Die vorliegende Erfindung betrifft Mittel und Verfahren zum Nachweis von Candida und Candida-verwandten Pilzzellen in klinischem Material.
Candida albicans ist ein Pilz der Candidagruppe, die zu den Hefepilzen gehören. Dieser Pilz ist bei Warmblütern (und daher auch Mensch) häufig auf den Schleimhäuten von Nase und Rachen und im Genitalbereichs sowie im Verdauungskanal zu finden. Bei etwa 75% aller gesunden Menschen kann er nachgewiesen werden (laut deutscher Gesellschaft für Ernährung). Er kann auch zwischen Fingern und Zehen vorkommen und auf den Finger- und Fußnägeln. Candida gehört zu den fakultativ pathogenen Erregern (nur unter bestimmten Bedingungen eine Krankheit auslösend) und ist als ein Saprophyt anzusehen, der in einem Gleichgewichtszustand mit anderen Mikroorganismen siedelt. Die Besiedelung durch diesen Pilz verursacht in der Regel kaum Beschwerden. Bei fehlender oder verminderter Immunität, zum Beispiel im Rahmen von anderen Grundkrankheiten und/oder Medikamentengaben, konvertieren diese Pilze jedoch zu pathogenen Keimen. Es kommt zu einer Candidain- fektion wie Candidose, Candidiasis, Candidamykose, Monoliasis oder Soor.
Meist tritt eine Candidainfektion auf bei Grunderkrankungen wie schwere Diabetes, Leukämien, AIDS, unter der Wirkung bestimmter Medikamente wie Kontrazeptiva, bei Medikamenten, die die Resistenzlage gezielt oder als Nebenwirkung herabsetzen, bei Antibiotika in hoher und häufiger Dosierung, bei hoch dosierten Corticoiden und Zytostatika, und/oder bei weiteren begünstigenden Bedingungen. Zu
den Risikogruppen zählen Tumorpatienten mit Neutropänie, Patienten nach Knochenmarktransplantation oder sonstiger Organtransplantation, immunsupprimierte Patienten, Patienten mit großen Wundflächen oder Verbrennungen, Polytraumatisierte und Frühge- borene. Weiterhin gibt es bei Intensivpatienten prädisponierende
Faktoren für eine systemische Candidainfektion
Der eigentliche pathophysiologische Mechanismus, der zur Ausbildung einer tiefen Candidose und nachfolgend zur lebensbedrohlichen Candida-Sepsis führt, ist noch nicht eindeutig geklärt. Die ge- websschädigende Wirkung beruht hauptsächlich auf toxischen, noch wenig erforschten Pilzprodukten.
Die Inzidenz von Candidämie und disseminierter Candidiasis bei Intensivpatienten hat in den letzten Jahren deutlich zugenommen. Angesichts der assoziierten hohen Morbidität und Mortalität wäre es wünschenswert, die Schwierigkeiten in der Diagnostik mit sicheren und frühen Nachweisen der Candidainvasion zu überwinden.
Die Diagnose einer Candidiasis wird im klinischen Routinelabor meist mikroskopisch gestellt. Schleimhautabstriche, Stuhlproben, Urin, eine positive Blutkultur oder anderes Untersuchungsmaterial aus sterilen Organkompartimenten (Liquor, Gewebebiopsie) können geeignet sein. Der sichere Nachweis einer Candidiasis gelingt dabei nur selten. Häufig sind allerdings falsch positive Ergebnisse, während gerade bei der Soorsepsis auch falsch negative Befunde auftreten. Die Kultivierung von Patientenproben ist außerdem sehr zeitin- tensiv, deshalb lassen sich Diagnosen oft erst zu spät erstellen.
Pilze sind lebende Antigenmosaike und können die verschiedenen Teile des Immunsystems stimulieren. Antigene der Pilzkapsel in
Form von Proteinen, Polysacchariden, Lipiden und chitinartigen Substanzen induzieren eine Antikörperbildung durch B-Zellen. Infolgedessen können im Serum pilzinfizierter Patienten entsprechende präzipitierende und komplementbindende Antikörper nachgewiesen werden. Bei klinischem Verdacht auf eine systemische Candidainfek- tion zeigt sich oft gleichzeitig bei serologischen Verlaufsuntersuchungen ein Anstieg im Titer von gegen Candida gerichteten Antikörpern.
Bekannte Antikörperassays basieren auf gegen Zellwandproteine gerichtete Antikörper, die meist an Trägerkügelchen (so genannte
„beads") immobilisiert sind. In einem einer Blutgruppenbestimmung ähnlichen Anordnung werden klinische Proben wie Blut mit den Anti- körperbeads in Verbindung gebracht. Liegen Candida spezifische Zellwandbestandteile in der Probe vor, so kommt es zu einer Ver- klumpung der beads, was in einer Tüpfelplatte beziehungsweise
Mikrotiterplatte sichtbar wird. Dieser Test ist als so genannter Häm- agglutinintest (HAT) bekannt. Diese Test sind in der Medizin allerdings aufgrund Ihrer geringen Sensitivität und Aussagekraft stark umstritten.
Durch eine schnelle, genauere und aussagekräftigere Detektion dieser Infektion, könnte eine wirkungsvolle, lebensrettende Behandlung schneller und zielgerichteter erfolgen. Der Erfolg einer Pilztherapie hängt nicht unwesentlich davon ab, wie rechtzeitig die Therapie begonnen wird. Auf der anderen Seite weisen die eingesetzten Anti- mykotika nicht unerhebliche Nebenwirkungen auf. Spezielle, neu entwickelte hoch effektive Antimykotika haben zwar weniger Nebenwirkungen, sind aber in Ihrer Anwendung erheblich teurer. Neben einem schnellen und sensitiven Nachweis von Candida, soll ein
Candida-Test also auch eine hohe Selektivität aufweisen, um die Zahl der falsch positiven Resultate, und damit die Zahl der unnötigen Therapien, zu minimieren.
Weiter soll ein Candida-Test in der klinischen Routinediagnostik schnell und sicher einsetzbar sein. Das heißt bei reduzierten Kosten des Einzeltests und geringem Aufwand an spezialisiertem Personal muss ein möglichst hoher Probendurchsatz ermöglicht werden. Dies kann regelmäßig erreicht werden durch den Einsatz automatisierter Auslesegeräte, die besonders unmittelbar mit den Patientendaten- banken in Verbindung stehen. Idealerweise ist eine hohe Zahl individueller Tests in einem einzigen Durchlauf zu realisieren. Weiter muss ein verbesserter Test die Möglichkeit bieten, in einem einzigen Ansatz mit anderen verwandten Tests, beispielsweise zum Nachweis anderer Erreger, durchgeführt zu werden.
Der vorliegenden Erfindung liegt das technische Problem zugrunde,
Mittel und Verfahren zum Nachweis von Candida und Candida- verwandten Pilzzellen in klinischem Material bereitzustellen, bei denen die im Stand der Technik bekannten Nachteile beseitigt sind, insbesondere eine erhöhte Sensitivität und Selektivität erreicht wird, und die zur Verwendung in automatisierten Screening- und Analytiksystemen, geeignet sind.
Die vorliegende Erfindung löst das ihr zugrunde liegende technische Problem durch die Bereitstellung eines Funktionselementes zum Nachweis von Candida, das heißt eines Candida-Diagnose-Chips, umfassend einen Träger mit einer Oberfläche und mindestens eine auf der Trägeroberfläche angeordneten Mikrostruktur mit molekülspezifischen Erkennungsstellen ausgewählt aus: spezifischen Anti-
körpern gegen das Protein TSA 1 , bevorzugt so genannte anti-TSA 1 IgG1 und dem Protein TSA 1 , die daran immobilisiert sind.
