Description
Foreliggende oppfinnelse vedrører fremstilling av nye cystinderivater med immunomodulerende aktivitet og som vil være nyttige i behandlingen av forskjellige sykdommer.
N-acetyl-L-cystein er en forbindelse som er utbredt benyttet for behandling av kroniske obstruktive luftveissykdommer/ kronisk bronkitt (for ytterligere referanser se Multicentre Study Group. Longterm oral acetylcysteine in chronic bronchitis. A double-blind controlled study. Eur. J. Respir. Dis. 1980, 61 (suppl. 111 ), 93-108; Boman, G. , Båcker, TJ., Larsson, S., Melander, B., og wahlander, L. Oral acetylcysteine reduces exacerbation rate in chronic bronchitis. N,N'-diacetyl-L-cystin er også kjent. Denne forbindelsen har tidligere kort blitt beskrevet i patentlitteraturen og også i den vitenskapelige litteratur (US 4.708.965; EP 300.100; US 4.724.239; US 4.708.965; DE 2.326.444; Wilson, I.D., and Nicholson, J.K. I US 4.827.016 angis forbindelsen å være effektiv for topisk behandling av dermale inflammasjoner som induseres og utvikles av leukotriener. Intet har imidlertid blitt rapportert eller er generelt kjent hva angår dens farmakologiske og/eller terapeutiske egenskaper med hensyn til immunologiske systemer og inflammatoriske sykdommer i lungen slik som kronisk bronkitt.
Tidligere har immunostimulerende egenskaper blitt rapportert for enkle disulfider slik som hydroksyetyldisulfid (HEDS, se: St. Georgiev, V. New synthetic immunomodulating agents.

