Description
Foreliggende oppfinnelse vedrører kjemiske forbindelser og anvendelse derav. L-glutamat og L-asparginat, de endogene, sure aminosyrer, er blitt fastslått som viktige eksitatoriske neuro-transmittere. Virkningen av disse eksitatoriske aminosyrer blir formidlet av flere klare reseptorundertyper, hvorav en av de mest studerte er N-metyl-D-asparginat(NMDA)reseptoren. For kraftig aktivering av NMDA-reseptorkomplekset kan forårsake neuronal overstimulering med patologiske konsekvenser. Eksperimentell evidens antyder at en forlenget agonistfremkalt ledeevne i en NMDA-gated ionekanal vil tillate en unormal økning av kalsiuminngangen, og det resulterende økte intra-cellulære kalsiumnivå spiller en avgjørende skadelig rolle ved eksitotoksisk neuronskade, neurodegenerering og forsinket neuromdød.
Eksitatoriske aminosyrer settes i sammenheng med neuro-patologi av traumatisk, endogent, genetisk og miljømessig opprinnelse. Hjerneskade i forbindelse med anoxia hypo-glycemia, traumatisk skade, slag, epilepsi, spesifikke metaboliske defekter og noen kroniske neurodegenerative syk-dommer er i stor utstrekning frembragt ved eksitotoksiske mekanismer.
Flere studier har vist at en blokkering av NMDA-under-gruppe reseptoren signifikant vil redusere en neuronskade og
-tap som foregår i dyremodeller som etterligner flere forskjellige neuropatologiske situasjoner. Disse observasjoner viser klart at NMDA antagonister tilbyr effektiv neurobeskytt-else i flere kliniske miljø. Midler som motvirker de eksitotoksiske effekter mediert av NMDA reseptoren er således fordelaktige ved behandling av ischemiske tilstander, slag, skader i hjernen eller ryggmargen, og generelt hos pasienter med økende nivå av eksitatoriske transmittere. Spesifikke anvendelser omfatter også terapi ved senil demens av Alzheimertypen, demenskompleks av Parkinsontypen, Huntingtons chorea, og andre dominante eller recessive spino-cerebellare degenerasjoner hvor NMDA antagonister forebygger eller
retarderer utviklingen av sykdommen.

Betegnelsen "lavere alkyl" og "lavere alkoksy" refererer til grupper som har 1 til 4 karbonatomer i karbonkjeden. Betegnelsen "halogen" refererer til fluor, klor, brom og iod.
Forbindelsene ifølge denne oppfinnelse er chirale, og følgelig omfatter forbindelsene ifølge denne oppfinnelse ikke bare de racemiske blandinger, men også de enkelte enantio-merer. Enantiomerene betegnes ifølge R/S-systemet ved anvendelse av sekvensregelen.
Forbindelsene ifølge denne oppfinnelse kan fremstilles ved flere synteseveier. Ifølge et foretrukket skjema, blir en beskyttet benzimidazolyl-D-alanin-forløper omsatt med en alkyl-fosfonatester etterfulgt av avbeskyttelse, for å gi de ønskede sluttprodukter:

Den utgående X-gruppe i alkylfosfonsyrereaktanten kan være halogen, metylsulfonyl, tolylsylfonyl eller trifluorsulfonyl, hvorav trifluorsulfonyl blir spesielt foretrukket. Den beskyttede benzimidazolyl-alanin-forløper kan oppnås i optisk ren form ved enantio-selektiv syntese ut ifra et R eller S, N-beskyttet asparaginsyre esterderivat ved fremgangsmåten til Nestor et al., J. Avbeskyttelsen av det alkylfos-fonylerte mellomprodukt kan utføres ved anvendelse av sur eller basisk hydrolyse, hydrogenolyse, og/eller trimetylsilyl-bromidbehandling, avhengig av den beskyttelsesgruppe som skal fjernes. Disse trinn kan permuteres eller kombineres som nødvendig og passende. Når R<3>er benzyl, R<4>er benzyloksy-karbonyl, ogR<5>er 4-nitrobenzyl, kan det utføres en samlet hydrogenolyttisk avbeskyttelse i e'tt trinn.