Unter „TSA" wird vorliegend das Protein „Thiol-specific-antioxidant- (like)-protein" von Candida, einem Mitglied der Peroxiredoxin- Enzymfamilie (EC 1.1 1.1.15) verstanden. Es handelt sich dabei um ein physiologisch wichtiges Antioxidans mit Disulfidbindung, welches mittels enzymatischer Aktivität schwefelhaltige Radikale abwehren kann. TSA 1 ist primär cytosolisch lokalisiert. TSA 1 hat die Aminosäuresequenz SEQ ID NO: 1
Bevorzugt wird TSA 1 in Form von rekombinantem TSA 1 eingesetzt.
Selbstverständlich kann gemäß der Erfindung ein Fragment oder ein Abkömmling (Derivat) von TSA 1 eingesetzt werden. Das Fragment oder der Abkömmling können durch Austausch und/oder Weglassen von 1 , 2, 3, 4, 5, 6, 7, 8, 9, 10, 1 bis 10, 1 bis 20, 1 bis 30, 1 bis 40 und/oder 1 bis 50 Aminosäuren aus dem Protein nach SEQ ID NO: 1 erhalten werden. Das Fragment oder der Abkömmling von TSA 1 weist ebenfalls Candida-spezifische Antigenizität auf und bindet spezifisch an Candida-spezifische anti-TSA 1 Antikörper (anti-TSA 1 IgG). In einer weiteren erfindungsgemäß bevorzugten Variante ist das TSA 1 Protein Bestandteil eines Candida-Zelllysats oder eines
Proteincocktails, der aus Candida Zellen gewonnen wird, beispielsweise cytosolische Proteine oder Zellwandproteine. Das Funktionselement weist auch dann die erfindungsgemäßen molekülspezifischen Erkennungsstellen auf, wenn neben dem TSA 1 Protein noch weitere Candida Proteine immobilisiert sind.
Bevorzugt ist die Mikrostruktur aus mehreren dreidimensional übereinander angeordneten Schichten von Nanopartikeln gebildet und
die Nanopartikel weisen die molekülspezifischen Erkennungsstellen auf.
Weiter bevorzugt sind Mikrostrukturen, die identische molekülspezifische Erkennungsstellen für Candida Antigene oder Antikörper auf- weisen. Weiter bevorzugt sind Mikrostrukturen, die auch nichtidentische molekülspezifische Erkennungsstellen für Candida Antigene oder Antikörper aufweisen. Diese Strukturen erlauben, mehrere unterschiedliche Candida Proteine in einem Test zu integrieren.
In bevorzugter Ausführung werden die Mikrostrukturen unter Ein- Schluss mindestens eines Biomolekül stabilisierenden Agens gebildet. Die, bevorzugt mehrdimensional angeordneten, Schichten von Nanopartikeln vergrößern in drastischer Art und Weise die für die erwünschten Nachweisreaktionen zur Verfügung gestellten Reaktionsflächen des Funktionselementes, wobei gleichzeitig durch den Einschluss des Protein stabilisierenden Agens in einer bevorzugten
Ausführungsform erreicht wird, dass bei Verwendung von TSA 1 Protein oder anti-TSA 1 Antikörpern die natürliche Struktur und Funktion der Proteine erhalten bleibt.
Bevorzugt sind die mehreren bevorzugt dreidimensional angeordne- ten Schichten von Nanopartikeln, in einer Dicke von 10 nm bis
10 μm, bevorzugt von 50 nm bis 2,5 μm, besonders bevorzugt von 100 nm bis 1 ,5 μm auf der Trägeroberfläche angeordnet. Der erfindungsgemäße Aufbau des Funktionselements ermöglicht eine hohe Nachweissensitivität auch bei Kleinstmengen an nachzuweisendem Analyten.
Vorzugsweise sind die erfindungsgemäß eingesetzten Funktionselemente zum Nachweis von Candida - in lateraler Strukturierung -
mit weiteren Funktionsschichten mit anderen molekülspezifischen Erkennungsstellen ausgestattet, die jeweils spezifisch adressierbar sind. So können ortsaufgelöst spezifische Analyten binden. Der parallele Nachweis von anderen Analyten neben Candida-spezifischen Molekülen auf einem einzigen Funktionselement, in einem einzigen
Nachweisverfahren wird dadurch ermöglicht. Als weitere molekülspezifische Erkennungsstellen werden - je nach Anwendungsgebiet - vorzugsweise Proteine und/oder Antikörper, die spezifisch für mikrobielle Erreger wie Pilzzellen sind eingesetzt; bevorzugt sind die Pilzzellen klinisch relevante Erreger wie Aspergillus, Cryptococcus
(Histoplasma, Blastomyces), Coccidioides immitis, Epidermophyton, Geotrichum, Paracoccidioides (Blastomyces). Weitere molekülspezifische Erkennungsstellen sind bevorzugt weitere ausgewählte isolierte Candida Antigene und/oder Antikörper, gerichtet gegen weitere Candida Antigene.
Die vorliegende Erfindung stellt also ein Funktionselement bereit, auf dessen Oberfläche ein oder mehrere Mikrostrukturen angeordnet sind, wobei jede Mikrostruktur bevorzugt aus vielen Nanopartikeln besonders bevorzugt in mehreren Schichten mit identischen oder nicht-identischen molekülspezifischen Erkennungsstellen besteht, wobei mindestens eine molekülspezifische Erkennungsstelle ausgewählt ist aus: spezifischen Antikörpern gegen das Protein TSA 1 , bevorzugt so genannte anti-TSA 1 IgG, und dem Protein TSA 1.
Im Unterschied zu im Stand der Technik bekannten Systemen, bei- spielsweise herkömmlichen Gen- oder Protein-Arrays, sieht die vorliegende Erfindung also vor, biologische Moleküle nicht direkt an eine planare Oberfläche zu binden, sondern an mehrere, bevorzugt dreidimensionale, Nanopartikeloberflächen zu immobilisieren, die vor
oder nach der Immobilisierung zur Bildung einer lateral strukturierten Mikrostruktur eingesetzt werden.
Auf den erfindungsgemäßen Funktionselementen werden die molekülspezifischen Erkennungsstellen kovalent und/oder nicht-kovalent an den Nanopartikeln gebunden. Die spezifischen Antikörper gegen das Protein TSA, beziehungsweise das Protein TSA kann sowohl ungerichtet als auch gerichtet an den Nanopartikeln immobilisiert werden, wobei nahezu jede gewünschte Ausrichtung der Biomoleküle möglich ist. Durch die Immobilisierung der Biomoleküle an den Nanopartikeln wird auch eine Stabilisierung der Biomoleküle erreicht.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem „Nanopartikel" eine partikuläre Bindematrix verstanden, die erste funktionelle chemische Gruppen umfassende molekülspezifische Erkennungsstellen aufweist. Die erfindungsgemäß verwendeten Na- nopartikel umfassen einen Kern mit einer Oberfläche, auf der die ersten funktionellen Gruppen angeordnet sind, die in der Lage sind, komplementäre zweite funktionelle Gruppen eines Biomoleküls kovalent oder nicht-kovalent zu binden. Durch Wechselwirkung zwischen den ersten und zweiten funktionellen Gruppen ist das Biomolekül an dem Nanopartikel und damit an der Mikrostruktur des Funktionselementes immobilisiert und/oder kann daran immobilisiert werden. Die erfindungsgemäß zur Bildung der Mikrostrukturen verwendeten Nanopartikel weisen eine Größe von kleiner 500 nm, vorzugsweise kleiner 150 nm auf.
