Description
実施例1:抗BGモノクローナル抗体の作製
DBA/2マウスに,complete freund adjuvantと共にCSBGを皮下投与して免疫し、追加免疫を行った後、抗BG抗体価をCSBG固相化ELISAプレート用いチェックした。抗体価の上昇したマウスの脾臓中の抗体産生細胞とミエローマ(骨髄腫細胞:X63)を細胞融合させ、ハイブリドーマ作製した。HAT選択培地による培養を行い、オリゴクローナルな抗体産生細胞を得た。得られたオリゴクローナル抗体産生細胞を96 well plateに、1,2,3 cells/wellになるように撒き、限界希釈を行った。得られた陽性クローンをさらに,0.3,0.5,1 cells/wellになるように撒き2回目の限界希釈を行った。抗BGモノクローナル抗体産生細胞のスクリーニングはCSBG固相化ELISA法により行い、抗BGモノクローナル抗体産生ハイブリドーマ2B8を得た。このハイブリドーマを共雑タンパクの少ないハイブリドーマメディウムで培養し、インテグラセルラインにて高密度培養を行った。
実施例2:抗BGモノクローナル抗体の精製
抗BGモノクローナル抗体産生ハイブリドーマ2B8培養上清をSpectra/Por(MW cut off : 20000)で透析した後、protein Gカラム(GE Healthcare Bio-Science AB)にて精製した。
実施例3:抗BGモノクローナル抗体(BG2B8)の反応特異性
抗BGモノクローナル抗体(BG2B8)の反応特異性を検討するため、様々な抗原やBGを固相化したELISA plateを用い、抗BGモノクローナル抗体(BG2B8)の希釈系列を作製し、反応性を検討した。BGとしては、Candida細胞壁由来のCSBG(β-1,6-グルカンと1,3-グルカンを構成成分とするBG)、Agaricus brasiliensis由来AgHWE(β-1,6-グルカンを主成分とするBG)、Aspergillus細胞壁由来ASBG (β-1,3-グルカンを主成分とするBG)、Grifola frondosa由来GRN(1分岐1,3-グルカン)、Yeast-mannanを使用した。
さらに、高次構造に対する反応特異性を検討するためにCSBG-OH、CSBG-autoclave、SPG、SPG-OH、SPG-autoclaveによる競合ELISA法を検討した。抗BGモノクローナル抗体(BG2B8)はCSBG添加で抑制され、いずれのSPGにおいても抑制されなかった。
実施例4:抗BGモノクローナル抗体(BG2B8)の真菌細胞への結合
抗BGモノクローナル抗体(BG2B8)が病原性真菌Candida albicans (CA)細胞壁に結合するか否かを検討するため、蛍光標識抗体を用い共焦点顕微鏡により観察した。
実施例5:抗BGモノクローナル抗体(BG2B8)のカンジダ細胞壁BG刺激PEC過酸化酸素産生への影響
過酸化酸素は抗菌活性に関与する重要なメディエーターの一つである。カンジダ細胞壁由来BG(不溶性:OX-CA、可溶性:CSBG)のPEC過酸化酸素産生における抗BGモノクローナル抗体の影響を検討した。
実施例6:抗BGモノクローナル抗体(BG2B8)の血中クリアランス
ICRマウスに抗BGモノクローナル抗体(2B8)(100μg/マウス)を腹腔内投与および静脈内投与し、投与後1、2、4、8、24、48、72時間に尾静脈より採血した。得られたサンプルの抗体価をCSBG固相化ELISA法にて測定した。
実施例7:抗BGモノクローナル抗体(BG2B8)のCSBG血管透過性亢進に与える影響
ICRマウスにCSBG(100μg/mouse)、抗BGモノクローナル抗体2B8(100μg/mouse)、エバンスブルー(色素)(2mg/mouse)を静脈内に同時投与し、投与後1時間のエバンスブルーの腹腔内への浸出を吸光度を測定することで検討した。

- Candida細胞壁から精製したβ-グルカン(CSBG)で免疫したマウス脾臓に由来するハイブリドーマBG2B8(FERM AP-21285)から産生され、β-1,6-グルカンに高い反応性を有し、β-グルカンによる血管透過性を亢進させるとともに、不溶性β-グルカンによるマクロファージの過酸化水素産生を上昇させることを特徴とする抗β-1,6-グルカン・モノクローナル抗体。
- 請求項1記載の抗β-1,6-グルカン・モノクローナル抗体を産生するハイブリドーマBG2B8(FERM AP-21285)。
- 請求項1記載の抗β-1,6-グルカン・モノクローナル抗体を含むβ-1,6-グルカンの測定試薬。
- 請求項1記載の抗β-1,6-グルカン・モノクローナル抗体を有効成分とするβ-1,6-グルカン関連疾患の治療薬。