Ifølge foreliggende oppfinnelse er det funnet at de nye 3,3'-ditiobis(propionsyre)-forbindelsene med den generelle formel:
hvor R er hydrogen eller en gruppe del -CO-R<1> hvor R<1> er metyl, etyl, n-propyl, n-butyl, n-pentyl, n-heksyl, n-heptyl, n-oktyl, n-nonyl, n-decyl, n-undecyl, isopropyl, 1-metylpropyl, tert.butyl, 3-metylbutyl eller 2-metylbutyl, R<2> er metyl, etyl, n-propyl, n-butyl, n-pentyl, n-heksyl, n-heptyl, n-octyl, n-nonyl, n-decyl, n-undecyl, isopropyl, 1-metylpropyl, tert.butyl, 3-metylbutyl eller 2-metylbutyl, og R<3> er hydrogen, metyl, etyl, propyl, isopropyl, butyl eller isobutyl, forutsatt at R<1> og R<2> ikke samtidig er metyl og videre forutsatt at når R<3> er hydrogen så er R<1> og R<2> ikke samtidig n-propyl eller n-heptyl, eller et fysiologisk akseptabelt salt og/eller stereokjemisk isomer derav, er et immunomodulerende, spesielt immunostimulerende middel.
Forbindelsene med formel I kan derfor benyttes for behandling av sykdommer hvor en defekt i immunsystemet og/eller et innefektivt vertforsvar forekommer eller kan mistenkes.
Eksempler på slike sykdommer er kronisk bronkitt og andre inflammatoriske sykdommer i luftveiene slik som astma og rhinitt, men også visse former for autoimmune sykdommer slik som diabetes og raumatoid artritt og/eller forskjellige ondartede sykdommer. HIV-infeksjon eller fullt utviklet AIDS kan behandles med forbindelsene. Også artereosklerotisk sykdom kan behandles med forbindelsene.
Effektive mengder av forbindelsene med formel I for bruk i behandlingen av de ovennevnte sykdommer er i området 0,5-500 mg, fortrinnsvis 5-50 mg, daglig dose.
Foretrukne forbindelser med formel I ovenfor er de hvor R er hydrogen eller en gruppedel -CO-R<1> hvor R<1> er metyl, etyl n-propyl, n-butyl, n-pentyl, n-heksyl, n-heptyl, n-oktyl, n-nonyl, n-decyl, n-undecyl, isopropyl, 1-metylpropyl, tert.butyl, 3-metyolbutyl eller 2-metylbutyl, R<2> er metyl, etyl, n-propyl, n-butyl, n-pentyl, n-heksyl, n-heptyl, n-oktyl, n-nonyl, n-decyl, n-undecyl, isopropyl, 1-metylpropyl, tert.butyl, 3-metylbutyl eller 2-metylbutyl, og R<3> er hydrogen, metyl eller etyl, forutsatt at R<1> og R<2> ikke samtidig er metyl og videre forutsatt at når R<3> er hydrogen så er R<1> og R<2> ikke samtidig n-propyl eller n-heptyl.
Foretrukne forbindelser med formel I når R<3> er hydrogen er de hvor R<1> og R<2> samtidig er isopropyl og tert.butyl, respektivt .
Foretrukne forbindelser med formel I ovenfor hvor R<3 >hydrogen er de hvor R<1> og R<2> inneholder 3-7 karbonatomer.
Særlig foretrukne forbindelser med formel I ovenfor når R<3> f hydrogen er de hvor R<1> og R<2> er isopropyl og R<3> er metyl; R<1 >og R<2> er n-pentyl og R<3> er metyl; R1 og R2 er n-heptyl og R<3 >er metyl eller etyl.
Den stereoisomere formen av forbindelsene med formel I som er særlig foretrukket er L-isomeren, det vil si forbindelser avledet fra L-cystin.
Et fysiologisk akseptabelt salt av forbindelsene med formel I når R<3> er hydrogen er et salt av natrium, ammonium, kalsium eller magnesium, sammen med de ikke-toksiske syreaddisjons-saltene derav. Også innbefattet er salter avledet fra arginin, lysin, histidin, etanolamin, dietanolamin, etylen-diamin og kolin, eller andre egnede organiske aminer.
Et fysiologisk akseptabelt salt av forbindelsene med formel I når R<3> ^ og R = hydrogen er hydrokloride, hydrobromide, hydrosulfate, oksalate, tartrate og så videre. Saltene kan også være i form av solvater, for eksempel hydrater.
Farmasøytiske preparater
De beskrevne aktive stoffene kan innbefattes i forskjellige doseringsformer, for eksempel tabletter, belagte tabletter, gelatinkapsler, oppløsninger og aerosoler.
For fremstilling av tabletter, belagte tabletter og gelatinkapsler kan den aktive substans kombineres med farmasøytisk akseptable materialer, for eksempel laktose, stivelse, dikalsiumfosfat, mikrokrystallinsk cellulose, polyvinyl-pyrrolidon, gelatin, cellulosederivater, kolloidalt silisium-dioksyd, talk og stearinsyre eller dens salter.
For fremstilling av orale oppløsninger er egnede eksipienser vann, sakkarose, glukose, sorbitol, fruktose og xylitol.
Doseringsformene kan foruten de nevnte eksipienser inneholde preservativer, stabilisatorer, viskositetsregulerende midler, emulgeringsmidler, søtningsmidler, fargemidler, aromamidler, tonisitetsregulerende midler, buffere eller antioksydasjons-midler. De kan også inneholde andre terapeutisk verdifulle stoffer.
De nye forbindelsene med formel I fremstilles ifølge oppfinnelsen ved de nedenfor angitte fremgangsmåte-alternativer A-H: A. Oksydasjon av et N-acylcysteinderivat med formelen:
hvor R<1> og R<3> har de ovenfor angitte betydninger eller når R<3 >er hydrogen eventuelt salt derav, til dannelse av en forbindelse med formelen:
Som oksydasjonsmiddel kan benyttes:
Hydrogenperoksyd, luft under alkaliske betingelser, alkyl-hydroperoksyder, persyrer, halogen, dinitrogenoksyd eller nitrogenoksyd, oksyderende metaller thallium (II), trialkyl-sulfoniumsalter, eller selen- eller telluriumoksyder. Oksydasjonen kan også utføres elektrokjemisk.
Et halogen er for eksempel klor, brom eller jod. Som salter i metode A kan følgende benyttes: natrium, kalium, kalsium, ammonium og så videre.
B. Omsetning, i nærvær av en egnet base, av en forbindelse med formelen:
eller et salt derav, hvor R<3> har den ovenfor angitte betydning, med en forbindelse med formelen:
R<1->COX
hvor R<1> har den ovenfor angitte betydning og COX er en reaktiv gruppe som kan reagere med en aminogruppe under dannelse av en amidgruppedel, til dannelse av en forbindelse med formelen:
Acyleringsmidlet R<1>COX kan for eksempel være et syre-halogenid, anhydrid, et amid, eller en aktivert syre eller ester.
Saltet i metode B og i hvilken som helst av de følgende metoder hvor et salt kan benyttes, kan være et hydroklorid, hydrobromid, hydrosulfat, oksalat, tartrat og så videre. Et salt i metode B kan også være hvilket som helst av de alkalisalter som er nevnt i metode A. c. Oksydasjon av en blanding av N-acylcysteinderivater med formlene:

