Description
A találmány tárgya új eljárás 4-amino-5-hexénsav előállítására termikus átrendezési reakciók alkalmazásával.
A 4-amino-5-hexénsav más néven vigabatrin vagy vinil-GABA egy GABA-T inhibitor, amelyet a SABRIL márkanév alatt forgalmaznak epilepszia kezelése céljából (lásd a vigabatrinról szóló összefoglaló közleményt: S.M. Grant és munkatársai, Drugs, 41(6): 889-926, 1991).
A találmány tárgya eljárás 4-amino-5-hexénsav és gyógyszerészetileg elfogadható sói előállítására, mely eljárás során (i) eritritolt hangyasav jelenlétében termikus reakció során dehidratálunk és formilezünk;
(ii) a kapott 4-(formil-oxi)-3-hidroxi-l-butént trietil-ortoacetáttal reagáltatjuk termikus reakciókörülmények között, a keletkezett etil-6-(formil~oxi)-4-hexenoátot etanol és sósav jelenlétében etil-6-hidroxi-4-hexanoáttá alakítjuk;
(iii) az így kapott etil-6-hidroxi-4-hexenoátot triklór-acetonitrillel végzett reakció segítségével átalakítjuk etil-6-(triklór-acetimidoil-oxi)-4-hexenoáttá, majd ezt a vegyületet termikusán átrendezzük és az így kapott éti 1-4-(triklór-acetamido)-5-hexenoátot hidrolízis segítségével a kívánt 4-amino-5-hexénsavvá alakítjuk át; és kívánt esetben a savat gyógyszerészetileg elfogadható sóvá alakítjuk.
A találmány szerinti eljárás lényegében ismert termikus reakciókon alapul eritritol kiindulási vegyületet alkalmazva. A termikus reakciók magukban foglalják az (1) eliminációs reakciót, amelynek során a kettős kötés keletkezik, a (2) Claisen átrendeződést és a (3) Overman átrendeződést. A találmány szerinti eljárást az A reakcióvázlatban mutatjuk be.

Az eljárás (a) reakciólépése ismert termikus átrendeződési reakció, amelynek során az (1) képletű eritritolból a (2) képletű 4-(formil-oxi)-3-hidroxi-l-butént állítjuk elő (lásd Prevost C., Ann. Chem. [10], 10, 398, 1928 közleményét).
Ugyan nem szükséges a reakcióelegy feldolgozása, azonban tisztább vegyület jobb hozamát érhetjük el, amennyiben a terméket desztillációval újra tisztítjuk. A (b) reakciólépésben egy második termikus átrendezési reakciót, majd ezt követő hidrolízist végzünk, amelynek során a 4-(formil-oxi)-3-hidroxi-l-butént 140-150 °C hőmérsékletre hevítjük felesleg menynyiségű ortoacetát jelenlétében (4:1) olyan reakciókörülmények között, amelyek során in situ eltávozik a képződő alkohol (lásd Johnson W. és Coll, J. Am. Chem. Soc. 92, 741, 1970). A felesleg ortoacetát eltávolítása, illetve a hidrolízis után az így kapott etil-6-(formil-oxi)-4-hexenoát ebben a formában is alkalmazható a következő reakciólépésben vagy kívánt esetben tisztítás céljából desztillációnak vethetjük alá, vagy szilikagélen gyorskromatográfia segítségével tisztíthatjuk. Más eljárás szerint a fenti termikus átrendeződést ekvivalens mennyiségű ortoacetát alkalmazásával inért oldószerben is végezhetjük, amely oldószer forráspontja kb. 140-150 °C közötti érték (pl. xilol). A fenti reakciók reakcióidejét az alkohol (metanol vagy etanol) mennyiségének mérésével követhetjük, ugyanis ez az alkohol a reakcióelegyből kidesztillál.
A (c) reakciólépésben a formiátot megfelelő alkohol vegyületté alakítjuk úgy, hogy a formiátot kb. 15-25 °C hőmérsékleten hagyjuk keveredni száraz etanolban, amelyhez katalitikus mennyiségű alkoholos sósavgázoldatot adagoltunk. A (d) reakciólépésben triklór-acetonitrilt reagáltatunk etil-6-hidroxi-4-hexenoáttal katalitikus mennyiségű nátrium-hidrid (kb. 0,1 ekvivalens) jelenlétében aprotikus száraz oldószerben (előnyösen száraz éterben) inért gáz atmoszférában, előnyösen nitrogén atmoszférában kb. 0 °C hőmérsékleten. így az (5) képletű imidát közbenső termék in situ keletkezik, amelyet ezután termikus átrendeződés segítségével a (6) képletű etil-4-(triklór-acetamido)-5-hexenoáttá alakítunk át. A termikus átrendeződési reakciót Overman L. J., Am. Chem. Soc. 98, 2901, 1976 közleményében leírt eljárás szerint végezzük. A végső reakciólépésben az imidátot hidrolizáljuk, előnyösen savas hidrolízis segítségével, azonban más eljárás szerint bázisos hidrolízist is alkalmazhatunk, és így a 4-amino-hexénsavat nyerjük sósavas só formában. A szabad sav vagy más gyógyszerészetileg elfogadható sók ebből standard eljárásokkal a szakirodalomban ismert módon állíthatók elő.
