Abstract
本發明提供一種骨質疏鬆症治療劑,其含有下述式(I)(式中,R1為可具有取代基之C1-6烷基;R2為氟原子、或氯原子;R3為氫原子、氟原子、或氯原子)所表示之醯基硫脲化合物或其鹽作為有效成分。

Description
本發明係關於一種將醯基硫脲化合物或其鹽作為有效成分之骨質疏鬆症之治療劑、及醫藥組合物。
關於骨質疏鬆症,於2000年之美國國立衛生研究所(NIH)之共識會議中,提出將骨質疏鬆症定義為「以骨強度之降低為特徵,且骨折之風險變得容易增大之骨骼疾病:A skeletal disorder characterized by compromised bone strength predisposing to an increased risk of fracture」,實際上為因骨密度之降低與骨質之劣化而導致骨強度降低之疾病(非專利文獻1)。骨質疏鬆症被分類為伴隨年齡增加、閉經或妊娠而使骨密度降低之原發性、及以某些疾病為背景因素之繼發性。
骨密度之降低係由於骨吸收之亢進超過骨形成而產生,但成骨細胞功能及與其相伴之骨形成之降低亦有干預(非專利文獻2)。即,基於骨吸收與骨形成之骨重塑之平衡失衡,骨吸收佔優勢之情況較深地干預病情。
目前,將腰椎與大腿骨頸部合計,推測在日本骨質疏鬆症之患者有1280萬人(2005年),大腿骨頸部骨折產生數量以每年14萬8100人急速增加,而正開發各種治療方法或預防劑。
目前之骨質疏鬆症治療劑主要使用有鈣吸收促進藥、骨形成促進藥、骨吸收抑制藥。然而,該等治療劑一方面顯示有效性,另一方
面亦報告有嚴重之副作用。例如鈣製劑中頻度最高之副作用為胃腸障礙,且於併用活性型維生素D3製劑之情形時,由於有產生高鈣血症之可能性,故而必須定期進行血液檢查(非專利文獻3)。
作為雙磷酸鹽製劑之主要之副作用,報告有胃腸障礙、顎骨壞死、大腿骨之非定形骨折、低鈣血症、腎損傷、及流行性感冒樣症狀等(非專利文獻4)。又,其半衰期較長,有於治療中止後數年間,殘存於骨骼中之可能性。持續長期地過度抑制骨重塑有引起因骨之鈣化度上升而導致之脆弱化或微小損傷之增加等不良作用之可能性。
又,針對閉經後之骨量減少,使用有雌激素製劑、或相較於雌激素製劑而對骨之雌激素受體選擇性地作用之選擇性雌激素調節劑(Selective estrogen receptor modulators;SERM)。然而,對於雌激素製劑而言,除子宮內膜出血、白帶之增加、乳房痛等對女性激素特有之副作用以外,報告有深部靜脈血栓栓塞症、乳癌及子宮內膜癌之發病風險之增加(非專利文獻5),又,SERM雖可抑制對乳房、子宮之副作用,但仍然報告有深部靜脈血栓栓塞症、熱潮紅、下肢痙攣(非專利文獻6)。
因此,近年來一直進行以組織蛋白酶K抑制劑、Src激酶抑制劑、抗硬化蛋白抗體(Anti-Sclerostin antibody)等為新目標之骨質疏鬆症治療劑之臨床開發(非專利文獻7、8、9),但現狀係作為骨質疏鬆症治療藥並不存在兼具優異之有效性與安全性之藥劑。
