Oppfinnelsens felt
Denne oppfinnelse er innenfor feltet for medisinsk kjemi og vedrører okso-2H-pyridin-, okso-2H-kinolin- og okso-lH-isokinolinderivater som angitt i krav 1 og farmasøytiske sammensetninger derav, som hemmer caspaser som medierer celleapoptose og inflammasjon. Oppfinnelsen vedrører også anvendelse av forbindelsene for fremstilling av et medikament for å behandle eller forebygge sykdommer hvor caspaseaktivitet er implisert. Videre vedrører oppfinnelsen en fremgangsmåte ved fremstilling av forbindelsene.
Oppfinnelsens bakgrunn
Apoptose, eller programmert celledød, er en prinsipiell mekanisme ved hvilken organismer eliminerer uønskede celler. Dereguleringen av apoptose, enten usedvanlig stor apoptose eller den manglende evne til å gjennomgå den, har blitt implisert i flere sykdommer slik som cancer, akutte inflammatoriske og autoimmune forstyrrelser, iskemiske sykdommer og visse nevrodegenerative forstyrrelser (se generelt Science, 1998, 281, 1283-1312; Ellis et al., Ann. Rev. Cell. Biol., 1991, 7, 663).
Caspaser er en familie av cystein-protease-enzymer som er nøkkelmediatorer i signalveiene for apoptose og celledisassemblering (Thornberry, Chem. Biol., 1998, 5, R97-R103). Disse signalveier varierer avhengig av celletype og stimulus, men alle apoptoseveier synes å konvergere ved en felles effektorvei som fører til proteolyse av nøkkelproteiner. Caspaser er involvert i både effektorfasen av signalveien og vider oppstrøm ved dens initiering. Oppstrøms caspaser involvert i initieringshendelser blir aktivert og aktiverer i sin tur andre caspaser som er involvert i de senere faser av apoptose.
Caspase-1, den første identifiserte caspase, er også kjent som interlevkinkonverterende enzym eller "ICE." Caspase-1 konverterer forløper interlevkin-ip ("pIL-iP") til den pro-inflammatoriske aktive form ved spesifikk spalting av pIL-ip mellom Asp-116 og Ala-117. Foruten caspase-1 er det også elleve andre kjente humane caspaser, hvorav alle spaltes spesielt ved aspartylresiduer. De er også observert å ha strenge krav til minst fire aminosyreresiduer på den N-terminale side av spaltingsstedet.
Caspasene har blitt klassifisert i tre grupper avhengig av aminosyresekvensen som er foretrukket eller primært kjent igjen. Gruppen av caspaser, som inkluderer caspaser 1, 4, 5 og 13, har blitt vist å foretrekke hydrofobe aromatiske aminosyrer i posisjon 4 på den N-terminale side av spaltingsstedet. En annen gruppe som inkluderer caspaser 2, 3 og 7, kjenne igjen aspartylresiduer i både posisjon 1 og 4 på den N-terminale side av spaltingsstedet, og fortrinnsvis en sekvens av Asp-Glu-X-Asp. En tredje gruppe, som inkluderer caspaser 6, 8, 9 og 10, tolererer mange aminosyrer i den primære gjenkjennelsessekvens, men synes å foretrekke residuer med forgrenede, alifatiske sidekjeder slik som valin og leucin i posisjon 4.
Caspasene har også blitt gruppert i henhold til deres oppfattede funksjon. Den første subfamilie består av caspaser-1 (ICE), 4, 5 og 13. Disse caspaser har blitt vist å være involvert i proinflammatorisk cytokinprosessering og spiller derfor en viktig rolle i inflammasjon. Caspase-1, det mest studerte enzym i denne klasse, aktiverer IL-ip-forløperen ved proteolytisk spalting. Dette enzym spiller derfor en nøkkelrolle i den inflammatoriske respons. Caspase-1 er også involvert i prosesseringen av interferon-y-induserende faktor (IGIF) som stimulerer produksjonen av interferon-gamma, en nøkkelimmunoregulator som modulerer antigen-presentasjon, T-celleaktivering og celleadhesjon.
