Claims (10)
1. Вариант арабинозоизомеразы, обладающий увеличенной активностью превращения D-галактозы в D-тагатозу, где вариант арабинозоизомеразы содержит замену аминокислоты в положении 275 аминокислотой кроме фенилаланина и замену аминокислоты в положении 469 пролином в аминокислотной последовательности, представленной в SEQ ID NO: 1.
2. Вариант арабинозоизомеразы по п.1, где замена аминокислоты в положении 275 осуществляется любой аминокислотой, выбранной из группы, состоящей из валина, метионина и изолейцина.
3. Полинуклеотид, кодирующий вариант арабинозоизомеразы по п. 1.
4. Полинуклеотид по п.3, который имеет нуклеотидную последовательность SEQ ID NO: 8, SEQ ID NO: 9 или SEQ ID NO: 10.
5. Микроорганизм рода Corynebacterium, трансформированный рекомбинантным вектором, включающим полинуклеотид по п. 3.
6. Микроорганизм по п.5, который представляет собой Corynebacterium glutamicum.
7. Микроорганизм по п.5, который представляет собой Corynebacterium glutamicum pFIS-1-TNAI-2 (KCCM 11378P), pFIS-1-TNAI-3 (KCCM 11379P) или pFIS-1-TNAI-4 (KCCM 11380P).
8. Способ получения D-тагатозы, включающий культивирование микроорганизма по любому из пп.5-7.
9. Способ по п.8, который включает реакцию раствора, содержащего D-галактозу, с источником ионов металла, выбранным из группы, состоящей из хлорида марганца, хлорида магния и хлорида цинка, в присутствии микроорганизма.
10. Способ по п.8, где хлорид марганца добавляют в концентрации 0,1-10 мМ.
