Description
Foreliggende oppfinnelse angår en ny syntese av 4-amino-5-heksensyre under anvendelse av termiske omleirings-reaksjoner, og nye mellomprodukter fremstilt derved.
4-amino-5-heksensyre, ellers kjent som vigabatrin eller vinyl-GABA, er en GABA-T-inhibitor markedsført under varemerket SABRIL<®> for behandling av epilepsi. (Se oversikts-artikkel om vigabatrin av S.M. Grant et al. i Drugs, 41 (6): 889-926, 1991).
Oppfinnelsen angår således en fremgangsmåte for fremstilling av 4-amino-5-heksensyre og dens farmasøytisk akseptable salter, hvilken fremgangsmåte er kjennetegnet ved at den omfatter trinnene: (a) termisk omleiring av erytritol til 4-formyl-3-hydroksy-l-buten, (b) termisk omleiring av 4-formyloksy-3-hydroksy-l-buten til etyl-6-formyloksy-4-heksenoat, etterfulgt av omdannelse av formiatet til dets tilsvarende alkohol, (c) omdannelse av det således fremstilte etyl-6-hydroksy-4-heksenoat til etyl-6-trikloracetimid-oksy-4-heksenoat ved omsetning med trikloracetonitril, etterfulgt av dets termiske omleiring til dets imidat, etyl-4-trikloracetoamido-5-heksenoat, som ved hydrolyse omdannes til den
ønskede 4-amino-5-heksensyre, og eventuell omdannelse av angitte syre til et farmasøytisk akseptabelt salt derav.
Denne prosess er basert på kjente termiske reaksjoner som starter fra erytritol; hvilke termiske reaksjoner er (1) en elimineringsprosess for dannelse av en dobbeltbinding, (2) en Claisen-omleiring og (3) en Overman-omleiring. Den invol-verte reaksjonssekvens er vist ved følgende reaksjonsskjerna.

Trinn (a) av prosessen innbefatter den kjente termiske omleiringsreaksjon for fremstilling av 4-formyloksy-3-hydroksy-l-buten (2) fra erytritol (1) (se Prevost, C, Ann. Chem. [10], 10, 398, 1928). Selv pm ingen opparbeidelse er nødvendig, kan bedre utbytter av en renere forbindelse erholdes hvis produktet redestilleres. Trinn (b) innbefatter en andre-termisk omleiringsreaksjon - etterfulgt av en hydrolyse
- hvori 4-formyl-3-hydroksy-l-buten varmes til 140-150 °C i nærvær av overskuddsmengder av ortoacetatet (4 til 1) under betingelser for fjerning av den ln situ fremstilte alkohol (se Johnson, W. og Coll, J. Am. Chem. Soc., 92, 741, 1970). Etter hydrolyse og fjerning av overskudd av ortoacetat kan det således fremstilte produkt etyl-6-formyloksy-4-heksenoat anvendes som sådant, eller det kan eventuelt underkastes en destillasjon for rensing, eller det kan underkastes flashkromatografi på Si02. Alternativt kan denne termiske omleiring utføres under anvendelse av én ekvivalent av ortoacetatet i et inert løsningsmiddel som koker rundt 140-150 "C (f.eks. xylen). Reaksjonstiden for disse reaksjoner kan overvåkes ved måling
av alkoholen (metanol eller etanol), som destilleres av.
Trinn (c) innbefatter omdannelse av formiatet til dets tilsvarende alkohol ved at formiatet tillates å bli om-rørt ved temperaturer på ca. 15-25 °C mens det foreligger i absolutt etanol, til hvilket katalytiske mengder av alkoholisk HCl-gass er blitt tilsatt. Trinn (d) innbefatter omsetning av trikloracetonitrilet med etyl-6-hydroksy-4-heksenoat i nærvær av katalytiske mengder NaH (-0,1 ekvivalent) i et aprotisk, vannfritt løsningsmiddel (fortrinnsvis vannfri eter) under en inertgass, fortrinnsvis nitrogen, ved ca. 0 "C, under dannelse av et in situ imidatmellomprodukt (5), som ved termisk omleiring omdannes til etyl-4-trikloracetoamido-5-heksenoat (6); idet omleiringen bevirkes under anvendelse av teknikkene ifølge Overman, L.J., Am. Chem. Soc, 98, 2901, 1976. Slutt-trinnet innbefatter hydrolyse av imidatet, fortrinnsvis ved syrehydrolyse, men alternativt under anvendelse av basiske hydrolysebetingelser, under dannelse av den ønskede 4-amino-heksensyre, som dets HCl-salt. Den frie syre eller andre far-masøytisk akseptable salter derav kan erholdes ved standard-prosedyrer vel kjent innen faget.
Fordelene ved denne fremgangsmåte kan oppsummeres som følger: (1) prosessen anvender ikke eller danner karsinogene materialer, heller ikke anvendes noen farlige reaktanter eller løsningsmidler, (2) utgangsmaterialet kan fremstilles fra et billig råmateriale (potetstivelse), (3) reaksjonssekvensen kan utføres med bare én rensing før den sluttelige hydrolyse, (4) et begrenset antall organiske løsningsmidler er nødvendig, (5) overskudd av reaktanter (f.eks. triklororto-acetat) og løsningsmidler (f.eks. xylen) kan gjenvinnes og resykliseres,
(6) mangel på uønskede biprodukter,
(7) reaksjonene er enkle uten problemer assosiert med temperaturkontroll, og produktene kan renses uten behov for kromatografisk opparbeidelse.
Det etterfølgende eksempel illustrerer den nye fremgangsmåte ifølge oppfinnelsen.

