Description
Den foreliggende oppfinnelse vedrører nye karboksylsyrederivater og deres anvendelse.
Endotelin er et peptid som er oppbygget av 21 aminosyrer og syntetiseres og frigis av vaskulær endotel. Endotelin eksisterer i tre isoformer, ET-1, ET-2 og ET-3.1 det følgende betyr "endotelin" eller "ET" én eller alle isoformer av endotelin. Endotelin er en sterk vasokonstriktor og har en sterk effekt på kar-tonusen. Det er kjent at denne vasokonstriksjon forårsakes av bindingen av endotelin på dets reseptor (Nature, 332, 411-415, 1988; FEBS Letters, 231, 440-444, 1988 og Biochem. Biofys. Res. Commun., 154, 868-875, 1988).
Forhøyet eller abnormal frigjøring av endotelin forårsaker en vedvarende kar-kontraksjon i perifere, renale og cerebrale blodkar, noe som kan føre til sykdommer. Det er angitt i litteraturen at forhøyede plasmanivåer av endotelin var blitt funnet i pasienter med hypertoni, akutt myokardinfarkt, pulmonær hypertoni, Raynaud-syndrom, aterosklerose og i luftveiene hos astmatikere (Japan J. Hypertension, 12, 79 (1989), J. Vascular Med. Biology 2, 207 (1990), J. Am. Med. Association 264, 2868 (1990)).
I henhold til dette skulle substanser som spesifikt inhiberer bindingen av endotelin til reseptoren, også antagonisere de ovenfor nevnte forskjellige fysiologiske effekter av endotelin og derfor være verdifulle medikamenter.
Det ble nå funnet at bestemte karboksylsyrederivater er gode hemmere for endotelinreseptorer.
Gjenstand for oppfinnelsen er karboksylsyrederivater med formelen I
hvori R betyr en gruppe i hvilken R<1> har betydningen en rest OR<8>, hvori R<8> betyr: hydrogen, kationet av et alkalimetall så som litium, natrium, kalium, eller
kationet av et jordalkalimetall så som kalsium, magnesium og barium, eller et fysiologisk godtagbart organisk ammoniumion så som tertiært CrC4-alkylammonium eller ammoniumionet;
C3-C8-cykloalkyl, som cyklopropyl, cyklobutyl, cyklopentyl, cykloheksyl og
cykloheptyl, cyklooctyl;
Ci-Cs-alkyl, særlig Ci-C4-alkyl som metyl, etyl, n-propyl, iso-propyl,
n-butyl, iso-butyl, tert.-butyl; CH2-fenyl,
og de øvrige substituenter har følgende betydning:
R<2> er CrC4-alkyl, Ci-C4-halogenalkyl, Ci-C4-alkoksy;
X er nitrogen eller CR<12>, hvori CR<12> danner sammen med CR2 eller CR3 en
fenylring
Y er nitrogen;
R<3> er hydrogen, Ci-C4-alkyl, Ci-C4-halogenalkyl, d-C4-alkoksy;
R<4> ogR<5>erfenyl;
R<6> er Ci-C8-alkyl;
Z er oksygen.
Forbindelsene og også mellomproduktene for deres fremstilling, som f.eks. II og IV, kan ha ett eller flere asymmetriske substituerte karbonatomer. Slike forbindelser kan foreligge i form av de rene enantiomerer eller rene diastereomerer eller som blandinger av disse. Foretrukket er anvendelsen av en enantiomer-ren forbindelse som virkestoff.
Gjenstand for oppfinnelsen er videre anvendelsen av de ovenfor nevnte karboksylsyrederivater for fremstilling av legemidler, særlig for fremstilling av hemmende stoffer for endotelin-reseptorer.
Fremstillingen av forbindelsene med den generelle formel IV, i hvilke Z er svovel eller oksygen, kan - også i enantiomer-ren form - finne sted som beskrevet i P 44 36 851.8. Forbindelsene ifølge oppfinnelsen, hvor substituentene har den betydning som er angitt under den generelle formel I, kan eksempelvis fremstilles ved å reagere karboksylsyrederivatene med den generelle formel IV, hvor substituentene har den angitte betydning, med forbindelser med den generelle formel V
hvor R<13> betyr halogen eller R<14->S02-, idet R<14> kan være Ci-C4-alkyl,
CrC4-halogenalkyl eller fenyl. Reaksjonen finner fortrinnsvis sted i et inert løsnings- eller fortynningsmiddel under tilsetning av en egnet base, dvs. en base som bevirker en deprotonering av mellomproduktet IV, i et
temperaturområde fra romtemperatur opptil kokepunktet for løsemidlet.