Die erfindungsgemäß bevorzugt eingesetzten Nanopartikel weisen eine Kern-Schale-Struktur auf. In bevorzugten Ausführungsformen besteht der Kern der Nanopartikel aus einem anorganischen Material
wie einem Metall, beispielsweise Au1 Ag oder Ni, Silizium, SiO2, SiO, einem Silikat, AI2O3, SiO2^AI2O3, Fe2O3, Ag2O, TiO2, ZrO2, Zr2O3, Ta2Oo, Zeolith, Glas, Indiumzinnoxid, Hydroxylapatit, einem Q-Dot oder einem Gemisch davon oder enthält dieses. In weiteren bevor- zugten Ausführungsformen besteht der Kern aus einem organischen
Material oder enthält dieses. Vorzugsweise ist das organische Polymer Polypropylen, Polystyrol, Polyacrylat, ein Polyester von Milchsäure oder ein Gemisch davon. Die Herstellung der Kerne der erfindungsgemäß verwendeten Nanopartikel kann unter Verwendung üblicher, auf dem Fachgebiet bekannter Verfahren, wie SoI-GeI-
Synthese-Verfahren, Emulsionspolymerisation, Suspensionspolymerisation usw. erfolgen.
In einer bevorzugten Ausgestaltung sind im Kern zusätzliche Funktionen verankert, die unter Verwendung geeigneter Nachweisverfah- ren eine einfache Detektion der Nanopartikelkeme und damit der
Mikrostrukturen ermöglichen. Bei diesen Funktionen kann es sich beispielsweise um Fluoreszenzmarkierungen, UV/Vis-Markierungen, superparamagnetische Funktionen, ferromagnetische Funktionen und/oder radioaktive Markierungen handeln. Geeignete Verfahren zum Nachweis von Nanopartikeln umfassen beispielsweise Fluoreszenz- oder UV-Vis-Spektroskopie-, Fluoreszenz- oder Licht- Mikroskopie-, MALDI-Massenspektrometrie-, Wellenleiterspektroskopie-, Impedanzspektroskopie-, elektrische und radiometrische Verfahren. Auch eine Kombination der Verfahren kann zur Detektion der Nanopartikel verwandt werden. In einer weiteren Ausführungsform ist vorgesehen, dass die Kern-Oberfläche durch Aufbringen zusätzlicher Funktionen wie Fluoreszenzmarkierungen, UV/Vis- Markierungen, superparamagnetischer Funktionen, ferromagneti- scher Funktionen und/oder radioaktiver Markierungen modifiziert
sein kann. Vorzugsweise weist die Oberfläche der Nanopartikelkeme separat oder zusätzlich lonenaustausch-Funktionen auf. Nanoparti- kel mit lonenaustausch-Funktionen sind insbesondere zur Optimierung der MALDI-Analyse geeignet, da dadurch störende Ionen ge- bunden werden können.
Weiter ist vorgesehen, dass die Kern-Oberfläche chemische Verbindungen aufweist, die zur sterischen Stabilisierung und/oder zur Verhinderung einer Konformationsänderung der immobilisierten Moleküle und/oder zur Verhinderung der Anlagerung weiterer biologisch aktiver Verbindungen an die Kern-Oberfläche dient. Vorzugsweise handelt es sich bei diesen chemischen Verbindungen um Polyethy- lenglykole, Oligoethylenglykole, Dextran oder ein Gemisch davon.
Erfindungsgemäß bevorzugt verwendete Nanopartikel besitzen einen Durchmesser von 5 nm bis 500 nm. Unter Verwendung solcher Na- nopartikel lassen sich daher Funktionselemente herstellen, die sehr kleine Mikrostrukturen beliebiger Form im Nanometer- bis Mikrometerbereich aufweisen. Die Verwendung der Nanopartikel zur Erzeugung der Mikrostruktüren erlaubt daher eine bislang nicht erreichte Miniaturisierung der Funktionselemente, die mit erheblichen Verbes- serungen signifikanter Parameter der Funktionselemente einhergeht.
Unter einer „Mikrostruktur" werden Strukturen im Bereich einzelner Mikrometer oder Nanometer verstanden. Insbesondere im Zusammenhang mit der vorliegenden Erfindung wird unter „Mikrostruktur" eine Struktur verstanden, die aus mindestens zwei Einzelkomponen- ten in Form von mehreren dreidimensional angeordneten Schichten von Nanopartikeln mit molekülspezifischen Erkennungsstellen besteht und auf der Oberfläche eines Trägers angeordnet ist, wobei ein
bestimmter Flächenabschnitt der Oberfläche des Trägers abgedeckt wird, der eine definierte Form und einen definierten Flächeninhalt aufweist und der kleiner als die Träger-Oberfläche ist. Erfindungsgemäß ist insbesondere vorgesehen, dass mindestens einer der Flächen-Längen-Parameter, der den durch die Mikrostruktur abgedeckten Flächenabschnitt bestimmt, im Mikrometerbereich liegt. Weist die Mikrostruktur beispielsweise die Form eines Kreises auf, liegt der Durchmesser des Kreises im Mikrometerbereich. Ist die Mikrostruktur als Rechteck ausgeführt, liegt beispielsweise die Breite dieses Rechteckes im Mikrometerbereich. Erfindungsgemäß ist insbesondere vorgesehen, dass der mindestens eine Flächen-Längen- Parameter, der den von der Mikrostruktur abgedeckten Flächenabschnitt bestimmt, kleiner als 999 μm ist. Da die Mikrostruktur erfindungsgemäß aus mindestens zwei Nanopartikeln besteht, liegt die untere Grenze dieses Flächen-Längenparameters bei 10 nm.
In einer bevorzugten Ausführung weisen dreidimensional angeordneten Schichten von Nanopartikeln eine Gesamtdicke von 10 nm bis 10 μm auf. Erfindungsgemäß wird eine Dicke von 50 nm bis 2,5 μm, besonders aber eine Dicke von 100 nm bis 1 ,5 μm, bevorzugt.
Die zur Bildung der Mikrostrukturen bevorzugt verwendeten Nano- partikel besitzen ein vergleichsweise sehr großes Oberfläche- Volumen-Verhältnis und können dementsprechend eine große Menge eines biologischen Moleküls pro Masse binden. Im Vergleich zu Systemen, bei denen biologische Moleküle direkt an einem planaren Träger gebunden sind, kann ein Funktionselement somit pro Flächeneinheit eine erheblich größere Menge der biologischen Moleküle binden. Die Menge der pro Flächeneinheit gebundenen Moleküle, das heißt die Packungsdichte, ist erfindungsgemäß deshalb so groß,
weil zur Erzeugung der Mikrostruktur auf der Trägeroberfläche mehrere Partikel-Lagen übereinander geschichtet werden. Eine weitere Erhöhung der pro Flächeneinheit gebundenen Menge biologischer Moleküle wird bevorzugt dadurch erreicht, dass die Nanopartikel zu- nächst mit Hydrogelen und dann mit den biologischen Molekülen beschichtet werden.
Im Zusammenhang mit der vorliegenden Erfindung wird unter einem „Funktionselement" ein Element verstanden, das entweder allein oder als Bestandteil einer komplexeren Vorrichtung, das heißt im Zusammenhang mit weiteren ähnlichen oder anders gearteten Funktionselementen, mindestens eine definierte Funktion ausübt. Ein Funktionselement umfasst mehrere Bestandteile, die aus gleichen oder unterschiedlichen Materialien bestehen können. Die einzelnen Bestandteile eines Funktionselementes können innerhalb eines Funktionselementes unterschiedliche Funktionen ausüben und können in unterschiedlichem Maße oder in unterschiedlicher Art und Weise zur Gesamtfunktion des Elementes beitragen. In der vorliegenden Erfindung umfasst ein Funktionselement einen Träger mit einer Trägeroberfläche, auf der definierte Schichten von Nanoparti- kein bevorzugt dreidimensional als Mikrostruktur(en) angeordnet ist/sind, wobei die Nanopartikel mit molekülspezifischen Erkennungsstellen ausgewählt aus: spezifischen Antikörpern gegen das Protein TSA 1 , bevorzugt so genannte anti-TSA 1 IgG, und dem Protein TSA 1 , zur Bindung Candida-spezifischer Moleküle, versehen sind.
