Det beskrives nye, aromatiske fluorglykosidderivater, legemidler som inneholder disse forbindelser og videre forbindelsens og legemidlenes anvendelse. Det beskrives særlig substituerte, aromatiske fluorglykosidderivater med formel I de restene har den angitte betydning, samt deres fysiologisk godtagbare salter og fremgangsmåter for deres fremstilling. Forbindelsene egner seg for eksempel som anti-diabetika.

Foreliggende oppfinnelse angår substituerte, aromatiske fluorglykosidderivativer, og deres fysiologisk godtakbare salter. Oppfinnelsen vedrører videre anvendelse av forbindelsene, legemidler inneholdende et eller flere slike forbindelser samt fremgangsmåte for fremstilling av legemidlet.
I litteraturen er det allerede kjent flere substansklasser med SGLT virkning. Alle disse strukturer hadde naturstoffet florizin som rettesnor. Fra dette ble følgende klasser avledet og hvis beskyttelsesretter er som angitt nedenfor: propiofenonglykosider fra Tanabe (WO 0280936, WO 0280935, JP 2000080041 og EP 850948)
2-(glukopyranosyloksy)benzylbenzener fra Kissei (WO 0244192, WO 0228872 ogWO 0168660)
glukopyranosyloksypyrazoler fra Kissei og Ajinomoto (WO 0268440,
WO 0268439, WO 0236602 og WO 0116147)
O-glykosidbenzamider fra Bristol-Myers Squibb (WO 0174835 og
WO 0174834) og C-arylglykosider fra Bristol-Myers Squibb (WO 0127128 og US 2002137903).
Alle de kjente strukturer inneholder glukose som meget viktig strukturelement.
Videre er diaiylsulfidforbindelser forbehandling av betennelses- og immunsykdommer kjent fra US 2002/132807.1 EP 0 953 357 Al beskrives generelt glykosidforbindelser som renalmedikamentbærer og i WO 95/23780 beskrives 4-hydroksyfenoksy-heterocykloalkylforbindelser som hudbleker.
Foreliggende oppfinnelse har til oppgave å tilveiebringe nye forbindelser som muliggjør forebyggelse eller behandling av diabetes type 1 og type 2. Det er nå overraskende funnet at aromatiske fluorglykosidderivater har denne virkning på SGLT. Disse forbindelser egner seg derfor for forebyggelse og behandling av diabetes type 1 og type 2.

Foreliggende oppfinnelse angår også forbindelser med formel I i form av racemater, racemiske blandinger og rene enantiomerer samt deres diastereomerer og blandinger derav.
Alkylrestene i substituentene R4, R5, R6, R7, R8 og R9 kan være rette eller forgrenet. Med halogen menes F, Cl, Br eller I, fortrinnsvis F eller Cl.
Farmasøytisk godtakbare salter er spesielt egnet for medisinske anvendelser på grunn av den høye vannoppløselighet i forhold til utgangs- hhv. basisforbindelsene. Saltene må oppvise et farmasøytisk akseptabelt anion eller kation. Egnede, farmasøytisk akseptable syreaddisjonssalter av forbindelsene ifølge oppfinnelsen er salter med uorganiske syrer som salt-, bromhydrogen-, fosfor-, metafosfor-, salpeter- eller svovelsyre samt organiske syrer som eddik-, benzensulfon-, benzo-, sitron-, etansulfon-, fumar-, glukon-, glykol-, isetion-, melke-, laktobion-, malein-, eple-, metansulfon-, rav-, p-toluensulfon-eller vinsyre.
Egnede, farmasøytisk akseptable basiske salter er ammoniumsalter, alkalimetallsalter, som natrium- eller kaliumsalter, jordalkalimetallsalter, som magnesium- eller kalsiumsalter, trometarnol (2-amino-2-hydroksymetyl-l,3-propanediol), dietanolamin, lysin eller etylendiarnin.
Salter med et ikke-farmasøytisk godtakbart anion, som for eksempel trifluoracetat ligger også innenfor oppfinnelsens ramme som nyttig mellomprodukt for fremstilling eller rensing av farmasøytisk godtakbare salter og/eller for anvendelse av ikke-terapeutisk, for eksempel in vitro anvendelser.
Forbindelsene ifølge oppfinnelsen kan også anvendes i form av promedikamenter, slike promedikarnenter beskrives for eksempel av T. Okada et al., Chem. Pharm. Bull. 1994, 42, 57-61. Slike promedikamenter kan metaboliseres in vivo til en forbindelse ifølge oppfinnelsen. Disse promedikamentene kan eventuelt være aktive per se. Foretrukket er karbonater på 6 posisjon av sukkeret (se WO 0280936 og WO 0244192), spesielt foretrukket er metylkarbonat og etylkarbonat.