Eksempler på slike løsemidler eller fortynningsmidler er alifatiske, alicykliske og aromatiske hydrokarboner, som i hvært tilfelle eventuelt kan være klorert, som foreksempel heksan, cykloheksan, petroleter, ligroin, benzen, toluen, xylen, metylenklorid, kloroform, karbontetraklorid, etylklorid og trikloretylen; eter, som for eksempel diisopropyleter, dibutyleter, metyl-tert.-butyleter, propylenoksyd, dioksan og tetrahydrofuran; nitriler, som for eksempel acetonitril og propionitril; syreamider, som for eksempel dimetylformamid, dimetylacetamid og N-metylpyrrolidon; sulfoksyder og sulfoner, som for eksempel dimetylsulfoksyd og sulfolan.
Forbindelser med formelen V er kjente, og tildels kan de kjøpes, eller de kan fremstilles ved hjelp av generelt kjente fremgangsmåter.
Som base kan det anvendes et alkali- eller jordalkalimetallhydrid som natriumhydrid, kaliumhydrid eller kalsiumhydrid, et karbonat som alkalimetallkarbonat, f.eks. natrium- eller kaliumkarbonat, et alkali- eller jordalkalimetallhydroksyd som natrium- eller kaliumhydroksyd, en metallorganisk forbindelse som butyllitium eller et alkaliamid som litiumdiisopropylamid.
Forbindelser med formelen I kan også fremstilles ved at man starter med de tilsvarende karboksylsyrer, dvs. forbindelser med formelen I, hvor R betyr COOH, og så først overføre sistnevnte på vanlig måte i en aktivert form, så som et syrehalogenid, et anhydrid eller imidazolid, og så reagere dette med en passende hydroksylforbindelse HOR<8>. Denne omsetning lar seg gjennomføre i de vanlige løsemidler og krever ofte tilsetning av en base, hvorunder de ovenfor nevnte kommer i betraktning. Dsse to trinn lar seg eksempelvis også forenkle ved at man lar karboksylsyren innvirke på hydroksylforbindelsen i nærvær av et vannavspaltende middel, så som et karbodiimid.
Dessuten kan forbindelser med formelen I også fremstilles ved at man går ut fra saltene av de tilsvarende karboksylsyrer, dvs. fra forbindelser med formelen I, i hvilke R står for en gruppe COR<1> og R<1> for OM, idet M kan være et alkalimetall-kation eller ekvivalenten av et jordalkalimetall-kation. Disse salter kan bringes til reaksjon med mange forbindelser med formelen R<1->A, hvor A betyr en vanlig nukleofug avspaltende gruppe, eksempelvis halogen, så som klor, brom, jod, eller eventuelt med halogen, alkyl eller halogenalkyl substituert aryl- eller alkylsulfonyl, som f.eks. toluensulfonyl og metylsulfonyl eller en annen ekvivalent avspaltende gruppe. Forbindelser med formelen R<1->A med en substituent A som er i stand til å reagere, er kjente eller kan lett oppnås ved hjelp av den generelle fagkunnskap. Denne omsetning lar seg gjennomføre i vanllige løsemidler og gjennomføres med fordel under tilsetning av en base, hvorav de ovenfor nevnte kommer i betraktning.
Med henblikk på den biologiske virkning er slike karboksylsyrederivater med den generelle formel I - både som rene enantiomerer og som rene diastereomerer eller som blandinger av disse - foretrukket, hvor substituentene har følgende
betydning:
R betyr COOH;
X betyr CH;
minst én av restene X og Y beyr nitrogen (N);
minst én av restene R2 og R3 betyr CrC4-alkyl.
Eksempler på foretrukne forbindelser er angitt i den følgende tabell.
Forbindelsene ifølge foreliggende oppfinnelse tilveiebringer et nytt tera-peutisk potential for behandlingen av hypertoni, pulmonalt høytrykk, myokardinfarkt, angina pectoris, akutt nyresvikt, nyreinsuffisiens, cerebrale vasospasmer, zerebral iskemi, subaraknoidalblødninger, migrene, astma, aterosklerose, endotoksisk sjokk, endotoksinindusert organsvikt, intravaskulær koagulasjon, restenose etter angioplasti, benign prostata-hyperplasi, iskemisk nyresvikt og nyresvikt forårsaket av intoksikasjon eller hypertoni, samt kreftsykdommer, og særlig prostatakrebs og hudkreft. En ytterligere gjenstand for oppfinnelsen er anvendelse av kombinasjonen av forbindelser med formelen I med hemmende stoffer fra renin-angiotensin-systemet (RAS). RAS-hemmere er eksempelvis kjent fra EP 634 175.
