Description
Foreliggende oppfinnelse vedrører en fremgangsmåte ved frem-' stilling av tieno [2,3-b] pyrrolderivater med den generelle formel:
hvor R^betyr et hydrogenatom eller rett- eller forgrenet alkyl-restkjede med fra 1 til 5 karbonatomer,
1*2 betyr:
- en cyanorest,
- en karboksyrest,
- en COOM gruppe hvori M betyr et alkalimetall, et jordalkalimetall, en ammoniumrest eller en mono-, di-, tri-eller tetra-alkylammonium eventuelt mono- eller poly-substiuert med en hydroksyrest,
- en COOR gruppe hvori R betyr en alkylrest med fra 1 til
5 karbonatomer, eller
- en - CO N K„ gruppe hvori R' og R" som er like eller forskjellige hver betyr et hydrogenatom, eller en rettkjedet eller forgrenet alkylrest med fra 1 til 5 karbonatomer;
R^betyr en rettkjedet eller forgrenet alkylrest med fra
1 til 5 karbonatomer, og
R^betyr en rettkjedet eller forgrenet alkylrest med fra
1 til 5 karbonatomer, eller en fenylrest som er eventuelt mono- eller polysubstituert med et halogenatom,
en alkyl eller alkoksyrest med hver fra 1 til 5 karbonatomer, en nitrorest eller en dialkylaminorest hvori hver alkylgruppe inneholder fra 1 til 5 karbonatomer og kan være rettkjedet eller forgrenet.

risk karbonatom. Det foreligger således to enantiomere hvis fremstilling også inngår i foreliggende oppfinnelse.
Foreliggende oppfinnelse vedrører også en fremgangsmåte ved fremstillingen av derivater med den generelle formel I,karakterisert vedat:
diazoderivatene med den generelle formel II:
hvori R-, har den forut angitte betydning, de-diazoteres til forbindelsen med den generelle formel III: hvori R^har den forut angitte betydning, den'således fremstilte forbindelse III acyleres ved Friedel-Craft metoden ved hjelp av et acylhalogenid med- formel X-CO-R^ hvori X betyr et halogenatom, spesielt et brom- eller kloratom og R^har den forut angitte betydning, til derivatet med den generelle formel IV: hvor R., og R^har de forut angitte betydninger, derivatene IV hydrolyseres til forbindelsene med den generelle formel V:
hvor R-j og R^ har de forut angitte betydninger,
disse forbindelser V dekarboksyleres til derivater med den generelle formel VI:
hvor R^og R^har de forut angitte betydninger, hvilke forbindelser VI så N-alkyleres ved hjelp av en a-halogensyre eller et derivat med formelen:
hvor R-^ og R2har de forut angitte betydninger og X betyr et halogenatom såsom klor eller brom.
Det er spesielt fordelaktig å utføre didiazotiseringen av derivatet II enten ved oppvarming i et egnet løsningsmiddel såsom f.eks. etanol, om nødvendig under katalyse av reaksjonen med spor av HCl, U.V. eller radikalinitiatorer, eller■ved reduksjon ved hjelp av reduksjonsmidler såsom f.eks. LiAlH^, NaBH^osv. På samme måte er det spesielt gunstig for acyleringen av forbindelsene III at den utføres i nærvær av en katalysator (Lewis syre) i et passende løs-ningsmiddel såsom diklormetan.
Hydrolysen av forbindelsene IV utføres lett ved oppvarming
av disse forbindelser IV i en base-etanol blanding, etterfulgt av surgjøring. Til slutt utføres dekarboksyleringen av forbindelser V med fordel ved oppvarming i et egnet løs-ningsmiddel såsom f.eks. kinolin i nærvær av kobberpulver som katalysator.
Utgangsmaterialene med den generelle formel II kan fremstilles ifølge den metode som er beskrevet av Wierzbicki M. et al., Bull Soc. Chim. France (1975) s. 1786-1792 fra derivater med den generelle formel
hvori R^har de forut angitte betydninger.
