新規な多糖誘導体及び分離剤
〔産業上の利用分野〕
本発明は、 例えば光学分割を行う機能材料として極めて有用な、 新規な多糖誘 導体及びその多糖誘導体からなる分雜剤に関する。
〔従来の技術〕
多糖誘導体を充塡剤に用いたカラムで、 液体クロマトグラフィーにより種々の ラセミ体が光学分割されることは既に知られている。 例えば、 Journal of Li qui d Chromatography, _9_(2 & 3), 313-340(1986)、 特開昭 60— 142930号又はそれに対応 する EP-A 147804 及び US-A 4818394、 特開昭 63 - 178101号又はそれに対応する EP -A 238044 に開示されている。
本発明は、 新規な多糖誘導体を用いた、 より分割能力に優れた分離剤を提供し ようとするものである。
〔発明の開示〕
本発明者らは上記課題を解決すベく鋭意研究の結果、 本発明を完成するに到つ た。
即ち本発明は、 多糖の有する水酸基又はァミノ基の一部又は全てを二種以上の 異なる置換基で置換してなる新規な多糖誘導体、 及び該多糖誘導体を含む分離剤 又は分離装置、 及びこの分離剤又は分離装置を用いてラセミ体を各光学異性体に 分離する方法を提供するものである。
以下、 本発明について詳細に説明する。
ぐ置換基 >
本発明において、 多糖の有する水酸基又はァミノ基を置換する置換基としては 、 下記に示すものが挙げられる。

上記のような置換基の中では芳香族基を有するもの、 特に 2位及びノ又は 3位 に電子供与性基を有する芳香族基を有するもの、 特に電子供与性基を有する芳香 族基を有するものと電子吸引性基を有する芳香族基を有するものとの組み合わせ が好ましい。 電子供与性基としては、 アルキル基等が挙げられる。 更に好ましい 置換基は、 下式 ( I ) で示される 3, 5 —ジメチルフヱ二ルカルバメ一ト基及び下 式 (I I) で示される 3, 5 -ジクロ口フエ二ルカルバメート基の組み合わせ である。

これら置換基の割合は 3,5 —ジメチルフヱ二ルカルバメート基 ( I ) 3,5 — ジクロロフヱ二ルカルバメート基 (II) の重量比で 9ノ 1〜 1ノ9の割合が好ま しい。
本発明の多糖誘導体は少なく とも 2個の異なる置換基を有し、 特に 2位及び Z 又は 3位の置換基と 6位の置換基が異なるものが好ましい。 本発明の多糖誘導体 の上記置換基による置換度は少なく とも 40%以上、 好ましくは 50%以上、 更に好 ましくは 85%以上である。
<多 糖 >
本発明における多糖とは、 合成多糖、 天然多糖及び天然物変成多糖のいずれか を問わず、 光学活性であればいかなるものでも良いが、 好ましくは結合様式の規 則性の高いものである。 具体的には、 α— 1,4 ーグルカン (アミロース、 アミ口 ぺクチン) 、 一1,6 —グルカン (デキス トラン) 、 ー 1,4 ーグルカン (セル ロース) 、 ;3— 1,6 一ゲルカン (ブスッラン) 、 /3— 1,3 一ゲルカン (例えば、 カー ドラン、 シゾフィ ラン等) 、 一 1,3—グルカン、 /3—1,2—グルカン(Crown Gall多糖) 、 3— 1,4 —ガラクタン、 ;3— 1,4 一マンナン、 な 一 1,6—マンナン、 3 -1,2 一フラクタン (ィヌ リ ン) 、 ;9一 2,6 —フラクタン (レノくン) 、 β - 1, 4 -キシラン、 ;6— 1,3 —キシラン、 β - 1,4 —キトサン、 ^一 1,4 -Ν ーァセ チルキトサン (キチン) 、 プルラン、 ァガロース、 アルギン酸等が挙げられ、 ァ ミロースを含有する澱粉なども含まれる。 特に好ましいものは高純度の多糖を容 易に得ることのできるアミロース、 セルロース、 3— 1,4 一キトサン、 キチン、 yS -1,4 一マンナン、 ー 1,4 ーキシラン、 ィヌ リ ン、 カー ドラン等である。
