Foreliggende oppfinnelse vedrører kondenserte 1,2,4-tiadiazinderivater, preparater som omfatter forbindelsene, anvendelsen av disse forbindelsene til fremstilling av medikamenter, og fremgangsmåte for fremstilling av medikamenter. Forbindelsene kan anvendes i terapi, f.eks. ved behandling av sykdommer i sentralnervesystemet, det kardiovaskulære system, det pulmonale system, mage- og tarmkanalsystemet og det endo-^ krinologiske system.
Oppfinnelsens bakgrunn
Kaliumkanaler spiller en viktig rolle i den fysio-logiske og farmakologiske kontroll av cellemembranspenning. Blant de forskjellige typer av kaliumkanaler er de ATP-sen-sitive (KATP-) kanaler som reguleres ved endringer i den intracellulære konsentrasjon av adenosintrifosfat. KATP-kanalene er blitt funnet i celler fra forskjellige vev, slik som hjerte-celler, bukspyttkjertelceller, skjelettmuskler, glattmuskler, sentralnerver og adenohypofyseceller. Kanalene er blitt asso-siert med forskjellige cellulære funksjoner, f.eks. hormon-utskillelse (insulin fra bukspyttkjertel-beta-celler, vekst-hormon og prolaktin fra adenohypofyseceller), vasodilasjon (i glattmuskelceller), hjerteslagspenningsvarighet, neurotrans-mitterfrigjørelse i sentralnervesystemet.
Modulatorer for K^p-kanalene er blitt funnet å være av betydning for behandlingen av forskjellige sykdommer. Visse sulfonylureaforbindelser som har vært brukt til behandlingen av ikke-insulinavhengig diabetes mellitus, virker ved å stimu-lere insulinfrigjørelse gjennom en inhibering av KATP-kanalene på bukspyttkjertel-beta-celler.
Kaliumkanalåpnerne som omfatter en heterogen gruppe av forbindelser, er blitt funnet å være i stand til å relaksere vaskulære glattmuskler og er derfor blitt brukt til behandlingen av hypertensjon.
I tillegg kan kaliumkanalåpnere anvendes som bronko-dilatorer ved behandlingen av astma og forskjellige andre sykdommer .
Dessuten er kaliumkanalåpnere blitt påvist å fremme hårvekst og har vært brukt til behandling av skallethet.
Kaliumkanalåpnere er også i stand til å relaksere urinblæreglattmuskel og kan derfor anvendes til behandling av urinveisinkontinens. Kaliumkanalåpnere som relakserer glatt-muskel i livmoren, kan anvendes til behandling av for tidlig fødsel. Ved å påvirke kaliumkanaler i sentralnervesystemet kan disse forbindelsene anvendes til behandling av forskjellige neurologiske og psykiatriske sykdommer, slik som Alzheimer, epilepsi og cerebral iskemi.
Forbindelsene er funnet å være anvendbare ved behandling av benign prostatahyperplasi, erektil dysfunksjon og i befruktningshindring.
Forbindelser ifølge foreliggende oppfinnelse som inhiberer insulinutskillelse ved å aktivere kaliumkanaler i beta-cellen, kan anvendes i kombinasjon med andre forbindelser som kan anvendes til å behandle ikke-insulinavhengig diabetes mellitus og insulinavhengig diabetes mellitus. Eksempler på slike forbindelser er insulin, insulinsensitiviseringsmidler, slik som tiazolidindioner, insulinsekretagoga, slik som repa-glinid, tolbutamid, glibenklamid og glukagonlignende peptid (GLPl), inhibitorer for a-glukosidaser og leverenzymer som er ansvarlige for biosyntesen av glukose.