Die erfindungsgemäßen Funktionselemente lassen sich auf einfache Weise unter Verwendung bekannter Verfahren herstellen. Es wird zur Herstellung und zu weiteren Ausführungsformen der Funktions-

elemente auf die deutsche nachveröffentlichte Patentanmeldung DE 102004 062 573 verwiesen, deren Offenbarungsgehalt hiermit vollinhaltlich einbezogen wird.
Beispielsweise lassen sich unter Verwendung geeigneter Suspen- diermittel aus Nanopartikeln sehr einfach stabile Suspensionen erzeugen. Nanopartikelsuspensionen verhalten sich wie Lösungen und sind dadurch kompatibel mit Mikrostrukturierungsverfahren. Nanopartikelsuspensionen können daher direkt, beispielsweise unter Verwendung herkömmlicher Verfahren wie Nadel-Ring-Printer, lithogra- phischer Verfahren, Tintenstrahlverfahren und/oder Mikrokontaktver- fahren, strukturiert auf geeignete Träger abgeschieden werden, die zuvor mit einem Verbindungsmittel zur festen Anhaftung der Nano- partikel vorbehandelt wurden. Durch eine geeignete Wahl des Verbindungsmittels kann die gebildete Mikrostruktur so ausgebildet wer- den, dass sie zu einem späteren Zeitpunkt teilweise oder vollständig von der Trägeroberfläche des Funktionselementes, beispielsweise durch Änderung des pH-Wertes oder der Temperatur, gelöst und gegebenenfalls auf die Trägeroberfläche eines anderen Funktionselementes übertragen werden kann.
Erfindungsgemäß bevorzugt ist in die Mikrostruktur mindestens ein
Biomolekül stabilisierendes Agens, insbesondere mindestens ein Protein stabilisierendes Agens, eingeschlossen. Durch solche Agenzien wird die Stabilisierung der Biomoleküle weiter verstärkt. Der Zusatz mindestens eines Biomolekül stabilisierenden Additivs, insbe- sondere mindestens eines proteinstabilisierenden Additivs, erhält die
Funktionalität Nanopartikel-gebundener biologischer Moleküle, insbesondere Peptide oder Proteine, innerhalb der Partikelschichten, wenn diese auf einem Substrat eingetrocknet werden, und garantiert
so die Lagerfähigkeit nanopartikulärer Funktionsschichten. Dabei beträgt die Lagerfähigkeit bis ein Jahr, bevorzugt bis 8 Monate, insbesondere 3 Monate. Der erfindungsgemäße Einschluss mindestens eines Biomolekül stabilisierenden Agens, insbesondere mindestens eines proteinstabilisierenden Agens, in die Mikrostruktur schützt also die Funktion, vor allem die biologische Funktion, und die Wirksamkeit der erfindungsgemäßen Funktionselemente. Unter „Biomolekül stabilisierenden Agenzien", insbesondere „proteinstabilisierenden Agenzien", werden erfindungsgemäß Agenzien verstanden, die die dreidimensionale Struktur von Proteinen, also Sekundär-, Tertiär-,
Quartärstruktur, unter Trockenstress stabilisieren und damit die Funktionalität der Proteine im Trockenzustand, das heißt nach Verdampfen des Lösungsmittels, erhalten. In einer bevorzugten Ausführungsform ist das Protein stabilisierende Agens ein Saccharid, ins- besondere Saccharose (Sucrose), Lactose, Glucose, Trehalose oder
Maltose, ein Polyalkohol, insbesondere Inositol, Ethylenglycol, GIy- zerol, Sorbitol, Xylithol, Mannitol oder 2-Methyl-2,4-pentandiol, eine Aminosäure, insbesondere Natriumglutamat, Prolin, alpha-Alanin, beta-Alanin, Glyzin, Lysin-HCI oder 4-Hydroxyprolin, ein Polymer, insbesondere Polyethylenglykol, Dextran, Polyvinylpyrrolidon, ein anorganisches Salz, insbesondere Natriumsulfat, Ammoniumsulfat, Kaliumphosphat, Magnesiumsulfat oder Natriumfluorid, ein organisches Salz, insbesondere Natriumacetat, Natriumpolyethylen, Natri- umcaprylat, Propionat, Lactat oder Succinat, oder Trimethylamin N- oxid, Sarcosin, Betain, gamma-Aminobuttersäure, Octopin, Alanopin,
Strombin, Dimethylsulfoxid oder Ethanol oder eine Mischung aus den genannten Stoffen.
Erfindungsgemäß besteht der Träger des Funktionselementes, insbesondere die Träger-Oberfläche, aus einem Metall, einem Metall-
oxid, einem Polymer, Glas, einem Halbleitermaterial oder Keramik. In bevorzugter Ausführungsform besteht der Träger des Funktionselementes aus Materialien, wie transparentem Glas, Siliciumdioxid, Metallen, Metalloxiden, Polymeren und Copolymeren von Dextranen oder Amiden, beispielsweise Acrylamid-Derivaten, Cellulose, Nylon, oder polymeren Materialien, wie Polyethylenterephthalat, Cellulose- acetat, Polystyrol oder Polymethylmethacrylat oder einem Polycar- bonat von Bisphenol A. Im Zusammenhang mit der Erfindung bedeutet dies, dass entweder der Träger vollständig aus einem der vorste- hend genannten Materialien besteht oder dieses im Wesentlichen enthält. Der Träger oder seine Oberfläche besteht dabei zumindest zu etwa 60%, bevorzugt zu etwa 70%, zu etwa 80% oder zu etwa 100% aus einem der vorstehend genannten Materialien oder einer Kombination solcher Materialien.
In bevorzugter Ausführungsform der Erfindung ist vorgesehen, dass zwischen der Trägeroberfläche und der Mikrostruktur mindestens eine Schicht eines Verbindungsmittels angeordnet ist. Das Verbindungsmittel dient zur festen Bindung der Nanopartikel an der Trägeroberfläche des Funktionselementes. Die Auswahl des Verbin- dungsmittels richtet sich nach der Oberfläche des Trägermaterials und den zu bindenden Nanopartikeln. Bei dem Verbindungsmittel handelt es sich vorzugsweise um geladene oder ungeladene Polymere. Die Verbindungsmittel sind vorzugsweise schwache oder starke Polyelektrolyte, das heißt ihre Ladungsdichte ist pH-abhängig oder pH-unabhängig. In einer bevorzugten Ausführungsform besteht das Verbindungsmittel aus Poly(diallyl-dimethyl-ammoniumchlorid), einem Natriumsalz der Poly(styrolsulfonsäure), einem Natriumsalz der Poly(vinylsulfonsäure), Poly(allylamin-hydrochlorid), linearem oder verzweigtem Poly(ethylenimin), Poly(acrylsäure), Poly(meth-
acrylsäure) oder einem Gemisch aus diesen. Bei dem Polymer handelt es sich bevorzugt um ein Hydrogel.
Weitere bevorzugte Verbindungsmittel sind ausgewählt aus funktionellen Silanen, besonders zur Aktivierung von Glasoberflächen, SiIi- ziumoberflächen oder ähnlichen, und funktionellen Thiolen, besonders zur Aktivierung von Goldoberflächen. Diese Moleküle bestehen im Wesentlichen aus einem „Anker", wie Silanol, Chlorsilan oder ähnlichen, einem „Spacer", wie Polyethylenglykol, Oligoethylenglykol, Kohlenwasserstoffketten, Kohlenhydratketten oder ähnlichen, und mindestens einer funktionellen Gruppe, vorzugsweise Aminogruppe,
Carboxygruppe, Hydroxygruppe, Expoxygruppe, Tosylchlorid, N- hydroxy-Succimidester, Maleimid und/oder Biotin.