これら多糖の数平均重合度 ( 1分子中に含まれるビラノース或いはフラノース 環の平均数) は 5以上、 好ましくは 10以上であり、 上限は 2000、 好ましくは 500 以下であることが取り扱いの容易さにおいて好ましい。
ぐ合成法 >
本発明の多糖誘導体の水酸基又はァミノ基を上記のような置換基で置換する方 法としては以下の方法が挙げられる。
(1) 水酸基の置換方法
Α) エステル置換
本発明に係る多糖の水酸基をエステル置換する方法は、 対応するカルボン 酸 0 2G00H)を塩化チォニル、 ォキサリルクロリ ドなどを用いて酸クロリ ドと した後、 この試薬をピリジン溶媒中で、 対応する多糖と前記酸クロリ ドとを 反応させることにより、 容易に置換することができる。
B) 力ルバメート置換
本発明に係る多糖の水酸基をカルバメート置換する方法は、 通常のアルコ ールとイソシァネートからウレタンを生ずる反応をそのまま適用できる。 例 えば、 適当な溶媒中で三級アミ ン等のルイス塩基、 又は錫化合物等のルイス 酸を触媒として、 対応するイソシァネート(R3NC0) と多糖とを反応させるこ とにより得ることができる。 また、 イソシァネートの合成は、 例えば、 対応 するァニリン誘導体のァミノ基にホスゲンを作用させることにより、 容易に 置換することができる。
C) エーテル置換
本発明に係る多糖の水酸基をエーテル置換する方法は、 対応するハロゲン 化物(R (X はハロゲンを示す))と相当する多糖とを、 例えばジォキサン又 はピリジン溶媒中で、 塩基として水酸化力リゥム、 カリウム t一ブトキシド を用いて反応させることにより容易に置換することができる。
(2) ァミノ基の置換方法
D) ビニリデン置換
本発明に係わる多糖のァミノ基をビニリデン置換する方法は、 通常のァミ ンとカルボニル化合物からシッフ塩基を生ずる反応をそのまま適用できる。 E) ァ ミ ド置換
本発明に係わる多糖のァミノ基をァミ ド置換する方法は、 通常のァミ ンと 酸クロリ ドからアミ ドを生ずる反応をそのまま適用できる。
F) 尿素置換
本発明に係わる多糖のアミノ基を尿素置換する方法は、 通常のアミ ンとィ ソシァネートから尿素を生ずる反応をそのまま適用できる。
本発明において、 多糖の有する水酸基又はァミノ基を 2種以上の異なる置換基 で置換するには、 上記のような反応剤の中の異なる 2種以上の反応剤を同時に使 用して反応させたり、 あるいは逐次的に反応させて特定の水酸基又はァミノ基に 特定の置換基を導入することもできる。
本発明の多糖誘導体の中で、 前記式 ( I ) で示される 3, 5 —ジメチルフヱニル カルバメート基及び前記式 (I I) で示される 3, 5 —ジクロ口フユ二ルカルバメー ト基で置換された多糖誘導体は、 3, 5 —ジメチルフヱ二ルイソシァネート及び 3, 5 ージクロ口フエ二ルイソシァネー トと多糖とを反応させることにより得ること ができる。 この反応は通常のアルコールとイソシァネートからウレタンを生ずる 反応をそのまま適用できる。 例えば、 適当な溶媒中で三鈒ァミ ン等のルイス塩基 、 又は錫化合物等のルイス酸を触媒として、 対応するイソシァネートと多糖を反 応させることにより得ることができる。
ぐ分離剤〉
本発明の多糖誘導体は、 機能材料として極めて有用な物質であり、 とくに光学 分割用充塡剤、 即ち分離剤として有用なものである。
本発明の多糖誘導体を分離剤として、 化合物の混合物や光学異性体混合物を分 離する目的に使用するには、 本発明の多糖誘導体を充塡したガスクロマトグラフ ィー、 液体クロマトグラフィー、 薄層クロマ トグラフィーなどのクロマ トグラフ ィ一法を用いるのが一般的であるが、 この他、 本発明の多糖誘導体を含む膜を成 形し、 これで膜分雜を行うこともできる。
本発明の多糖誘導体を分離剤として液体クロマトグラフィ一法に応用するには、 その粉体としてカラムに充塡する方法が簡便である。 本発明の多糖誘導体を粉砕 するかビーズ状にすることが好ましく、 粒子は多孔質であることがより好ましい。 更に分離剤の耐圧能力の向上、 溶媒置換による膨潤、 収縮の防止、 理論段数の向 上のために多糖誘導体を担体に担持させることも好ましい。