Nylig er det blitt påvist at diazoksid (7-klor-3-metyl-2H-l,2,4-benzotiadiazin-l,1-dioksid) og visse 3-(alkyl-amino)-4H-pyrido[4,3-e]-1,2,4-tiadiazin-l,1-dioksidderivater inhiberer insulinfrigjørelse ved hjelp av en aktivering av KATp-kanaler på bukspyttkjertel-beta-celler (Pirotte B. et al., Biochem. Pharmacol., 47, 13 81-1386 (1994); Pirotte B. et al., J. Med. Chem., 36, 3211-3213 (1993)). Diazoksid er dessuten blitt påvist å forsinke utbruddet av sukkersyke hos BB-rotter (Vlahos WD et al., Metabolism 40, 39-46 (1991)). Hos fete zucker-rotter er diazoksid blitt påvist å redusere insulinutskillelse og øke insulinreseptorbinding og følgelig forbedre glukosetoleranse og redusere vektøkning (Alemzadeh R. et al., Endocrinol. 133, 705-712, 1993). Det forventes at forbindelser som aktiverer KATP-kanaler, kan anvendes til behandling av sykdommer som er kjennetegnet ved en overproduksjon av insulin, og til behandling og forhindring av sukkersyke.
I EP 618 2 09 beskrives en klasse av pyridotiadiazin-derivater med en alkyl- eller en alkylaminogruppe i 3-stilling i tiadiazinringen. Disse forbindelsene kreves å være agonister for AMPA-glutamatreseptoren.
I J. Med. Chem. 1980, 23, 575-577 er syntesen av 4(5)-amino- og formylaminoimidazo-5(4)-karboksamid og deres egenskaper som midler med kjemoterapeutisk verdi beskrevet. Spesielt er forbindelsene 3-aminoimidazo[4,5-e]-1,2,4-tiadiazin-1,1-dioksid og N-benzoylaminoimidazo[4,5-e]-1,2,4-tiadiazin-1,1-dioksid vist.

Forbindelsene ifølge foreliggende oppfinnelse reagerer med kaliumkanalene og virker følgelig som åpnere eller blokkeringsmidler for de ATP-regulerte kaliumkanaler, noe som gjør dem anvendbare ved behandlingen av forskjellige sykdommer i det kardiovaskulære system, f.eks. cerebral iskemi, hypertensjon, iskemiske hjertesykdommer, angina pectoris og koron-are hjertesykdommer; pulmona1systernet; mage- og tarmkanalsystemet; sentralnervesystemet og det endokrinologiske system.
Ettersom noen KATP-åpnere er i stand til å antagon-isere vasospasmer i basilare eller cerebrale arterier, kan forbindelsene ifølge foreliggende oppfinnelse anvendes til behandling av slike vasospastiske forstyrrelser som subarachnoid blødning og migrene.
Forbindelsene ifølge foreliggende oppfinnelse vil også kunne anvendes til behandling av sykdommer som er forbundet med redusert skjelettmuskelblodstrømning, slik som Rey-nauds sykdom og intermitterende smerter i leggmuskel.
Videre kan forbindelsene ifølge oppfinnelsen anvendes til behandling av kroniske luftveissykdommer, inkludert astma, og til behandling av detrusormuskelustabilitet sekundært til urinblæreutstrømningsobstruksjon og derfor til nyrestener ved å hjelpe til med deres passering langs urinlederen.
De foreliggende forbindelser kan også anvendes til behandling av tilstander som er forbundet med forstyrrelser i mage- og tarmkanalmobilitet, slik som irritabelt tarmsyndrom. I tillegg kan disse forbindelsene anvendes til behandling av for tidlig fødsel og dysmenoré.
Kaliumkanalåpnere hyperpolariserer neuroner og inhiberer neurotransmitterfrigjørelse, og det forventes at slike forbindelser kan anvendes til behandling av forskjellige sykdommer i sentralnervesystemet, f.eks. epilepsi, iskemi og neurodegenerative sykdommer, og til håndteringen av smerte.
Videre fremmer kaliumkanalåpnere hårvekst, og forbindelsene ifølge foreliggende oppfinnelse kan derfor anvendes til behandling av skallethet.
Kaliumkanalåpnere relakserer også urinblæreglattmuskel, og forbindelsene ifølge foreliggende oppfinnelse kan således anvendes til behandling av urinveisinkontinens.