I de tilfeller hvor substitusjonsmønsteret for R<1>og R<2>på benzimidazolringen er asymetrisk, hvorved alkylering med alkylfosfonatester-reaktanten i reaksjonssekvensene ovenfor ville gi en diastereomer blanding av produkter, ville det være nødvendig med fraksjonert krystallisasjon eller kromotografi for å skille produktene fra hverandre. Dette kan unngås ved anvendelse av egnede utgangsmaterialer som gir de ønskede for-løpermaterialer. Et hensiktsmessig substituert o-nitroanilin eller o-halogen-nitrobenzen blir således omsatt med henholds-vis en alkylfosfonatester-reaktant eller en aminofosfonat-esterreaktant, for å gi det hensiktsmessige mellomprodukt som deretter, omsettes videre ved metoden til Nestor et al. etterfulgt av avbeskyttelse for å gi de ønskede sluttprodukter:
Utgangsmaterialene i sekvensene ovenfor er alle enten kommersielt tilgjengelige eller kan fremstilles ved konvensjonelle metoder som er kjent og rapportert i den kjemiske litteratur.
Forbindelsene ifølge denne oppfinnelse kan danne farmakologisk akseptable salter av farmakologisk akseptable organiske eller uorganiske syrer såsom saltsyre, hydrogen-bromid-syre, sulfonsyre, svovelsyre, fosforsyre, salpetersyre, maleinsyre, fumarsyre, benzosyre, askorbinsyre, 4,4 '-metylen-bis[3-hydroksy-2-naftalenkarboksylsyre], ravsyre, metansulfon-syre, eddiksyre, propionsyre, vinsyre, sitronsyre, melkesyre, eplesyre, mandelsyre, kanelsyre, palminitsyre, itakonsyre og benzensulfonsyre. Forbindelsene ifølge denne oppfinnelse som fosfonokarboksylsyrene, er istand til å danne alkalimetall- og jordalkalimetall-karboksylater og karboksylater av farmakologisk akseptable kationer avledet av ammoniakk eller et basisk amin. Forbindelsene ifølge denne oppfinnelse er konkurrerende NMDA antagonister som kan anvendes ved behandling av konvulsjoner, cerebral ischemia, slag, hjerne- eller ryggmargs-skade, CNS forstyrrelser såsom senil demens, Alzheimers sykdom, Huntingtons chorea, og andre dominante eller recessive spino-cerebellare degenerasjoner. De nevnte forbindelser kan være spesielt nyttige som pre-anesthetika og neurobeskyttel-sesmidler under høyrisikokirurgi, såsom hjernekirurgi og rygg-margskirurgi, eller som resultat av trauma, hvor risikoen for hjerte- eller lungestans kan forårsake delvis, midlertidig eller fullstendig tap av blodstrømmen til hjernen. Ytter-ligere fordeler ved anvendelse av forbindelsene ifølge denne oppfinnelse som pre-anesthetika beror på deres milde aksiolyttiske/sedative egenskaper, deres korttidshukommelses-svekkende egenskap (korttids amnesi) og på deres evne til å forsterke virkningen av anesthetika slik at disse kan anvendes i lavere dose..
Oppfinnelsen tilveiebringer også anvendelse av en NMDA antagonist med formelen
hvori R er hydrogen, lavere alkyl, benzyl eller pivaloyloksymetyl; R<1>og R2 er, uavhengig hydrogen, lavere alkyl, lavere alkoksy, trifluormetyl, trifluormetoksy, metansulfonylamino, acetylamino eller halogen, eller R<1>og R<2>tilsammen representerer en metylendioksygruppe; og de farmakologisk akseptable salter derav, til fremstilling av et farmasøytisk preparat for behandling og forebyggelse av nevropatologiskek forstyrrelser indusert ved overstimulering av eksitatoriske aminosyrereseptorer.
Som sådanne kan forbindelsene ifølge denne oppfinnelse administreres rene eller sammen med en farmasøytisk bærer, og således kan de formuleres til orale doseringsformer såsom tabletter, kapsler o.l. Forbindelsene kan administeres ved å kombinere dem med konvensjonelle bærere, såsom magnesium-karbonat, magnesiumstearat, talk, sukker, laktose, pektin, dekstrin, stivelse, chelatin, tragakant, metylcellulose, natriumkarboksybutylcellulose, lavtsmeltende voks, kakaosmør o.l. Fortynningsmidler, smaksmidler, solubilisatorer, smøre-midler, oppslemmingsmidler, bindemidler, tablettsprengnings-midler o.l. kan anvendes. Forbindelsene kan også injiseres parenteralt, i hvilket tilfelle de anvendes i form av en steril løsning inneholdende andre løste stoffer, f.eks. tilstrekkelig saltløsning eller glukose til å gjøre løsningen isotonisk.
