Abstract

X-isini kristalografisi , bir hedef biyomolekülünün ligandlari bilinmeyen birlesikleri için, bunlarin hedefe baglanma yetenegini görüntülemek ve teshis etmek için kullanilabilmektedir. Yöntem hedef biyomolekülün kristalinin elde edilmesini; hedefi, hedefin potansiyel ligandlari olan bir veya fazla test nümunesine tabi tutmayi, ligand/biyomolekül kompleksinin meydana gelip gelmediginin saptanmasini içermektedir. Hedef, potansiyel ligandlara, kristalin bir veya daha fazla potansiyle ligandlar çözeltisinde islatilmasini veya biyomolekülün bir veya daha fazla potansiyel ligandlarin mevcudiyetinde ortak-kristallestirilmesini içeren, çesitli yöntemlerle maruz birakilmaktadir. Bulunan ligand/almaç komplekslerinden saglanan yapisal bilgi, daha siki, daha spesifik baglanan, daha iyi biyolojik etkinlige sahip yeni ligandlar tasarlamakta kullanilmaktadir.