另一方面,已知如4-[2-氟-4-[[[(2-苯基乙醯基)胺基]硫酮基甲基]胺基]-苯氧基]-7-甲氧基-N-甲基-6-喹啉甲醯胺般於喹啉環之6位具有胺基羰基且於7位具有烷氧基之醯基硫脲化合物為顯示出減輕副作用之強力之c-Met(肝細胞生長因子受體)抑制效果及VEGF(Vascular Endothelial Growth Factor,血管內皮生長因子)受體抑制之抗腫瘤劑(專利文獻1),又,與其他抗腫瘤劑併用時顯示出優異之增強抗腫瘤
效果之作用(專利文獻2)。
然而,全然不知該化合物對骨質疏鬆症有效。

本發明之目的在於提供一種對骨質疏鬆症發揮優異之預防或治療效果之骨質疏鬆症治療劑及醫藥組合物。
本發明者等人為解決上述問題而努力研究,結果發現下述式(I)所表示之醯基硫脲化合物或其鹽藉由對破骨細胞抑制其分化形成而阻礙骨吸收,且對成骨細胞具有異形骨形成抑制作用,進而於骨質疏鬆模型小鼠中顯示骨量恢復效果,故而對骨質疏鬆症之預防或治療有用。
即,本發明提供一種骨質疏鬆症治療劑,其含有下述式(I)所表示之醯基硫脲化合物或其鹽作為有效成分。
又,本發明提供一種醫藥組合物,其係用以治療骨質疏鬆症,且含有下述式(I)所表示之醯基硫脲化合物或其鹽、及藥學上所容許之載體。
又,本發明提供一種骨質疏鬆症之治療方法,其特徵在於:對
患者投予含有下述式(I)所表示之醯基硫脲化合物或其鹽及藥學上所容許之載體之醫藥組合物。
(式中,R1表示可具有取代基之C1-6烷基,該取代基表示羥基、C3-10環烷基、可具有取代基之C1-6烷氧基、可具有取代基之C1-6烷基胺基、C1-6烷醯基胺基、C1-6烷基磺醯基、可具有取代基之C6-14芳香族烴基、可具有取代基之飽和或不飽和雜環基、可具有取代基之C1-6烷基胺基羰基、可具有取代基之飽和或不飽和雜環羰基中之任一者,R2表示氟原子、或氯原子,R3表示氫原子、氟原子、或氯原子)
本發明化合物主要發揮起因於抑制破骨細胞之分化形成之骨吸收抑制效果,進而發揮成骨細胞之異形骨形成抑制效果,且顯示骨量恢復效果,藉此對骨質疏鬆症發揮優異之效果。因此,根據本發明,可有效地治療骨質疏鬆症。
圖1係人類前列腺癌株PC-3之小鼠脛骨移植部位之μCT圖像。

關於本發明之通式(I)所表示之醯基硫脲化合物(稱為「本發明化合物」),於記為「可具有取代基」之情形時,係指有於該結構上之化學上可能之位置上具有1個或2個以上之「取代基」之情形。
該結構上所存在之取代基之種類、取代基之個數、取代位置並無特別限定,於存在2個以上之取代基之情形時,其等可相同,亦可不同。作為「取代基」,可例示鹵素原子、羥基、氰基、硝基、C1-6烷醯基、C1-6烷基、C3-10環烷基、C2-6烯基、C1-6烷氧基、胺基、C1-6烷基胺基、C1-6烷醯基胺基、C1-6烷基胺基羰基、C1-6烷基磺醯基、C6-14芳香族烴基、飽和或不飽和雜環基、飽和或不飽和雜環羰基、側氧基等,於存在上述取代基之情形時,其個數典型的是1~3個。