Preparatene på denne basis egner seg for behandling av slike sykdommer, som også forbindelsene med formelen I viser virkning for, særlig for behandling av hypertoni og kronisk hjertesvikt.
Den gode virkning av forbindelsene lar seg vise i de følgende forsøk:
Reseptorbindingsstudier
For bindingsstudier ble klonede humane ETA-reseptor-uttrykkende CHO-celler og marsvin-lillehjerne-membraner med > 60 % ETB sammenliknet med ETA-reseptorer. x
Membranpreparering
De ETA-reseptor-uttrykkende CHO-celler fikk formere seg i Fi2-medium med 10 % føtalt kalveserum, 1 % glutamin, 100 E/ml penicillin og 0,2 % streptomycin (Gibco BRL, Gaithersburg, MD, USA). Etter 48 timer ble cellene vasket med PBS og inkubert med 0,05 % trypsinholdig PBS i 5 min. Deretter ble det nøytralisert med F12-medium og cellene ble samlet ved sentrifugering ved 300 x g. For lysing av cellene ble pelleten kort vasket med lysings-buffer (5 mM tris-HCI, pH 7,4 med 10 % glycerol) og deretter inkubert i en konsentrasjon på 10<7> celler/ml lysingsbuffer i 30 min. ved 4°. Membranene ble sentrifugert ved 20 000 x g i 10 min. og pelleten lagret i flytende nitrogen.
Marsvin-lillehjerner ble homogenisert i en Potter-Elvejhem-homogenisator og utvunnet ved differensiell sentrifugering i 10 min. ved 1.000 x g og gjentatt sentrifugering av det overstående i 10 min. ved 20 000 x g.
Bindingstester
For ETa- og ETB-reseptorbindingstesten ble membranene suspendert i inkubasjonsbuffer (50 mM tris-HCI, pH 7,4 med 5 mM MnCI2, 40 ug/ml bacitracin og 0,2 % BSA) i en konsentrasjon på 50 ug protein pr. testblanding og inkubert ved 25°C med 25 pM [125JJ-ET! (ETA-reseptortest) eller 25 pM [125J]-RZ3 (ETB-reseptortest) i nærvær og i fravær av testsubstans. Den ikke-spesifikke binding ble bestemt med 10" 7 M ETi. Etter 30 min. ble den frie og den bundne radioligand fraskilt ved filtrering over GF/B glassfiberfilter (Whatman, England) på en Skatron-cellesamler (Skatron, Lier, Norge) og filtrene ble vasket med iskald tris-HCI-buffer, pH 7,4 med 0,2 % BSA. Den radioaktivitet som var samlet på filtrene ble kvantifisert med en Packard 2200 CA væske-scincillasjonsteller.
Funksjonelt in vitro testsystem for å søke etter endotelinreseptor (subtype A)-antagonister
Dette testsystem er en funksjonell, cellebasert test for endotelinreseptorer. Bestemte celler viser, dersom de blir stimulert med endotelin 1 (ET1), en stigning i den intracellulære kalsiumkonsentrasjon. Denne stigning kan måles i intakte celler som er påført kalsium-sensitive farvestoffer.
1-Bindevevsceller som var blitt isolert fra rotter og hvor det var blitt påvist en endogen endotelinreseptor av A-subtype, ble belastet med fluorescensfarvestoffet Fura 2-an som følger: Etter trypsinering ble cellene resuspendert i buffer A (120 mM NaCI, 5 mM KCI, 1,5 mM MgCI2, 1 mM CaCI2, 25 mM HEPES, 10 mM glukose, pH 7,4) inntil en densitet på 2 x 106/ml og inkubert i 30 min. ved 37°C i mørke med Fura 2-am (2 uM), Pluronics F-127 (0,04 %) og DMSO (0,2 %). Deretter ble cellene vasket to ganger med buffer A og resuspendert til 2 x 10<6>/ml.
Fluorescens-signaletfra 2 x 10<5> celler pr. ml ved Ex/Em 380/510 ble ved 30°C registrert kontinuerlig. Testsubstansene ble satt til cellene, og etter en inku-basjonstid på 3 min. med ET1 ble den maksimale endring i fluorescensen bestemt. Cellenes respons på ET1 uten tilsetning av en testsubstans på forhånd tjente som kontroll og ble satt lik 100 %.