Doseringskravene vil variere med de spesielle blandinger som anvendes, administrasjonsmåten, graden av de gitte symptomer og det spesielle individ som behandles. Generelt vil man begynne behandlingen med små doser, mindre enn den optimale dosen av forbindelsen. Deretter blir doseringen øket inntil den optimale effekt under de gitte omstendigheter blir oppnådd. Synthesis 1988 62] og pyridin (0,87 g, 11,0 mmol) i diklormetan (50 ml) blir behandlet ved -10°C til -20°C med trifluormetansulfonsyre-anhydrid (3,1 g, 11,0 mmol) og omrørt ved -10°C i 1 time. Benzyl N-(benzyloksykarbonyl)-3-(2-bezimidazoyl)-D-alaninat (30 g, 0,07 mol) (fremstilt ifølge fremgangsmåten til Nestor et al., J. Med. Chem., 1984, 27, 320), trifluormatan-sulfonsyre [dietoksyfosfinyl]metylester (23,04 g, 0,076 mol) og pulverisert vannfritt kaliumkarbonat (37 g, 0,268 mol) omrøres i acetonitril (5 00 ml) ved romtemperatur i 20 timer.Blandingen filtreres, filtratet inndampes, og resten løses i diklormetan. Acetonitrilen fordampes, og resten ristes med diklormetan (100 ml) og H20 (2 x 100 ml). Diklormetansjiktet vaskes med 5% NaHC03(2 x 100 ml), saltløsning (100 ml), og tørkes over MgS04. A) Under tørr nitrogen løses N-Boc-D-asparaginsyre o;-benzylester (13,4 mmol, 4,34 g) i tørr tetrahydrofuran (67 ml) og avkjøles til -10°C. I rekkefølge tilsettes trietylamin (13,4 mmol, 1,86 ml) og etylklorformiat (13,4 mmol, 1,28 ml)
og reaksjonsblandingen omrøres i 10 minutter ved -10°C, hvoretter det tilsettes langsomt en løsning av kommersiell 4-klor-1,2-fenylendiamin (14,7 mmol, 2,1 g) i tørr tetrahydrofuran (27 ml).Blandingen får varme seg opp langsomt til omgivende temperatur. Den hentes deretter over i iskald saltløsning (150 ml), ekstraheres med etylacetat (2 x 100 ml). Forbindelsene ifølge denne oppfinnelse blir testet for sin konkurrerende NMDA antagonistaktivitet ved sin evne til å erstatte tritsiert 3-(2-karboksypiperazin-4-yl)propyl-l-fosfonsyre (CPP), en kjent konkurrerende NMDA antagonist, i homogenater av frontal kortex hos rotte i in vitro [<3>H]CPP bindingsundersøkelser.
Denne undersøkelse utføres som følger:
Rottene halshugges og deres hjerner fjernes umiddelbart, veies og plasseres i ca 15 volumer av iskald 10% sukrose. Hver hjerne homogeniseres ved anvendelse av en Potter Elvehjem glass homogenisator (12 slag ved 84 0 opm) utstyrt med en Teflon pistill. Homogenatet sentrifugeres deretter 1,000 x g i 10 minutter. Den resulterende pellet blir kastet, og supernatanten sentrifugeres ved 20,000 x g i 20 minutter. Den rå mitokondrial pellet slemmes opp igjen i iskaldt destillert vann og dispergeres ved anvendelse av en Brinkmann Polytron (PT-10 ved en skalaverdi på 6 i 30 sekunder). Oppslemmingen sentrifugeres ved 8,000 ganger g i 20 minutter. Den resulterende supernatant og "buffy coat" sentrifugeres ved 48,000 x g i 20 minutter. Den endelige rå synapse membran pellet slemmes opp igjen i iskaldt destillert vann og sentrifugeres ved 48,000 g i 20 minutter.

Når testforbindelsene undersøkes for et doseresponsforhold, blir resultatene avsatt som logit til %-binding mot log til testmedikamentkonsentrasjonen. Denne undersøkelse utføres som følger:
Sveitsiske albinohannmus (CD-1 mus, Charles River) 18-22 g, etter en sulteperiode på 18 timer, blir vant til et observasjonskammer i 3 0 minutter forbehandles med den representative testforbindelse etterfulgt 30 minutter senere med en NMDA, 195 mg/kg i.p., som er en dose som vanligvis forårsaker 90% mortalitet som fraskriver seg motoranfall omfattende ukontrollerbar bakbenkloring eller rykking i lemmene og/eller i torsomuskelen med tap av opprettingsrefleks etterfulgt av død innenfor observasjonsperioden på 30 minutter etter en NMDA administrasjon. og de farmakologiske akseptable salter derav.