Forbindelsene ifølge oppfinnelsen kan også foreligge i forskjellige, polymorfe former, for eksempel som amorfe eller krystallinske, polymorfe former. Alle polymorfe former av forbindelsene ifølge oppfinnelsen Ugger innen rammen av oppfinnelsen.
Nedenfor skal alle henvisninger til "forbindelsefr) i henhold til formel I" eller tilsvarende vise til en eller flere forbindelser med formel I som beskrevet ovenfor samt deres salter, og solvater som her beskrevet.
Forbindelsen(e) med formel (I) kan også administreres i kombinasjon med ytterligere aktive stoffer.
Foreliggende oppfinnelse omfatter videre et legemiddel, kjennetegnet ved at det inneholder en eller flere forbindelser som omtalt ovenfor.
Mengden av en forbindelse i henhold til formel I, som er nødvendig for å oppnå den ønskede, biologiske effekt, er avhengig av en rekke faktorer, for eksempel den valgte, spesifikke forbindelse, den tilsiktede anvendelse, typen administrering og pasientens kliniske tilstand. Generelt ligger dagsdosen i området 0,3 mg til 100 mg, karakteristisk 3 mg til 50 mg, pr. dag pr. kilo kroppsvekt, for eksempel 3-10 mg/kg/dag. En intravenøs dose kan for eksempel ligge i området 0,3 mg til 1,0 mg/kg, en dose som er egnet som en infusjon kan administreres i en mengde på 10 ng til 100 ng per kilogram og per minutt. Egnede infusjonsoppløsninger for dette formål kan for eksempel inneholde fra 0,1 ng til 10 mg og karakteristisk 1 ng til 10 mg, per milliliter. Enkeltdose kan for eksempel inneholde fra 1 mg til 10 g av den aktive bestanddel. Derved kan ampuller for injeksjon for eksempel inneholde 1 mg til 100 mg, og oralt administrerbare enkeltdosefbrmuleringer, som tabletter eller kapsler, kan for eksempel inneholde 1,0 til 1000 mg, og karakteristisk 10 til 600 mg. For behandling av de ovenfor nevnte tilstander kan forbindelsene i henhold til formel I anvendes som forbindelse per se, fortrinnsvis foreligger de dog sammen med en godtakbar bærer i form av et farmasøytisk preparat. Bæreren må naturligvis også være godtakbar dit hen at den er kompatibel med de andre bestanddeler i preparatet og ikke er sunnhetsskadelig for pasienten. Bæreren kan være et faststoff eller en væske eller begge deler og formuleres fortrinnsvis sammen med forbindelsen som enkeltdose, for eksempel som tablett, som kan inneholde fra 0,05 til 95 vekt-% aktiv bestanddel. Ytterligere farmasøytisk aktive stoffer kan likeledes være tilstede, inkludert ytterligere forbindelser med formel I. De farmasøytiske preparatene ifølge oppfinnelsen kan fremstilles i henhold til en av de kjente, farmasøytiske metoder som i det vesentlige består i at bestanddelene blandes med farmakologisk godtakbare bærer- og/eller hjelpestoffer.
Ifølge oppfinnelsen fremstilles legemidlet ved at den aktive bestanddel blandes med en farmasøytisk egnet bærer og at blandingen bringes til en form egnet for administrering.
De farmasøytiske preparatene ifølge oppfinnelsen er slike som er egnet for oral, rektal, topisk, peroral (for eksempel sublingual) og parenteral (for eksempel subkutan, intermuskulær, intradermal eller intravenøs) administrering eller, selv om den best egnede aammistreringsmåte bestemmes i hvert enkelt tilfelle avhengig av type og alvor av tilstanden som behandles og typen av den i hvert tilfelle anvendte forbindelse i henhold til formel I. Også dragerte formuleringer og dragerte retardformuleringer ligger innenfor rammen av oppfinnelsen. Foretrukket er syre- og mavesaftresistente formuleringer. Egnede, magesaftresistente belegg omfatter celluloseacetatftalat, polyvinylacetatftalat, hydroksypropylmetylcelluloseftalat og anioniske polymerer av metakrylsyre og metylmetacrylat.
