Abstract
本發明之松露固態發酵數週製程可取代傳統四至七年栽培的松露子實體。松露是塊菌屬,為地下生長的食用真菌,今利用穀物或豆類如:小麥、燕麥、糙米、薏仁和大豆,或混合基質作為松露塊菌固態發酵之基質,並控制水分含量為30~60%,將此基質裝於太空包或其他塑膠、玻璃容器中,先進行殺菌,冷卻後即可注入1~10%已預活化的松露塊菌液或埋入1~10%新鮮的松露菌絲,在溫度控制在25~30℃下進行培養數週後會長滿菌絲,以提升松露固態發酵產物的有效成分和增加抗氧化活性。此松露固態發酵產物經射頻加熱、殺菌釜殺菌或超高壓殺菌後,再進行射頻熱風乾燥、磨粉。此松露固態發酵產物可直接製備保健食品的膠囊、飼料添加物及食品如:穀液、沖泡穀粉、麵條、餅乾、醬料和調味料等原料或松露固態發酵產物可進一步萃取、純化生物活性成分,如:多醣及三萜等。

本專利技術乃以不同穀物包含小麥、燕麥、糙米、薏仁和大豆或混合穀物作為固態基質,然後加入預活化培養的松露塊菌液,再進行固態發酵數週,並以射頻加熱方式殺菌及乾燥,此松露固態發酵產物具有抗氧化成分和抗氧化活性,可進一步萃取、純化生化活性成分作為保健食品膠囊,或直接作為飼料及添加於穀液、沖泡穀粉、麵條、餅乾、松露醬料和松露調味料等食品中。
松露為一種地底下真菌,屬於塊菌屬,具有獨特風味及迷人香氣,和與魚子醬和鵝肝醬並列為世界著名的三大名菜。松露由於產地不同,也有不同的品種,如:義大利的紳士塊菌(白松露)(Tuber magnatum)、法國的黑孢塊菌(黑松露)(Tuber melanosporum)、地中海地區的瘤孢塊菌(Terfeziaboudieri)又稱為沙漠松露;在中國著名的松露品種為夏塊菌(Tuber aestivum)、印度塊菌(Tuber indicum)、中國塊菌(Tuber sinense)(胡,2010)。
松露富含多醣、三萜、多酚、類黃酮、γ-胺基丁酸、牛磺酸、神經醯胺、α-雄烷醇、植物固醇、多元不飽和脂肪酸等生理活性代謝物,並且具有抗氧化活性、降血糖、抗發炎、抗菌活性、抗腫瘤活性、免疫調節和舒緩神經等作用。
過去松露培養以植栽為主,它可以和松科或殼斗科的樹木根部形成外生菌,由地下挖出松露的子實體,但這需要花四至七年的時間,產量稀少且價格非常昂貴。近年來有學者研究松露的液態發酵,探討葡萄糖、麥芽糖、蔗糖、乳糖等不同碳源(Tanget al.,2008a)和酵母萃取物、蛋白棟、牛肉萃取物、尿素、硝酸鉀、硝酸鈉、硝酸銨、氯化銨、硫酸銨不同氮源(Liu et al.,2008)和不同金屬離子(Ca2+、Na+、K+、Mg2+)(Tanget al.,2008b)對松露細胞生長影響,則一週內即可取得液態發酵菌絲體和多醣代謝產物,且其活性成分含量和生理功能並不低於子實體。
生物體在正常代謝過程中會產生自由基與活性氧,導致氧化傷害,當自由基不正常地累積時,就會使身體機能受到傷害而導致疾病產生。將不同的5%穀物作為松露液態發酵培養液,在溫度25℃下進行搖瓶培養一週內即可提高獲得多醣和抗氧化物質含量和提升抗氧化活性(徐等,2015)。然而松露液態發酵的後段製程,卻需要花較多的能耗和乾燥時間以完成,故進一步開發以全穀物作為松露固態發酵基質以製造無汙染的松露產物。
射頻是一種電磁波,如同微波加熱屬介電加熱的一種,可克服固態物質的熱傳礙障,由於射頻的穿透深度較微波深且加熱均勻,故射頻熱風可用於松露固態發酵產物的殺菌和乾燥,以縮短後段製程時間(陳,2013),或甚至利用微波萃取有效成分以縮短萃取時間及提高萃取率(陳和陳,2013)。