Disse sistnevnte derivater kan i seg selv fremstilles ifølge den metode som er beskrevet av Gewald K. et al., Chem. Ber.
(1966), 99, s. 94 og 2712 ut fra utgangsmateriåler med den generelle formel: hvor R3har de ovenfor angitte betydninger, hvilke utgangs-materialer behandlet med WN-Cr^-CN, svovel., et sekundært amin og etanol, cykliseres under dannelse av derivater med den generelle formel:.
hvor R^er som forut definert og disse derivater N-acetyleres med.CH3COCl/CH3COOH.
Derivatene med den generelle formel I har interessante farmakologiske egenskaper, spesielt vesentlig analgetiske egenskaper og i mindre grad anti-inflammatoriske egenskaper .
Toksisiteten deres er lav, deres LD^q, bestemt oralt på mus er mellom 250 og 2000 mg/kg.
Således er produktet fra eksemplene 7 og 9 i det følgende LD^g oralt hos mus henholdsvis 480 til 680 mg/kg, mens
under de samme betingelser er de henholdsvis 1150 go 14 00 mg/kg for glafenin og klormetacin som er velkjente analgetiske midler og er tatt som sammenligningsprodukter.
Den analgetiske virkningen til forbindelsene fremstilt
ved fremgangsmåten ifølge oppfinnelsen er påvist spesielt ved forsøket til Hendershot, L.C., Forsaith, J.J., J. Pharmacol. Exp. Ther. (1959), 125, 237 som vedrører mage-kramper indusert ved fenylbenzokinon, og ved forsøket til Koster R., Anderson M. , de Beer E.S., Fed.. Proe. (1959), 18, 412 som vedrører "Writhing" indusert av eddiksyre.
For hver av disse prøver ligger den gjennomsnittlige effektive dose ED^q for forbindelsene ifølge oppfinnelsen mellom 5<:>og 100 mg/kg ved oral administrering til sveisiske musi Således ble det funnet at ifølge forsøket til Hendershot som tidligere er nevnt var de gjennomsnittlige effektive doser ED^g en time etter behandlingen for forbindelsene
fra eksempel 7 og 9 i det følgende 21,6 og 26,7 mg/kg henholdsvis, mens den var 43 mg/kg for glafenin. Videre er varigheten av den analgetiske virkning for forbindelsene ifølge oppfinnelsen meget større enn den som ble observert med de analgetiske sammenligningsforbindelser. Med fenylbenzokinon-forsøket ifølge Hendershot et al observeres således hos mus at behandlingen oralt ved en dose på 25 mg/kg med forbindelsen for eksempel 7 i det følgende får man en beskyt-telsesgrad som når 60% og opprettholdelse i 5 timer, mens under de samme betingelser med klormetacin er beskyttelses-graden bar 4 6% og denne aktivitet forsvinner etter 3 timer.
Den antiinflammatoriske aktiviteten til derivatene fremstilt
■ifølge oppfinnelsen er studert i forskjellige forsøk.
Spesielt ligger ifølge forsøket til La Belle A. og Tislow
R., J. Pharmacol. Exp. Ther. (1950), 98, 19 vedrørende smerten forårsaket ved artritis indusert med sølvnitrat den gjennomsnittlige effektive dose (ED<-g) til produktene i foreliggende oppfinnelse mellom 50 og 200 mg/kg ved oral administrering til rotter.
Ved L.F. Sancilio forsøket, J. Pharmacol. Exp. Ther. (1969), 168, 199, vedrørende pleurisy indusert av carrageenin,
viser produktene fremstilt ifølge oppfinnelsen meget tydelig aktivitet mellom 12,5 og 200 mg/kg gitt oralt til sveitsiske mus.
Også ifølge prøven til Winter C.A., Riseley E.A., og Nuss. G. W., J. Pharmacol. Exp. Ther. (1963), 141, 369 vedrørende granuloma indusert av bomull, hadde ingen av produktene, fremstilt ifølge oppfinnelsen noen betydelig aktivitet ved doser under 50 mg/kg gitt oralt til S.D. rotter.