式(I)中,R1所示之「可具有取代基之C1-6烷基」之「C1-6烷基」表示碳數1~6之直鏈狀或支鏈狀烷基,可例示甲基、乙基、正丙基、異丙基、正丁基、第二丁基、第三丁基、正戊基、異戊基、正己基、異己基,更佳為C1-4烷基,尤佳為甲基、乙基、正丙基、異丙基、正丁基、第二丁基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「C3-10環烷基」表示碳數3~10之環烷基,可例示環丙基、環丁基、環戊基、環己基,更佳為環己基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「可具有取代基之C1-6烷氧基」之「C1-6烷氧基」表示碳數1~6之直鏈狀或支鏈狀烷氧基,可例示甲氧基、乙氧基、正丙氧基、異丙氧基、正丁氧基、第二丁氧基、第三丁氧基、正戊氧基、正己氧基,尤佳為甲氧基、乙氧基、異丙氧基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示

式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「可具有取代基之C1-6烷基胺基」之「C1-6烷基胺基」表示經上述C1-6烷基單取代或二取代之胺基,可例示甲胺基、乙胺基、二甲胺基、甲基乙基胺基、正丙胺基、異丙胺基、正丁胺基、第二丁胺基、第三丁胺基、正戊胺基、正己胺基,更佳為二乙胺基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「C1-6烷醯基胺基」可列舉甲醯基、乙醯基、丙醯基、丁醯基等,更佳為乙醯胺基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「C1-6烷基磺醯基」表示經上述C1-6烷基取代之磺醯基,更佳為甲磺醯基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「可具有取代基之C6-14芳香族烴基」之「C6-14芳香族烴基」表示碳數6~14之芳香族烴基,可例示苯基、萘基等,更佳為苯基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「可具有取代基之飽和或不飽和雜環基」之「飽和或不飽和雜環基」表示具有1個或2個氧原子、氮原子、硫原子中之任一原子之單環性或二環性之飽和或不飽和雜環基,例如可列舉吡咯啶基、哌啶基、哌基、嗎啉基、硫代嗎啉基、高哌啶基、四氫噻吩基、咪唑基、噻吩基、呋喃基、吡咯基、唑基、異唑基、噻唑基、異噻唑基、吡唑啉基、三唑基、四唑基、吡啶基、吡基、嘧啶基、嗒基、吲哚基、異吲哚基、吲唑基、亞甲基二氧基苯基、伸乙基二氧基苯基、苯并呋喃基、二氫苯并呋喃基、苯并咪唑基、苯并唑基、苯并噻唑基、嘌呤基、喹啉基、異喹啉基、喹唑啉基、喹啉基等。更佳為含有1~4個氮原子及/或氧原子之5~7員之雜環基,尤佳為吡咯啶基、
嗎啉基、二氧雜環戊烷、四氫哌喃基、吡啶基、四唑基。該飽和或不飽和雜環基亦可進而具有取代基,作為取代基,較佳為C1-6烷基(尤其是甲基)、側氧基。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「可具有取代基之C1-6烷基胺基羰基」之「C1-6烷基胺基羰基」表示具有上述C1-6烷基胺基之羰基,更佳為乙基胺基羰基、二甲胺基、甲基丁基胺基。該C1-6烷基胺基羰基亦可進而具有取代基,作為取代基,較佳為羥基、C1-6烷氧基(尤其是甲氧基)。