Resten av caspasene utgjør den andre og tredje subfamilie. Disse enzymer er av sentral viktighet i de intracellulære signalveier som fører til apoptose. En subfamilie består av enzymene involvert i initiering av hendelser i den apoptotiske vei, inklusive transduksjon av signaler fra plasma-membranen. Medlemmer av denne subfamilie inkluderer caspaser-2, 8, 9 og 10. Den andre subfamilie, bestående av effektorcapsasene 3, 6 og 7, er involvert i de siste nedstrøms spaltingshendelser som resulterer i den systematiske nedbrytning og død av cellen ved apoptose. Caspaser involvert i oppstrøms signaltransduksjonen aktiverer nedstrøms-caspasene, som deretter ødelegge DNA-reparasjonsmekanismer, fragment DNA, tar fra hverandre celle cytoskjelettet og til sist fragmenterer cellen.
Kunnskap om fire-aminosyresekvensen primært kjent igjen av caspasene har blitt anvendt for å designe
caspaseinhibitorer. Reversible tetrapeptidinhibitorer har blitt fremstilt som har strukturen CH3CO-[P4]-[P3]-[P2]-CH(R)CH2C02H hvor P2 til P4 representerer en optimal aminosyre-gjenkjennelses-sekvens og R er et aldehyd, nitril eller keton i stand til å binde til caspase-cystein-sulfhydrylen. Irreversible inhibitorer basert på den analoge tetrapeptid gjenkjennelsessekvens har blitt fremstilt hvor R er et acyloksymetylketon -COCH2OCOR' . R' er eksemplifisert ved en eventuelt substituert fenyl slik som 2,6-diklorbenzoyloksy og hvor R er COCH2X hvor X er en utgående gruppe slik som F eller Cl. Anvendbarheten til caspaseinhibitorer for å behandle en variasjon av pattedyrs sykdomstilstander assosiert med en økning i cellulær apoptose har blitt demonstrert ved å anvende peptidiske caspaseinhibitorer. For eksempel har caspaseinhibitorer i gnagermodeller blitt vist å redusere infarktstørrelse og hemme kardiomyocyttapoptose etter myokardinfarkt, redusere lesjonsvolum og nevrologisk mangel resulterende fra slag, redusere post-traumatisk apoptose og nevrologisk mangel i traumatisk hjerneskade, være effektiv i behandling av fulminant leverdestruksjon, og til forbedret overlevelse etter endotoksisk sjokk. Generelt er de peptidiske inhibitorer beskrevet over svært potente mot noen av caspaseenzymene. Imidlertid har ikke denne potens alltid blitt reflektert i cellulære apoptosemodeller. I tillegg er peptidinhibitorer typisk karakterisert ved uønskede farmakologiske egenskaper slik som dårlig oral absorpsjon, dårlig stabilitet og rask metabolisme. Ved å erkjenne behovet for å forbedre de farmakologiske egenskaper til de peptidiske caspaseinhibitorer, har peptidomimetiske inhibitorer blitt rapportert. Blant disse har inhibitorer hvor P3-aminosyren har blitt erstattet med derivater av 3-aminopyridin-2-oner og 5-aminopyrimidin-4-oner fått mye oppmerksomhet (US-patent 5,756,466 (Bemis et al.); Dolle et al. J. Med. Chem. 39, 2438, (1996); Golec et al. Bioorg. Med. Chem. Lett. 7, 2181, (1997); Semple et al, Biorg. Med. Chem. Lett. 7, 1337, (1997)) og ført til forbindelser med generell struktur:
hvor R<1> er hydrogen eller forskjellige grupper, R<2> er hydrogen, metyl eller etyl, R3 er alkyl, fenyl eller fenalkyl og R<4> er forskjellige grupper.
På grunn av de iboende problemer med de peptidiske inhibitorer, fortsetter det å være et behov for små-molekyl, ikke-peptid caspaseinhibitorer som er potente, stabile og penetrerer membraner for å gi effektiv hemming av apoptose in vivo. Slike forbindelser vile være ekstremt nyttige i behandlingen av de ovennevnte sykdommer hvor caspaseenzymer spiller en rolle.