Egnede farmasøytiske forbindelser for oral administrering kan foreligge i separate enheter som kapsler, oblatkapsler, sugetabletter eller tabletter som i hvert tilfelle inneholder en bestemt mengde av forbindelsen med formel I; som pulver eller granulat; som oppløsninger eller suspensjoner i en vandig eller ikke-vandig væske; eller som en olje-i-vann- eller vann-i-oljeemulsjon. Disse preparater eller blandinger kan, som allerede nevnt, tilberedes i henhold til en hvilken som helst egnet, farmasøytisk metode som omfatter et trinn der den aktive bestanddel og bæreren (som kan bestå av en eller flere ytterligere bestanddeler) bringes i kontakt. Generelt blir preparatet fremstilt ved jevn og homogen blanding av aktive bestanddeler med en flytende og/eller finfordelt fast bærer hvoretter produktet, hvis ønskelig eller nødvendig, formgis. Således kan det for eksempel fremstilles en tablett idet et pulver eller granulat av forbindelsen presses eller formes, eventuelt sammen med en eller flere ytterligere bestanddeler. Pressede tabletter kan fremstilles ved tablettering av forbindelsen i frittrislende form som for eksempel et pulver eller granulat, eventuelt blandet med et bindemiddel, glidemiddel, en inert fortynner og/eller (ett eller flere) overflateaktive/dispergerende midler i en egnet maskin. Formgitte tabletter kan fremstilles ved forming av det pulverformige og med et inert, flytende fortynningsmiddel fuktede forbindelse i en egnet maskin.
Farmasøytiske preparater som er egnet for en peroral (sublingual) administrering omfatter sugetabletter som inneholder en forbindelse med formel I sammen med et smaksstoff, vanligvis sakkarose og gummiarabikum eller tragakantgummi og pastiller, som omfatter forbindelsen i en inert basis som gelatin eller glyserin eller sakkarose og gummiarabikum.
Egnede, farmasøytiske preparater for parenteral administrering omfatter fortrinnsvis sterile, vandige tilberedninger av en forbindelse med formel I som fortrinnsvis er isotonisk med blodet hos den tilsiktede mottager. Disse preparater administreres fortrinnsvis intravenøst, selv om administreringen også kan skje subkutant, intramuskulært eller intradermalt som injeksjon. Disse preparater kan fortrinnsvis fremstilles ved at forbindelsen blandes med vann og den således oppnådde oppløsning gjøres steril og isotonisk med blodet. Injiserbare preparater ifølge oppfinnelsen inneholder generelt fra 0,1 til 5 vekt-% av den aktive forbindelse.
Egnede farmasøytiske preparater for rektal administrering foreligger fortrinnsvis som enkeltdosetapper. Disse kan fremstilles ved at en forbindelse av formel I blandes med en eller flere konvensjonelle, faste bærere, som kakaosmør og den dannede blanding så gis egnet form.
Egnede, farmasøytiske preparater for topisk anvendelse på huden foreligger fortrinnsvis som salve, krem, lotion, pasta, spray, aerosol eller olje. Som bærer kan det anvendes vaselin, lanolin, polyetylenglykoler, alkoholer eller kombinasjoner av to eller flere av disse stoffer. Den aktive bestanddel foreligger generelt i en konsentrasjon fra 0,1 til 15 vekt-% av preparatet, for eksempel 0,5 til 2%.
Også en transdermal administrering er mulig. Egnede, farmasøytiske preparater for transdermal anvendelse kan foreligge som enkeltplaster som er egnet for en lang, nær kontakt med pasientens epidermis. Slike plastere inneholder på egnet måte den aktive bestanddel i en eventuelt bufret, vandig oppløsning, oppløst og/eller dispergert i et vedheftingsmiddel eller dispergert i en polymer. En egnet aktiv stoffkonsentrasjon er for eksempel 1 til 35% og fortrinnsvis ca. 3 til 15%. Som en spesiell mulighet kan den aktive bestanddel settes fri ved elektrotransport eller iontoforese, som for eksempel beskrevet i Pharmaceutical Research, 2(6): 318 (1986).

De viste skjemaer for fremgangsmåtene A-F taler for seg selv og fagmannen kan utføre disse som de står. Ytterligere enkeltheter er dog gitt i den eksperimentelle del. I henhold til fremgangsmåtene A-F oppnås forbindelsene i eksemplene 1 til 24. Andre forbindelser med formel I kan oppnås på tilsvarende måte eller i henhold til kjente metoder.
Forbindelsen(e) med formel I kan også adniinistreres i kombinasjon med andre aktive bestanddeler.
Som ytterligere aktive stoffer for kombinasjonspreparater egner seg:
Alle antidiabetika som er nevnt i "Rote Liste" 2001, kapittel 12. Disse kan kombineres med forbindelsene ifølge oppfinnelsen med formel I, særlig for synergistisk virkningsforbedring. Amninistreringen av aktivstoffkombinasjoner kan skje enten ved separat administrering av aktivbestanddelene til pasientene eller i form av kombinasjonspreparater der flere aktive bestanddeler foreligger i et farmasøytisk preparat. De fleste av de nedenfor anførte aktive bestanddeler er beskrevet i "USP Dictionary of US AN og International Drug Names", US Pharmacopeia, Rockville 2001.
