Description
Foreliggende oppfinnelse vedrører . fremstilling av nye terapeutisk aktive (S)enantiomerer av nye substituerte-2-aminotetraliner og salter derav.
Et formål ved oppfinnelsen er å tilveiebringe forbindelser for terapeutisk bruk, spesielt forbindelser som har en terapeutisk aktivitet via sentralnervesystemet (CNS). Et ytterligere formål er å tilveiebringe forbindelser som har en selektiv effekt på 5-hydroksytryptamin-reseptorene i pattedyr inkludert mennesket.
Terapeutisk nyttige 3-aminodihydro-[1]-benzopyraner og-benzotiopyraner som har virkning på 5-hydroksytryptamin-nøyronene i pattedyr, er beskrevet i EP 0222 996. Disse forbindelsene er definert ved formelen:
hvor Z er 0 eller S;
R er hydrogen eller lavere alkyl;
R^ er hydrogen, lavere alkyl eller aryl lavere alkyl;
R2 er hydrogen, lavere alkyl eller aryl lavere alkyl;
eller R^°S ^2 sammen danner en ring med 4-6 karbonatomer;
R3 er hydrogen, hydroksy, lavere alkoksy, aryl lavere alkoksy, acyloksy eller aryloksy når Z er S og R3 er hydroksy, lavere alkoksy, aryl lavere alkoksy, acyloksy eller aryloksy når Z er 0 og R3 er i 5-eller 8-stilling når Z er 0;
R4 og R5 er uavhengig hydrogen, lavere alkyl eller halogen, og mono- eller di-S-oksydene derav når Z er S, og farmasøytisk akseptable salter derav. Formålet med foreliggende oppfinnelse er å oppnå nye forbindelser som har en høy affenitet til 5-hydroksytryptamin-reseptorene i sentralnervesystemet samtidig som de virker som agonister, partielle agonister eller antagonister på serotoninreseptorene.
De nye forbindelsene med formel A som angitt nedenfor og salter derav er således nyttige i terapeutisk behandling av 5-hydroksytryptamin-formidlede tilstander og forstyrrelser slik som depresjon, angst, anorexi, senil demente, Alzheimers sykdom, migrene, termoregulator- og seksuelle forstyrrelser, og kan anvendes for bekjempelse av smerte og ved modulasjon av hjerte-karsystemet.
De nye terapeutisk aktive substituerte 2-aminotetralinene som fremstilles ifølge oppfinnelsen har den generelle formelen:
eller et farmasøytisk akseptabelt salt derav,
hvor R-^ er H, eller C^-C^alkyl,
R2 er E, eller C1-C6alkyl,
R3 er H, eller C^^-C^alkyl,
og hvor Ci-C^alkyl representerer rette, forgrende og cykliske alkylgrupper med 1-6 karbonatomer.
De nevnte rette, forgrende og cykliske alkylgrupper med 1-6 karbonatomer, kan utgjøres av metyl, etyl, n-propyl, i-propyl, n-butyl, i-butyl, t-butyl, n-pentyl, i-pentyl, t-pentyl, n-heksyl, i-heksyl, cyklopropyl, cyklobutyl, cyklopentyl, cykloheksyl, metylcyklopropyl, etylcyklopropyl, metylcyklobutyl. Foretrukne alkylgrupper har 1-4 karbonatomer .
Spesielt foretrukne er forbindelser av formel A hvor R^ , R2 og R3 er valgt blant metyl, etyl og n-propyl.
Den aboslutte konfigurasjon for (+)-X-HBr har ved røntgen-krystallografi (Ingeborg Csoregh, upubliserte resultater) blitt fastslått å være R.
Det er S-enantiomerene av forbindelsene formel A som viser 5-HT1A reseptor-antagonisteffekt. Dette fremgår fra det faktum at (S)-X i rottemoddelen inhiberer (R)-XXIV ((R)-8-hydroksy-2-(dipropylamino)-tetralin, (R)-8-0H DPAT) induserte biokjemiske og adferdsendringer på en doseavhengig måte. Både (S)-XXIV og (R)-XXIV er kjent for å være sterktvirkende 5-FTia reseptoraggonister L.J. Cornfield et al, Mol. Pharmacol, 1991, 39, 780-787. I motsetning til dette er racemisk X inaktiv i funksjonelle analyser hvilket skulle skyldes at (R)-X viser farmakologiske egenskaper som er felles for andre tetralinbaserte reseptoragonister.
