明 細 書
免疫系活性化剤 技術分野
本発明は、 免疫系活性化剤に関し、 更に詳しくは、 二次的に糖尿病質者あるい は癌患者などの体調を調整し、 症状の進行を抑制する免疫系活性化剤に関する。 背景技術
癌患者あるいは I I型糖尿病質などの生活習慣病を患つている者は、 抗癌剤あ るいは糖尿病治療薬などの投薬と共に、 健康食品の管理などを併用して体調を調 整し、 症状の緩和を行うのが通常である。 投薬が激しい副作用を併発して危険な 場合、 あるいは投薬が既に効果を失っている場合などには、 投薬などは行わずに 食品管理だけを行う場合もある。
癌患者や糖尿病質などの生活習慣病患者などに対する飲食物管理で効果がある と推奨される健康食品としては、 たとえば、 緑茶、 ァガリクス茸、 マイタケ、 見 布、 キャベツ、 カイアポィモなどが挙げられる。 これらは、 民間伝承的にその有 効性が経験上知られてきた。
民間伝承的に経験上知られてきた健康食品の場合は、 科学的定量的な根拠が乏 しい。 優れた効果が認められるものもあるが、 中には実際はほとんど効果が認め られないものも混在している。
また、 非常に優れた効果が認められた食品であっても、 個々の患者のケースで は、 期待した効果が十分発揮されない場合もある。 この理由として、 薬効成分が 不明であるために、 体調に応じた摂取量あるいは摂取頻度の合理的な管理が困難 で、 患者一人一人の長年の勘と経験に基づかざるを得な 、のが現状であることが 挙げられる。
各種遊離ァミノ酸を所定の割合で配合したグルタミン産生剤により免疫能改善 に言及したもの (特開平 7— 2 6 7 8 5 5号公報) もあるが、 これはタンパク質 合成を介した一般的栄養改善に伴う機能改善を予測したものであって、 具体的な 効果を証してはいない。 またこのグルタミン産生剤は少なくとも 1 0種類の遊離
了ミノ酸を含むものであり、 特定の遊離ァミノ酸 1種類のみによる効果に言及し てはいない。
そこで本発明の目的は、 癌あるいは糖尿病などに代表される生活習慣病の患者 などの体調を調整し、 症状の進行を抑制することができ、 し力、も、 投薬と共にあ るいは投薬治療とは別に行う飲食物管理を合理的かつ適切に行いうる安全度の高 、免疫系活性ィ匕剤を提供することにある。 発明の開示
本発明者は、 茶、 ァガリ クス茸、 マイタケ、 昆布、 AHC C (活性化へミセ 口一ス関連ィ匕合物く A c t i v e H e x o s e C o r r e l a t e d C omp o u n d ) 、 カイアポィモなど、 生活習慣病に効能があると言われて ヽる健康食品には L一フヱニルァラニンが共通に遊離状態で含まれていることを 独自に見い出した (第 1図および参考実験例参照) 。 そこで、 マウスによる実験 や生活習慣病を患つている者に対し、 L—フヱニルァラニンを水溶液形態で投与 したところ、 体調改善に優れた効果のあることが確認され、 本発明を完成するに 至った。
即ち、 本発明は、 L—フヱニルァラニン又はその生理学的に許容しうる塩若し くはエステルを有効成分とする免疫系活性化剤である。 本発明の免疫系活性化剤 は、 癌抑制型免疫系活性化剤及び糖尿病抑制型免疫系活性化剤として、 好適に用 いることができる。
L—フヱニルァラニンはヒ 卜の必須ァミノ酸の一つであり、 動物タンパク質構 成アミノ酸として肉類などを通じ、 日常的に大量に食用に供されている。 当然、 人体にも含まれている。 したがって、 安全性も極めて高いことが認められる。 図面の簡単な説明