式(I)中,作為R1之「可具有取代基之C1-6烷基」之取代基而表示之「可具有取代基之飽和或不飽和雜環基羰基」之「飽和或不飽和雜環基羰基」表示具有上述飽和或不飽和雜環基之羰基,更佳為具有1~2個氮原子及/或氧原子之5~7員之飽和雜環羰基,尤佳為吡咯啶基羰基、嗎啉基羰基。該飽和或不飽和雜環羰基亦可進而具有取代基,作為取代基,較佳為鹵素原子(尤其是氟原子)、可具有羥基之C1-6烷基(尤其是甲基)。
作為R1所示之可具有取代基之C1-6烷基之C1-6烷基,較佳為甲基、乙基、正丙基、異丙基、正丁基、或第二丁基,作為該烷基上之取代基,較佳為羥基、環己基、甲氧基、乙氧基、異丙氧基、二乙胺基、乙醯胺基、甲磺醯基、苯基、吡咯啶基、嗎啉基、二氧雜環戊烷基、四氫哌喃基、吡啶基、三唑基、乙基胺基羰基、二甲胺基羰基、甲基丁基胺基羰基、吡咯啶基羰基、或嗎啉基羰基,該烷氧基亦可進而具有羥基作為取代基,該雜環基亦可進而具有甲基、或側氧基作為取代基,該烷基胺基羰基亦可進而具有羥基、或甲氧基作為取代基,該雜環羰基亦可進而具有氟原子、可具有羥基之甲基作為取代基。
作為R1所示之可具有取代基之C1-6烷基之C1-6烷基,較佳為甲基、乙基、正丙基、異丙基、正丁基、或第二丁基,作為該烷基上之
取代基,較佳為羥基、甲氧基、或嗎啉基。
作為R1,更佳為甲基、甲氧基乙基、嗎啉基乙基、嗎啉基羰基甲基、2-羥基-正丁基、2-羥基-2-甲基正丙基、1-羥基正丁烷-2-基,於1-羥基正丁烷-2-基之情形時,較佳為S體。
作為R2之取代位置,較佳為2位、或3位,尤佳為2位。又,作為R2,較佳為氟原子或氯原子,進而較佳為氟原子。
作為R3之取代位置,較佳為2位、或4位。又,作為R3,更佳為氫原子或氟原子。
本發明化合物中較佳為如下者:R1為可具有取代基之甲基、乙基、正丙基、異丙基、正丁基、或第二丁基,且該烷基上不存在取代基,或取代基為羥基、甲氧基、或嗎啉基,R2為氟原子,R3為氫原子或氟原子。
進而,作為本發明化合物之較佳之具體例,可列舉如下化合物。
(1)4-(2-氟-4-(3-(2-苯基乙醯基)硫代脲基)苯氧基)-7-甲氧基-N-甲基喹啉-6-甲醯胺
(2)4-(2-氟-4-(3-(2-苯基乙醯基)硫代脲基)苯氧基)-7-甲氧基-N-(2-嗎啉基乙基)喹啉-6-甲醯胺
(3)(S)-4-(2-氟-4-(3-(2-(4-氟苯基)乙醯基)硫代脲基)苯氧基)-N-(1-羥基丁烷-2-基)-7-甲氧基喹啉-6-甲醯胺
其中,進而較佳為下述式(IA)所示之4-(2-氟-4-(3-(2-苯基乙醯基)硫代脲基)苯氧基)-7-甲氧基-N-甲基喹啉-6-甲醯胺[別名:(4-[2-氟-4-[[[(2-苯基乙醯基)胺基]硫酮基甲基]胺基]-苯氧基]-7-甲氧基-N-甲基-6-喹啉甲醯胺)]。

作為本發明化合物之鹽,只要為藥學上所容許之鹽即可,例如可列舉與鹽酸、氫溴酸、氫碘酸、硫酸、硝酸、磷酸等無機酸、或甲酸、乙酸、丙酸、草酸、丙二酸、琥珀酸、反丁烯二酸、順丁烯二酸、乳酸、蘋果酸、檸檬酸、酒石酸、碳酸、苦味酸、甲磺酸、對甲苯磺酸、麩胺酸等有機酸之酸加成鹽;與鈉、鉀、鎂、鈣、鋁等無機鹼、或甲基胺、乙基胺、葡甲胺、乙醇胺等有機鹼、或離胺酸、精胺酸、鳥胺酸等鹼性胺基酸之鹽或銨鹽。