Weitere bevorzugte Verbindungsmittel sind auch Polymere, die Aktivester enthalten, wie Phenyldimethyl-sulfoniummethylsulfat Grup- pen, photoaktive Crosslinker, Proteine, wie Streptavidin, BSA und ähnliche, sowie Nucleinsäuren.
Bevorzugt sind auch Kombinationen mindestens zweier vorgenannter Verbindungsmittel.
Im Zusammenhang mit der vorliegenden Erfindung bedeutet „adres- sierbar", dass die Mikrostruktur nach dem Auftragen der Nanoparti- kel auf die Trägeroberfläche wieder auffindbar und/oder nachweisbar ist. Wird die Mikrostruktur beispielsweise unter Verwendung einer Maske oder eines Stempels auf die Trägeroberfläche aufgetragen, resultiert die Adresse der Mikrostruktur einerseits aus den Koordina- ten x und y des von der Maske oder dem Stempel vorgegebenen
Bereiches der Trägeroberfläche, auf den die Mikrostruktur aufgetragen wird. Andererseits resultiert die Adresse der Mikrostruktur aus
den molekülspezifischen Erkennungsstellen auf der Oberfläche der Nanopartikel, die ein Wiederauffinden beziehungsweise einen Nachweis der Mikrostruktur erlauben.
Die vorliegende Erfindung betrifft weiter die Verwendung des erfin- dungsgemäßen Funktionselementes zum Nachweis von Candida und Candida-verwandten Pilzellen in, also besonders zur Diagnose von Candidosen in menschlichen oder tierischen Körpern.
Unter „klinischem Material" oder „Probe eines klinischen Materials" wird eine Probe wie Vollblut, Blutserum, Lymphe, Gewebeflüssigkeit, Bronchiallavage, gastrointestinale Spülflüssigkeit, Stuhl, Zervikal- schleim, Schleimhautabstrich verstanden. Ebenso wird darunter auch eine Biopsie oder Gewebeprobe, die einem lebenden oder toten Organismus, Organ oder Gewebe entnommen wurde, verstanden. Eine Probe kann jedoch auch ein Kulturmedium, beispielsweise ein Fermentationsmedium, sein, in dem Organismen, beispielsweise
Mikroorganismen, oder menschliche, tierische oder pflanzliche Zellen kultiviert wurden. Eine solche Probe kann bereits Aufreinigungsschritten, wie Proteinisolation, unterworfen worden sein, kann aber auch ungereinigt vorliegen.
Die erfindungsgemäße Verwendung des erfindungsgemäßen Funktionselementes macht sich die spezifische Antigen-Antikörper- Bindung zwischen den molekülspezifischen Erkennungsstellen, ausgewählt aus spezifischen Antikörpern gegen das Protein TSA 1 und dem Protein TSA 1 , mit entsprechenden Candida-spezifischen MoIe- külen, die in der zu untersuchenden Probe des klinischen Materials vorkommen, zunutze.
Der durch Inkontaktbringen des Funktionselements mit dem bereitgestellten klinischen Material sich ausbildende Antigen-Antikörper- Komplex kann in an sich bekannter Weise nachgewiesen werden. Bekannte Verfahren der Immunhistologie lassen sich, entsprechend angepasst, auf die Funktionselemente anwenden. Erfindungsgemäß bevorzugt werden zum Nachweis des Antigen-Antikörper-Komplexes auf dem Funktionselement markierte Antigen-Proteine beziehungsweise markierte primäre oder markierte sekundäre Antikörper eingesetzt, die die im Antigen-Antikörper-Komplex spezifisch gebundenen Candida-spezifischen Moleküle aus der Probe durch eine weitere spezifische Antigen-Antikörper-Bindung markieren. Als Markierungsmittel wird bevorzugt die Fluoreszenzmarkierung oder die Metallmarkierung verwendet. Zum Nachweis dieser Markierung werden bevorzugt MALDI-Massenspektrometrie, Fluoreszenz- oder UV-VIS- Spektroskopie, Fluoreszenz- oder Lichtmikroskopie, Wellenleiterspektroskopie, elektrische Verfahren wie Impedanzspektroskopie oder eine Kombination dieser Verfahren eingesetzt.
Wird ein Fluoreszenznachweisverfahren eingesetzt, wird ein fluoreszenzmarkierter Analyt und/oder fluoreszenzmarkiertes biologisch aktives an dem Nanopartikel gebundenes Detektionsmolekül mit
Licht angeregt und mit Licht ausgelesen. Erfindungsgemäß bevorzugt ist bei der Verwendung eines Fluoreszenzverfahrens der Analyt und/oder das molekülspezifische Detektionsmolekül und/oder ein weiteres sekundäres Nachweismolekül wie ein sekundärer Antikör- per, Streptavidin etc. fluoreszenzmarkiert.
Besonders bevorzugt findet der Nachweis des markierten Antigen- Antikörper-Komplexes in automatisierter Form beispielsweise in Scannern statt.
Die vorliegende Erfindung betrifft daher auch ein Verfahren zur Identifizierung und/oder zum Nachweis von Candida und Candida- verwandten Pilzellen insbesondere in klinischem Material, also besonders ein Verfahren zur Diagnose von Candidosen in menschli- chen oder tierischen Körpern. In einem Verfahrensschritt a) wird eine
Probe, besonders aus klinischem Material, bereitgestellt. In einem weiteren Verfahrensschritt b) wird ein erfindungsgemäßes Funktionselement, also ein Candida Diagnose Chip, bereitgestellt, das in einem weiteren Verfahrensschritt c) mit der Probe unter Bedingun- gen in Kontakt gebracht wird, die eine spezifische Antikόrper-
Antigen-Bindung ermöglichen, wobei Candida-spezifische Moleküle aus der Probe an die molekülspezifischen Erkennungsstellen des Funktionselements, ausgewählt aus spezifischen Antikörpern gegen das Protein TSA 1 und dem Protein TSA 1 , in einem Antigen- Antikörper-Komplex gebunden werden. In einem weiteren Verfahrensschritt f) wird der auf dem Candida-Diagnose-Chip gebildete An- tigen-Antikörper-Komplex in an sich bekannter Weise, bevorzugt mittels fluoreszenzmarkierten Antigenen beziehungsweise Antikörpern, nachgewiesen. In Schritt e) werden daher bevorzugt die auf dem Caπd/da-Diagnose-Chip gebundenen Candida-spezifischen Moleküle mit fluoreszenzmarkierten Molekülen, wie markierte Antikörper, markierte sekundäre Antikörper, markierte rekombinante Proteine, etc., gebunden. In einer weiteren bevorzugten Verfahrensform wird als Nachweisverfahren ein MALDI-Massenspektrornetrieverfahren angewendet.
Bevorzugt werden nach Schritt c) und vor dem Nachweis in Schritt f) in einem zusätzlichen Schritt d) nicht gebundene Candida- spezifische Moleküle und auch unspezifische Moleküle von dem Funktionselement durch Waschen mit einer biokompatiblen Wasch-
flüssigkeit entfernt. Die biokompatible Waschflüssigkeit ist vorzugsweise Wasser und/oder Puffer, z. B. Phosphat-buffered Saline: PBS, und/oder Puffer mit Zusatz eines Detergenz, z. B. TritonX-100. In bevorzugter Ausführungsform der Erfindung wird der Träger bei Raumtemperatur sequentiell in Wasser und Puffer, gegebenenfalls mit einem Detergenz, oder Puffer, gegebenenfalls mit einem Detergenz, und Wasser z.B. je 30 min gewaschen werden.