A találmány szerinti eljárás előnyei az alábbiak:
(1) az eljárásban nem alkalmazunk, illetve nem képződik karcinogén anyag, továbbá nem alkalmazunk veszélyes reaktánsokat vagy oldószereket, (2) a kiindulási anyag olcsó nyersanyagból (burgonyakeményítő) állítható elő, (3) a reakciósorozat csak egyetlen tisztítási lépés alkalmazásával hajtható végre egészen az utolsó hidrolízis lépésig, (4) az alkalmazott szerves oldószerek csak kis számúak, (5) a reaktánsok (például triklór-ortoacetát) és oldószerek (például xilol) feleslege kinyerhető, illetve rccirkuláltatható, (6) nem kelezkeznek nem kívánatos melléktermékek, (7) a reakciók végrehajtása könnyű, nem szükséges különös hőmérséklet szabályozás és a termékek kromatográfiás eljárás alkamazása nélkül tisztíthatok.
A találmány szerinti eljárást az alábbi példán részletesen is bemutatjuk.
1. példa
4-amino-5-hexénsav (a) reakciólépés: 4-(formil-oxi)-3-hidroxi-l-butén előállítása: 50 g (0,5 mól) eritritol 150 g (75 t%-os) vizes hangyasavban készült oldatát 100 °C hőmérséklet fölé melegítjük 12 órán át, majd a vizet és a hangyasavat a reakcióelegyből kidesztilláljuk és a reakcióelegyet 200 °C hőmérséklet fölé melegítjük Bunsen-égő segítségével. A terméket desztilláció segítségével nyerjük ki, (F.p.: 230 °C, 30 g) és desztilláció segítségével tisztítjuk. (F.p.: 90 °C/15 Hgmm, 1995 Pa).
’H-NMR spektrum (90 MHz) (CDC1·,, TMS) őppm. 3.23 (s, 1H, OH), 3.6 (m, 1H, CH), 4.23 (t, 2H; CH2), 5.33 (m, 2H, CH2=), 5.83 (m, 1H, -CH=), 8.16 (s, 1H, HCO2).
(b) reakciólépés: etil-6-(formil-oxi)-4-hexenoát előállítása: 1,06 g (10 mmol) 4-(formil-oxi)-3-hidroxi-l2
HU 213 374 Β
-butén és 1 csepp propionsav 6 g (40 mmol) tríetil-ortoacetátban készült elegyét 140 °C hőmérsékletre melegítjük úgy, hogy közben az etanolt desztilláció segítségével eltávolítjuk. 2 óra elteltével a felesleg etil-ortoacetátot vákuumdesztilláció segítségével eltávolítjuk. A maradékot víz segítségével hidrolizáljuk, majd a vizes elegyet etil-acetáttal extraháljuk. A terméket gyorskromatográfia segítségével szilikagélen 2 : 8 térfogatarányú etil-acetát : hexán eluens alkalmazásával tisztítjuk, 1 g terméket nyerünk, 60% termelés. Alkalmazhatunk előnyösen desztillációs tisztítási lépést is, amennyiben nagyobb mennyiségű terméket állítunk elő.
'H-NMR spektrum (90 MHz) (CDC13), TMS) δρρηι. 1.26 (t, 3H, CH,, J = 6 Hz), 2.4 (s, 4H, (CH2)2), 4.1 (q, 2H, CH2, J = 6HZ), 4.6 (d, 2H, CH2-C=, J = 6 Hz), 5.73 (m, 2H, CH=CH), 8.06 (s, 1H, HCO2).
(c) reakciólépés: etil-6-hidroxi-4-hexenoát előállítása:
0,9 g (5 mmol) etil-6-(formil-oxi)-4-hexenoát 10 ml száraz etanolban készült oldatához néhány csepp telített alkoholos hidrogénklorid oldatot adagolunk, majd az elegyet 2 órán át 20 °C hőmérsékleten tartjuk. Ezután az oldószert vákuumban elpárologtatjuk és a maradékot a következő reakciólépésben tisztítás nélkül felhasználjuk (0,7 g kvantitatív termelés). A vegyület szilikagélen végzett gyorskromatográfia során részben bomlik.