其中較佳為與有機酸之鹽,更佳為甲磺酸鹽,進而較佳為單甲磺酸鹽。
又,本發明化合物中亦包含光學異構物,且亦包含水合物、各種溶劑合物及多晶型。
本發明化合物亦可為藥理學上所容許之前藥之形態。所謂藥理學上所容許之前藥,只要為藉由於活體內之生理學條件下、例如利用胃酸或酵素進行水解、氧化、還原反應而轉化為通式(I)者,則可為任意,例如可例示將羧基改質之甲酯、乙酯、丙酯、苯酯、羧氧基甲酯、乙氧羰酯等酯型化合物。形成該等前藥之代表性者可列舉如廣川書店1990年刊『醫藥品之開發』第7卷,163頁至198頁所記載之於生理學條件下轉化為化合物(I)之化合物等。
本發明化合物為公知化合物,例如可依據國際公開第2009/125597號(上述專利文獻1)記載之方法而製造。
如下述實施例所示,本發明化合物於移植有癌細胞株之小鼠脛
骨內移植模型中,藉由抑制骨破壞亢進與異常之弱骨之形成,而維持骨量正常(試驗例1及2)。又,於作為骨質疏鬆症之典型性模型所知之卵巢摘除模型動物(OVX小鼠),顯示顯著之骨量恢復效果(試驗例3)。
因此,本發明化合物或其鹽作為適用於骨質疏鬆症之患者,對該骨質疏鬆症發揮優異之預防或治療效果之醫藥有用。進而,對類風濕性關節炎、牙周病、骨折等之治療亦有用。
關於本發明之骨質疏鬆症治療劑及用以治療骨質疏鬆症之醫藥組合物,所謂「治療」係指用以預防及治療疾病,以及減輕症狀及防止復發的維持療法。
本說明書中,所謂「骨質疏鬆症」係指以因骨量之減少與骨質之劣化而導致骨強度惡化,從而骨折之風險容易增加為特徵之原發性及繼發性之骨疾病。作為可利用本發明之骨質疏鬆症治療劑及用以治療骨質疏鬆症之醫藥組合物而治療之骨質疏鬆症,例如可列舉閉經後骨質疏鬆症、男性骨質疏鬆症、突發性骨質疏鬆症(妊娠後骨質疏鬆症等)、以及骨發育不全症等。
本發明化合物或其鹽可製備成經口或非經口之任一投予形態,可使用藥學上所容許之載體,藉由公知之方法而製造成醫藥組合物。作為此種醫藥組合物之製劑形態並無特別限制,可例示片劑、包衣片劑、丸劑、散劑、顆粒劑、膠囊劑、液劑、懸浮劑、乳劑等經口劑;注射劑、栓劑等非經口劑等。
於成形為片劑之形態時,作為載體,例如可使用乳糖、白糖、氯化鈉、葡萄糖、脲、澱粉、碳酸鈣、高嶺土、結晶纖維素、矽酸等賦形劑;水、乙醇、丙醇、玉米澱粉、單糖漿、葡萄糖液、澱粉液、明膠溶液、羧甲基纖維素、蟲膠、甲基纖維素、羥丙基纖維素、羥丙基甲基纖維素、磷酸鉀、聚乙烯吡咯啶酮等結合劑;乾燥澱粉、海藻酸鈉、瓊脂末、昆布糖末、碳酸氫鈉、碳酸鈣、聚氧乙烯山梨醇酐脂
肪酸酯類、月桂基硫酸鈉、硬脂酸單甘油酯、乳糖等崩解劑;白糖、硬脂酸、可可脂、氫化油等崩解抑制劑;四級銨鹽、月桂基硫酸鈉等吸收促進劑;甘油、澱粉等保濕劑;澱粉、乳糖、高嶺土、膨潤土、膠體狀矽酸等吸附劑;精製滑石、硬脂酸鹽、硼酸末、聚乙二醇等潤滑劑等。進而,片劑可視需要製成施加有通常之包衣之片劑,例如糖衣藥丸、明膠包衣錠、腸溶包衣錠、膜衣錠、雙層錠、多層錠等。
於成形為丸劑之形態時,作為載體,例如可使用葡萄糖、乳糖、澱粉、可可脂、氫化植物油、高嶺土、滑石等賦形劑;阿拉伯膠末、黃耆膠末、明膠、乙醇等結合劑;昆布糖、瓊脂等崩解劑等。膠囊劑係依照慣例與上述所例示之各種載體混合並填充至硬質明膠膠囊、軟質膠囊等而製備。