Eine weitere erfindungsgemäße Verwendung des Funktionselements ist die Isolierung eines mit den immobilisierten molekülspezifischen Erkennungsstellen, ausgewählt aus spezifischen Antikörpern gegen das Protein TSA 1 und dem Protein TSA 1 , in Wechselwirkung tretenden Proteinen aus einer Probe.
Die vorliegende Erfindung betrifft schließlich auch die Verwendung des Funktionselementes zur Entwicklung und Herstellung von phar- mazeutischen Präparaten für die Diagnose und Therapie von Candidose und verwandten Pilzinfektionen des menschlichen oder tierischen Körpers.
Weitere vorteilhafte Ausgestaltungen der Erfindung ergeben sich aus den Unteransprüchen.

Die Erfindung wird anhand der folgenden Figuren und Beispiele näher erläutert.
Figur 1 zeigt das Ergebnis der Detektion von Rabbit-anti-
TSA 1 -Antikörpern. Der Antikörper (35 ng/ml) wird mit Hilfe nanopar- tikulärer Affinitäts-Schichten nachgewiesen. Die Sensorschichten bestehen aus funktionellen Nanopartikeln, die Candida-Zelllysat auf ihrer Oberfläche gebunden haben. Der Nachweis der Anbindung ge- schieht durch einen fluoreszenzmarkierten anti-Rabbit-Antikörper.
Figur 2 zeigt das Ergebnis der Detektion von fluoreszenzmarkiertem Candida-Antigen. Das rekombinante Antigen wird mit Hilfe nanopartikulärer Affinitäts-Schichten in einer Konzentration von 40 pmol/l nachgewiesen. Die Sensorschichten bestehen aus funktionel- len Nanopartikeln, die anti-TSA 1 -Antikörper auf ihrer Oberfläche gebunden haben.
Figur 3 zeigt das Ergebnis der Detektion von Candida-Antigen mit Hilfe der Sandwich Technik. Das rekombinante Antigen wird mit Hilfe nanopartikulärer Affinitäts-Schichten in einer Konzentration von
100 pmol/l nachgewiesen. Die Sensorschichten bestehen aus funktionellen Nanopartikeln, die anti-TSA 1-Antikörper auf ihrer Oberfläche gebunden haben. Der Nachweis geschieht durch einen fluoreszenzmarkierten anti-TSA 1 -Antikörper.
Beispiel 1 : Nachweis von anti-Caπd/'da a/fo/cans-Antikörpern in klinischem Material
In diesem Beispiel wird ein Antikörper detektiert, der gegen ein gegen das Antigen TSA 1 von Candida albicans gerichtet ist. Der Nachweis von anti-Candida Antikörpern in einer Probe wird geführt, indem Candida-Zelllysat auf funktionellen Silica-Nanopartikeln immobilisiert wird und diese bioaktiven Nanopartikel als Affinitäts- Beschichtung auf einem Substrat abgeschieden werden. In der Probe befindliche anti-Candida-Antikörper binden an Candida-Antigen TSA, das in dreidimensional nanostrukturierten Affinitätsschichten immobilisiert ist. Der Nachweis der Bindung wurde geführt mittels fluoreszenzmarkiertem sekundärem Antikörper.
1.1 Herstellung von Nanopartikel-basierten Candida Diagnose- Chips
Substrat:
Zur Herstellung von Nanopartikel-basierten Candida Diagnosechips, die für die Fluoreszenz-Auslese geeignet sind, werden exemplarisch Glas-Substrate verwendet. Die Haftung der Nanopartikel auf Oberflächen wird hierbei größtenteils durch elektrostatische Wechselwir- kung vermittelt. Zur Adsorption proteinbeschichteter Nanopartikel
auf dem Substrat sind meist positiv geladene Oberflächen erforderlich. Kommerziell erhältliche Glasobjektträger, die positive Gruppen an den Oberflächen aufweisen, werden ohne weitere Vorbehandlung mit proteinbeschichteten Nanopartikeln bedruckt.
Herkömmliche Glasoberflächen werden in einer 2 Vol.-% wässrigen
HELLMANEX®-Lösung 90 min bei 40 0C gereinigt. Nach Waschen in MiIIiQ-H2O (entionisiertes Wasser, 18 MΩ) werden die Glasobjekträ- ger in einer 3:1 (v/v) N H3ZH2O2-Lo" sung 40 min bei 70 0C hydroxyliert (NH3 puriss. p.a. -25% in Wasser und H2O2 zur Analyse, 30%, ISO Reag., stabilisiert).
Nach gründlichem Waschen in MilliQ-Wasser werden die Substrate bei Raumtemperatur für 20 min in einer wässrigen Polykationlösung (0,02 mol/l Poly(allylamin) (bezogen auf das Monomer), pH 8,5) inkubiert, 5 min in MilliQ-Wasser gewaschen und anschließend tro- cken zentrifugiert.
Synthese von Kern-Schale-Partikeln:
Zu 200 ml Ethanol werden 12 mmol Tetraethoxysilan und 90 mmol NH3 gegeben. Dann wird 24 h bei Raumtemperatur gerührt. Anschließend werden die Partikel durch mehrfache Zentrifugation ge- reinigt. Es resultieren 650 mg Kern-Schale-Partikel mit einer mittleren Partikelgröße von 125 nm.
Amino-Funktionalisierung von Kern-Schale-Partikeln:
Eine 1 Gew.-% wässrige Suspension der Kern-Schale-Partikel wird mit 10 Vol.-% 25%igem Ammoniak versetzt. Dann werden 20 Gew.-% Aminopropyltriethoxysilan, bezogen auf die Partikel, zu-
gegeben und es wird 1 h bei Raumtemperatur gerührt. Die Partikel werden durch mehrfache Zentrifugation gereinigt und tragen funktionelle Aminogruppen auf ihrer Oberfläche (Zeta-Potential in 0, 1 mol/l Acetat-Puffer: + 35 mV).
Carboxy-Funktionalisierung von Kern-Schale-Partikeln:
10 ml einer 2 Gew.-% Suspension Amino-funktionalisierter Nanopar- tikel werden in Tetrahydrofuran aufgenommen. Dazu werden 260 mg Bernsteinsäureanhydrid gegeben. Nach einer 5 min Behandlung mit Ultraschall wird 1 h bei Raumtemperatur gerührt. Die Partikel werden durch mehrfache Zentrifugation gereinigt und tragen funktionelle
Carboxygruppen auf ihrer Oberfläche (Zeta-Potential in 0,1 mol/l Acetat-Puffer: - 35 mV). Die mittlere Partikelgröße beträgt 170 nm.
1.2 Anbindung der molekülspezifischen Erkennunqsstellen an die Kern-Schale-Partikel - Anbindung von Candida-Zelllysat enthaltend TSA 1 Protein
1 mg Carboxy-funktionalisierter Kern-Schale-Partikel wird mit 30 μl eines Candida Zelllysats, welches das Antigen TSA 1 enthält, und 10 μl einer EDC-Lösung (N-(3-Dimethylaminopropyl)-N'-ethylcarbo- diimid-HCI; 3,8 mg /ml) versetzt und mit MES-Puffer (pH 4,5) auf 1 ml aufgefüllt.
Es wird über Nacht (ca. 10 h) bei 4 0C geschüttelt. Anschließend werden die Partikel durch mehrfache Zentrifugation gereinigt.
Es werden mit Zelllysat von Candida albicans- Wildtyp beladene Na- nopartikel hergestellt. Als Kontrolle dienen mit Zelllysat von Candida albicans-TSA 1 -Knockout beladene Nanopartikel.
Konservierung der Proteinfunktion in Nanopartikelschichten:
Zur Stabilisierung der Funktion Nanopartikel-gebundener Fänger- Proteine in Nanopartikelschichten werden die Partikel zur Beschich- tung in 5% (w/v) wässriger Trehalose-Lösung suspendiert.