Antidiabetika omfatter insulin og insulinderivater, som for eksempel Lantus<®>(se www.lantus.com) eller HMR1964, hurtigvirkende insuliner (se US 6,221,633), GLP-1 derivater som for eksempel de i WO 98/08871 i navnet Novo Nordisk A/S, samt oralt virksomme, hypoglykemiske aktive bestanddeler.
De oralt virksomme, hypoglykemiske aktive bestanddeler omfatter fortrinnsvis sulfonylurear, biguamdiner, meglitimder, oksadiazolidindioner, tiazolidindioner, glukosidaseinhibitorer, glucagonantagonister, GLP-1 agonister, kandiumkanalåpnere, som for eksempel de som er beskrevet i WO 97/26265 og WO 99/03861 i navnet Novo Nordisk A/S, insulinsensitiserere, inhibitorer av leverenzymer som er involvert i stimuleringen av glukonogenese og/eller glykogenolyse, modulatorer av glukoseopptaket, fettstoffskifteforandrende forbindelser som antihyperlipidemisk aktivbestanddeler og antilipidemisk aktivbestanddeler, forbindelser som reduserer næringsinntaket, PP AR og PXR agonister og aktivbestanddeler som virker på den ATP-avhengige kaliumkanal av betaceller.
Forbindelsene med formel I kan for eksempel administreres i kombinasjon med en HMGCoA reduktaseinhibitor som simvastatin, fluvastatin, pravastatin, lovastatin, atorvastatm, cerivastatin, rosuvastatin.
Videre kan forbindelsene med formel I administreres i kombinasjon med en kolesterolresepsjonsinhibitor, som for eksempel ezetimib, tiquesid, pamaquesid.
Forbindelsene med formel I kan også administreres i kombinasjon med en PP AR gamma agonist, som for eksempel rosiglitazon, pioglitazon, JTT-501, GI 262570.
Forbindelsene med formel I kan videre administreres i kombinasjon med en PP AR alfa agonist, som for eksempel GW 9578, GW 7647.
Videre kan forbindelsene med formel I administreres i kombinasjon med en blandet PP AR alfa/gamma agonist, som for eksempel GW 1536, AVE 8042, AVE 8134, AVE 0847, AVE0897 eller som beskrevet i WO 00/64888, WO 00/64876, WO 03/20269.
Forbindelsene med formel I kan også administreres i kombinasjon med et fibrat, som for eksempel fenofibrat, clofibrat, bezafibrat.
Videre kan forbindelsene med formel I administreres i kombinasjon med en MTP inhibitor, som for eksempel implitapid, BMS-201038, R-103757.
Videre kan forbindelsene med formel I administreres i kombinasjon med en gallesyreresorpsjonsinhibitor (se for eksempel US 6,245,744 eller US 6,221,897), som for eksempel HMR 1741.
Forbindelsene med formel I kan også administreres i kombinasjon med en CETP inhibitor, som for eksempel JTT-705.
Forbindelsene med formel I kan likeledes administreres i kombinasjon med en polymer gallesyreadsorber, som for eksempel kolestyramin, kolesevelam.

Forbindelsene med formel I kan videre administreres i kombinasjon med en aktiv bestanddel som virker på den ATP-avhengige kaliumkanal av betaceller, som for eksempel tolbutamid, glibenclamid, glipizid, glimepirid eller repaglinid.
Forbindelsene med formel I kan videre adrninistreres i kombinasjon med mer enn en av de ovenfor nevnte forbindelser, for eksempel i kombinasjon med et sulfonylurea og metformin, et sulfonylurea og akarbose, repaglinid og metformin, insulin og et sulfonylurea, insulin og metformin, insulin og troglitazon, insulin og lovastatin, og så videre.

Kombinasjonen med Caromax<®>kan skje i ett preparat eller ved separat administrering av forbindelsene med formel I og Caromax<®>. Caromax<®>kan derved også administreres i form av næringsmidler, som for eksempel i bakevarer eller i muesliprodukter. Forbindelsene med formel I utmerker seg ved gunstig virkning på glukosestoffskifte, de reduserer særlig blodsukkernivået og er egnede for behandling av diabetes type I og type II. Forbindelsene kan derfor anvendes alene eller i kombinasjon med ytterligere blodsukkersenkende aktive bestanddeler (antidiabetika).
Foreliggende oppfinnelse omfatter følgelig også anvendelse av forbindelsene omtalt ovenfor for fremstilling av et medikament for behandling av diabetes type 1 og type 2.
Videre omfatter oppfinnelsen anvendelse av forbindelsene omtalt ovenfor for fremstilling av et medikament for blodsukkersenkinng.