Ved slike sykdommer som nesidioblastose og insulinoma hvor en hypersekresjon av insulin forårsaker alvorlig hypogly-kemi, kan forbindelsene ifølge foreliggende oppfinnelse anvendes til å redusere insulinutskillelse. Ved fedme støter man svært ofte på hyperinsulinemi og insulinresistens. Denne til-stand kan føre til utviklingen av ikke-insulinavhengig sukkersyke (NIDDM). Det forventes at kaliumkanalåpnere og følgelig forbindelsene ifølge foreliggende oppfinnelse kan anvendes til å redusere hyperinsulinemien og derved forhindre sukkersyke og redusere fedme. Ved åpenbar NIDDM kan behandling av hyperinsulinemi med kaliumkanalåpnere og følgelig de foreliggende forbindelser være gunstig for å gjenopprette glukosesensiti-vitet og normale insulinutskillelser.
Ved tidlige tilfeller av insulinavhengig sukkersyke (IDDM) eller ved prediabetiske tilfeller kan kaliumkanalåpnere og følgelig de foreliggende forbindelser anvendes til å indu-sere bukspyttkjertelcellehvile, noe som kan forhindre progre-sjonen av autoimmunsykdommen.
Kaliumkanalåpnerne ifølge foreliggende oppfinnelse kan administreres i kombinasjon med et immunoundertrykkende middel eller med et middel som nikotinamid, som vil redusere autoimmun degenerasjon av beta-celler.
Kombinasjon av beta-cellehvile med en behandling som beskytter beta-cellene mot cytokinformidlet beta-cellesvekk-else/cytotoksisitet, er et annet aspekt av denne oppfinnelsen. Insulinkrevende sukkersyke eller type 1-sukkersyke (IDDM) samt IDDM med sen oppstart (også kjent som type 1.5., f.eks. ikke-insulinkrevende type 2- (NIIDM) pasienter med autoreaktivitet mot beta-celleepitoper som senere viser seg å være insulinkrevende) har autoreaktive monocytter/lymfocytter i omløp som finner frem til øycellene/beta-cellene og frigjør deres cyto-kiner. Noen av disse cytokinene (f.eks. interleukin-lb (IL-lb), tumornekrosefaktor a (TNFa) og interferon g (IFNg)) er spesifikt toksiske overfor beta-cellene, f.eks. gjennom induk-sjonen av nitrogenoksid (NO) og andre frie radikaler. Inhibering av denne cytotoksisiteten, f.eks. ved hjelp av samadminis-trering av nikotinamid (NA), et derivat herav eller andre cytokinbeskyttende forbindelser til de prediabetiske/diabet-iske pasienter behandlet med PCO-forbindelsen, er et eksempel på dette aspektet. Nikotinamid tilhører B-vitaminfamilien og avledes fra nikotinsyre ved amidering av karboksylgruppen. Det har ingen av nikotinets farmakologiske egenskaper. NA omdannes til NAD+ som virker som et koenzym for proteiner involvert i vevsrespirasjon. NA er blitt foreslått å påvirke flere av de putative intracellulære, molekylære hendelser som følger etter immunangrep på beta-cellene. Dyreforsøk og tidlige ikke-blind-ede forsøk hos mennesker har indikert en beskyttende rolle for denne forbindelsen mot IDDM samt i cytokin/immun-formidlet beta-celledestruksjon. Forbindelser ifølge foreliggende oppfinnelse som virker som blokkeringsmidler for KRTP-kanaler, kan anvendes til behandlingen av NIDDM.
Forbindelsene ifølge foreliggende oppfinnelse vil fortrinnsvis kunne anvendes til behandling eller forhindring av sykdommer i det endokrinologiske system, slik som hyperinsulinemi og sukkersyke.
Ved et annet aspekt vedrører følgelig oppfinnelsen en av forbindelsene ifølge oppfinnelsen eller et farmasøyt-isk akseptabelt syreaddisjonssalt derav for anvendelse som et terapeutisk akseptabelt stoff, fortrinnsvis for anvendelse som et terapeutisk akseptabelt stoff ved behandling av hyperinsulinemi og behandling eller forhindring av sukkersyke.