ARBEIDSEKSEMP LER
Eksempel 1 (R ,R)-N,N-dipentanoyl-3,3'-ditiobis(2-amino-propionsyre)dimetylester: En suspensjon av L-cystindimetyl-esterdihydroklorid (1,0 g, 3 mmol) i THF (20 ml) (hvit oppslemming) ble omrørt og avkjølt til 0°C. Til reaksjonsblandingen ble det tilsatt 4 ekvivalenter (2,0 ml) av N-etyldiisopropylamin og 2,2 ekvivalenter (0,8 ml, 6,6 mmol) av pentanoylklorid. Blandingen ble omrørt i 4 timer på et isbad og det hvite bunnfallet av N-etyldiisopropylammoniumklorid ble fjernet ved filtrering. Oppløsningsmidlet ble fjernet ved inndampning og råproduktet ble gjenoppløst 1 diklormetan. Etter vasking med vann ble den organiske fasen tørket over natriumsulfat. Filtrering og inndampning av oppløsningsmidlet ga det urene tittelproduktet som ble omkrystallisert fra etylacetat.

Eksempel 2 (R ,R )-N ,N * -dipropionyl-3,3 ' -ditiobis( 2-amino-propionsyre)dimetylester: Forbindelsen ble fremstilt i overensstemmelse med fremgangsmåten beskrevet i eksempel 1 ved anvendelse av propionylklorid som acyleringsmiddel.

Eksempel 3 (R,R )-N,N'-dipentaoyl-3,3 *-ditiobis(2-amino-propionsyre)dietylester: Forbindelsen ble fremstilt i overensstemmelse med fremgangsmåten beskrevet i eksempel 1 ved å starte fra L-cystindietylesterdihydroklorid og ved anvendelse av pentanoylklorid som acyleringsreagens.