Testing av ET-antagonister i vivo
SD-hannrotter med vekt 250 - 300 g ble narkotisert med amobarbital, fikk kunstig åndedrett, og ble vagotomisert og despinalisert. Arteria carotis og vena
jugularis ble katetisert.
I kontrolldyrfører intravenøs administrering av 1 ug/kg ET1 til en tydelig blodtrykkstigning, som bibeholdes over et lengre tidsrom.
Testdyrene fikk en i.v.-injeksjon av testforbindelsene (1 ml/kg) 5 min. før administrering av ET1. For bestemmelse av de ET-antagonistiske egenskaper ble blodtrykkstigningen i testdyrene sammenliknet med blodtrykkstigningen i kontroll-dyrene.
Endotelin-1 indusert "plutselig død" for mus
Testprinsippet besetår i hemning av den plutselige hjertedød for mus forårsaket av endotelin, og som sannsynligvis er betinget av sammentrekning av koro-narkarene, ved forbehandling med endotelin-reseptorantagonisten. Etter intravenøs injeksjon av 10 nmol/kg endotelin i et volum på 5 ml/kg kroppsvekt dør dyrene i løpet av få minutter.
Den letale endotelin-1-dosis etterprøves i hvert tilfelle på et lite dyrekollektiv. Dersom testsubstansen administreres intravenøst, finner i de fleste tilfeller den for kontrollgruppen letale endotelin-1-injeksjon sted 5 minutter senere. Ved andre administreringsmåter forlenges tidene før administrering, eventuelt opptil flere timer.
Overlevelsesgraden noteres og den effektive dose, som beskytter 50 % av dyrene (ED 50) i 24 timer eller lenger mot endotelin-indusert hjertedød, blir bestemt.
Funksjonell kartest for endotelin-reseptorantagonister
På aortasegmenter fra kanin blir det etter en for-spenning på 2 g og en relaksasjonstid på 1 time i Krebs-Henseleit-løsning ved 37°C og en pH-verdi mellom 7,3 og 7,4 først utløst en K<+->kontraktur. Etter utvasking blir det laget en endotelin-dosisvirkningskurve opptil maksimum.
Potensielle endotelin-antagonister blir administrert til andre preparater i det samme kar 15 minutter før begynnelsen av endotelin-dosisvirkningskurven. Virk-ningene av endotelinet blir beregnet i % av K<+->kontrakturen. Ved virksomme endotelin-antagonister opptrer høyreforskyvning av endotelin-dosisvirkningskurven.
Forbindelsene ifølge oppfinnelsen kan på vanlig måte administreres oralt eller parenteralt (subkutant, intravenøst, intramuskulært, intraperotonealt). Administrering kan også finne sted med damper eller spray gjennom nese-hals-området.
Doseringen avhenger av pasientens alder, tilstand og vekt, samt av admi-nistreringsmåten. Vanligvis utgjør den daglige virkestoffdosis mellom ca. 0,5 og 50 mg/kg legemsvekt ved oral administrering og mellom ca. 0,1 og 10 mg/kg legemsvekt ved parenteral administrering.
De nye forbindelser kan anvendes i de vanlige galeniske faste eller flytende former, f.eks. som tabletter, filmtabletter, kapsler, pulvere, granulater, dragéer, suppositorier, løsninger, salver, kremer eller spray. Disse fremstilles på vanlig måte. Virkestoffene kan bearbeides med de vanllige galeniske hjelpemidler så som tablett-bindemidler, fyllstoffer, konserveringsmidler, tablett-sprengmidler, flytreguleringsmidler, myknere, overflateaktive midler, dispergeringsmidler, emul-gatorer, løsemidler, retarderingsmidler, antioksydanter og/eller drivgasser (se. H.. Suckeretal.: Pharmazeutische Technologie, Thieme-Verlag, Stuttgart, 1991). De således oppnådde anvendelsesformer innholder normalt virkestoffet i en mengde på 0,1 til 90 vekt%.
Syntese-eksempler
Eksempel 1
2-Hydroksy-3-metoksy-3,3-difenylpropionsyremetylester
5 g (19,6 mmol) 3,3-difenyl-2,3-epoksypropionsyremetylester ble oppløst i 50 ml absolutt metanol og ved 0°C blandet med 0,1 ml bortrifluorid-eterat. Det ble omrørt i 2 timer ved 0°C og ytterligere 12 timer ved romtemperatur. Løsemidlet ble avdestillert, resten tatt opp med eddiksyreester, vasket med natriumhydro-genkarbonat-løsning og vann og tørket over magnesiumsulfat. Etter avdestillering av løsemidlet ble det igjen 5,5 g (88 %) av en svakt gul olje.