Dessuten vedrører oppfinnelsen også anvendelsen av forbindelsene ifølge oppfinnelsen til fremstilling av medikamenter som er nyttige for behandling av hyperinsulinemi og behandling eller forhindring av sukkersyke.
Forbindelsene ifølge oppfinnelsen kan fremstilles ved hjelp av kjente fremgangsmåter, slik det fremgår av eksemplene nedenunder.

Farmakologiske metoder
Evnen til forbindelsene når det gjelder å reagere med kaliumkanaler, kan bestemmes ved hjelp av forskjellige metoder. Når det anvendes "patch-clamp"-teknikker (Hamill O.P., Marty A., Nefer E., Sakman B. og Sigworth F.J., Plugers Arch., 391, 85-100 (1981)), kan ionestrømmen gjennom en enkeltkanal i en celle måles.
Aktiviteten til forbindelsene som kaliumkanalåpnere, kan også måles som relaksasjon av rotteaortaringer i henhold til den følgende fremgangsmåte: Et snitt av rottetoraksaorta mellom aortabuen og diafragmaet ble dissekert ut og montert som ringpreparater, som beskrevet av Taylor P.D. et al., Brit. J. Pharmacol., 111, 42-48 (1994).
Etter en 45 minutters ekvilibreringsperiode under en spenning på 2 g ble preparatene trukket sammen, hvorved man oppnådde 80% av den maksimale respons under anvendelse av den påkrevde fenylefrinkonsentrasjon. Når fenylefrinresponsen nådde et platå, ble potensielle vasodilatormidler tilsatt kumulativt til badet i små volum under anvendelse av halv-logaritmiske, molare økninger ved 2 minutters intervaller. 86Rb+- utstrømning fra en p- cellelinje
RIN 5F-cellelinjen ble dyrket i RPMI 1640 med "Gluta-max I", supplert med 10% kalvefosterserum (fra Gibco BRL, Skottland, Storbritannia) og holdt ved en atmosfære av 5% C02/95% luft ved 37 °C. Cellene ble løsnet med en trypsin-EDTA-oppløsning (fra Gibco BRL, Skottland, Storbritannia), på nytt oppslemmet i medium, tilsatt 1 mCi/ml <86>Rb<+> og platet ut på nytt i mikrotiterplater (96-brønners "duster 3596", sterile, fra Costar Corporation, MA, USA) ved en tetthet på 50 000 celler/brønn i 100 ul/brønn og dyrket 24 timer før anvendelse i analysen.
Platene ble vasket fire ganger med Ringer-buffer (150 mM NaCl, 10 mM Hepes, 3,0 mM KCl, 1,0 mM CaCl2, 2 0 mM sukrose, pH 7,1). 80 (il Ringer-buf f er og 1 ul kontroll- eller testforbindelse oppløst i DMSO ble tilsatt. Etter inkubasjon i 1 time ved romtemperatur med et lokk ble 50 ul av supernatan-ten overført til "PicoPlates" (Packard Instrument Company, CT, USA) og 100 ul "MicroScint 40" {Packard Instrument Company, CT, USA) tilsatt. Platene ble tellet i "TopCount" (Packard Instrument Company, CT, USA) i 1 minutt/brønn ved <32>P-program-met.
Beregningen av EC50 og E^ka. ble gjort ved hjelp av "SlideWrite" (Advanced Graphics Software, Inc., CA, USA) under anvendelse av en fire-parameters logistikkurve: y =
(a-d)/(1+(x/c)<b>)+d, hvor a = aktiviteten beregnet ved konsentrasjon null, b = en helningsfaktor, c = konsentrasjonen midt på kurven, og d = aktiviteten beregnet ved uendelig konsentrasjon. EC50 = c og Emaka. = d når kurven avbrytes ved uendelige konsentrasj oner.
Forbindelsene ifølge oppfinnelsen er effektive over et bredt doseområde. Generelt oppnås tilfredsstillende res-ultater med doseringer fra 0,05 til 1 000 mg, fortrinnsvis fra 0,1 til 500 mg pr. dag. Den mest foretrukne dosering er 50 mg til 200 mg pr. dag. Den nøyaktige dosering vil avhenge av ad-ministreringsmåten, formen som den administreres i, individet som skal behandles og kroppsvekten til individet som skal behandles, og preferansen og erfaringen til den behandlende lege eller veterinær.