此發明技術乃以穀類或豆類作為松露固態基質,先進行純種松露塊菌預活化液態發酵或生產新鮮松露塊菌絲塊,再將1~10%松露菌絲
液或菌絲塊植入已殺菌穀類或豆類固態培養基中,溫度控制在25~30℃下進行固態發酵數週,利用不同發酵時間可控制發酵期間的抗氧化或有效成分的含量,再進行射頻熱風殺菌和乾燥、磨粉後,即可得到具有保健功能的松露固態發酵產物直接添加於食品、醬料和調味料,或可以利用微波萃取有效成分及純化生物活性物質。

松露固態發酵產物的製造流程如圖一所示,是利用薏仁、糙米、小麥、燕麥等穀類或以大豆等豆類作固態發酵的基質加水,使基質的水分含量控制在30%~60%,並裝於太空包或塑膠、玻璃容器中,先進行殺菌、冷卻後,再植入1~10%已預活化的松露(塊菌)絲液或新鮮松露(塊菌)絲塊,在25~30℃下進行固態發酵數週可長滿菌絲,可提升抗氧化物質(粗多醣、粗三萜、總多酚和類黃酮)的含量(圖二)和抗氧化活性(清除DPPH能力、螯合亞鐵離子能力和還原力)(圖三),再將該發酵產物經射頻熱風殺菌、乾燥及磨粉以製成松露固態發酵產物,可添加原料應用於穀液、沖泡穀粉、麵條、餅乾等食品、醬料和調味調中或作為進一步萃取、純化松露生物活性的原料作為具有保健功能的食品膠囊。
Claims (7)
- 一種松露固態發酵產物之製造方法,該方法包括:(a)預活化松露菌液,(b)製備穀類或豆類的固態發酵基質,(c)固態發酵培養數週後,使松露菌絲長滿後,(d)再經殺菌、乾燥和磨粉以製成松露固態發酵產物。
- 如請求項1所述的製造方法,其中(a)預活化松露菌液的製備方法,可將長滿於馬鈴薯葡萄糖洋菜培養基(PDA)之各種松靈的塊菌學名如:白松露(Tuber magnatum,紳士塊菌)、黑松露(Tuber melanosporum,黑孢塊菌)、沙漠松露(Terfeziaboudieri,瘤孢塊菌)、(Tuber aestivum,夏塊菌)、(Tuber indicum,印度塊菌)或(Tuber sinense,中國塊菌)取出菌塊,接種於裝有已殺菌冷卻馬鈴薯葡萄糖洋菜培養液(PDB)中,在25~30℃進行搖瓶或攪拌培養3~7天。
- 如請求項1所述的製造方法,其中(b)製備穀類或豆類的固態發酵基質,係以穀類或豆類作為固態基質,包括:薏仁、糙米、小麥、燕麥或黃豆等原料,或前述之組合固態基質,並加水調控固態基質的水分含量在30~60%,裝入太空包或容器中,且開品處需塞棉花或包裏不織布,在121℃殺菌釜中殺菌1小時後、冷卻,在無菌操作台中,植入1~10%前述(a)預活化松露菌液,並和固態基質混合均勻。
- 如請求項1所述的製造方法,其中(c)固態發酵培養數週後,使松露菌絲長滿後,其中固態發酵的培養的條件在25~30℃,進行松露固態發酵2~8週後,此時松露菌絲會長滿於固態基質,即可準備收成。
- 如請求項1所述的製造方法,其中(d)再經殺菌、乾燥和磨粉以製成松露固態發酵產物,殺菌方法可採用121℃殺菌釜殺菌1小時以上,或利用射頻加熱發酵產物,數分鐘即可立即升溫至90℃以上,或以超高壓達500MPa以上殺菌5min以上,以達到松露菌的死滅效果;而後可採的乾燥方法包括即可保藏,或利用磨粉機進行磨粉、過篩而成松露固態發酵 產物粉末。
- 一種松露固態發酵產物,係依請求項1至5中的方法製得,其中的乾燥松露固態發酵穀粉或豆粉可添加於食品、飼料,其中食品包括穀液、沖泡穀粉、麵條、餅乾、醬料或調味料。
- 如請求項6所述松露固態發酵產物可進一步進行萃取、純化、乾燥松露多醣、三萜、具有抗氧化的生物活性物質,以作為保健食品原料;其中萃取可採用下列任一方法,溶劑迴流、超音波、超臨界二氧化碳萃取或微波萃取。