Eksempel 8 (S,S)-N,N'-di(2-metylpropionyl)-3,3'-ditiobis(2-(2-aminopropionsyre)dimetylester: N,N'-di i sobutyry1-D-cystin (1,86 g, 4,9 mmol) ble oppløst i 10 ml metanol inneholdene en dråpe saltsyre. Eksempel 15 ( R,R)-N-acetyl-N*-heksanoyl-3,3'-ditiobis( 2-aminopropionsyre)dimetylester: (R,R)-3,3'-ditiobis(2-aminopropionsyre)dimetylesterdihydroklorid (690 mg, 2 mmol) ble omrørt sammen med 1,39 ml (10 mmol) trietylamin i 20 ml THF i en 100 ml rundbunnet kolbe. Den uklare oppløsningen ble avkjølt til 0°C på et isbad, før en oppløsning av acetylklorid (142 jjL, 2 mmol) og heksanoylklorid (276 jjL 2, mmol) i 3 ml THF ble tilsatt dråpevis. Den uklare oppløsningen ble omrørt i 1 time ved 0°C. Et hvit bunnfall av 1 g (100 %) Et3NHCl ble frafiltrert og filtratet ble inndampet til oppnåelse av en oljeaktig rest som ble skilt mellom 10 ml H2O og 10 ml CHCI3. Etter separering av fasene så ble den vandige fasen ekstra-hert med 3 x 10 ml CHCI3. Eksempel 16 (R,R)-N,N'- di(2-metylpropionyl)-3,3'-ditiobis ( 2-aminopropionsyre ) : N-isobutyryl-L-cystein(9,5 g, 50 mmol) ble oppløst i 50 ml vann. Hydrogenperoksyd (30 %, 3,1 ml, 30 mmol) ble tilsatt og reaksjonsblandingen ble omrørt ved romtemperatur i 6 timer. Etter inndamping av oppløsnings-midlet under redusert trykk ble det oppnådd en hvit krystallisert olje (9,8 g). Omkrystallisering fra etylacetat ga tittelforbindelsen som et hvitt faststoff som ble tørket i vakuum.
Utbytte: 4,8 g (50 %).

Eksempel 17 (R,R)N-acetyl-3,3'-ditiobis(2-aminopropionsyre): L-cystien (2,42 g, 20 mmol) og N-acetyl-L-cystein (3,26 g, 20 mmol) ble oppløst i 25 ml vann. pH-verdien til denne oppløsningen var 2,6 ifølge lakmuspapir. Vandig hydrogenperoksyd (30 %, 2,3 ml, 21 mmol) ble tilsatt og reaksjonsblandingen ble hensatt ved romtemperatur natten over. Et hvitt bunnfall ble frafiltrert (1,26 g). Det gulaktige filtratet som inneholdt den ønskede forbindelsen og de to symmetriske forbindelsene (R,R)-N,N'-diacetylcystin og (mindre mengder) cystin ble separert på en preparativ HPLC ved bruk av en Dynamax C^8-kolonne (8 pm, 60 Å, 21,4 x 250 mm) med en Dynamax C^8-"guard"-kolonne (8 um, 21,4 x 350 mm) og med en dobbelt Gilson-oppløsningsmiddelavleveringssystem (305 pumpe, pumpehode 100 SC som virket som en avlever ingspumpe for oppløsningsmiddel, pumpehode 5 SC som virket som en prøve-injektor, 806 manometrisk modul, 811B dynamisk blander, 115 UV-detektor, 201 fraksjonsoppsamler, 201-202 fraksjons-regulator). De benyttede oppløsningsmidler var A=10 mM H0Ac/H20 og B=MeOH, med et 95:5 forhold for A:B. Oppløsnings-middelstrømmen var 10 ml/min og separeringen ble registrert ved 230 nm. Etter deteksjon av hver fraksjon på TLC (Merck 16483, plastark-silisiumdioksydgel 60 WF 254s, elueringsmiddel <n>BuOH/HOAc/H20 1:1:1, I2 som visualiseringsmiddel), ble fraksjonene med akseptabel renhet kombinert og inndampet til oppnåelse av en oljeaktig rest. Resten ble oppløst i aceton (pa) og oppløsningen ble inndampet til oppnåelse av tittelprodktet som hvite krystaller.

Eksempel 18 (R , R )-N-acetyl-N ' - ( 2-me ty lpropionyl )-3,3'-ditiobis(2-aminopropionsyre): En blanding av N-acetyl-L-cystein (0,652 g, 4 mmol) og N-isobutyryl-L-cystein (0,764 g, 4 mmol) i 10 ml MeOH ble omrørt mens hydrogenperoksyd (30 %, 0,60 ml, 5 mmol) ble tilsatt dråpevis. Omrøringen ble fortsatt i 3 timer ved romtemperatur hvoretter oppløsnings-midlet ble fjernet på en rotasjonsfordamper. Tilsetning av 25 ml aceton og gjentatt inndampning ga 1,5 g råmateriale som en olje som størknet ved henstand. Den ønskede tittelforbindelsen ble isolert fra blandingen ved preparativ HPLC som beskrevet i eksempel 17. Utbytte: 31 H>.