Oppsummering av oppfinnelsen
Det har nå blitt funnet at forbindelser av denne oppfinnelse og farmasøytiske sammensetninger derav er effektive som inhibitorer av caspaser og cellulær apoptose. Disse forbindelser har den generelle formel IA som er angitt i det videre.
Forbindelsene av denne oppfinnelse har potente hemningsegenskaper over en rekke caspasemål med god virkning i cellulære apoptosemodeller. I tillegg er disse forbindelser forventet å ha forbedret cellepenetrasjon og farmakokinetiske egenskaper og, som en følge av deres potens, ha forbedret virkning mot sykdommer hvor caspaser er implisert.
Detaljert beskrivelse av oppfinnelsen
Denne oppfinnelse tilveiebringer nye forbindelser og farmasøytisk akseptable derivater derav, som er nyttige som caspaseinhibitorer. Disse forbindelser har den generelle formel IA:
hvor R<1> er R eller -CH2Y;
R er Ci-6-alkyl eller fenyl eventuelt substituert med halogen;
Y er halogen eller -OC=0(R);

Forbindelser av denne oppfinnelse hvor R<2> er COOH og y = 1 er gamma- -ketosyrer som kan eksistere i løsning, enten som den åpne form 1 eller den cykliserte hemiketalform 2. Fremstillingen heri av enten den ene eller den andre isomere form er ment å inkludere den andre. Likeledes vil de være åpenbart for fagmannen at visse forbindelser av denne oppfinnelse kan eksistere i tautomere former eller hydratiserte former, alle slike former av forbindelsene er innenfor rammen av oppfinnelsen. Med mindre annet er angitt, er strukturer avbildet heri også ment å inkludere alle stereokjemiske former av strukturen; dvs. R- og S-konfigurasjonene for hvert asymmetriske senter. Derfor er enkelte stereokjemiske isomerer samt enantiomere og diastereomere blandinger av de foreliggende forbindelser innenfor rammen av oppfinnelsen. En annen utførelse av denne oppfinnelse vedrører forbindelser med formel IB hvor R4 og R<5> sammen med karbonatomene til hvilke de er bundet danner en fenylring. Eksempler på lB-forbindelser er vist i tabell 2.
En annen utførelse av denne oppfinnelse vedrører forbindelser med formel IC hvor X2 er karbon, bindingen mellom X2 og det nærliggende CR4 er en dobbeltbinding, og R<6 >og R7 danner sammen med karbonatomene til hvilke de er bundet en fenylring. Eksempler på IC-forbindelser er vist i tabell 3.

En omrørt løsning av lfZ-Pyridin-2-on (500 mg, 5,2 6 mmol) i vannfri THF (50 ml) ved romtemperatur ble behandlet porsjonsvis med 60% NaH (231 mg, 5,78 mmol). Reaksjonsblandingen ble holdt i 10 min deretter tilsatt over 5 min til en løsning av (R)-2-trifluormetansulfonyloksy)-propionsyre-etylester (1315 mg, 5,2 6 mmol) i vannfri THF (2,5 ml) ved romtemperatur. En omrørt blanding av (S)-2-(2-okso-2if-pyrid-1-yl)-propionsyre-etylester (364 mg, 1,87 mmol), THF (2 ml), vann (1 ml) og natriumhydroksid (90 mg, 2,24 mmol) ble holdt ved romtemperatur i 1 time, deretter konsentrert under redusert trykk. Residuet ble løst i eter og den resulterende løsning vasket med vann. En omrørt blanding av (S) -2- (2-okso-2f/-pyridin-l-yl) -
propionsyre (70 mg, 0,42 mmol), 3-amino-5-fluor-4-hydroksy-pentansyre-tert-butylester (91 mg, 0,44 mmol), HOAt (63 mg, 0,46 mmol) og DMAP (59 mg, 0,48 mmol) og vannfri THF (5 ml) ble avkjølt til 0°C deretter ble EDC (88 mg, 0,46 mmol)