Eksempel 2
2-(2,6-Dimetoksy-pyrimidin-4-yloksy)-3-metoksy-3,3-difenyl-propionsyremetylester
1.15 g (4 mmol) 2-hydroksy-3-metoksy-3,3-difenyl-propionsyremetylester ble oppløst i 10 ml dimetylformamid og blandet med 276 mg (2 mmol) kaliumkarbonat. Det ble så tilsatt 524 mg (3 mmol) 2,6-dimetoksy-4-klorpyrimidin og omrørt i 6 timer
ved 100°C. Så ble det forsiktig hydrolysert med 10 ml vann, med sitronsyre innstilt på en pH-verdi på 5, ekstrahert med eddiksyreester, den organiske fase ble vasket med-vann, tørket over magnesiumsulfat og løsemidlet avdestillert. Den oljeaktige rest (1,9
g) ble kromatografert på silikagel, hvorved det ble oppnådd 617 mg av en lett uren
olje.
Eksempel 3
2-(2,6-Dimetoksy-pyrimidin-4-yloksy)-3-metoksy-3,3-difenyl-propionsyre
550 mg (1,3 mmol) 2-(2,6-dimetoksy-pyrimidin-4-yloksy)-3-metoksy-3,3-difenyl-propionsyremetylester ble oppløst i 5 ml dioksan, blandet med 2.6 ml 1 n KOH-løsning og omrørt i 3 timer ved 100°C. Løsningen ble fortynnet med 300 ml
vann og ekstrahert med eddiksyreester for fjerning av ikke omsatt ester. Deretter ble vannfasen med fortynnet saltsyre innstilt på pH 1-2 og det ble ekstrahert med eddiksyreester. Etter tørking over magnesiumsulfat og avdestillering av løsemidlet ble den skumformige rest (425 mg) renset over MPLC, hvorved det ble oppnådd 205 mg (40%) av et skumformig produkt.
Eksempel 4
2-(4-Metylchinolin-2-yloksy)-3-metoksy-3,3-difenyl-propionsyremetylester
5.7 g (20 mmol) 2-hydroksy-3-metoksy-3,3-difenyl-propionsyremetylester ble oppløst i 90 ml dimetylformamid og blandet med 0,98 g (22 mmol) natriumhydrid (55% i olje). Det ble omrørt i 15 minutter og deretter tilsatt 3,9 g (22 mmol) 2-klor-4-metylchinolin. Den mørkerøde løsning ble omrørt natten over ved romtemperatur, så forsiktig hydrolysert med 20 ml vann, og deretter ekstrahert med eddiksyreester. Eksempel 5
2-(4-Metylchinolin-2-yloksy)-3-metoksy-3,3-difenyl-propionsyre
1.8 g (4,2 mmol) 2-(4-metylchinolin-2-yloksy)-3-metoksy-3,3-difenyl-propionsyremetylester ble oppløst i 25 ml dioksan, blandet med 8,4 ml 1 n KOH-løsning og omrørt i 20 timer ved 60°C. Løsningen ble fortynnet med 300 ml vann og ekstrahert med eddiksyreester for fjerning av ikke omsatt ester og forurensninger. Deretter ble vannfasen med fortynnet saltsyre innstilt på pH 1-2 og det ble ekstrahert med eddiksyreester. Etter tørking over magnesiumsulfat og avdestillering av løsemidlet ble det oppnådd 1,0 g av et skunr Dette ble oppløst i en liten mengde eddiksyreester og fikk stå over natten ved romtemperatur. Derved oppsto 265 mg av
produktet i form av hvite krystaller. Gjentakelse av denne prosedyre med moderluten ga ytterligere 166 mg av produktet.