Administreringsveien kan være hvilken som helst vei som effektivt transporterer den aktive forbindelse til det passende eller ønskede virkningssted, slik som oral eller parenteral, f.eks. rektal, transdermal, subkutan, intravenøs, intramuskulær eller intranasal, idet den orale vei er fore-trukket .
Typiske preparater omfatter en forbindelse med formel I eller et farmasøytisk akseptabelt syreaddisjonssalt derav, forbundet med en farmasøytisk akseptabel eksipiens som kan være en bærer eller en fortynner, eller som kan være fortynnet med en bærer, eller innelukket i en bærer som kan være i form av en kapsel, liten pose, papir eller annen beholder. Ved fremstilling av preparatene kan det anvendes vanlige teknikker for fremstillingen av farmasøytiske preparater. For eksempel vil den aktive forbindelse vanligvis bli blandet med en bærer, eller fortynnet med en bærer, eller innelukket i en bærer som kan være i form av en ampulle, kapsel, liten pose, papir eller annen beholder. Preparatene ifølge oppfinnelsen kan formuleres slik at de gir hurtig, langvarig eller forsinket frigjørelse av den aktive bestanddel etter administrering til pasienten ved å anvende fremgangsmåter som er godt kjent innen tek-nikken.
De farmasøytiske preparater kan steriliseres og, om ønsket, blandes med hjelpemidler, emulgeringsmidler, salt for påvirkning av osmotisk trykk, bufferstoffer og/eller farge-stoffer og lignende, som ikke på skadelig måte reagerer med de aktive forbindelser.
For parenteral applikasjon er det særlig egnet med injiserbare oppløsninger eller suspensjoner, fortrinnsvis vandige oppløsninger med den aktive forbindelse oppløst i polyhydroksylert ricinusolje.
Tabletter, dragéer eller kapsler med talkum og/eller en karbohydratbærer eller et bindemiddel eller lignende, er særlig egnet for oral applikasjon. Foretrukne bærere for
tabletter, dragéer eller kapsler omfatter laktose, maisstiv-else og/eller potetstivelse. En sirup eller eliksir kan anvendes i tilfeller hvor det kan gjøres bruk av en søtet bærer.
Generelt dispenseres forbindelsene i enhetsform omfattende fra 1 til 100 mg i en farmasøytisk akseptabel bærer pr. enhetsdosering.

a) 4- cyan- 2- hydrazinokarbonyl- 5- metylt iofen- 3 - sulfonamid
Metyl-4-cyan-5-metyl-3 -sulfamoylt iofen-2-karboksylat
(2,9 g), fremstilt fra metyl-3-amino-4-cyan-5-metyltiofen-2-karboksylat analogt med kjente fremgangsmåter, f.eks. F. Junquera et al., Eur. J. Med. Chem. 23, 329 (1988), ble tilsatt, og blandingen ble omrørt i 1 time ved romtemperatur og så inndampet. Oljeresten (3,3 g) ble triturert med 10 ml vann, og de utfelte krystaller ble samlet opp ved filtrering, vasket med vann og tørket, hvorved man fikk tittelforbindelsene som gule krystaller (utbytte 1,62 g) ; smp. 186-91 °C, """H-NMR (DMSO-d6) , 5 (ppm) : 7,25 (br, 5H, NH/NH2) , 2,51 (s, 3H, CH3) .

