Рекомбинантная плазмидная ДНК pFGM17, кодирующая полипептид гранулоцитарно-макрофагального колониестимулирующего фактора человека (ГМ-КСФ), имеющая молекулярную массу 2,04 Md (3,133 т.п.о.) и состоящая из KpnI/EcoRI-фрагмента ДНК плазмиды pSPFl, длиной 2,686 т.п.о., содержащего lac-промотор Е.coli, ген bla β-лактамазы, фрагмент гена lacZ Е.coli, участок ori инициации репликации и искусственную последовательность ДНК, кодирующую сигнальный пептид белка Caf1 Yersinia pestis; а также KpnI/EcoRI-фрагмента промежуточной плазмиды pSK-GM длиной 0,447 т.п.о., включающего синтетический ген ГМ-КСФ человека, последовательность которого приведена на фиг.2, и содержащая уникальные сайты узнавания рестрикционньми эндонуклеазами, имеющие следующие координаты: Kpn I-280, PstI-293, BglII-309, NcoI-333, XhoI-474, BamHI-688, SalGI-700, Cla I-710, Eco RI-727.1. Recombinant plasmid DNA pFGM17 encoding a polypeptide of a granulocyte macrophage colony stimulating human factor (GM-CSF), having a molecular weight of 2.04 Md (3.133 kb) and consisting of a KpnI / EcoRI DNA fragment of the plasmid pSPFl, length 2.666 kbp containing an E. coli lac promoter, a β-lactamase bla gene, an E. coli lacZ gene fragment, a replication initiation ori site and an artificial DNA sequence encoding a signal peptide of the Yersinia pestis Caf1 protein; as well as the KpnI / EcoRI fragment of the intermediate plasmid pSK-GM with a length of 0.447 kbp, including the synthetic human GM-CSF gene, the sequence of which is shown in figure 2, and containing unique recognition sites by restriction endonucleases having the following coordinates: Kpn I-280, PstI-293, BglII-309, NcoI-333, XhoI-474, BamHI-688, SalGI-700, Cla I-710, Eco RI-727. 2.