(S)-X (32 pmol/kg, s.c.) påvirker ikke i betydelig grad 5-HTP-nivåer i rotter som ikke er forbehandlet med reserpin eller adferden til reserpinforbehandlede rotter. Den fortrenger imidlertid (±)-XXIV fra 5-HT^^-reseptorer (tabell 1).
I tillegg blir de adferdsmessige effektene av (R)-XXIV (}jmol/kg, s.c.) i reserpinbehandlede rotter fullstendig blokkert av forbehandling med (S)-X (10 jjmol/kg, s.c, gitt 10 min før). Forbehandling med (S)-X 2 timer før demper den (R)-XXIV-induserte adferd, men ingen blokkering observeres når (S)-X gis 4 timer før (R)-XXIV. Denne antagonismen er like effektiv etter forbehandling med Dg-resptorantagonisten haloperindol (2 mg/kg, i.p.).

Farmasøytiske preparater
Forbindelsene med formel A vil normalt bli administrert oralt, rektalt eller ved injeksjon, i form av farmasøytiske preparater omfattende den aktive bestanddel enten som en fri base eller et farmasøytiks akseptabelt ikke-toksisk salt, for eksempel hydroklorid, hydrobromid, laktat, acetat, fosfat, sulfat, sulfamat, citrat, tartrat, oksalat og lignende i en farmasøytisk akseptabel doseringsform. Doseringsformen kan være et fast, halvfast eller flytende preparat. Vanligvis vil den aktive substans utgjøre mellom 0,1 og 99 vekt-# av preparatet, mere spesielt mellom 0,5 og 20 vekt-# for preparater beregnet for injeksjon og mellom 0,2 og 50 vekt-# for preparater egnet for oral administrasjon.
For å fremstille farmasøytiske preparater inneholdene en forbindelse med formel I i form av doseringsenheter for oral anvendelse, kan den valgte forbindelsen blandes med en fast eksipiens, for eksempel laktose, sakkarose, sorbitol, mannitol, stivelser slik som potetstivelse, maisstivelse eller amylopektin, cellulosederivater, et bindemiddel slik som gelatin eller polyvinylpyrrolidon, og et smøremiddel slik som magnesiumstearat, kalsiumstearat, polyetylenglykol, vokser, paraffin og lignende, og deretter presses til tabletter. Dersom belagte tabletter er nødvendig så kan kjernene fremstilt som beskrevet ovenfor belegges med en konsentrert sukkeroppløsning som kan inneholde for eksempel gummiarabikum, gelatin, talk, titandioksyd og lignende. Tabletten kan alternativt belegges med en polymer som er kjent for fagmannen på området, oppløses i et lett flyktig organisk oppløsningsmiddel eller blanding av organiske oppløsningsmidler. Fargestoffer kan tilsettes til disse beleggene for lett å kunne skjelne mellom tabletter som inneholder forskjellige aktive substanser eller forskjellige mengder av de aktive forbindelsene.
For fremstilling av myke gelatinkapsler så kan den aktive substans blandes med for eksempel en vegetabilsk olje eller polyetylenglykol. Harde gelatinkapsler kan inneholde granuler av den aktive substans ved anvendelse av enten de ovennevnte eksipienser for tabletter, for eksempel laktosek, sakkarose, sorbitol, mannitol, stivelser (for eksempel potetstivelse, maisstivelse eller amylopektin), cellulosederivater eller gelatin. Væsker eller halvfaste stoffer som inneholder legemidlet kan også fylles i harde gelatinkapsler.
Doseringsenheter for rektal administrasjon kan være opp-løsninger eller suspensjoner eller kan fremstilles i form av suppositorier omfattende den aktive substans i blanding med en nøytral fettbase, eller rektale gelatinkapsler omfattende den aktive substans i blanding med vegetabilsk olje eller praffinolje.
Flytende preparater for oral administrasjon kan være i form av siruper eller suspensjoner, for eksempel oppløsninger inneholdene fra 0,2 til 20 vekt-# av den aktive substans som heri beskrevet, hvor resten utgjøres av sukker og/eller blandinger av etanol, vann, glyserol og porpylenglykol. Slike flytende preparater kan eventuelt inneholde fargemidler, aromastoffer, sakkarin og karboksymetylcellulose som fortykningsmidler eller andre eksipienser som er kjent for fagmannen på området.