Forbindelsene med formel I egner seg videre for forebyggelse og behandling av diabetiske senskader, som nefropati, retinopati, neuropati samt syndrom X, fedme, hjerteinfarkt, myokardialt infarkt, perifere, arteri elle lukkesykdommer, tromboser, arteriosklerose, betennelser, immunsykdommer, autoimmunsykdommer, som for eksempel AIDS, astma, osteoporose, kreft, psoriasis, Alzheimer's, schizofreni og infeksjonssykdommer, fordelaktig er behandling av diabetes type 1 og type 2 samt forebyggelse og behandling av diabetisk senskader, syndrom X og fedme.
Virksomheten for forbindelsene ble testet som følger:
Preparering av børstesømmembranvesikler fra tynntarm fra kaniner, rotter og svin.
Prepareringen av børstesømmembranvesikler fra tarmcellene fra tynntarmen skjedde med den såkalte Mg<2+>presipiteringsmetode. Shmhinne fra tynntarmen ble skrapet av og suspendert i 60 ml iskald Tris/HCI buffer (pH 7,1)/300 mM mannitol, 5 mM EGTA. Etter fortynning til 300 ml med iskald, destillert vann ble det hele homogenisert med en Ultraturrax (18 stav, JKA Werk Staufen, FRG) ved 75% maksimal ytelse i 2 x 1 minutt under isavkjøling. Etter tilsetning av 3 ml IM MgCl2oppløsning (sluttkonsentrasjon 10 mM), ble det hele satt hen nøyaktig 15 minutter ved 0°C. Tilsetning av Mg<2>"<1->aggregerte cellemembranene og presipiterte dem med unntak av børstesømmembranene. Etter en 15 minutters senrrifugering ved 3 000 x g (5 000 omdreininger, SS-34 rotor) ble presipitatet kastet og supernatanten som inneholdt børstesømmembranene sentrifugert i
30 minutter ved 26 700 x g (15 000 omdreininger, SS-34 rotor). Supernatanten ble kassert og presipitatet rehomogenisert i 60 ml 12 mM Tris/HCI buffer (pH 7,l)/60 mM mannitol, 5 mM EGTA med en Potter Elveihem homogenisator (Braun, Melsungen, 900 omdreininger, 10 slag). Etter tilsetning av 0,1 ml IM MgCl2oppløsning og 15 minutters inkubering ved 0°C ble det sentrifugert igjen i 15 minutter ved 3 000 x g. Supernatanten ble deretter sentrifugert nok en gang i 30 minutter ved 46 000 x g (20 000 omdreininger, SS-34 rotor). Presipitatet ble tatt opp i 30 ml 20 mM Tris/Hepes buffer (pH 7,4)/280 mM mannitol og resuspendert homogent ved 20 slag i en Potter Elveihem homogenisator ved 1 000 omdreininger pr. minutt. Etter 30 minutters sentrifugering ved 48 000 x g (20 000 omdreininger, SS-34 rotor) ble presipitatet tatt opp i 0,5 til 2 ml Tris/Hepes buffer (pH 7,4)/280 mM mannitol (sluttkonsentrasjon 20 mg/ml) og resuspendert ved hjelp av en tuberkulinsprøyte med en 27 gauge nål.
Vesiklene ble enten anvendt umiddelbart etter preparering for markerings- eller transportundersøkelser eller oppbevart ved -196°C i 4 mg porsjoner i flytende nitrogen.
For preparering av børstesømmembranvesiklene fra rottetynntarm ble 6 til 10 Wistar-hannrotter (avlet ved Kastengrund, Aventis Pharma) avlivet ved servikal dislokasjon, tynntarmen ble tatt ut og vasket med kald, isotonisk kokesaltoppløsning. Tarmene ble kuttet opp og slimhinnen skrapet av. Opparbeidingen for isolering av børstesømmembranene skjedde som beskrevet ovenfor. For separering av cytoskjelettandelene ble børstesømmembranvesiklene fra rottetynntarm behandlet med KSCN som kaotropt ion.
For preparering av børstesømmembraner fra kanintynntarm ble kaninene avlivet ved intravenøs injeksjon av 0,5 ml av en vandig oppløsning av 2,5 mg tetrakain HC1,
100 mg m-butramid og 25 mg mebezoniumjodid. Tynntarmene ble tatt ut vasket med iskald, fysiologisk læksdtoppløsning og frosset inn i plastposer under nitrogen ved -80°C og lagret 4 til 12 uker. For preparering av membranvesiklene ble de frosne tarmer tint opp ved 30°C i et vannbad og deretter ble slimhinnen skrapet av. Opparbeiding av membranvesiklene skjedde som beskrevet ovenfor.
For preparering av børstesømmembranvesiklene fra svinetarm ble jejunumsegmenter fra et nyslaktet svin vasket med iskald, isotonisk koksaltoppløsning og frosset inn i plastposer under nitrogen ved -80°C. Preparering av membranvesiklene skjedde som beskrevet ovenfor.