Videre er i varmplateforsøket ifølge metoden beskrevet av Chen I.Y.P. og Beekman H., Science (1951), 113, 631 av for bindelsene fremstilt ifølge den oppfunnede fremgangsmåte I høyere enn 50 mg/kg ved subkutan administrering til NMRI mus, hvilket viser mangel på sentral aktivitet for de forbindelser som ble prøvet på denne måte.
Anvendelse av forsøket beskrevet av Flower R.J., Chung H.S. og Cushman D.W., Prostaglandines (1973), (4), 325 på forbindelsene fremstilt ifølge oppfinnelsen og vist at visse av disse forbindelser er inhibitorer for enzymatiske systemer for prostaglandinsyntese ved gjennomsnittlige inhiberende doser (ID^q) som kan gå med til 10 ^M, hvilket skulle vise at disse produktene har en virkningsmåte som må ansees å være perifer.
De ovenfor beskrevne farmakologiske egenskaper samt den lave toksisiteten til forbindelsene med den generelle formel I gjør dem terapeutisk anvendelige, hovedsakelig ved behand-ling av akutt eller kronisk smerte og spesielt en smerte'
i forbindelse med en inflammatoriske prosess (rheumatisk smerte ved.arthrose, arthritis, lumbosciatica, osv.), traumatisk, post-traumatisk eller post-operativ smerte, smerte i ORL, mage- eller genital-urinalområdene og likeledes visse neuralgiske, hode-, migrene- og kreftsmerter.
Foreliggende oppfinnelse vedrører også fremstillingen av farmasøytiske preparater hvilke som aktiv bestanddel inneholder en forbindelse med den generelle formel I i blanding eller i kombinasjon med en passende farmasøytisk eksipient som f.eks. destillert vann, glukose, laktose, stivelse, talkum, etylcellulose, magnesiumstearat eller kokosolje.
De farmasøytiske preparater som fremstilles slik er generelt i doseringsform og kan inneholde fra 25 til 250 mg aktiv bestanddel. De kan f.eks. anta form av tabletter, drageer, gelatinovertrukne piller, suppositorier, injiserbare eller drikkbare løsninger eller salver og, avhengig av det foreliggende tilfelle, kan de gis oralt, rektalt, parenteralt eller lokalt i en dose fra 25 til 250 mg fra 1 til 4 gangier pr. dag. j De følgende eksempler illustrerer oppfinnelsen, smelte-punktene, med mindre det motsatte er angitt, bestemmes med Kofler varmeplate.
Eksempel 1:
2, 5- dietoksykarbonyl- 3- metyltieno[ 2, 3- b] pyrrol
1 mol av 2-acetamido-3-cyano-5-etoksykarbonyl-4-metyltiofen (fremstilt i henhold til metoden beskrevet av Gewald K. et al., Chem. Ber. (1966), 99, s. 94 og 2712, fra etylacetat) bringes til tilbakeløp i 24 timer i 1,6 1 aceton med 187 g etylbromacetat og 250 g kaliumkarbonat. Deretter felles blandingen i 6 liter isvannblanding (1:1) . Det hele røres kraftig, den resulterende utfelling filtreres, tørkes med luft, vaskes med 500 ml av en iskald blanding vann og etanol (60:40), tørkes igjen og vaskes til slutt med 700 ml av en blanding av cykloheksan og benzen (6:1). Det resulterende produkt (320 g - 0,95 mol) røres så i 1,2 liter r^SO^
på 30% holdt på en temperatur under 10°C i en nitrogenstrøm..