Samlet utbytte: 431 mg ^ 25%

Eksempel 6
2-(2-7e/f-butyl-6-trifluormetyl-pyrimidin-4-yloksy)-3-metoksy- 3,3-difenyl-propionsyre-benzylester
2,0 g (5.5 mmol) 2-hydroksy-3-metoksy-3,3-difenyl-propionsyrebenzylester (se P44 36 851.8) ble oppløst i 20 ml dimetylformamid og blandet med 0,76 g (5,5 mmol) kaliumkarbonat. Det ble så tilsatt 1,32 g (5,5 mmol) 2-tert-butyl-6-trifluormetyl-4-klorpyrimidin, omrørt over natten ved romtemperatur, og så 5 timer ved 100°C. For å gjøre reaksjonen fullstendig ble ytterligere 0,5 g pyrimidin samt 0,5 g NaH tilsatt og det ble omrørt ytterligere 6 timer ved 60°C. Til slutt ble reaksjonsblandingen helt på isvann, ekstrahert med eddiksyreester, den organiske fase ble vasket med mettet NaCI-løsning, tørket over magnesiumsulfat og løsemidlet avdestillert. Den oljeaktige rest (3,5 g) krystalliserte i n-heptan/eddiksyreester (5%). Fellingen ble frafiltrert og tørket.
Utbytte: 1,6 g = 52%

Eksempel 7
2-(2-rerr-butyl-6-trifluormetyl-pyrimidin-4-yloksy)-3-metoksy- 3,3-difenyl-propionsyre
2,0 g (5,5 mmol) 2-(2-terr-butyl-6-trifluormetyl-pyrimidin-4-yloksy)-3-metoksy-3,3-difenyl-propionsyrebenzylester ble oppløst i 250 ml metanol, blandet med 120 mg palladium på aktivkull (5%) og under hydrogen omrørt ved romtemperatur i 2 timer. Etter avsluttet hydrogenopptak ble katalysatoren frafiltrert over Celite og løsemidlet trukket av i rotasjonsfordamperen.
Utbytte: 1,0 g 98%
1H-NMR (360 MHz, i CDCI3, angivelse i ppm): 1.35 (s, 9H); 3.3 (s, 3H); 6.3 (s, 1H); 6.8 (s, 1H); 7.15 - 7.45 (m, 10H) -
På samme måte kan de i tabell 1 oppførte forbindelser fremstilles.

I henhold til den ovenfor beskrevne bindingstest ble det for de nedenfor angitte forbindelser målt reseptorbindingsdata.
Resultatene er vist i tabell 2.
Claims (7)
1. Karboksylsyrederivater med formelen I
karakterisert ved at R betyr en gruppe
i hvilken R<1> har betydningen en rest OR<8>, hvori R<8> betyr: hydrogen, kationet av et alkalimetall så som litium, natrium, kalium, eller
kationet av et jordalkalimetall så som kalsium, magnesium og barium, eller et fysiologisk godtagbart organisk ammoniumion så som tertiært C1-C4-alkylammonium eller ammoniumionet; C3-C8-cykloalkyl, som cyklopropyl, cyklobutyl, cyklopentyl, cykloheksyl og
cykloheptyl, cyklooctyl; Ci-Cs-alkyl, særlig CrC4-alkyl som metyl, etyl, n-propyl, iso-propyl,
n-butyl, iso-butyl, tert.-butyl; CH2-fenyl,
og de øvrige substituenter har følgende betydning: R<2> er CrC4-alkyl, CrC4-halogenalkyl, CrC4-alkoksy; X er nitrogen eller CR<12>, hvori CR<12> danner sammen med CR2 eller CR3 en
fenylring Y er nitrogen; R<3> er hydrogen, Ci-C4-alkyl, CrC4-halogenalkyl, Ci-C4-alkoksy; R<4> og R5 er fenyl;R<6> er d-Cs-alkyl; Z er oksygen.
2. Karboksylsyrederivater ifølge krav 1,
karakterisert ved at R betyr COOH.
3. Karboksylsyrederivater ifølge et av de foregående krav,
karakterisert ved at X betyr CH.
4. Karboksylsyrederivater ifølge et av de foregående krav,
karakterisert ved at minst én av restene X, Y betyr nitrogen (N).
5. Karboksylsyrederivater ifølge et av de foregående krav,
karakterisert ved at minst én av restene R2, R3 betyr CrC4-alkyl.
6. Anvendelse av forbindelser ifølge krav 1 - 5 for fremstilling av medikamenter for behandling av hypertoni, pulmonalt høytrykk, aktutt og kronisk nyresvikt, kronisk hjertesvikt, cerebral ischemi, restenose-etter angioplasti, prostatakreft.
7. Anvendelse av en kombinasjon av en forbindelse ifølge krav 1-5 med en renin-angiotensin-system (RAS) -inhibitor for fremstilling av medikamenter for behandling av hypertoni, pulmonalt høytrykk, aktutt og kronisk nyresvikt, kronisk hjertesvikt, cerebral ischemi, restenose etter angioplasti, prostatakreft.