Oppløsninger for parenteral administrasjon ved injeksjon kan fremstilles i en vandig oppløsning av et vannoppløselig farmasøytisk akseptabelt salt av den aktive substans, fortrinnsvis i en konsentrasjon fra 0,5 til 10 vekt-$. Disse oppløsningene kan også inneholde stabiliseringsmidler og/eller buffermidler og kan hensiktsmessig tilveiebringes i forskjellige doseringsenhetampuller.
Egnede daglige doser av forbindelsene av formel A ved terapeutisk behandling av mennesker er 0,01-100 mg/kg legemsvekt ved peroroal administrasjon og 0,01-100 mg/kg legemsvekt ved parenteral administrasjon.
Forbindelsene med formel (A) ovenfor fremstilles ifølge foreliggende oppfinnelse ved
a) alkylering av en forbindelse med formelen:
hvor R3' er C^-C^alkyl og en av R4 og R5 er benzyl og den
andre er H eller C^-C^alkyl
61) i det tilfellet R4 eller R5 er H, reduktiv eliminering av benzylgruppen, eller 62) i det tilfellet R4 eller R5 er CC^alkyl, hydrogenerende eliminering av benzylgruppen,
og eventuelt alkylering av den i aj) eller ag) oppnådde forbindelse,
b) og i det tilfellet det er ønsket en forbindelse (I) hvor R3 er H, dealkylering ved koking med konsentrert
hydrobromsyre,
og/eller, om ønsket, omdannelse av en oppnådd forbindelse til et farmasøytisk akseptablet salt derav.
Forbindelsene av formel A kan således oppnås blant annet fra p-fluorfenol via de tilsvarende 3-klorpropionater eller akrylater (for eksempel forbindelsen I) som gjennomgår Fries-omleiring fulgt av en ringlukning av Friedel-Crafts-typen for oppnåelse av 7-hydroksy-l-indanoner (for eksempel forbindelsen II). Indanonene blir O-metylert for oppnåelse av metyletere (for eksempel forbindelsen III) og blir ved en Wittig-reaksjon omdannet til de tilsvarende 2-metylenindanene (for eksempel forbindelse XVII) som oksydativt omleires under katalyse med tallium(III)nitrat for oppnåelse av 5-fluor-8-metoksy-2-tetraloner (for eksempel forbindelse V). Tetralonene omdannes til 2-benzylaminotetraliner (for eksempel forbindelse VI) ved reduksjon med natriumcyanobor-hydrid av deres Schiff-baser oppnådd ved kondensasjon med benzylamin. Herfra kan flere veier følges til 5-fluor-8-metoksy-2-(dialkylamino)tetralinene (for eksempel forbindelse VIII).
En første vei leder via de tilsvarende 2-benzylalkylamino-tetralinene (for eksempel forbindelse XI) ved hydrogenerende fjerning av benzylgruppen til monoalkylaminotetraliner (for eksempel forbindelse IX) og derfra ved uttømmende alkylering av de respektive dialkylaminotetralinene.
En annen vei med reduktiv fjerning av benzylsubstituenten leder til 2-aminotetralinene (for eksempel forbindelse VII) som dialkyleres til oppnåelse av nevnte dialkylaminotetra-liner.
Det siste trinnet i syntesen medfører fjerning av O-metyl-gruppen ved koking av konsentrert hydrobromsyre hvilket gir de respektive 5-fluor-8-hydroksy-2-(dialkylamino)tetralinene (for eksempel forbindelse X).
Forbindelsene av formel A oppnås enten i fri form eller i form av deres salter. Hver base kan overføres til det tilsvarened syreaddisjonssalt, fortrinnsvis ved bruk av en terapeutisk akseptabel syre eller et ioneutvekslingsmiddel. Salter som oppnås kan overføres til de tilsvarende frie baser, for eksempel ved bruk av en sterkere base eller et ioneutvekslingsmiddel. Siden det er et så nært kjemisk slektskap mellom den frie basen og dens salter så er det naturlig å betrakte begge selv om bare en av dem er identifi-sert. Forbindelser av formel A oppnås enkelte ganger ved krystallisasjon i form av deres hydrater med varierende vanninnhold.