Preparering av børstesømmembranvesikler fra nyrekorteks fra rottenyre.
Preparering av børstesømmembranvesikler fra korteks fra rottenyrer skjedde i henhold til metoden ifølge Biber et al. Nyrene fra 6 til 8 rotter (200 til 250 g) ble tatt ut og fra hver nyre hentet man korteks som et ca. 1 mm tykt sjikt. Nyrene ble tatt opp i 30 ml iskald 12 mM Tris/HCI buffer (pH 7,4)/300 mM mannitol og homogenisert under isavkjøling i 4 x 3 sekunder med en Ultraturrax stav (trinn 180 volt). Etter tilsetning av av 42 ml iskaldt, destillert vann ble det tilsatt 850 [ d av en IM MgCk oppløsning. Etter 15 minutters inkubering ved 0°C ble det sentrifugert i 15 minutter ved 4 500 omdreininger pr. minutt (Sorvall SS-34 rotor). Bunnfallet ble kassert og supernatanten sentrifugert i 30 minutter ved 16 000 omdreininger pr. minutt. Etter resuspendering av bunnfallet i 60 ml 6 mM Tris/HCI buffer (pH 7,4)/150 mM mannitol/2,5 mM EGTA ved 10 slag i en Potter-Elveihem homogenisator (900 omdreininger pr. minutt) ble det hele inkubert i 15 minutter ved 0°C etter uUsetning av 720 ul 1 mM MgCfe oppløsning. Etter 15 rninutters sentrifugering ved 4 500 omdreininger pr. minutt (SS-34 rotor) ble den resulterende supematant sentrifugert i 30 minutter ved 16 000 omdreininger pr. minutt. Supernatanten ble homogenisert ved 10 slag i 60 ml 20 mM Tris/Hepes buffer (pH 7,4)/280 mM mannitol, og den dannede suspensjon deretter sentrifugert i 30 minutter ved 20 000 omdreininger pr. minutt. Bunnfallet ble resuspendert ved hjelp av en tuberkulinsprøyte med en 27 gauge nål i 20 mM Tris/HCI buffer (pH 7,4)/280 mM mannitol og innstilt til en proteinkonsentrasjon på 20 mg/ml.
Måling av glukoseopptaket ved børstesømmembranvesikler.
Opptaket av [ l4C]-markert glukose i børstesømmembranvesiklene ble målt ved hjelp av membranfiltreringsmetoden. 10 ul av børstesømmembranvesikkelsuspensjonen i 10 mM Tris/Hepes buffer (pH 7,4)/300 mM mannitol ble ved 30°C satt til 90 ul av en oppløsning av 10 uM [<14>C]D-glukose og den tilsvarende konsentrasjon av den angjeldende inhibitor (5-200 uM) i 10 mM Tris/Hepes buffer (pH 7,4)/100 mM NaCl/100 mM mannitol.
Etter 15 sekunders inkubering ble transportprosessen stanset ved tilsetning av 1 ml iskald stoppoppløsning (10 mM Tris/Hepes buffer (pH 7,4)/150 mM KC1) og vesikkelsuspensjonen umiddelbart sugd av ved et undertrykk på 25 til 35 mbar over et membranefilter av cellulosenitrat (0,45 um, 25 mm diameter, Schleicher & Schtill). Filtratet ble vasket med 5 ml iskald stoppoppløsning. Hvert målepunkt ble utført som dobbel- eller trippelbestemmelse. For måling av opptaket av radioaktivt merket substrat ble membranfilteret oppløst i 4 ml av en tilsvarende scintillator (Quickszint 361, Zinsser Analytik GmbH, Frankfurt am Main), og radioaktiviteten bestemt ved væskescintillasjonsmåling. De målte verdier ble, etter justering av apparatet ved hjelp av standardprober og etter korrektur av eventuell tilstedeværende kjemilurninescens, oppnådd som dpm (desintegrasjon per minutt).