1.3 Herstellung der Mikroarrays
Zur Herstellung Fluoreszenz-auslesbarer Candida Diagnosechips werden die mit Candida Zelllysat beladenen Nanopartikel mit Hilfe eines Pin-Ring Spotters auf das vorbehandelte Glas-Substrat übertragen. Die Konzentration der verwendeten Partikel-Suspensionen beträgt 2% (w/v). Pro Nadel-Kontakt mit der Oberfläche werden ca.
50 pl Suspension übertragen, es wird fünf Mal pro Spot gedruckt. Der Spof-Durchmesser beträgt ca. 150 μm. Die Platzierung der einzelnen Spots auf dem Substrat ist frei programmierbar.
1.4 Einsatz der Candida Diagnose-Chips
Antikörperherstellung:
Je 3 mg der synthetisierten Peptide HPGDETIKPS (SEQ ID NO: 2) und EASKEYFNKVNK (SEQ ID NO: 3) Ge 20 mg synthetisiert, >70 % Reinheit; Thermo Electron Corporation, Ulm) in 300 μl PBS wurde an 3 mg Keyhole limpet hemocyanin (KLH, Sigma Aldrich, Taufkirchen) in 300 μl Wasser gekoppelt. Die Kopplung erfolgte zunächst bei Raumtemperatur durch fünfmalige Zugabe von je 2,4 μl 5 % Glutaraldehyd (Endkonzentration ca. 10 mmol/l) in Abständen von 5 min. Der Reaktionsmix wurde 30 min auf Eis inkubiert. Die Blockierung erfolgte mit 24 μl 1 mol/l Glycin pH 8,5.
Die gekoppelten Peptide wurden vereinigt und je zur Hälfte pro Tier verwendet. Zwei Kaninchen wurden im Abstand von 30 Tagen insgesamt viermal immunisiert (Pineda, Berlin). Präimmunserum, Serum vom Immunisierungstag 61 , 90 und 120 wurde charakterisiert.
Immobilisierte Peptide für die Affinitätsreinigung der TSA1 -Antikörper wurden mittels CNBr-aktivierter Sepharose 4B (Amersham Bioscien- ces, Freiburg) nach Angaben der Firma hergestellt. 0,3 g CNBr- aktivierte Sepharose 4B wurde in ein Röhrchen gegeben und 15 min in 1 mmol/l HCl quellen gelassen, so dass die Kügelchen bedeckt waren. Anschließend wurde die Sepharose mehrmals mit insgesamt
300 ml 1 mmol/l HCl und danach mit 7,5 ml 100 mmol/l NaHCO3 0,5 mol/l NaCI pH 8,3 (Bindungspuffer) gewaschen.
Je 2,5 mg von Peptid 10 und Peptid 12 wurden in 2 ml Bindungspuffer gelöst, zu der gewaschenen Sepharose gegeben und über Nacht bei 4 0C auf dem Rotierrad inkubiert. Überschüssiges Peptid wurde durch einmaliges Waschen mit 5 ml Bindungspuffer entfernt und die noch verbliebenen aktiven Gruppen mit 1 mol/l Ethanolamin pH 8,0 für 2 h blockiert. Die Sepharose wurde abwechselnd jeweils mindestens dreimal mit dem fünffachen Gelvolumen mit 0,1 mol/l Na-Acetat 0,5 mol/l NaCI pH 4 und 0,1 mol/l Tris-HCI 0,5 mol/l NaCI pH 8,0 gewaschen. Die Affinitätsmatrix wurde noch zweimal in PBS pH 7,4 gewaschen und bei 4 0C mit 0,02 % (w/v) Azid gelagert.
Inkontaktbringen mit der Probe:
Pro Aufreinigung wurden 3 ml Serum der TSA1 -Antikörper verwen- det. Inkubation erfolgte rotierend über Nacht bei 4 0C, dreimal wurde mit PBS pH 7,4 gewaschen, anschließend mit 0, 1 mol/l Glycin pH 2,8 eluiert. Das Eluat wurde in 1 ml Fraktionen in 1 ,5 ml Reak-
tinsgefäßen gesammelt, in denen je 50 μl 1 mol/l Tris-HCI pH 8,8 vorgelegt wurde. Insgesamt wurden zehn Fraktionen gesammelt. Diese wurden bei 280 nm in einer Quarz-Küvette vermessen und die Fraktionen 1-3 vereinigt und gegen PBS pH 7,4 dialysiert. Die Dialy- se erfolgte einmal für 2 h und einmal über Nacht bei 4 0C in je 2 I
PBS. Die affinitätsgereinigten und dialysierten TSA1 -Antikörper wurden mit 0,02 % (w/v) Azid versetzt und bei 4 0C gelagert.
Die Nanopartikeloberflächen werden zunächst 1 h mit einer 3%igen (w/v) Lösung von BSA in PBS-Puffer geblockt. Anschließend wird für 1 ,5 h im Dunkeln bei Raumtemperatur mit einer Probe aufgereinigtem anti-TSA 1 Antikörper (ca. 230 pmol/l oder 5 μg auf 100 ml PBS + 1% BSA) enthält, inkubiert. Danach wird jeweils 30 min in PBS gewaschen.
Als Kontrolle dienen funktionelle Nanopartikel, auf denen das ZeIIIy- sat eines Candida-Stammes immobilisiert, das das Antigen TSA 1 nicht enthält, weil das Gen dieses Antigen stillgelegt ist (Knockout- Stamm).
Markierung der gebundenen anti-TSA '\-Antiköφer:
Mit einem fluoreszenzmarkierten sekundären Antikörper gegen die Spezies, aus der die Antikörper stammen, im tierexperimentellen
Ansatz hier anti-Rabbit-Antikörper (bei diagnostischen Test anti- Human-Antikörper), wird die Anbindung nachgewiesen. Der fluoreszenzmarkierte sekundäre Antikörper wird in einer 1 %igen BSA- Lösung in PBS/Tween (0,1 %) gelöst (0,7 μg auf 100 ml). Die Chips werden 1 h bei Raumtemperatur im Dunkeln damit inkubiert und anschließend je 30 min in PBS/0,1 %TritonX 100, in PBS und in MiIIiQ-
Wasser gewaschen. Alle Schritte werden in Glas- Objektträgerständern durchgeführt.
Auslesen des Chips:
Das Fluoreszenz-Signal der gebundenen anti-Candida-Antikörper, anti-Rabbit-Antikörper wird in einem kommerziellen Chip-Reader
System der Firma ArrayWorx, detektiert. Die Belichtungszeiten liegen zwischen 0,1 s und 2 s und werden innerhalb eines Experiments konstant gehalten. Die Signalintensitäten werden in Form von Grauwertabstufungen gespeichert. Die Auswertung der Daten erfolgt mit Hilfe des Programms Aida der Firma Raytest, Berlin. Die Ergebnisse sind in Figur 1 dargestellt.
Beispiel 2: Nachweis eines fluoreszenzmarkierten rekombinanten Candida a/fe/'cans-Antiqens mittels Candida Diagnose-Chip
Der Nachweis des Candida spezifischen Antigens TSA 1 in einer
Probe wird geführt, indem Antikörper gegen TSA 1 auf funktionellen Silica-Nanopartikeln immobilisiert werden und diese bioaktiven Na- nopartikel als Affinitäts-Beschichtung auf einem Substrat abgeschieden werden. In der Probe befindliche TSA 1 -Antigene (im Experi- ment beispielhaft gewählt: TSA 1-Maltosebindingprotein Fusions- konstrukt (TSA 1-MPB) binden an den anti-TSA 1 -Antikörper, der in dreidimensionalen nanostrukturierten Affinitätsschichten immobilisiert ist. In diesem Beispiel wurde rekombinantes, fluoreszenzmarkiertes TSA 1-MPB-Fusionsprotein als Candida-Antigen eingesetzt.