以下、 本発明の実施の形態を説明する。
本発明の免疫系活性化剤では、 通常、 下記式、
で表わされる遊離形態の L一フヱニルァラニンが用いられるが、 特に遊離形態で ある必要はなく、 薬理学的に許容される塩、 例えば、 ナトリウム塩、 カリウム塩 等、 塩酸塩、 硫酸塩等の鉱酸塩、 酢酸塩、 乳酸塩、 リンゴ酸塩等の有機酸塩等の 形態で、 又は生体内で加水分解されて遊離アミノ酸に変換されるエステルの形態 で用いることもできる。
健康食品中に L—フヱニルァラニンが共通に含まれることは以下のような方法 で確認することができる。 すなわち、 L一フエ二ルァラニンを含有する抽出源を 食塩水中で粉砕する。 抽出源としては、 例えば、 マイタケ、 緑茶、 A H C C、 ァ ガリクス茸などが挙げられる。 粉砕する際に使用する食塩水は、 0 . 3モル/ 1 0— 3 m 3程度の濃度が好ましい。 粉砕にあたっては、 例えば、 家庭用ミキ サ一などで約 5分間程度撹拌するとよい。 次いで、 ろ過で固形成分を除去し、 続 いて、 ロータリ一エバポレーターで吸引減圧し、 成分を濃縮する。 これで、 L— フエ二ルァラニンを含む抽出液を得ることができる。
得られた抽出液を逆相クロマトグラフで分離精製する。 逆相クロマトグラフに よる精製は次のようにして行う。 固定相としては、 例えば、 S o u r c e R P C (フアルマシア社販売) を用い、 移動相としては、 例えば、 水ーァセトニ
卜リル混合液を用いる。
常温又は室温下で 3 0分程度かけ、 降下によって分画する。 分画成分中、 ァセ トニトリル 0 . 5 %付近で溶出する。 濃縮後、 再クロマトグラフィーによって精 製する。 実際に、 ァガリクス茸について得られた分画チャートを第 1図に示す。 本発明においては、 このような方法で得られる天然 L一フヱニルァラニンの 他、 L—フヱニルァラニンという商品名で、 東京化成 (株) 、 和光純薬 (株) 、 その他の製薬会社などから販売されている化学合成による市販品を好適に用いる ことができる。
本発明の L一フエ二ルァラ二ン含有免疫系活性ィ匕剤は、 適当量摂取することに より、 例えば、 癌患者、 糖尿病質者などはその進行を抑制する方向に体調が改善 化され、 病気に対する抵抗力も強ィ匕される。
本発明の免疫系活性ィ匕剤の投与形態は、 特に制限されるものではないが、 経口 投与に適した液剤又は用時に溶解して使用する粉末製剤とすることができる。 あ るいは、 粉末製剤を食品添加物として使用してもよい。 これら製剤の調製は常法 に従い、 適当な薬理担体及び希釈剤を用いることにより行うことができる。 担体 及び希釈剤としては、 製剤分野で慣用されるものを 、ずれも使用することができ る。 かかる希釈剤として飲用水を使用して経口投与にて摂取することは、 調製及 び投与の容易さ並びに摂取管理の観点から好ましい。
本発明の免疫系活性化剤の投与量は、 これを投与される患者の病理状態、 栄養 状態、 年齢、 体重等に応じて適宜に増減させ、 特に規定されるものではない。 例 えば、 患者の体力回復剤、 体調改善薬を目的とする場合、 水 1 0 0 X 1 0 _ 0 m 3中に 0 . 0 5 ~ 1 . 0グラム、 特には 0. 1〜0 . 5グラムの遊離 L—フ ヱ二ルァラニンの粉末を溶解した水溶液を一回の摂取量とし、 一日 1〜6回、 特 には 2〜 5回程度摂取するようにし、 症状ある t、は体調により 1週間〜数ケ月間 連続的に摂取する。 これにより、 一般に病気の進行が減速し、 あるいは体力の回 復が図られる。 以下、 本発明を実施例に基づき説明する。
く実験例 1 :糖尿病動物 >
遺伝性糖尿病モデルマウス (C 57 BL/Ks J— dbZdb J c l) 、 6 匹 (ォス 3、 メス 3) に L一フヱニルァラニンを経口投与した。
投与方法は、 遊離 L—フ 二ルァラニン (和光純薬 (株) 製) の粉末を飲用水 に下記の第 1表に示す濃度で溶解し、 この水溶液を檻の中のマウスが適宜自由に 飲用できる状態にした。 当初、 0. 1 wZv%の濃度で 2001年 6月 8日から 投与を開始したが、 同年 6月 25日からは投与濃度を 0. 5 w/v%に上げた。 検査個体に現れた血糖値を各検査日に検査した。
平均値とその上下幅を統計値とし、 統計値を併せて第 1表及び第 2表に示す。 第 1表にォス、 第 2表にメスを表示する。

また、 L—フエ二ルァラニンを投与しない同一種マウスの個体についてその血 糖値変ィ匕を公開データに基づき同図中に破線で示した。
第 1表、 第 2表及び第 2図、 第 3図から、 L一フヱニルァラニンは、 個体差は
あるものの濃度 0. 1 w_/v%でも血糖値抑制効果が認められ、 0. 5 w/ V % では一層明確に血糖値抑制効果が認められた。
このことから、 約 0. 3 w/ V %程度の濃度値以上から血糖値の抑制に際だつ た効果を期待できることが分かつた。
<実験例 2 :皮膚癌動物 >
実験用マウス (S q l 6) 3匹それぞれに皮膚癌を皮下接種し、 生存日数を調 ベた。 生存期間中、 0. 4w/v%濃度で L一フヱニルァラニン (和光純薬 (株) 製) の粉末を水に溶解した飲用水を、 適宜各マウスが飲用できる状態に置 いた。
死亡後、 内臓各部の重量を測定した。 接種前の体重を 100%とし、 増減を併 せて示す。
結果を第 3表に示す。

