Abstract

細菌由来のチトクロームP-450遺伝子を組み込んだ大腸菌を用いた微生物変換系においてみられる変換効率の低さを改善する手段の提供を目的とする。大腸菌に、アシネトバクター・エスピー(Acinetobacter sp.) OC4株由来の電子伝達系をコードする遺伝群aciBCを組み込むとともに、細菌由来のチトクロームP-450遺伝子の5'末端側に、アシネトバクター・エスピー(Acinetobacter sp.) OC4株由来のアルカン酸化性チトクロームP-450であるAciAのN末端48アミノ酸等をコードするポリヌクレオチドを付加した組換え大腸菌を作製し、これを微生物変換に使用することにより、疎水性または両親媒性の基質化合物を非常に効率よくその目的化合物に変換することができる。