'H-NMR spektrum (90 MHz) (CDC13), TMS) őppm. 1.26 (t, 3H, CH3, J = 6 Hz), 2.4 (s, 4H, (CH2)2), 2.83 (s, 1H, OH), 4.1 (s, 2H, CH2-C=) 4.16 (q, 2H, CH3CH2, J = 6 Hz) 5.7 (s, 2H, CH=CH).
(d) és (e) reakciótépés: etil-4-(triklór-acetamido)-5-hexenoát előállítása:
0,7 g (5 mmol) etil-6-hidroxi-4-hexenoát és 0,6 g (5 mmol) triklör-acetonitril 50 ml száraz éterben készült oldatához nitrogén atmoszférában 0 °C hőmérsékleten 0,03 g (50 t%-os olajos diszperzió, 0,5 mmol) nátrium-hidridet adagolunk. Az elegyet 1 órán át keverjük, majd a keverékhez 0,5 mmol etanolt adagolunk és az oldószert vákuumban elpárologtatjuk. Az imidát képződést NMR analízis segítségével ellenőrizzük (NH körülbelül
8,5 ppm). A nyers imidát 30 ml xilolban készült oldatát 48 órán át visszafolyatás mellett forraljuk. Ezután az oldószert vákuumban elpárologtatjuk és a maradékot gyorskromatográfia segítségével szilikagélen 2 : 8 térfogatarányú etil-acetát: hexán, eluens alkalmazásával tisztítjuk. 1,1 g címbeli vegyületet kapunk (kb. 70% termelés).
'H-NMR spektrum (90 MHz) (CDC13, TMS) őppm. 1.23 (t, 3H, CH,, J = 6 Hz), 2.0 (t, 2H, CH2-CH2-CO2, J = 5 Hz), 2.36 (s, 2H, CH2CO2), 4.1 (q, 2H, CH3CH2, J = 6 Hz), 4.4 (t, 1H, CH-CH2, J = 5 Hz), 5.1 (m, 2H, CH2), 5.76 (m, 1H, CH=CH2), 7.2 (s, 1H, NH).
Egy minta mennyiségű anyagot analízis céljára desztillációval tisztítunk (F.p.: 150 °C, 0,5 Hgmm, 67 Pa). Elemenalízis a CioH|4N03C13 képlet alapján számított: C 39.69; H 4.66; N 4.64 mért: C 39,87; H 4.62; N 4.49 (f) reakciólépés: 4-amino-5-hexénsav előállítása:
0,3 g(l mmol) etil-4-(triklór-acetamido)-5-hexenoát 10 ml 6n sósavban készült oldatát 6 órán át visszafolyatás mellett forraljuk. Ezután az elegyet vákuumban bepároljuk, majd a maradékot 10 ml vízzel hígítjuk. A vizes elegyet etil-acetáttal kétszer extraháljuk, majd az anyagot vákuumban megszárítjuk. így a címbeli vegyületet kapjuk (0,18 g 100% termelés).
Az NMR spektrum a vékonyrétegkromatográfiás analízis (NH4OH:EtOH (3 : 7 eluens) azonos az autentikus minta 4-amino-5-hexénsav adataival.
' H-NMR spektrum (90 MHz) (D2O), őppm. (TMS) 1.83 (m, 2H, CH2CO2), 2.33 (m, 2H, CH2CH2) 3.66 (m, 1 H, CJ-C=), 5.35 (m, 3H, CH2 = CH ).
Claims (1)
- SZABADALMI IGÉNYPONT1. Eljárás 4-amino-5-hexénsav és gyógyszerészetileg elfogadható sói előállítására, azzal jelemezve, hogy (i) eritritolt hangyasav jelenlétében termikus reakció során dehidratálunk és formilezünk;(ii) a kapott 4-(formil-oxi)-3-hidroxi-l-butént trietil-ortoacetáttal reagáltatjuk termikus reakciókörülmények között, a keletkezett etil-6-(formil-oxi)-4-hexenoátot etanol és sósav jelenlétében etil-6-hidroxi-4-hexanoáttá alakítjuk;(iii) az így kapott etil-6-hidroxi-4-hexenoátot triklór-acetonitrillel végzett reakció segítségével átalakítjuk etil-6-(triklór-acetimidoil-oxi)-4-hexenoáttá, majd ezt a vegyületet termikusán átrendezzük és az így kapott éti 1-4-(triklór-acetamido)-5-hexenoátot hidrolízis segítségével a kívánt 4-amino-5-hexénsawá alakítjuk át; és kívánt esetben a savat gyógyszerészetileg elfogadható sóvá alakítjuk.