En oppløsning av natriumnitritt (0,47 g) i 5 ml vann
ble tilsatt dråpevis til en omrørt oppløsning av 4-cyan-2-hydrazinokarbonyl-5-metyltiofen-3-sulfonamid (1,6 g) i 38 ml 1 M saltsyre ved 0 °C. Den resulterende blanding ble omrørt i 30 minutter ved 0 °C og så filtrert. Filterkaken ble vasket med vann og tørket under vakuum, hvorved man fikk 1,35 g av tittelforbindelsen, Hi-NMR (DMS0-d6) , 6 (ppm) : 7,75 (br, 1H, NH2) , 2,75 (s, 1H, CH3) .
c) 4- cyan- 2- e toksykarbonylamino- 5- me tylt i of en- 3 - sulfonamid
4-cyan-5-metyl-3-sulfamoyltiofen-2-karbonylazid (1,35 g) ble tilsatt i små porsjoner i løpet av 5 minutter til 50 ml absolutt etanol ved reflukstemperatur. Den resulterende oppløsning ble kokt under tilbakeløpskjøling i 5 minutter og så inndampet. Resten krystalliserte når den ble triturert med 20 ml etylacetat. Krystallene ble frafiltrert, skyllet med etylacetat og tørket, hvorved man fikk tittelforbindelsen, <1>H-NMR (DMSO-de) , 8 (ppm): 9,45 (s, 1H, NH) , 7,82 (br, 2H, NH2) , 4,23 (q, 2H, CH2) , 2,51 (s, 3H, CH3) , 1,25 (t, 3H, CH3) .
d) N-( 4- cyan- 2- etoksykarbonylamino- 5- metyl- 3-tienylsulfonyl)- N'- isopropyltiourea
En blanding av 4-cyan-2-etoksykarbonylamino-5-metyltiofen-3-sulfonamid (0,50 g), kaliumkarbonat (0,36 g) og isopropylisotiocyanat (300 ul) i 10 ml tørt aceton ble varmet opp ved 55 °C i 18 timer og så inndampet til tørrhet. Resten ble oppløst i 10 ml vann, og pH ble regulert til 2 ved dråpevis tilsetning av 1 M saltsyre. Til en omrørt oppløsning av N-(4-cyan-2-etoksykarbonylamino-5-metyl-3-tienylsulfonyl)-N<1->isopropyltiourea (0,3 g) og trietylamin (320 ul) i 10 ml tørt THF ved 0 °C ble det tilsatt 750 (al av en 2 0% oppløsning av fosgen i toluen. Blandingen ble omrørt ved 0 °C i 1 time og så inndampet til tørr-het. Resten ble triturert med 10 ml vann, frafiltrert, skyllet på filteret med vann og tørket, hvorved man fikk 0,24 g uren tittelforbindelse; smp. 116-119 °C. Produktet ble brukt til det neste trinn uten rensing.
f) 7- cyan- 3- isopropylamino- 6- metyl- 4H- tieno[ 2, 3- e]-1, 2, 4- tiadiazin- l, 1- dioksid
En blanding av etyl-7-cyan-3-isopropylamino-6-metyl-4H-tieno[2,3-e]-1,2,4-tiadiazin-4-karboksylat-l,1-dioksid (0,15 g) og 5 ml 1 M vandig natriumhydroksid ble omrørt ved romtemperatur i 1 time. Så ble blandingen filtrert, og pH ble regulert til 1-2 ved dråpevis tilsetning av 4 M saltsyre. Kalium-tert.-butoksid (0,49 g, 4,4 mmol) ble tilsatt til en oppløsning av 3-amino-5-klortiofen-2-sulfonamidhydro-klorid (0,5 g, 2,0 mmol) i tørt N,N-dimetylformamid (5 ml) med omrøring på et isbad. Etyl-3-(6-klor-l,4-dihydro-l,1-dioksotieno[3,2-e]-lX6, 2,4-tiadiazin-3-ylamino)butanoat (0,5 g, 1,42 mmol) ble hydrolysert til syren ved omrøring i 5 ml 2 N natriumhydroksid i 2 timer ved romtemperatur. Oppløsningen ble behandlet med avfargende aktivtkarbon, filtrert og surgjort med 4 M