於製成經口用液體製劑之情形時,可使用矯味/矯臭劑、緩衝劑、穩定劑等,依照慣例而製造內服液劑、糖漿劑、酏劑等。於此情形時,作為矯味/矯臭劑,可列舉白糖、橙皮、檸檬酸、酒石酸等,作為緩衝劑,可列舉檸檬酸鈉等,作為穩定劑,可列舉黃耆膠、阿拉伯膠、明膠等。
於成形為栓劑之形態時,作為載體,例如可使用聚乙二醇、可可脂、高級醇、高級醇之酯類、明膠、半合成甘油酯等。
於製成注射劑之情形時,較佳為將液劑、乳劑及懸浮劑殺菌,且與血液等張,於成形為該等形態時,作為稀釋劑,例如可使用水、乳酸水溶液、乙醇、丙二醇、聚乙二醇、乙氧化異硬脂醇、聚氧乙烯化異硬脂醇、聚氧乙烯山梨醇酐脂肪酸酯類等。
再者,於此情形時,可使醫藥製劑中含有對於製備等張性之溶液而言充分之量之食鹽、葡萄糖或甘油,又,亦可添加通常之溶解助劑、緩衝劑、舒緩劑等。進而,於上述各製劑中,亦可視需要調配著色劑、防腐劑、香料、調味劑、甜味劑等、或其他醫藥品。
本發明之骨質疏鬆症治療劑、及用以治療骨質疏鬆症之醫藥組合物之投予方法係根據各種製劑形態、患者之年齡、性別及其他條件、患者之症狀之程度等而適當決定。例如片劑、丸劑、散劑、顆粒劑、膠囊劑、液劑、懸浮劑及乳劑係經口投予。注射劑係單獨或與葡萄糖、胺基酸等通常之補液混合而被投予至靜脈內,進而視需要單獨地被投予至動脈內、肌內、皮內、皮下或腹腔內。栓劑係被投予至直腸內。
上述各投予單位形態中所應調配之本發明化合物或其鹽之量根據應適用其之患者之症狀、或其劑形等而不固定,但通常較理想為於每投予單位形態中,若為經口劑,則設為約0.005~1,000mg,若為注射劑,則設為約0.001~500mg,若為栓劑,則設為約0.01~1,000mg。又,具有上述投予形態之藥劑之每天之投予量根據患者之症狀、體重、年齡、性別等而有所不同,無法一概決定,通常設為成人每天約0.005~5,000mg、較佳為0.01~1,000mg即可,較佳為將其每天1次或分為2~4次左右投予。
以下,列舉實施例、試驗例對本發明進行更詳細之說明,但本發明並不限定於該等。
4-[2-氟-4-[[[(2-苯基乙醯基)胺基]硫酮基甲基]胺基]-苯氧基]-7-甲氧基-N-甲基-6-喹啉甲醯胺(化合物IA)之合成
依據國際公開第2009/125597號(上述專利文獻1)之實施例6之記載,合成表記化合物IA(本發明化合物)。
試驗例1 對人類前列腺癌株PC-3之小鼠脛骨內移植模型之異形骨形成及骨吸收之抑制效果
將人類前列腺癌細胞株PC-3以2×106cells/15μL移植至小鼠脛骨
內。於移植第二天以使各組中之小鼠之平均體重成為均等之方式進行分組,將化合物IA以200mg/kg/天連續10天進行經口投予。於自移植起第11天進行移植部位之μCT(Micro Computed Tomography,微電腦斷層掃描)攝影,實施骨病變之評價。
如圖1所示,確認於脛骨移植有人類前列腺癌株PC-3之小鼠中,於其移植部位之骨內部形成異形骨。本發明化合物明顯地抑制該異形骨形成。