Trinn A: 4- formvloksv- 3- hvdroksv- l- buten
En løsning av 50 g (0,5 mol) erytritol i 150 g (75 %) vandig maursyre ble oppvarmet til over 100 °C i 12 timer, hvorpå vann og maursyre ble destillert fra, og reaksjons-blandingen ble oppvarmet til over 200 °C med en Bunsen-brenner. Trinn B: Etvl- 6- formvloksv- 4- heksenoat
En løsning av 1,06 g (10 mmol) 4-formyloksy-3-hydroksy-l-buten og 1 dråpe propionsyre i 6 g (40 mmol) trietyl-ortoacetat ble oppvarmet til 140 "C under betingelser for destillasjonsmessig fjerning av etanol. Etter 2 timer ble overskudd av etylortoacetat fjernet ved destillasjon i vakuum. Residuet ble hydrolysert med vann og ble ekstrahert med AcOEt. Produktet ble renset ved flashkromatografi på Si02 (elueringsmiddel AcOEtiheksan, 2:8) (1 g, 60 %), men rensing ved destillasjon er foretrukket når større mengder er involvert.
<X>H NMR (90 MHz) (CDC13, TMS) 6 ppm, 1,26 (t, 3 H, CH3, J = 6 Hz), 2,4 (s, 4 H, (CH2)2), 4,1 (q, 2 H, CH2, J = 6 Hz), 4,6 (d, 2 H, CH2-C=, J = 6 Hz), 5,73 (m, 2 H, CH=CH), 8,06 (s, 1 H, HC02).
Trinn C: Etyl- 6- hydroksy- 4- heksenoat
En løsning av 0,9 g (5 mmol) 6-formyloksy-6-heksenoat i 10 ml absolutt EtOH inneholdende noen få dråper av en mettet løsning av alkoholisk HCl-gass fikk stå i 2 timer ved 20 °C. Løsningsmidlet ble fjernet i vakuum, og residuet ble anvendt i neste trinn uten ytterligere rensing (0,7 g, kvantitativt). Denne forbindelsen ble funnet å bli delvis spaltet ved flashkromatografi på Si02.

Trinn D: Etvl- 4- trikloracetoamido- 5- heksenoat
Natriumhydrid (0,03 g av en 50 % dispersjon i olje, 0,5 mmol) ble tilsatt til en løsning av 0,7 g (5 mmol) etyl-6-hydroksy-4-heksenoat og 0,6 g (5 mmol) trikloracetonitril i 50 ml vannf r i eter under N2 ved 0 °C. Etter 1 time ble 0,5 mmol etanol tilsatt, og løsningsmidlet ble fjernet i vakuum. Dan-nelsen av imidatet ble regulert ved NMR (NH, ~8,5 ppm). En løsning av det urene imidat i xylen (30 ml) ble oppvarmet til tilbakeløpskoking i 48 timer. Løsningsmidlet ble deretter fjernet i vakuum, og residuet ble renset ved flashkromatografi på Si02, (elueringsmiddel AcOEtrheksan, 2:8) under dannelse av tittelproduktet (1,1 g, -70 %).
<X>H NMR (90 MHz) (CDC13, TMS) 6 ppm, 1,23 (t, 3 H, CH3, J = 6 Hz), 2,0 (t, 2 H, CH2-CH2-C02, J = 5 Hz), 2,36 (s, 2 H, CH2C02), 4,1 (q, 2 H, CH3CH2, J = 6 Hz), 4,4 (t, 1 H, CH-CH2, J = 5 Hz), 5,1 (m, 2 H, CH2), 5,76 (m, 1 H, CH=CH2), 7,2 (s, 1 H,
NH).
En prøve ble destillert for analyse (k.p. 150 °C, 0,5 mm Hg).
Analyse beregnet for C10H14N03C13:
Trinn E: 4- amino- 5- heksensyre
En suspensjon av 0,3 g (1 mmol) etyl-4-trikloracetoamido-5-heksenoat i 10 ml 6 N HC1 ble oppvarmet under tilbake-løpskjøling i 6 timer. Blandingen ble deretter konsentrert i vakuum, ble fortynnet med 10 ml vann, ble vasket to ganger med AcOEt, og ble tørket i vakuum under dannelse av tittelproduktet (0,18 g, 100 %). NMR, TLC (NH40H:EtOH, 3:7) er identisk med dem av en autentisk prøve av 4-amino-5-heksensyre.

Claims (4)
1. Fremgangsmåte for fremstilling av 4-amino-5-heksensyre og dens farmasøytisk akseptable salter, karakterisert ved at den omfatter trinnene: (a) termisk omleiring av erytritol til 4-formyl-3-hydroksy-1-buten, (b) termisk omleiring av 4-formyloksy-3-hydroksy-l-buten til etyl-6-formyloksy-4-heksenoat, etterfulgt av omdannelse av formiatet til dets tilsvarende alkohol, (c) omdannelse av det således fremstilte etyl-6-hydroksy-4-heksenoat til etyl-6-trikloracetimid-oksy-4-heksenoat ved omsetning med trikloracetonitril, etterfulgt av dets termiske omleiring til dets imidat, etyl-4-trikloracetoamido-5-heksenoat, som ved hydrolyse omdannes til den ønskede 4-amino-5-heksensyre, og eventuell omdannelse av angitte syre til et farmasøytisk akseptabelt salt derav.