En omrørt løsning av 5-fluor-4-hydroksy-3-[(S)-2-(2-okso-2ff-pyridin-l-yl) -propionylamino] -pentansyre-tert-butylester (135 mg, 0,38 mmol) i vannfri DCM (5 ml) ble behandlet med 1,1,1-triacetoksy-1,1-dihydro-l,2-benziodoksol-3(1H) -on (193 mg, 0,4 6 mmol) ved 0°C. Den resulterende blanding ble holdt ved 0°C i 1,5 time, fortynnet med etylacetat, deretter helt i en 1:1 blanding av mettet vandig natriumhydrogenkarbonat og mettet vandig natriumtiosulfat. En omrørt løsning av 2,6-diklor-benzosyre 4- tert-butoksykarbonyl-2-hydroksy-3- [2- (l-okso-lff-isokinolin-2-yl)-propionylamino]-butylester (183 mg, 0,32 mmol) i vannfri DCM (3 ml) ble behandlet med 1,1,1-triacetoksy-1,1-dihydro-1,2-benziodoksol-3(1H)-on (147 mg, 0,34 mmol) ved 0°C. Trifluoreddiksyre (2 ml) ble tilsatt til en omrørt iskald løsning av 2,6-diklor-benzosyre 4-tert-butoksykarbonyl-2-okso-3-[2-(l-okso-lf/-isokinolin-2-yl) -propionylamino] - butylester (110 mg, 0,19 mmol) i vannfri diklormetan (2 ml). Assayene for caspasehemming er basert på spaltingen av et fluorgent substrat av rekombinante, rensede humane caspaser -1, -3, -7 eller -8. Hemming av IL- ip- sekresjon fra blandet populasjon av perifere blod mononukleære celler ( PBMC)
Prosessering av pre-IL-ip av caspase-1 kan måles i cellekultur ved å anvende en variasjon av cellekilder. Humane PBMC erholdt fra friske donorer gir en blandet populasjon av lymfocytt- og mononukleære celler som produserer et spektrum av interlevkiner og cytokiner i respons på mange klasser av fysiologiske stimulatorer.
Eksperimentell prosedyre
Testforbindelsen løses i dimetylsulfoksid (DMSO, Sigma #D-2650) for å gi en 100 mM stamløsning. Anti- Fas- indusert apoptoseassay
Cellulær apoptose kan induseres ved bindingen av Fas-ligand (FasL) til dens reseptor, CD95 (Fas). CD95 er en av en familie med beslektede reseptorer, kjent som selvmordsreseptorer, som kan utløse apoptose i celler via aktivering av caspase-enzymkaskaden. Prosessen inititieres ved bindingen av adaptormolekylet FADD/MORT-1 til det cytoplasmiske domene av CD-95-reseptor-ligand komplekset. Caspase-8 binder deretter FADD og blir aktivert, hvilket initierer en kaskade av hendelser som involverer aktiveringen av nedstrøms caspaser og deretter cellulær apoptose. Supernatanten fjernes og de kombinerte cellepelleter resuspenderes i 25 ml komplett medium. Cellene telles og tettheten justeres til 2xl0<6> celler/ml med komplett medium.
Testforbindelsen løses i dimetylsulfoksid (DMSO)(Sigma No.
D-2650) for å gi en 100 mM stamløsning. Denne fortynnes til 400 uM i komplett medium, fortynnes deretter serielt i en 96-brønns plate før tilsetning til celleassayplaten.
100 ul av cellesuspensjonen (2xl0<6> celler) tilsettes til hver brønn i en steril 96-brønns rundbunnet clusterplate (Costar No. 3790). 50 ul forbindelsesløsning ved den passende fortynning og 50 ul anti-Fas-antistoff, klon CH-11 (Kamiya No.MC-060) ved en sluttkonsentrasjon på 10 ng/ml tilsettes til brønnene. Kontrollbrønner settes opp minus antistoff og minus forbindelse, men med en seriell fortynning av DMSO som vehikkelkontroll. Platene inkuberes i 16-18 timer ved 37°C i 5% C02 og 95% fuktighet.