粉体として用いる場合の粒子の大きさおよび担体の大きさは使用するカラムの 大きさによって異なるが、 1 !〜 1画であり、 好ましくは 1 〃m〜300 m で ある。 担体は多孔質であることが好ましく、 その平均孔径は 10A〜 100 / m であ り、 好ましくは、 50A〜 50000Αである。 担体に担持させる多糖誘導体の量は担 体に対して 1〜100 重量%、 好ましくは 5〜50重量%である。
多糖誘導体を担体に担持させる方法は化学的方法でも物理的方法でもよい。 物 理的方法としては、 多糖誘導体を可溶性の溶剤に溶解させ、 担体と良く混合し、 減圧または加温下、 気流により溶剤を留去させる方法や、 多糖誘導体を可溶性の 溶剤に溶解させ、 担体と良く混合した後、 多糖誘導体に対し不溶性の溶剤に分雜 させることによつて可溶性溶剤を拡散させてしまう方法もある。 この様にして得 られた分雜剤は、 加熱、 溶媒の添加、 洗浄などの適当な処理を行うことによって、 その分雜能を改善することも可能である。
用いる担体としては多孔質有機担体または多孔質無機担体があり、 好ましくは 多孔質無機担体である。 多孔質有機担体として適当なものは、 ポリスチレン、 ポ リアクリルアミ ド、 ポリアクリレート等からなる高分子物質が挙げられる。 多孔 質無機担体として適当なものは、 シリカ、 アルミナ、 マグネシア、 ガラス、 カオ リン、 酸化チタン、 ゲイ酸塩などであり、 これらの表面に、 多糖誘導体との親和 性を良く したり、 担体自身の表面の特性を改質するために処理を施したものを用 いても良い。 表面処理の方法としては有機シラン化合物によるシラン化処理ゃプ ラズマ重合による表面処理方法等がある。
液体クロマトグラフィーあるいは薄層クロマトグラフィーを行う場合の展開溶 媒としては多糖誘導体を溶解またはこれと反応するものを除いて特に制約はない。 多糖誘導体を化学的方法で担体に結合したり、 架橋により不溶化した場合にはこ れと反応するものを除いて特に制約はない。
—方、 薄層クロマトグラフィ一を行う場合には、 0. 1 u rn〜0. 1 mm程度の粒子 からなる該分雜剤と、 必要であれば少量の結合剤より成る厚さ 0. 1 〃π! 〜 100瞧 の層を支持板上に形成すれば良い。 又、 膜分離を行う場合には中空糸あるいはフィルムとして用いる。
〔実施例〕
以下、 実施例により本発明を更に詳細に説明するが、 本発明はこれらの実施例 に限定されるものではない。
実施例 1
ピリジン (単蒸留、 K0H 乾燥) 35mlにセルロース(Avicel, Merc! (社製) 0.722 g, 3,5 ージメチルフエ二ルイソシァネート 0.988 g (6.71nmiol, 50%当量) 、 3,5 —ジクロ口フエ二ルイソシァネート 1.243 g (6.68瞧 ol, 50%当量) を加え、 約 100 °Cで 15時間反応させた。 この反応溶液にさらに 3, 5 —ジメチルフヱニルイソ シァネート 0.485g (3.30,01) 、 3,5 —ジクロ口フエ二ルイソシァネ一 ト 0.613 g (3.30mmol) を加えた。 110 でで 10時間攪拌した後、 反応物はメタノール中に 沈澱させ、 得られたセルロース誘導体をグラスフィルタ一に集めた。 メタノール でよく洗浄してから 60でで 2時間真空乾燥した。 収量 2.761 g、 収率 93.2%。 こ れをセルロース誘導体(1) という。
実施例 2
ピリジン (単蒸留、 K0H 乾燥) 35mlにセルロース(Avicel, Merck社製) 1.01 g、 3, 5 —ジメチルフエ二ルイソシァネート 1.83 g (12.45mmol, 67 %当量) 、 3, 5 —ジクロ口フエ二ルイソシァネート 1.16g(6.24隱 ol, 33%当量) を加え、 約 100 てで 15時間反応させた。 この反応溶液にさらにピリジン 15ml、 3,5 —ジメチルフ ェニルイソシァネート 0.54g (3.73mniol) 、 3,5 ージクロ口フエ二ルイソシァネ —ト 0.34g (1.83隱 ol) を加えた。 100 でで 24時間攪拌した後、 反応物はメタノ ール中に沈澱させ、 得られたセルロース誘導体をグラスフィルタ一に集めた。 メ 夕ノールでよく洗浄してから 60eCで 2時間真空乾燥した。 収量 3.520g、 収率 100 96。 これをセルロース誘導体(2) という。