85 g kaliumnitrit i 180 ml vann tilsettes gradvis mens tempe- . råturen holdes. Når tilsetningen er avsluttet, fortsetter man å røre noen få minutter, og tilsetter så 7,2 liter av
en isvannblanding. Det hele røres kraftig. Den erholdte gule utfelling vakuumfUtreres raskt og renses med vann til nesten nøytral reaksjon. Den bringes deretter til tilbakeløp i 3 liter etanol inntil nitrogenutviklingen er fullstendig.
Så konsentreres reaksjonsblandingen til 1 liter ved destillasjon, 300 ml tilsettes og det hele røres kraftig. Så snart krystallisasjonen er ferdig filtreres utfellingen og tørkes. På denne måten oppnås 205 g (0,73 mol) 2,5-dietoksykarbonyl 3-metyl-tieno [2,3-b] pyrrol, smp.: 139°C. Den følgende forbindelse ble fremstilt på samme måte: 2,5-dietoksykarbonyl-3-n-propyltieno[2,3-b]pyrrol, smp.:13 7-138°C.
Eksempel 2: 2, 5- dietoksykarbonyl- 3- metyl- 4- paraklorbenzoyl- tieno[ 2, 3- b] pyrrol
70 g (0,25 mol) av 2 , 5-dietoksykarbonyl-3-metyl-tieno [ 2 , 3'-b]
1 pyrrol røres med 65 g paraklorbenzoylklorid og 120 g tinn-tetraklorid i 1,2 liter diklormetan. Det hele holdes under forsiktig tilbakeløp i 20 timer. Blandingen hydrolyseres så med 500 ml 4N saltsyreløsning og 500 g knust is. Det hele røres kraftig inntil hydrolysen er fullstendig. Den vandige fase dekanteres- og vaskes tre ganger med 200 ml diklormetan hver gang. De samlede organiske faser vaskes med vann, og deretter med en fortynnet løsning av natriumkarbonat og tilslutt fordampes løsningsmiddelet. På denne måten får man 105 g rått 2,5-dietoksykarbonyl-3-4-paraklorbenzoyltieno[2,3-b]pyrrol som deretter brukes uten videre rensing.
De følgende forbindelser ble fremstilt på samme måte:-
a) 2,5-dietoksykarbonyl-3-mety1-4-acetyltieno-[2,3-b]pyrrol, b) 2,5-dietoksykarbonyl-3-metyl-4-ortoklorbenzoyltieno [2,3-b]pyrrol, c) 2,5-dietoksykarbonyl-3-metyl-4-(2,6-diklorbenzoyl)-tieno[2,3-b]pyrrol, d) 2,5-dietoksykarbonyl-3-metyl-4-parafluorbenzoyltieno [2,3-b]pyrrol, e) 2,5-dietoksykarbonyl-3-metyl-4-metaklorbenzoyltieno [2,3-b]pyrrol, f) 2,5-dietoksykarbony1-3-mety1-4-benzoyltieno[2,3-b]-pyrrol, g) 2,5-dietoksykarbonyl-3-metyl-4-parametoksybenzoyltieno [2,3-b]pyrrol, h) 2,5-dietoksykarbonyl-3-n.propy1-4-parafluorbenzoyltieno [2,3-b]pyrrol,
i) 2 , 5-dietoksykarbonyl-3-n.propy'l-4-ortoklorbenzoyltieno [2,3-b]pyrrol,
j) 2,5-dietoksykarbonyl-3-metyl-4-ortometylbenzoyltieno
[2,.3-b]pyrrol,
k) 2,5-dietoksykarbonyl-3-metyl-4-metametylbenzoyltieno
[2,3-b]pyrrol,
1) 2,5-dietoksykarbonyl-3-metyl-4-parametylbenzoyltieno [2,3-b]pyrrol.