飲用水にし—フヱニルァラニンを溶解しなかつた外は上記実験例 2と同様の実 験を行った。 結果を第 4表に示す。

本発明にしたがい、 L—フヱニルァラニンを投与すると、 対照に比べて寿命が 1. 3日 (= 1 8. 3日一 17日) 延び、 体全体で重量が平均約 1. 48倍に成 長が持続していることが分かつた。 この癌は肺に転移する特徴があり、 肺水腫状 態を引き起こし二次的原因で死期が早まる。 しかし、 投与群では、 特に免疫機能 に関係があるリ ンパ節が Lーフヱ二ルァラ二ン投与群が対照に比べて 2. 5〜 4. 6倍以上と顕著に重量を保持していることが分かった。
すなわち、 L一フヱニルァラニンは、 癌性のマウスの免疫系機能を活性ィ匕して いることが分かった。
<実験例 3 :癌細胞移植動物 >
実験用マウス (BALBZcA J c l— n u) 2匹それぞれに肉腫癌細胞 (サルコ—マ 180) を移植し、 生存日数を調べた。 移植後 9日経過後の皮膚の 膨らみが顕著になった時点で、 0. 6 w/v%濃度で L一フヱニルァラニン (和 光純薬 (株) 製) の水溶液を飲用水として与えた。
2匹の実験用マウスの生存日数は、 それぞれ 28日と 29日であり、 平均で 28. 5日であった。
く実験例 3の対照実験 >.
対照群マウスは 3匹とし、 飲用水に L—フヱニルァラニンを溶解しなかつた外 は上記実験例 3と同様の実験を行った。 3匹の対照群マウスの生存日数は、 それ ぞれ 8日、 10日および 1 1日であり、 平均で 9. 7日であった。
本発明にしたがい、 L—フエ二ルァラニン (和光純薬 (株) 製) を投与する と、 寿命が 1. 93倍 (= (28. 5-9. 7) ÷9. 7日) 延びることが分か つた (第 4図参照) 。
実験動物では L一フヱニルァラニンを経口投与開始から約 20日経過時点から 腫瘍が黒色に変化し始め、 縮小し、 最終的に空洞化した。 く臨床例 1 >
抗癌剤の投与を停止している肺癌女性 (88歳 末期) に L一フヱニルァラ二 ン水溶液を経口投与した。
濃度 2wZv%で、 1日に 200 X 10— 6m3づっ 2回経口投与し続け た。 10日経過後、 及び 30 S経過後の本人の所見を下記の第 6表に示す。 第 6表
過去に癌発症と治療を二度ほど経験していて現在は抗癌剤を停止している女性 (50代) に L—フヱニルァラニン水溶液を経口投与した。
濃度 2w/v%で、 1日に 20 O x 10一 0 m3づっ 2回経口投与し繞 け、 1ヶ月間様子を見た。
30日経過後、 症状の悪化が見られず。 症状は小康状態を継続していると判断 できた。
<臨床例 3〉
6月に肺ガンを検認、 その後脳に転移が認められた癌患者に、 本人の申し出に より L一フエ二ルァラニンを投与した。 L—フエ二ルァラニンの投与は、 8月 9 日に濃度 0. 25 w/v% (希釈液) で開始した。 8月 1 5日から濃度を 0. 5w/v% (表中、 「原液」 と称する) に上げた。
なお、 この患者は、 7月以降、 医師から抗癌剤を処方されて一時体調を崩し、 8月 9日からは以下のような処方を医師から受けてその指示にも従っていた。
8月 9日から デパケン 200 1 TX 3回
リンデロン 0. 5 2 TX 3回
アシノンカプセル 75 1カプセル X 2回
8月 21日から メイアク ト l O Omg 1 TX 3 (5日分) 追加 8月 20日には 7ナイフ手術を受けた。
患者付添人による病体経過記録を第 7表に示す

L一フヱニルァラニンを対照試料とし、 この対照試料とともに、 カイアポィ
モ ·緑茶 'ァガリクス茸 · AHC C ·尿 ·グレープフルーツについて、 これらの 抽出液をペーパークロマトグラフィ一で比較分析した。
対照試料は 2種類用意した。 一つは、 濃度 lmgZ10_ om3の L—フヱニ ルァラニン水溶液とした。 もう一つは、 L一フヱニルァラニンとチロシンの混合 水溶液で、 両者とも濃度を lmgZl 0一6 m3とした。
結果のクロマトグラムを第 5図に写真図で示し、 第 5図の模式図による説明を 第 6図に示す。 第 6図中、 手書きの楕円形表示部分は、 L一フヱニルァラニンと 同一の移動度の成分を示す。
いずれの食品等にも L—フヱニルァラニンと同一移動度の共通成分の存在が追 認できた。 産業上の利用可能性
本発明の免疫系活性ィ匕剤は、 免疫機能の昂進を介して癌あるいは糖尿病などに 代表される生活習慣病の患者などの体調を調整し、 症状の進行を抑制することに 優れた効果を奏する。 また、 癌あるいは糖尿病などに代表される生活習慣病で投 薬と共に、 あるいは投薬治療とは別に行う飲食物管理を定量ィ匕し、 合理的に行う ことができる。