Siden mesteparten av amino-mellomproduktene samt slutt-produktene er følsomme overfor oksygen, så utføres reaksjoner vanligvis under nitrogen. Forbindelsene av formel A blir fortrinnsvis lagret i form av deres addisjonssalter, først og fremst deres hydroklorider eller hydrobromider.
Forbindelsene formel A som inneholder et asymmetrisk karbonatom tilstøtende til aminogruppen (som kan være ikke-alkylert, monoalkylert eller dialkylert), kan spaltes i deres optiske antipoder ved hjelp av kjente metoder. Dette har blitt eksemplifisert ved spalting av racemisk forbindelse VI med L-vinsyre.
EKSEMPEL 1
4'- fluorfenyl- 3- klorpropionat ( I)
3- klorpropionylklorid (391,5 g) og 3 dråper trietylamin tilsettes til 4-fluorfenol (308,3 g) under omrøring ved 60°C. Etter oppvarming til 100°C i 1,5 timer så gir destillasjon 544,7 g av (I) som koker ved 126-130°C, 10 mm Hg.
4- fluor- 7- hvdroksy- l- indanon ( II)
Forbindelse I (219 g) tilsettes langsomt til vannfritt AICI3 (720 g) under nitrogen og holdes ved romtemperatur under omrøring. Omrøring stoppes når blandingen blir for viskøs. Etter oppvarming til 120°C i 1 time gjennopptas omrøring og temperaturen økes ytterligere til 180°C (2 timer) etter avkjøling og tilsetning av overskudd vann blir produktet dampdestillert. Ekstraksjon med kloroform og inndampning gir 127 g II-råprodukt.
4- f luor- 7- metoksv- l- indanon ( III)
I en nitrogenatmosfære blir det til 300 g pulverf ormig kaliumkarbonat i 1,5 1 aceton tilsatt 121 g II under omrøring, fulgt av 83 ml dimetylsulfat. Etter oppvarming til tilbakeløp i 2 timer blir oppløsningsmidlet avdestillert, vann tilsettes og blandingen tilbakeløpskokes i ytterligere 1 time. Ekstraksjon med metylenklorid og inndampning gir 125 g III-råprodukt som renses ved krystallisasjon fra EtOAc, smp. 118-120°C.
5- fluor- 8- metoksv- 2- tetralon ( V)
NaH (6,8 g) suspendert i dimetylsulfoksyd (40 ml) under nitrogen oppvarmes til 80° C i 1 time. Ytterligere 40 ml dimetylsulfoksyd og deretter 80 g metyltrifenylfosfonium-bromid tilsettes i porsjoner. Etter omrøring i 15 min blir 20 g III som er delvis suspendert i 40 ml dimetylsulfoksyd, tilsatt og blandingen oppvarmes til 70"C natten over. Helling av blandingen på is + heksan fulgt av ekstraksjon med heksan gir 17,6 g uren 4-f luor-7-metoksy-l-metylenindan (IV) som oppløses i 70 ml MeOH. Denne oppløsningen tilsettes til en omrørt oppløsning av trihydratet av talliumtrinitrat (43,9 g) i 400 ml MeOH og HC(0CH3)3. Etter omrøring i 1 min tilsettes 200 ml kloroform på en gang. Vasking med vånding natriumbi-karbonat, tørking og konsentrasjon gir et råprodukt som renses ved kromatografi (silitiumdioksyd, dietyleter-heksan 1:1). Den resulterende blanding av V og dets dimetylketal hydrolyseres med 1 M HC1 - dietyleter til oppnåelse av 11,8 g V. 1-H NMR (kloroform-d-L) å 7,02 - 6,64 (m, 2H); 3,79 (s, 3H); 3,51 (s, 2H); 3,10 (t, 2H); 2,56 (t, 2H).
( ±)- 2- benzylamino- 5- fluor- 8- metoksytetralin l"( ±1- VIl
Benzylamin (13,4 ml) tilsettes til 500 ml benzen og 11,8 g V under Ng•
Etter holding ved tilbakeløp natten over blir vann fjernet ved aceotrop destillasjon. Den urene reaksjonsblandingen inndampes og oppløses deretter i 500 ml metanol. pH-verdien justeres til 3-4 ved tilsetning av HCl/MeOH.