saltsyre til pH 2. Toluen (5 ml) ble avkjølt til 10 °C og litiumalumin-iumhydrid (114 mg, 3 mmol) ble tilsatt, etterfulgt av 0,53 ml tetrahydrofuran. Etyl-3-(6-klor-l,4-dihydro-l,1-dioksotieno-[3,2-e]- lX6, 2,4-tiadiazin-3-ylamino)butanoat (352 mg, 1 mmol) ble tilsatt til den avkjølte oppløsning, og blandingen ble omrørt i 2 timer ved 0 °C og så ved romtemperatur over natten. a) N-[ 5- klor- 3-( isopropyltiokarbamoyl) sulfamoyltiofen- 2-yl] acetamid
Kaliura-tert.-butoksid (135 mg, 1,2 mmol) ble tilsatt til en oppløsning av N-(5-klor-3-sulfamoyltiofen-2-yl)acetamid {255 mg, 1,0 mmol) i tørt N,N-dimetylformamid (5 ml) med om-røring på et isbad. Etter 5 minutter ble isopropylisotiocyanat (0,128 ml, 1,2 mmol) tilsatt dråpevis, og oppløsningen ble omrørt i 30 minutter ved 0 °C og så ved romtemperatur i 3 timer. a) Cyanmetansulfonamid
Tørt THF (200 ml) ble mettet med ammoniakk ved
-10 °C. Så ble en oppløsning av cyanmetansulfonylklorid (13,9 g, fremstilt i henhold til Sammes et al., J. Chem. Soc, 1971, 2151-5155) i 10 ml tørt THF tilsatt i små porsjoner med omrøring i løpet av 20 minutter ved -10 °C. Etter tilsetningen ble temperaturen økt til 0 °C, og reaksjonsblandingen ble filtrert. Fjerning av oppløsningsmidlet fra filtratet og rensing av resten ved hjelp av kolonnekromatografi på silikagel eluert med etylacetat, ga tittelforbindelsen som beige krystaller; smp. 97-99 °C; IR (KBr) , 1) (cm"1) : 3316, 3328 (N-H) , 2266 (CsN) , 1371, 1151 (S02) .
b) 2- amino- 4- metyltiofen- 3- sulfonamid
En blanding av cyanmetansulfonamid (1,0 g), 2,5-dihydroksy-2,5-dimetyl-l,4-ditian (0,75 g) og trietylamin (100 ul) i absolutt etanol (7 ml) ble varmet opp ved 45-50 °C under nitrogen i 3 timer. Så ble oppløsningsmidlet avdampet og resten fordelt i vann (30 ml) og etylacetat (50 ml). Vannfasen ble ekstrahert med 4 x 50 ml etylacetat. De kombinerte etyl-acetatfaser ble tørket over natriumsulfat og inndampet. Resten ble renset ved hjelp av kolonnekromatografi på silikagel eluert med etylacetat. Tittelforbindelsen ble erholdt som et gult, fast stoff, utbytte 0,65 (41%); smp. 142-144 °C; 1H-NMR (CD3OD) , 8 (ppm): 7,19 (s, 1H, C(5)H), 4,88 (br, 4H, S02NH2 + H20) , 4,71 (s, 2H, NH2) , 2,41 (s, 3H, CH3) ; MS: 192 (M+) , 112 (M<+> - S02NH2) , 72 (CH3-C2HS+) .
c) N-( 2- amino- 4- metyl- 3- tienylsulfonyl)- N'- isopropyl-tiourea
En blanding av 2-amino-4-metyltiofen-3-sulfonamid (0,30 g), kaliumkarbonat (0,32 g) og isopropylisotiocyanat (272 ul) i tørt aceton (5 ml) under nitrogen ble varmet opp ved 50-55 °C over natten. Så ble oppløsningsmidlet avdampet, og resten ble oppløst i vann (15 ml); pH ble regulert til 2 ved tilsetning av 4 M saltsyre, og blandingen ble omrørt i 30 minutter. De utfelte krystaller ble frafiltrert, skyllet med en liten mengde vann og tørket, hvorved man fikk 0,24 g (utbytte 54%) av tittelforbindelsen, smp. 118-120 °C.