2.1 Herstellung von Nanopartikel-basierten Candida Diagnosechips
Entspricht Beispiel 1.1.
2.2 Anbindung der molekülspezifischen Erkennungsstellen an die Kern-Schale-Partikel - Anbindung von anti-TSA 1-IgG
Die exemplarisch eingesetzten Rabbit-anti-TSA 1-lgG-Moleküle können a) ungerichtet, kovalent an die funktionellen Nanopartikel angebunden werden, oder gerichtet über b) Protein G oder c) Anti-Rabbit- IgG:
a) kovalent, ungerichtet:
1 mg Carboxy-funktionalisierter Silica-Partikel wird mit 66 μl Rabbit- anti-TSA 1 IgG Lösung (0,7 mg/ml) und 10 μl einer EDC-Lösung (N- (3-Dimethylaminopropyl)-N'-ethyl-carbodiimid-HCI; 3,8 mg/ml) versetzt und mit MES-Puffer (pH 4,5) auf 1 ml aufgefüllt. Die Mischung wird über Nacht bei 4 0C geschüttelt, anschließend werden die Partikel durch mehrfache Zentrifugation gereinigt.
b) über Protein G:
1 mg Carboxy-funktionalisierter Silica-Partikel wird mit 10 μl ProteinG Gamma Bind Typ 2 (Pierce) (3 mg/ml) und 10 μl einer EDC-Lösung (N-P-DimethylaminopropyO-N'-ethyl-carbodiimid-HCI; 3,8 mg /ml) versetzt und mit MES-Puffer (pH 4,5) auf 1 ml aufgefüllt. Die Mischung wird über Nacht bei 4 0C geschüttelt, anschließend werden die Partikel durch mehrfache Zentrifugation gereinigt.
500 μg ProteinG-Partikel werden mit 26 μl anti-TSA 1 IgG-Lösung (0,7 mg/ml) versetzt und mit PBS auf 500 μl aufgefüllt. Die Mischung wird über Nacht bei 4°C geschüttelt, anschließend werden die Partikel durch mehrfache Zentrifugation gereinigt.
c) über Anti-Rabbit-IgG:
1 mg Carboxy-funktionalisierter Silica-Partikel wird mit 66 μl anti- Rabbit IgG Lösung (0,7 mg/ml) und 10 μl einer EDC-Lösung (N-(3- Dimethylaminopropyl)-N'- ethyl-carbodiimid-HCI; 3,8 mg /ml) versetzt und mit MES-Puffer (pH 4,5) auf 1 ml aufgefüllt. Die Mischung wird über Nacht bei 4 0C geschüttelt, anschließend werden die Partikel durch mehrfache Zentrifugation gereinigt.
500 μg anti-Rabbit IgG Partikel werden mit 26 μl anti-TSA 1- IgG Lösung (0,7 mg/ml) versetzt und mit PBS auf 500 μl aufgefüllt. Die Mischung wird über Nacht bei 4 0C geschüttelt, anschließend werden die Partikel durch mehrfache Zentrifugation gereinigt.
Stabilisierung der Protein funktion:
Zur Konservierung/Stabilisierung der Proteinfunktion der an die Na- nopartikel gebundenen Proteine in Nanopartikelschichten werden die Partikel zur Beschichtung in 5% (w/v) wässriger Trehalose-Lösung suspendiert.
2.3 Herstellung der Mikroarrays

2.4 Einsatz der Candida Diagnose-Chips
TSA 1- Maltosebindingprotein - Fusionskonstrukt
Beispielhaft wurde als Probe (TSA 1 -Antigen) ein Fusionsprotein eingesetzt. Das Fusionsprotein (SEQ ID NO: 4) wurde kloniert, um die Aufreinigung über Maltosebindungsprotein (MBP; SEQ ID NO: 5) bewerkstelligen zu können. TSA 1 (SEQ ID NO: 1) ist mit dem C-terminalen Ende von MBP (SEQ ID NO: 5) über einen Linker (SEQ ID NO: 6) verknüpft.
Als Überexpressionsvektor wurde pMAL-p2X (Fa. NEB) eingesetzt. Die Proteinaufreinigung wurde in an sich bekannter Weise nach den
Angaben des Herstellers durchgeführt.
Inkontaktbringen mit der Probe:
Die Nanopartikeloberflächen werden zunächst 1 h mit einer 3%igen (w/v) Lösung von BSA in PBS-Puffer geblockt. Anschließend wird für 1 h im Dunkeln bei Raumtemperatur mit einer Lösung des fluoreszenzmarkierten rekombinanten TSA 1-MBP-Fusionsproteins- Antigens (40 pmol/l in PBS) inkubiert. Anschließend werden die Chips je 30 min in PBS/0,1%TritonX 100, in PBS und in MiIIiQ- Wasser gewaschen. Alle Schritte werden in Glasobjektträgerstän- dem durchgeführt.
Als Negativ-Kontrollen werden Anti-Rabbit-IgG-, anti-Goat IgG- und/oder Streptavidin-beschichtete Nanopartikel eingesetzt.
Auslesen des Chips:
Siehe Beispiel 1.4. Die Ergebnisse sind in Figur 2 dargestellt.
Beispiel 3: Nachweis von Candida albicans-Antiqen mittels Sandwich-Technik auf Nanopartikel-basiertem Candida-Diaqnose-Chip
Der Nachweis von Candida albicans Antigenen in einer Probe wird geführt, indem Antikörper gegen ein TSA 1 auf funktionellen Silica- Nanopartikeln immobilisiert werden und diese bioaktiven Nanoparti- kel als Affinitäts-Beschichtung auf einem Substrat abgeschieden werden. In der Probe befindliche TSA 1 -Antigene binden an den an- ti-Candida-Antikörper, der in den dreidimensionalen nanostruk- turierten Affinitäts-Schichten immobilisiert ist. Mit Hilfe eines fluores- zenzmarkierten Detektionsantikörpers wird die Bindung nachgewiesen (Sandwich). Als Negativ-Kontrollen werden anti-Goat IgG- beschichtete Nanopartikel eingesetzt.
3.1 Herstellung von Nanopartikel-basierten Candida Diagnosechips

3.2 Anbindung der molekülspezifischen Erkennungsstellen an die Kern-Schale-Partikel - Anbindung von Rabbit-anti-Candida IgG
Die Anbindung von Rabbit-anti-Candida IgG an Kern-Schale- Nanopartikeln erfolgt kovalent, ungerichtet; entsprechend Beispiel 2.2. Die Proteine werden stabilisiert wie in Beispiel 2.2.
3.3 Herstellung der Mikroarrays
Entspricht Beispiel 1.3.
3.4 Einsatz der Candida Diagnose-Chips
Die anti-Candida-Nanopartikel-Oberflächen werden zunächst 1 h mit einer 3%igen (w/v) Lösung von BSA in PBS-Puffer geblockt und dann zunächst für 1 h bei RT mit einer Lösung des rekombinanten TSA 1-MBP-Fusionsprotein-Antigens (100 pmol/l in PBS) inkubiert.
Anschließend werden die Chips je 30 min in PBS/0,1%TritonX 100 und PBS gewaschen, dann wieder 30 min in BSA-Lösung blockiert.
Sie werden dann 1 h bei Raumtemperatur im Dunkeln mit einer Lösung des fluoreszenzmarkierten Detektionsantikörpers (40 pmol/l in PBS) inkubiert und schließlich je 30 min in PBS/0, 1 %TritonX 100, in
PBS und in MilliQ-Wasser gewaschen. Alle Schritte werden in Glasobjektträgerständern durchgeführt.
Die Ergebnisse sind in Figur 3 dargestellt.