Apoptose av cellene måles ved kvantifisering av DNA-fragmentering ved å anvende et ^Celledød-deteksjonsassay' fra Boehringer-Mannheim, No. 1544 675. Etter inkubering i 16-18 timer sentrifugeres assayplatene ved 100xg ved romtemperatur i 5 minutter. 150 ul av supernatanten fjernes og erstattes med 150 ul nytt komplett medium. Cellene høstes deretter og 200 ul av lysebufferen supplert i assaykittet tilsettes til hver brønn. Cellene tritureres for å sikre fullstendig lyse og inkuberes i 30 minutter ved 4°C. Platene sentrifugeres deretter ved 1900xg i 10 minutter og supernatantene fortynnes 1:20 i anskaffet inkubasjonsbuffer. 100 ul av denne løsning analyseres deretter eksakt i henhold til produsentens instruksjoner levert med kittet. OD^snm måles 20 minutter etter tilsetning av sluttsubstratet i en SPECTRAmax Plus plateleser (Molecular Devices). OD405nm plottes versus forbindelseskonsentrasjon og ICso-verdiene for forbindelsene beregnes ved å anvende kurvetilpasningsprogrammet SOFTmax Pro (Molecular Devices) ved å anvende fire-parameter tilpasningsvalget.
Tabell 12 viser resultatene av aktiviteten til valgte forbindelser av denne oppfinnelse i det FAS-induserte apoptoseassay.
Selv om vi har beskrevet flere utførelser av denne oppfinnelse, er det åpenbart at våre basiseksempler kan endres for å gi andre utførelser som anvender forbindelsene og fremgangsmåtene av denne oppfinnelse. Derfor vil det bli verdsatt at rammen av denne oppfinnelse defineres av de vedlagte krav heller enn av de spesifikke utførelser som har blitt representert eksempelvis.
Claims (16)
1. Forbindelse med formel IA:
hvor R<1> er R eller -CH2Y;
R er Ci-6-alkyl eller fenyl eventuelt substituert med halogen;
Y er halogen eller -OC=0(R);
R2 er C02H;
R3 er hydrogen eller et C1-6 rettkjedet eller forgrenet alkyl;
hver av R4-R6 er uavhengig valgt fra hydrogen eller R; eller R<4> og R<5> eller R6 eller R7 sammen med karbonatomene til hvilke de er bundet danner en fenylring og R7 er valgt fra hydrogen eller fenyl.
2. Forbindelse ifølge krav 1, hvor R<1> er CH2Y og Y er F.
3. Forbindelse ifølge krav 2, hvor R<3> er hydrogen eller C1-3 alkyl.
4. Forbindelse ifølge krav 1, hvor R<1> er CH2Y og Y er F eller -OC=0(R); R<2> er C02H; R3 er hydrogen eller C1-3 alkyl, hver av R4-R6 er uavhengig valgt fra hydrogen eller R; og R<7 >er hydrogen eller fenyl.
5. Forbindelse valgt fra dem listet i tabell 1 under:

10. Anvendelse av en forbindelse ifølge et hvilket som helst av kravene 1-5 for fremstilling av et medikament for å minske IGIF- eller IFN-y-produksjon.
11. Anvendelse av en forbindelse ifølge et hvilket som helst av kravene 1-5 for fremstilling av et medikament for behandling av komplikasjoner assosiert med koronararteri-bypass-grafter.
12. Anvendelse av en forbindelse ifølge et hvilket som helst av kravene 1-5 for fremstilling av en sammensetning for å bevare celler.
13. Anvendelse av en forbindelse ifølge et hvilket som helst av kravene 1-5 for fremstilling av et medikament for å bevare organer i forbindelse med en organtransplantasjon eller for bevaring av blodprodukter.

og (c) å konvertere intermediat III-A til forbindelse I-A, hvori Y<1> er hydrogen eller Ci-6-alkyl;

R er en Ci-6-alkyl eller fenyl eventuelt substituert med halogen;