NaCNBHs (2,36 g) tilsettes og blandingen omrøres under Ng i 2 timer idet pH-verdien holdes ved 3-4 ved tilsetning av ECl/MeOH eller trietylamin. Etter konsentrering blir 5 M vandig EC1 tilsatt og bunnfallet frafUtrert og vasket med dietyleter. Omdannelse til den frie basen (±)-VI bevirkes ved behandling med 1 M NaOH/dietyleter. Rensing ved kromatografi på aluminiumoksyd (dietyleter/lett petroleum 1:2) gir 12,4 g av den frie basen (±)-VI.
Spalting av ( ±)- VI
(+)-L-vinsyre (9,08 g) tilsettes til (±)-VI (17,25 g) i 1050 ml varm 95 1o etanol. Oppløsningen holdes ved romtemperatur natten over og de resulterende krystaller (27,25 g) om-krystalliseres fra EtOH. Behandling med 5 M NaOH gir det frie aminet som ekstraheres med dietyleter. Det omdannes til hydrokloridet som omkrysalliseres fra MeOH/dietyleter til oppnåelse av 3,88 g (+)-VI-HCl. <1>E NMR (metanol-d4); S 7,60-7,40 (m, 5H), 7,05-6,65 (m, 2H), 4,35 (s, 2H), 3,82 (s, 3E), 3,7-1,6 (m, 7H). Optisk rotasjon ([a]D 22, MeOE, som i alle følgende bestemmelser): +61,4° (c, 1,0). Enantiomert overskudd bestemt ved kapillaer-GC av (R )-2-metoksy-2-fenylacetamidene (ee) er 99,7 %.
Det frie aminet (11,77 g) isolert fra modervæskene fra fremstillingen av (+)-VI tilsettes (-)-D-vinsyre (6,19 g). Fremgangsmåten benyttet for oppnåelse av (-)-VI'ECl 4,93 g er vesentlig den samme som ovenfor.
(+)- 2- amino- 5- fluor- 8- metoksytetralinhydroklorid r(+)- VII- HCll
(+)-VI'HCl (1,63 g) oppløses i 100 ml MeOH. Katalytisk hydrogener ing over Pd/C gir 1,14 g ( +)-VII "EC1, smp. 261-263°C. Optisk rotasjon: +67,8° (c, 1,0).
(-)- 2- amino- 5- fluor- 8- metoksytetralinhydroklorid f ( - l- VII ' ECU
Fremstilt som beskrevet for (+)-VII'ECl men med utgangspunkt i (-)-VII-ECl. Utbytte 99 %, smp. 262-264°C; optisk rotasjon: -67,2°C (c. 1,0).
(+)- 5- fluor- 8- metoksy- 2( dipropylamino) tetralinhvdroklorid r(+)- VIII- HC11
1-jodpropan (0,89 ml) tilsettes til en blanding av (+)-VIII-CE1 (1,01 g) og pulverformig kaliumkarbonat i 30 ml MeCN under nitrogen. Blandingen omrøres ved romtemperatur i 10 dager og i løpet av denne tiden tilsettes 2 porsjoner 0,3 ml 1-jodpropan. Etter tilsetning av dietyleter, filtrering og inndampning renses råproduktet ved kromatografi på aluminiumoksyd (dietyleter/lett petroleum, 1:4) til oppnåelse av 0,90 g (+)-VIII*ECl og, ved ytterligere eluering, 0,23 g (+)-5-fluor-8-metoksy-2-propylaminotetralinhydroklorid, (+)-IX'HCl.
Utbytte 71 #. Gjentatt omkrystallisering gir rent ( + )-VIII, smp. 134-135<0>C. Optisk rotasjon: +78,9° (c 1,0).
( - ) - 5 - f luor- 8- me toks y- 2- ( dipropylamino He tr al inhydroklor id r(-)- VIII- HCl
Fremstilt som beskrevet for ( +)-VIII"HC1, men med utgangspunkt i (-)-VII-HCl. Utbytte 80 %, smp. 134-135°C. Optisk rotasjon: 78,4° (c 1,0).