実施例 3
トリチルセルロース (トリチル基置換率 43%) 1.68gをピリジン 20mlに溶かし た。 これに 3, 5 —ジメチルフヱ二ルイソシァネート 1.75g (残 OH基に対して過剰 量) を加えた。 約 100 でで 9時間反応させた。 反応物はメタノール中に沈澱させ、 グラスフィルターに集めメタノールでよく洗浄した。 次にこのセルロース誘導体 1.45gをメタノール 40mlに懸濁した。 この混合物に 濃塩酸 4滴を加え、 室温で 20時間攪拌した。 トリチル基を除去したセルロース誘 導体はグラスフィルターに集め、 メタノールで洗浄してから、 60°Cで 2時間真空 乾燥した。 収量 0.770 g、 収率 92.3%。
次にピリジン 15mlに上記で得られたセルロース誘導体 0.77 g、 3,5 ージクロ口 フエ二ルイソシァネート 1.10g (5.93mmoK 過剰量) を溶かした。 約 100 てで 15 時間反応させてから、 反応物をメタノール中に沈鍛させ、 遠沈によりポリマーを 集めてメタノ一ルで洗浄を繰り返した。 ポリマーはデシケーター内で約 3時間乾 燥してから、 60でで 2時間真空乾燥をした。 収量 1.045 g、 収率 86.3%。
得られたセルロース誘導体は 6位に 3,5 —ジクロ口フエ二ルカルバメ一ト基、 2,3 位に 3, 5 —ジメチルフエ二ルカルバメ一ト基を選択的に修飾したセルロース 誘導体であった。 これをセルロース誘導体(3) という。
実施例 1〜3で得られたセル π—ス誘導体(1) 〜(3) の元素分析値を表 1に示 す。 また参考品としてセルローストリス(3, 5—ジクロロフヱ二ルカルバメート) (参考品 1 ) 及びセルローストリス(3,5—ジメチルフヱ二ルカルバメー ト)(参考 品 2) の元素分析値も表 1に示す。

実施例 1〜3で得られたセルロース誘導体(1) 〜(3) 及び参考品 1, 2 0.75 g をテトラヒ ドロフラン 15mlにそれぞれ溶かした。 これらの溶液を 3—アミノブ口 ピル ト リエトキシシラン処理したシリカゲル(Merck社製: SI4000) 3.0 gに担持 して各種充填剤を得た。
これらの充填剤を長さ 25cm、 内径 0.46cmのカラムにへキサン一流動パラフィ ン ( 2 : 1 ) を用いて充填した。
これらのカラムを用いて表 2及び表 3に示す種々のラセミ体化合物を分離した 。 その結果を表 2及び表 3に示す。
尚、 表中の容量比( )、 分離係数 ( ) は、 それぞれ下式により定義される。

Claims

請 求 の 範 囲
1. 多糖の有する水酸基又はァミノ基の一部又は全てを二種以上の異なる置換基 で置換してなる新規な多糖誘導体。
2. 2位及びノ又は 3位の置換基と 6位の置換基が異なる請求項 1記載の多糖誘 導体。
3. 芳香族基を有する置換基を持つ請求項 1記載の多糖誘導体。
4. 電子供与性基を有する芳香族基を有する置換基を持つ請求項 1記載の多糖誘 導体。
5. 2位及び/又は 3位に電子供与性基を有する芳香族基を有する置換基を持つ 請求項 1記載の多糖誘導体。
6. 電子供与性基を有する芳香族基を有する置換基、 及び罨子吸引性基を有する 芳香族基を有する置換基を持つ請求項 1記載の多糖誘導体。
7. 電子供与性基がアルキル基である請求項 5記載の多糖誘導体。
8. 置換基として、 下式 ( I ) で示される 3. 5 —ジメチルフヱ二ルカルバメート 基及び下式 (I I ) で示される 3, 5 —ジクロロフヱ二ルカルバメ一ト基を持つ請 求項 1記載の多糖誘導体。