Aktivitetssaminenligningen for de aktive bestanddelene ble gjennomført ved hjelp av IC50data som ble oppnådd i transportanalyse på lynntarm-børstesømmembranvesikler for kaniner for valgte substanser (absoluttverdiene kan være specie- og forsøksavhengige)
Nedenfor skal fi-emstilling av forskjellige eksempler beskrives detaljert, de øvrige forbindelser med formel I ble oppnådd på analog måte:

Reaksjonsskjema: Syntese av a-bromoglykosider 1 -Bromo-4-deoksy-4-fluor-2,3,6-tri-0-acetyl-alfa-D-glukose 2 5 g (27,5 mmol) 4-deoksy-4-fluor-D-glukopyranose 1 (Apollo) ble suspendert i 50 ml pyridin og 50 ml eddiksyreanhydrid. Reaksjonsoppløsningen ble omrørt ved 45°C i 4 timer. Derved oppnådde man en klar reaksjonsoppløsning som så ble dampet inn. Man oppnådde 12 g råprodukt. Dette råproduktet ble oppløst i 160 ml 33% HBr i iseddiksyre og satt hen i 2 timer ved romtemperatur. Reaksjonsoppløsningen ble så helt i en blanding av 300 g is og 300 ml etylacetat. Den organiske fase ble vasket to ganger med vandig NaCl oppløsning, filtrert over litt kieselgel og dampet inn. Resten ble separert ved kromatografi på kieselgel med etylacetat:heptan =1:1). Man oppnådde 8,19 g (80% over 2 trinn) 2 som lysegult faststoff. l-Bromo-4-deoksy-4-fluor-2,3,6-tri-0-acetyl-alfa-D-gaIaktose 4 100 mg (0,55 mmol) 3 ble analogt fremstillingen av forbindelse 2 omsatt med 3,5 ml pyridin og 3,5 ml eddiksyreanhydrid. Man oppnådde 89 mg (44%) 4 som amorft faststoff. l-Bromo-3-deoksy-3-fluor-2,4,6-tri-0-acetyl-alfa-D-glukose 6 335 mg (1,84 mmol) 5 ble analogt fremstillingen av forbindelse 2 omsatt med 10 ml pyridin og 10 ml eddiksyreanhydrid. Man oppnådde 628 mg (92%) 6 som amorft faststoff. Eksempel 1 (forbindelse 9)
100 mg (0,47 mmol) 2-(4-metoksybenzyl)fenol 7 og 370 mg (1,17 mmol) bromid 2 ble oppløst i 6 ml metylenklorid. Til denne oppløsning ble det etter hverandre satt 160 mg BusBnNCl (PTC = faseoverføringskatalysator), 320 mg K2C03og 0,4 ml vann og det hele omrørt i 20 timer ved romtemperatur. Reaksjonsoppløsningen ble fortynnet med 20 ml etylacetat og filtrert over kieselgel. Filtratet ble dampet inn og resten separert ved kromatografi på kieselgel (etylacetat/heptan = 1/1). Man oppnådde 72 mg 8 som et fargeløst faststoff. De oppnådde 72 mg 8 ble tatt opp i 4 ml metanol, og tilsatt 1 ml IN NaOMe/MeOH. Etter en time ble det hele nøytralisert med metanolisk HC1 og dampet inn og resten separert ved kromatografi på kieselgel (metylen

1,0 g (6,0 mmol) l-(2,6-dihydroksy-4-metylfenyl)etanon 17 og 1,0 g (2,7 mmol) bromid 2 ble oppløst i 30 ml metylenklorid. Til denne oppløsning ble det under kraftig omrøring etter hverandre tilsatt 800 mg benzyltributylammoniurnklorid (PTC), 1,6 g kaliumkarbonat og 1,5 ml vann. Denne suspensjonen omrøres under beskyttelse mot lys (aluminiumsfolie) i 18 timer og fortynnes så med 150 ml etylacetat og 150 mln-heptan. De faste bestanddeler filtreres fra over litt kieselgel og det hele dampes inn. Resten separeres ved kromatografi på kieselgel (etylacetat/heptan = 1/2). Det ble oppnådd 430 mg 18, vanskelig separerbart fra et like godt eluerende biprodukt, derfor kun ca. 50% renhet. Biproduktet kan lett separeres i det neste trinn.

200 mg forbindelse 18 (renhet rundt 50%) og 225 mg anisaldehyd (Fluka) ble oppløst i 10 ml metanol. Etter tilsetning av 5 ml IN NaOMe/MeOH oppløsning lar man reaksjonsoppløsningenkoke under refluks i 12 timer. Reaksjonsoppløsningen
nøytraliseres med metanolisk HC1, dampes inn og resten separeres ved kromatografi på kieselgel (metylen klorid/metanol/konsentrert ammoniakk, 30/5/1). 60 mg av 19 ble oppnådd som et gult faststoff. 60 mg (0,13 mmol) kalkon 19 og 50 mg Pd/C (10% Pd) ble suspendert i 15 ml metanol og hydrert under en 5 bar hydrogenatmosfære ved romtemperatur i 5 timer. Reaksjonsoppløsningen ble dampet inn og resten renset ved flasKkromatografi (metylen klorid/metanol/konsentrert ammoniakk, 30/5/1).