Eksempel 3: 2, 5- dikarboksy- 3- metyl- 4- paraklorbenzoyltieno[ 2, 3- b] pyrrol
105 g rått 2,5-dietoksykarbonyl-3-metyl-4-paraklorbenzoyltieno[2,3-b]pyrrol oppvarmes ved tilbakeløp i en blanding av 1 liter etanol og 1 liter av en normal løsning natriumhydroksyd til forsåpningen er fullstendig. Etanolen fjernes over destillasjon på et vannbad. Den vandige løsning filtreres og surgjøres deretter. Den resulterende utfelling samles ved filtrering, vaskes med vann og tørkes med luft og så i vakuum. På denne måten får man 92 g rått 2,5-dikar-.boksy-3-metyl-4-paraklorbenzoyltieno[2,3-b]pyrrol som brukes i etterfølgende synteser uten ytterligere rensing. De følg-ende forbindelser ble fremstilt på samme måte:
a) 2,5-dikarboksy-3-metyl-4-acetyltieno[2,3-b]pyrrol,
b) 2,5-dikarboksy-3-metyl-4-ortoklorbenzoyltieno[2,3-b] pyrrol, c) 2,5-dikarboksy-3-metyl-4-(2,6-diklorbenzoyl)-tieno [2,3-b]pyrrol, d) . 2,5-dikarboksy-3-metyl-4-parafluorbenzoyltieno[2,3-b] pyrrol, e) _ 2 , 5-dikarboksy-3-metyl-4-metaklorbenzoyltieno [ 2 , 3-b] pyrrol,
■ f) 2,5-dikarboksy-3-raetyl-4-benzoyltieno[2,3-b]-pyrrol,
g) 2,5-dikarboksy-3-metyl-4-parametoksybenzoyltieno[2,3-b] pyrrol, h) 2,5-dikarboksy-3-n.propy1-4-parafluorbenzoyltieno [2,3-b]pyrrol,
i) 2,5-dikarboksy,3-n.propyl-4-ortoklorbenzoyltieno[2,3-b]
pyrrol,
j) 2,5-dikarboksy-3-metyl-4-ortometylbenzoyltieno[2,3-b]

Eksempel 4:
3- metyl- 4- paraklorbenzoyltieno[ 2, 3- b] pyrrol
92 g 2,5-dikarboksy-3-metyl-4-paraklor-benzoyltieno[2,3-b]
pyrrol oppvarmes nær tilbakeløp i 900 ml kinolin med 300 mg kobberpulver inntil CC>2utvikles. Straks utviklingen er ferdig, får det hele vende tilbake til romtemperatur og under kjøling tilsettes så 200 ml av en 4N løsning saltsyre til blandingen. Det hele røres meget kraftig, den fine erholdte utfelling isoleres ved filtrering, vaskes to ganger med 50 ml av en normalløsning saltsyre hver gang og så med vann. De samlede vandige løsninger ekstraheres tre ganger med
150 ml diklormetan hver gang.
Diklormetanløsningen som derved oppnås vaskes med vann og inndampes så til tørrhet. Den resulterende rest opptas i et minimum diklormetan og gir en krystallisert forbindelse
i som tilsettes den tidligere. Det hele omkrystal.liseres fra
vandig etanol. På denne måten får man 56,5 g 3-metyl-4-paraklorbenzoyltieno[2,3-b]pyrrol, smp.: 223°C.
De følgende ble fremstilt på samme måte:
a) 3rmetyl-4-acetyltienot2,3-b]pyrrol, smp.: 183°C,
b) 3-metyl-4-benzoyltieno[2,3-b]pyrrol, smp.: 230,5°C,
c) 3-metyl-4-(2,6-diklorbenzoyl)-tieno[2,3-b]-pyrrol, smp. : 20 9°C,
d) 3-n.propyl-4-ortoklorbenzoyltieno[2,3-b]-pyrrol,
smp.: 108-109°C,
e) 3-metyl-4-metaklorbenzoyltieno[2,3-b]-pyrrol, smp.: 221-222°C, f) 3-metyl-4-ortoklorbenzoyltieno[2,3-b]-pyrrol, smp.: 186°C, g) 3-metyl-4-parafluorbenzoyltieno[2,3-b]-pyrrol, smp.: 245°C, h) 3-n.propyl-4-parafluorbenzoyltieno[2,3-b]-pyrrol, smp.: 157°C,
i) 3-metyl-4-parametoksybenzoyltieno[2,3-b]-pyrrol,
smp.: 215°C,
j) 3-me.tyl-4-ortometylbenzoyltieno [ 2 , 3-b]-pyrrol,
k) 3-metyl-4-metametylbenzoyltieno[2,3-b]-pyrrol,
1) 3-metyl-4-parametylbenzoyltieno[2,3-b]-pyrrol.
