 100mg、CuSO4・5H2O 40mg、MnCl2・4H2O 8mg、ZnSO4・7H2O 200mgを水100mlに溶解及び懸濁した液):0.1ml、水:100mlを含む500ml容バッフル付三角フラスコに接種して、28℃、7日間振とう培養した。7日目に培養液を濾過し、上清液を得た。
トリコデルマ・リーセイ培養上清液とアスペルギルス・カワチ培養上清液の混合割合を変化させ、糖化効率を比較した。両培養上清液は実施例1と同様にして調製したものを用いた。
濾過を行わないこと以外は実施例1と同様にして、トリコデルマ・リーセイ培養液及びアスペルギルス・カワチ培養液を得た。次いで、両培養液の混合割合を変化させた各種混合物を調製し、糖化効率を比較した。
トリコデルマ・リーセイ培養上清液とアスペルギルス・カワチ培養上清液の混合割合を変化させ、糖化効率を比較した。両培養上清液は実施例1と同様にして調製したものを用いた。
種々のアスペルギルス属糸状菌培養上清液について、トリコデルマ・リーセイ培養上清液と組み合わせて用い、糖化効率の相乗効果を比較した。トリコデルマ・リーセイ培養上清は実施例1と同様にして調製したものを用いた。
トリコデルマ属由来のセルラーゼについて、アスペルギルス・カワチ由来のセルラーゼとの相乗効果を調べた。トリコデルマ・リーセイ由来のセルラーゼ製剤(シグマ社)とトリコデルマ・ビリデ由来のセルラーゼ製剤(シグマ社)をそれぞれ水に溶解し、1%セルラーゼ液を調整した。脱リグニンバガス:0.5g、1%セルラーゼ液:4.65ml、アスペルギルス・カワチ培養上清液:4.65ml、1M酢酸バッファー(pH4.8):0.2mlからなる液(バガス5%液)を50℃、pH4.8、24時間、振とうさせて糖化し、生成したグルコースを測定した。アスペルギルス・カワチ培養上清液は実施例1と同様にして調製したものを用いた。
実施例1と同様の方法でトリコデルマ・リーセイ培養上清液を得た。
Claims (9)
- セルロース分解活性を示す、トリコデルマ(Trichoderma)属糸状菌培養液または上清液とアスペルギルス・カワチ(Aspergillus kawachii)培養物との混合物、またはその乾燥物。
- 前記アスペルギルス・カワチ培養物が、培養液または上清液である請求項1記載の混合物、またはその乾燥物。
- 前記アスペルギルス・カワチ培養物が、固体培養物または抽出液である請求項1記載の混合物、またはその乾燥物。
- 前記トリコデルマ属糸状菌培養液または上清液のセルラーゼ活性が50U/ml以上であり、前記アスペルギルス・カワチ培養液、上清液または固体培養物抽出液のヘミセルラーゼ活性が50U/ml以上である請求項1~3のいずれか記載の混合物、またはその乾燥物。
- 前記トリコデルマ属糸状菌培養液または上清液と前記アスペルギルス・カワチ培養液、上清液または固体培養物抽出液との混合割合が、体積比で10:90~90:10の範囲である請求項1~4のいずれか記載の混合物、またはその乾燥物。
- 前記トリコデルマ属糸状菌がトリコデルマ・リーセイ(Trichoderma reesei)又はトリコデルマ・ビリデ(Trichoderma viride)である請求項1~5のいずれか記載の混合物、またはその乾燥物。
- 前記トリコデルマ属糸状菌がトリコデルマ・リーセイである請求項1~5のいずれか記載の混合物、またはその乾燥物。
- 請求項1~7のいずれか記載の混合物または乾燥物を用いてセルロース原料を糖化する工程を包含する、グルコースの製造方法。
- 前記糖化は30~70℃及びpH3~7の条件で行われる請求項8記載の方法。