3,46 g (10 mmol) l-(2,6-bisbenzyloksy-4-metylfenyl)etanon (36) ble oppløst i 150 ml etanol, og 1,34 ml p-anisaldehyd ble tilsatt. Deretter ble det dryppet til 7 ml vandig kaliunmydroksidoppløsning. Reaksjonen ble omrørt i 12 timer ved romtemperatur. Halvparten av oppløsningsmidlet ble så trukket av på en rotasjonsfordamper. l-(2,6-Dihydroksy-4-metylfenyl)-3-(4-metoksyfenyl)propan-l -on 38
1,50 g (3.23 mmol) l-(2,6-bisbenzyloksy-4-metylfenyl)etanon (37) ble oppløst i 40 ml eddiksyreetylester og hydrert under argon med 400 mg 10% palladium på aktiv kull i en hydreringsautoklav ved 3 bar og romtemperatur i 2 timer. Katalysatoren ble så rrafiltrert, det hele vasket med eddiksyreetylester og den oppnådde oppløsning dampet inn på en rotasjonsfordamper. Råproduktet ble renset ved søylekromatografi over Si02med etylacetat/n-heptan 1:3. Det ble isolert 600 mg av produkt 38 (65%) som et fargeløst faststoff.

174,4 mg (0,61 mmol) av forbindelse 38 ble oppløst i 50 ml toluen, og det ble tilsatt 340 mg (0,61 mmol) av bromid 60 og 421 mg kadmiumkarbonat (2,44 mmol). Etter 3 timer ved romtemperatur ble det fortynnet med 300 ml eddikester/n-heptan (1:1) og vasket 1 x med NaHC03oppløsning og 1 x med Na2S203oppløsning. Den organiske fase ble filtrert over kieselgel og dampet inn. Resten ble separert ved kromatografi på kieselgel (etylacetat/n-heptan 1:1). Det ble oppnådd 2,9 g (79%) keton. Dette ble oppløst i 30 ml metylenklorid, under argon ble det tildryppet 4 ml BAST (Aldrich). Etter 20 timer ved romtemperatur ble det fortynnet med 200 ml eddikester og det ble vasket forsiktig med kaldNaHC03oppløsning (sterk brusing). Den organiske fase ble filtrert over kieselgel og dampet inn. Resten ble separert ved kromatografi på kieselgel (etylacetat/n-heptan 1:1). 2,6 g (85%) av 45 ble oppnådd som fargeløst olje. 4-Deoksy-4,4-dilfuor-l,2,3,6-tetra-0-acetyl-aIfa-D-glnkose 46
2,3 g (4,7 mmol) 45 og 2 g Pd/C (10% Pd) ble oppløst i 150 ml metanol og 10 ml eddiksyre og hydrert under en 5 bar hydrogenatomsfære i 16 timer ved romtemperatur.
Reaksjonsoppløsningen ble inndampet og resten renset ved flashkromatografi (metylen klorid/metanol/konsentrert ammoniakk, 30/5/1). Det ble oppnådd et utbytte på 850 mg (83%) l-metoksy-4-deoksy-4,4-difluor-alfa-D-glukose som et hvitt amorft faststoff. C7H12F2O5(214.17) MS(DCI): 215.4 (M + H<4>).
Av dette ble 700 mg (3,3 mmol) oppløst i 3,5 ml eddiksyre og 6,3 ml eddiksyreanhydrid. Etter tilsetning av 0.2 ml konsentrert H2SC>4 ble det hele omrørt i 5 timer ved 60°C. Reaksjonsoppløsningen ble så helt på en blanding av 30 g is og 30 ml etylacetat. Den organiske fase ble vasket to ganger til med vandig NaCl oppløsning, filtrert over litt kieselgel og dampet inn. Alternativt kan oksidasjonen skje med Jones reagens:
155 mg (4-metoksyfenyl)(2-metoksyfenyl)metanol 51 ble oppløst i 10 ml aceton, og 2 ml Jones reagens ble tilsatt dråpevis. Etter 2 timer ved romtemperatur ble det tilsatt 501 MTB eter og 30 ml vann. Den organiske fase ble vasket flere ganger med vann, den organiske fase ble ekstrahert med mettet NaCl oppløsning, det hele ble tørket over natriumsulfat og dampet inn til tørr tilstand. Det således oppnådde produkt i en mengde av 126 mg har en tilstrekkelig renhet for den videre anvendelse.

2,61 g (4-metoksyfenyl)(2-metoksyfenyl)metanon 52 ble oppløst i diklormetan. Satsen ble avkjølt i et isbad og det ble tilsatt 3,71 g borontribrornid-dimetylsulfidkompleks. 2-(4-Etylfenoksy)fenol 71
710 mg 2-(4-etylfenoksy)metoksybenzen 70 ble oppløst i 5 ml absolutt diklormetan. 0,6 ml borontribromid (1 M diklormetan) ble tildryppet og oppløsningen ble omrørt i 6 timer.