d) 3- isopropylamino- 7- metyl- 4H- tieno[ 2, 3- e]- l, 2, 4-tiadiazin- 1, 1- dioksid
En 20% oppløsning av fosgen i toluen (715 ul) ble tilsatt til en omrørt og avkjølt oppløsning av N-(2-amino-4-metyl-3-tienylsulfonyl)-N<1->isopropyltiourea (0,22 g) og trietylamin (313 ul) i tørt THF (5 ml), idet temperaturen ble holdt under 5 °C. Etter omrøring i 1,5 time ble blandingen inndampet og triturert med 10 ml vann. Utfellingen ble samlet opp ved filtrering og tørket, hvorved man fikk 0,14 g råprodukt. Krystallene ble varmet opp ved 70 °C med 5 ml etylacetat, og den resulterende blanding ble avkjølt til 0 °C i 10 minutter og så filtrert. 6- cyan- 3- isopropylamino- 4H- tieno[ 3, 2- e]- 1, 2, 4- tiadiazin- l,1-dioksid
Til en oppløsning av 6-brom-3-isopropylamino-4H-tieno[3,2-e]-l,2,4-tiadiazin-l,1-dioksid (243 mg, 0,75 mmol) i tørt N,N-dimetylformamid (2 ml) ble det tilsatt kobber(I)-cyanid (13 5 mg, 1,5 mmol), og blandingen ble varmet opp ved 150 °C i 2 timer under nitrogen. Den mørke blanding fikk avkjøles til romtemperatur, og vann ble tilsatt. Suspensjonen ble gjort basisk ved tilsetning av 1 N natriumhydroksid, filtrert, og filtratet ble surgjort ved tilsetning av 4 M saltsyre. 6- brom- 3- syklopentylamino- 4H- tieno[ 3, 2- e]- 1, 2, 4- tiadiazin- l, 1-dioksid
Brom (0,12 ml, 2,3 mmol) ble tilsatt dråpevis til en oppløsning av 6-klor-3-syklopentylamino-4H-tieno[3,2-e]-1, 2,4-tiadiazin (305 mg, 1,0 mmol) i eddiksyre (10 ml), og blandingen ble omrørt i 21 timer ved 100 °C i en lukket kolbe. Den avkjølte blanding ble inndampet til tørrhet, og resten ble triturert med vann, hvorved man fikk et fast stoff som ble rekrystallisert fra etylacetat og etanol, hvorved man fikk tittelforbindelsen (166 mg, 47%) forurenset med 33% av utgangsmaterialet. Rensing ved hjelp av preparativ HPLC ga tittelforbindelsen (65 mg, 18%) forurenset med 3% av utgangsmaterialet. ^-NMR (DMSO) : 5 10,7 (s, 1H, NH) , 7,35 (br s, 1H, NH), 7,13 (s, 1H, H-7), 4,00 (sekstett, 1H), 1,9 (m, 2H), 1,7 til 1,4 ppm (m, 6H). Dekomp.: 287-294 °C.

6- klor- 3-( 2- metylallyl) amino- 4H- tieno[ 3, 2- e]- 1, 2, 4- tiadiazin-1, 1- dioksid
En oppløsning av 3,6-diklor-4H-tieno[3,2-e]-1,2,4-tiadiazin-1,1-dioksid (128 mg, 0,5 mmol) i metallylamin
(0,5 ml) ble omrørt i 48 timer ved 90 °C i en lukket kolbe. Den avkjølte oppløsning ble konsentrert under vakuum, og resten ble omrørt med vann (3 ml), etterfulgt av regulering til pH 2 med 4 M saltsyre. Produktet ble isolert ved filtrering og vasket med vann, hvorved man fikk 92 mg (64%) av den rene tittelforbindelse; smp. 224-226 °C (dek.); <1>H-NMR (DMSO-d6) : 8 1,72 (s, 3H), 3,75 (d, 2H) , 4,83 (s, 2H) , 7,05 (s, 1H), 7,45 (br s, 1H), 11,0 (br s, 1H).

5. Farmasøytisk preparat ifølge krav 2 eller 3, karakterisert ved at forbindelsen administreres som en dose i et område fra 0,05 til 1 000, fortrinnsvis fra 0,1 til 500, og spesielt i området fra 50 til 200 mg pr. dag.