I EKSEMPEL 5: 3- metyl- 4- paraklorbenzoyl- 6- ( c/- karboksyetyl) tieno [ 2 , 3- b] pyrrol 34 g 3-metyl-4-paraklorbenzoyltieno[2,3-b]-pyrrol settes til en løsning av natriumetoksyd fremstilt ved å oppløse 5,66 g natrium i 400 ml etanol. Så tilsettes dråpevis .under røring 44 g etyl -brompropionat i 150 ml etanol, og det hele holdes ved tilbakeløp i 4 timer. Den erholdte blanding hydrolyseres og tilsettes uten rensing 150 ml av en normal natriumhydrok-.sydløsning og oppvarmes ved tilbakeløp i flere minutter.
Etanolen fordampes ved destillasjon. Blandingen fortynnes
med halve volumet vann og det hele ekstraheres med 500 ml diklormetan. Diklormetanfasen vaskes to ganger med.25 ml normal natriumhydroksydløsning hver gang og den vandig fase vaskes to ganger med 50 ml diklormetan hver gang. Denne vandige fase, surgjort til en pH. < 1, ekstraheres igjen med diklormetan. Den organiske fasen, vasket med vann og så behandlet med beinkull, fordampes deretter. Den krystalliserte erholdte rest vaskes med 200 ml av en blanding av benzen og cykloheksan 1:1. På denne måten får man 36 g 3-metyl-4-paraklorbenzoyl-6-(«i-karboksyetyl)-tieno [ 2 , 3-b] pyrool, smp.: . 149°C.

De følgende forbindelser ble fremstilt på samme måte som i eksempel .5: 6) 3-metyl-4-paraklorbenzoyl-6-karboksymetyltieno[2,3-b] pyrrol, smp.: 119°C, 7) 3-metyl-4-ortoklorbenzoyl-6-(Okarboksy-etyl)-tieno 2,3-b pyrrol, smp.: 188°C, 8) 3-mety 1-4-( 2 , 6-diklorbenzoyl)-6-(<.-karboksyetyl)-tieno[2,3-b]pyrrol, smp.: 186°C, 9) 3-metyl-4-benzoyl-6 - (o<i-karboksyetyl) -tieno- [ 2 , 3-b]
pyrrol, smp.: 152°C,

Forbindelsene i eksempel 14 til 16 er vanskelige å krys-tallisere eller oljeaktige. Natriumsalt av hver ble fremstilt ved å oppløse syren i en støkiometrisk mengde N/10 natriumhydroksyd, fordampe vannet og tørke den oppnådde rest.
Produktene som fremstilles på denne måten er amorfe eller hygroskopisk faste stoffer som er blitt prøvet i denne form.