(+ ) - 5 - f luor- 8- hydroksy- 2-( dipropylamino Hetralinhydrobromid r(+)- X' HBr1
Alt utstyr blir nøye vasket med konsentrert svovelsyre før reaksjonens start. (+)-VIII'HCl (98 mg) tilsettes til (47 %) vandig HBr og tilbakeløskokes under Ng i 2 timer. Syren avdrives i vakuum ved 100°C. Deretter tilsettes dietyleter to ganger og fjernes ved inndampning. Omkrystallisering fra MeOH/dietyleter gir 75 g (69 %) av (+)-X-HBr, smp. 186-187°C. Optisk rotasjon: +69,4° (c 0,9). (-)- 5- f luor- 8- hydroksy- 2-( dipropylamino) tetralinhydrobromid.
f(+)- XI- HBrl
Fremstilt som beskrevet for ( + )-X*HBr. Utbytte 85 , smp. 186-187°C. Optisk rotasjon: -69,3° (c 1,0).
( + )- 2-( N- benzyl- N- propylamino)- 5- fluor- 8- metoksytetral in-h<y>droklorid. (+)- XI' HCl
0,13 ml 1-jodpropan tilsettes til en blanding av (+)-VI (0,41
g) og pulverformig kaliumkarbonat (1,22 g) i 10 ml CH3CN. Blandingen omrøres ved romtemperatur under nitrogen i 31
dager. Ytterligere 1-jodpropan (1,6 ml) tilsettes i porsjoner i løpet av denne perioden. Opparbeidelse som beskrevet for (+)-VIII gir 211 mg (+)-XI'HCl, smp. 152-153°C ved omkrystallisering fra metanol/dietyleter. Optisk rotasjon: +78,7° (c 1,0). (-)- 2-( N- benzyI- N- propylamino)- 5- fluo r- 8- me tok sy tet ral in-hydroklorld. (- 1- XI' HCl syntetiseres på samme måte som ( + )-XI*HC1, men med (-)-VI som utgangsmateriale. (-)- IX- HCl
(-)-XI-HCl (1,93 g) ble oppløst i 100 ml MeOH og hydrogenert over Pd/C. Utbytte av (-)-IX-HCl 0,75 g, smp. 208-210°C (EtOH/dietyleter). Optisk rotasjon: -71,5° (c 1,0).
(+)-IX*HCl ble fremstilt på samme måte, men med utgangspunkt i (+J-XI-HC1, smp. 207-210°C (EtOH/dietyleter).
(-)- VIII- HCl fra (-)- IX' HCl
Reaksjonen ble utført vesentlig på samme måte som alky-leringen av (-)-VI med jodpropan som beskrevet ovenfor.
EKSEMPEL 2
( ± )- 5- f luor- 8- hydroksy- 2-( dipropylamino Hetralinhydrobromid.
(±) -X ble fremstilt analogt med hydrobromidene av (+)-X og (-)-X fra forbindelse (±)-VI. Smp. 201-203°C etter krystalli-sering fra EtOH/dietyleter. Mellomprodukt (+)-VII ble omdannet uten forutgående rensing til (±)-VIII som krystalli-serte med 1/4 molekyl vann fra EtOH/dietyleter, smp. 146-147°C.
Claims (2)
1.
Analogifremgangsmåte for fremstilling av en terapeutisk aktiv S-enantiomer av substituerte 2-aminotetraliner med formelen:
eller et farmasøytisk akseptabelt salt derav,
hvor R1 er H, eller C^-Cfcalkyl,
R2 er H, eller C1-CÉ)alkyl,
R3 er E, eller C^-Cfcalkyl,
og hvor C^-C^alkyl representerer rette, forgrende og cykliske alkylgrupper med 1-6 karbonatomer, karakterisert veda) alkylering av en forbindelse med formelen:
hvor R3' er C^-C^alkyl og en av R4 og R5 er benzyl og den andre er H eller C^-C^alkyl
a-jj i det tilfellet R4 eller R5 er H, reduktiv eliminering av benzylgruppen, eller
a2) i det tilfellet R4 eller R5 er C.C^alkyl, hydrogenerende eliminering av benzylgruppen,
og eventuelt alkylering av den i a^) eller a2 ) oppnådde forbindelse, b) og i det tilfellet det er ønsket en forbindelse (I) hvor R3 er E, dealkylering ved koking med konsentrert hydrobromsyre,
og/eller, om ønsket, omdannelse av en oppnådd forbindelse til et farmasøytisk akseptablet salt derav.

Analogifremgangsmåte ifølge krav 1 for fremstilling av (S)-5-fluor-8-hydroksy-2-(dipropylamino)tetralin eller et salt derav, karakterisert ved anvendelse av tilsvarende substituerte utgangsmaterialer.