根據以上內容可明確,本發明化合物未顯示死亡或體重減少,而是藉由抑制人類前列腺癌細胞於脛骨移植系統中之代謝性之骨破壞亢進與異常之弱骨之形成,而維持骨量正常。
試驗例2 對人類肺癌株A549-luc-BM1之小鼠脛骨內移植模型之異形骨形成及骨吸收之抑制效果
將導入有螢光素酶基因之人類肺癌細胞株A549移植至裸小鼠之左心室,藉此建立骨選擇性地轉移並增生之肺癌細胞株A549-luc-BM1。將該細胞以2×106cells/15μL移植至小鼠脛骨內。於移植後第7天利用活體成像系統(in vivo imaging system)測定移植部位之螢光素酶活性,並以使各組之平均螢光素酶活性成為相等之方式進行分組,將化合物IA以200mg/kg/天連續28天進行經口投予。治療對照組係使用作為第三代之雙磷酸鹽製劑之唑來膦酸,以0.2mg/kg/天每週2次進行皮下投予。骨病變之評價係於移植後第36天進行移植部位之μCT攝影。
針對脛骨移植有人類肺癌株A549-luc-BM1之小鼠,將其移植部位之骨病變之評價示於圖2。於移植後第36天,對照組中因異常之骨破壞與異形骨形成而脛骨大幅變形。另一方面,本發明化合物顯著地抑制骨破壞與異形骨形成,而維持接近於正常骨(normal bone)之形態。又,關於作為治療對照藥之唑來膦酸,雖抑制骨破壞,但確認有
異形骨形成,而產生骨之變形。
根據以上內容可明確,本發明化合物之骨代謝(骨吸收與骨形成)異常之抑制效果較唑來膦酸優異,作為骨質疏鬆症治療劑有用。
試驗例3 對利用卵巢摘除小鼠(OVX小鼠)之骨質疏鬆症模型之骨量降低抑制效果
於麻醉下摘除小鼠之卵巢,並於當天以使小鼠之平均體重成為均等之方式進行分組。作為治療陽性對照組,將雌二醇(E2)以3μg/kg/天連續4週每天投予。又,將化合物IA以200mg/kg/天連續4週每天進行經口投予。於各藥劑投予後摘除小鼠之大腿骨,利用骨密度測定裝置(雙能X射線吸收測量法,DEXA法)及μCT測定骨量並進行評價。將大腿骨之骨量(BMD)、大腿骨之μCT圖像、及體重變化分別示於圖3、4及5。
如圖3、4所示,OVX小鼠與Sham(假處理)組相比呈現大腿骨之骨量降低,雌二醇投予組對OVX小鼠顯示出抑制骨量降低。由此可確認,本模型系統可適當地進行卵巢摘除,而成立骨質疏鬆症模型(CLINICAL CALCIUM,2011,21(2),p.164)。
本發明化合物投予組對OVX小鼠發揮顯著之骨量恢復效果(圖3、4)。除總骨量(Total BMD)之恢復效果以外,於骨質疏鬆症中尤其被要求改善之遠端部(Distal BMD),骨量亦恢復(圖4)。另一方面,未確認對體重之影響(圖5)。
根據以上結果可明確,本發明化合物作為安全性較高之骨質疏鬆症治療劑有用。

- 一種骨質疏鬆症治療劑,其含有下述式(I)所表示之醯基硫脲化合物或其鹽作為有效成分:
- 一種醫藥組合物,其係用以治療骨質疏鬆症,且含有下述式(I)所表示之醯基硫脲化合物或其鹽及藥學上所容許之載體:
- 如請求項1之骨質疏鬆症治療劑或如請求項2之醫藥組合物,其中式(I)所表示之醯基硫脲化合物或其鹽為4-[2-氟-4-[[[(2-苯基乙醯基)胺基]硫酮基甲基]胺基]-苯氧基]-7-甲氧基-N-甲基-6-喹啉甲醯胺單甲磺酸鹽。