EKSEMPEL 20 til 24:
Saltene av de sure forbindelsene med formel I fremstilles

Syreforbindelsen i løsning i et passende løsningsmiddel såsom f.eks. etanol, metylenklorid eller benzen nøytraliseres med en støkiometrisk mengde av en base såsom f.eks. natriumhydroksyd, karbonater og bikarbonater, aminalkylaminer og hydroksylaminer. Produktet som oppnås ved fordampning av løsningsmiddelet omkrystalliseres i et passende løsnings-middel eller blanding av løsningsmidler såsom f.eks. etanol, eter, benzen eller cykloheksan. Ved denne metode ble de følgende forbindelser .fremstilt: 20) natriumsalt av 3-metyl-4-benzoyl-6-(a-karboksyetyl)-tieno[2,3-b]pyrrol, 21) tert.butylaminsalt av 3-metyl-4-benzoyl-6-(a-karboksyetyl ) -tieno [ 2 , 3-b] pyrrol , smp.: 14 9°C. 22) dimetylaminoetanolsalt av 3-metyl-4-benzoyl-6(a-karboksyetyl ) -tieno [ 2 , 3-b] pyrrol , smp.: 112°C. 23) cholinsalt av 3-metyl-4-benzoyl-6(a-karboksyetyl)-tieno[2,3-b]pyrool, smp.: 155°C. 24) dimetylaminoetanolsalt av 3-metyl-4-ortoklorbenzoyl-6(a-karboksyetyl)-tieno[2,3-b]pyrrol, smp.: 101°C.
Claims (5)
1. Fremgangsmåte ved fremstilling av tieno[2,3-b]pyrrolderivater og enantiomerene derav med formel:
hvor
R betyr hydrogen, rettkjedet eller forgrenet alkyl med fra 1 til 5 karbonatomer,
R2 betyr:
- en cyanorest,
- en karboksyrest,
- en COOM gruppe hvor M betyr et alkalimetall, et jordalkalimetall, en ammoniumrest eller en mono-, di-, og tri- eller tetra-alkylammoniumrest eventuelt mono- eller poly- substituert med hydroksylrest,
- en COOR hvor R betyr en alkylrest med fra 1 til 5 karbonatomer, eller
- CO N^R „ gruppe hvori R' og R" som er like eller forskjellige hver betyr et hydrogenatom, eller rettkjedet eller forgrenet alkylrest med fra 1 til 5 karbonatomer;
R^ betyr en rettkjedet eller forgrenet alkylrest med fra 1 til 5 karbonatomer, og
R^ betyr en rettkjedet eller forgrenet alkylrest med fra 1 til 5 karbonatomer eller en fenylrest som er eventuelt mono- eller poly-substituert med et halogenatom, en alkyl eller alkoksyrest som hver har fra 1 til 5 karbonatomer,
en nitrorest eller en dialkylaminorest hvor hver alkylgruppe inneholder fra 1 til 5 karbonatomer og kan være rettkjedet eller forgrenet, karakterisert ved at:diazoderivatene med den generelle formel II:
hvor R har den ovenfor angitte betydning de-diazoteres til forbindelsen med den generelle formel III:

og den således erholdte forbindelse III acyleres ved Friedel-Craft reaksjonen ved hjelp av et acylhalogenid med formel X-CO-R4 hvor R^ har ovenfor angitte betydning og X betyr et halogenatom, til derivatet med den generelle formel IV:
hvor R3 og R^ har de forut angitte betydninger, derivatene IV hydrolyseres til forbindelsene med den generelle formel V:

hvilke forbindelser VI og så N-alkyleres ved hjelp av en-halogensyre eller et derivat, med formelen:

2. Fremgangsmåte ved fremstilling av forbindelser med formel I ifølge krav 1, karakterisert ved at dediazoteringen av derivatene II utføres ved oppvarming i et passende løsningsmiddel, om nødvendig under katalyse av reaksjonen ved spor av HC1, U.V. eller radi-kale initiatorer eller ved innvirkning av et reduksjons-middel .
3. Fremgangsmåte ifølge krav 1 og 2, karakterisert ved at acyleringen av forbindelsene III utføres i nærvær av en katalysator i et passende løsningsmiddel.
4. Fremgangsmåte ifølge krav 1 til 3, karakterisert ved at hydrolysen av forbindelsene IV utføres ved å oppvarme forbindelsene IV i en base-etanolblanding, etterfulgt av surgjøring.
5. Fremgangsmåte ifølge krav 1 til 4, karakter tL- sert ved at dekarboksyleringen av forbindelsene V utføres ved å oppvarme i et passende løsningsmiddel i nærvær av kobberpulver som katalysator.