Eksempel 23 (R, R )-N ,N' -diheksanoyl-3,3 * -di tiobis( 2-amino-propionsyre ): En suspensjon av en ekvivalent 3,3'-ditiobis(2-aminopropion-syre )dimetylesterdihydroklorid i tetrahydrofuran (hvit oppslemming) ble omrørt og avkjølt til 0°C. Til reaksjonsblandingen ble det tilsatt 4 ekvivalenter N-etyldiisopropyl-amin og 2,2 ekvivalenter heksanoylklorid. Blandingen ble omrært i 4 timer på et isbad og det hvite bunnfallet av N-etyldiisopropylamminoiumklorid ble frafiltrert. Oppløsnings-midlet ble fjernet ved inndampning under redusert trykk og råproduktet ble gjenoppløst i diklormetan. Etter vasking med vann ble den organiske fasen tørket over natriumsulfat. Filtrering og inndamping av oppløsningsmidlet ga uren (R,R)-N ,N ' - diheksanoyl-3 ,3'-ditiobis( 2-aminopropionsyre )dimetyl-ester, som ble omkrystallisert fra metanol/vann og triturert med dietyleter.
En hvit oppslemming (0,1 M) av den ovenfor dannede dimetyl-ester og 0,5 M natriumhydroksyd i 10 % metanol ble omrørt kraftig ved romtemperatur. Etter ca 48 timer ble pH-verdien til den klare oppløsningen justert til 2 og det dannede hvite bunnfall av råprodukt ble frafiltrert og omkrystallisert fra aceton/heksan til oppnåelse av tittelforbindelsen som hvite krystaller. Totalt utbytte: 28 %.

Den innledningsvis dannede (R,R)-N,N'-di(1-okso-dodecanyl)-3,3'-ditiobis(2-aminopropionsyre)dimetylester ble omkrystallisert fra etylacetat. Tittelforbindelsen ble omkrystallisert fra touluen/diklormetan til oppnåelse av hvite krystaller. Totalt utbytte 23 %.

[oGd<25>: -113° (c=0,507, MeOH). ^H-NMR (300 MHz, DMS0-d26), S 8,19 (2H, d, NH), 4,48 (2H, m, CHN), 3,15 (2H, dd, CH2S), 2,89 (2H, dd, CH2S), 2,11 (4H, t, CH2C0), 1,49 (4H, m, CH2CH2C0), 1,25 (32H, m, (CH2)8), 0,87 (6H, t, CH3). Analyse beregnet for C30<H>56<N>206S2: C, 59,6; H, 9,3; N, 4,6; S, 10,6. Funnet: C, 59,2; H, 9,4; N, 4,6; S, 10,3.
Eksempel 26 (S,S)-N,N'-di(2-metylpropionyl)-3,3'-ditiobis(2-aminopropionsyre): Kaliumkarbonat (17,6 g, 127 mmol) ble oppløst i 40 ml vann og 40 ml metylenklorid under nitrogen. Oppløsningen ble avkjølt (-10°C) og 2-metylpropionylklorid (5,6 ml, 40 mmol) og D-cysteinhydrokloridmonohydrat (8,7 g, 49,5 mol) ble hurtig tilsatt. Reaksjonsblandingen ble omrørt ved romtemperatur i 4 timer hvoretter saltsyre ble tilsatt inntil blandingens pH-verdi ble mindre enn 1 ved bruk av lakmuspapir. Det vandige laget ble kassert og lettpetroleum (40-60° ) ble tilsatt til det organiske laget hvorved N-isobutyryl-2-cystein ble utfelt som hvite krystaller.
N-isobytyrl-D-cystein (1 g, 5,2 mmol) ble oppløst i 10 ml metanol. Hydrogenperoksyd (30 %, 0,3 ml, 2,6 mmol) ble tilsatt og reaksjonsblandingen ble omrørt ved romtemperatur i 6 timer. Etter inndampning av oppløsningsmidlet under redusert trykk ble det oppnådd en hvit krystallisert olje. Omkrystallisering fra etylacetat ga tittelforbindelsen som et hvit fast stoff ble tørket i vakuum.
Utbytte: 0,55 g (55 %)

Effekter av forbindelsene med formel I i en
modell av typen med forsinket hypersensitivitet i mus.
Egenskapen til forbindelsene med formel I til å stimulere immunresponser er illustrert ved deres effektivitet i en modell i mus av typen med forsinket hypersensitivitets-reaksjon (DTH).
Balb/c-mus av både han- og hunnkjønn oppnådd fra Bomhotsgaard (Danmark) og Charlie Rivers (England), ble benyttet ved en vekt på 18-20 g. 4-etoksymetylen-2-fenyloksazolon (OXA) ble innkjøpt fra BDH (England) og tjente som et antigen i denne testen.
Musene ble sesitivert, dag 0, ved epicutan anvendelse av 150 ul absolutt etanol/aceton (3:1) -oppløsning inneholdene 3 % oksa på det barberte brystet og buken. Behandling med forbindelsen som skulle undersøkes, DiNAC (som en positiv kontroll) eller bærer (fosfatbuffer, pH 7,0) ble initsiert ved oral tilførsel umiddelbart etter sensitivering og ble fortsatt en gang om dagen til dag 6. Syv dager (dag 6) etter sensitiveringen ble begge ørene til alle musene påvirket på begge sider ved topisk påføring av 20 ul 1 % oksa oppløst i olivenolje. Øretykkelse ble målt før og 24 eller 48 timer etter påvirkning ved bruk av en Oditest fjerbelastet passer. Påvirkninger og målinger ble utført ved lett pentobarbital-bedøvelse. Intensiteten til DTH-reaksjonene ble uttrykt ifølge formelen: T-t24/48_^t0 >jm enheter, hvor t0 og t24/48 representerer øretykkelsen før og 24 eller 48 timer etter påvirkning, respektivt, i individuell test (T). Resultatene ble uttrykt som middel+/-S.E.M. Nivået av signifikans mellom middel for gruppene ble oppnådd ved Students tohale-t-test. Tabell 1 viser representative resultater fra 24 og 48 timers målinger uttrykt som prosent økning i øretykkelse i forhold til den hos ikke-påvirket referanseøre. Et tall på 100 viser således en fordoblet øretykkelse.

hvor R er hydrogen eller en gruppedel -CO-R<1> hvor R<*> er metyl, etyl, n-propyl, n-butyl, n-pentyl, n-heksyl, n-heptyl, n-oktyl, n-nonyl, n-decyl, n-undecyl, isopropyl, 1-metylpropyl, tert.butyl, 3-metylbutyl eller 2-metylbutyl, R<2> er metyl, etyl, n-propyl, n-butyl, n-pentyl, n-heksyl, n-heptyl, n-octyl, n-nonyl, n-decyl, n-undecyl, isopropyl, 1-metylpropyl, tert.butyl, 3-metylbutyl eller 2-metylbutyl, og R<3> er hydrogen, metyl, etyl, propyl, isopropyl, butyl eller isobutyl, forutsatt at R<1> og R<2> ikke samtidig er metyl og videre forutsatt at når R<3> er hydrogen så er R<1> og R<2> ikke samtidig n-propyl eller n-heptyl, eller et fysiologisk akseptabelt salt og/eller stereokjemisk isomer derav, karakterisert vedA.
