Description
Fremgangsmåte til fremstilling av rene preparater av kallikrein-inaktivatoren.
Fra tysk utlegningsskrift nr. 146 146
er det kj ent at man kan fremstille et tungt-oppløselig derivat av kallikrein-inaktivatoren ved at man omsetter kallikrein-inaktivatoren med metafosforsyre.
Det er nu blitt funnet at man kan
også få rene preparater av kallikrein-inaktivatoren ved, at man utfeller kallikrein-inaktivatoren fra dets oppløsninger med
polyfosforsyrer eller deres vannoppløselige
salter under unngåelse av et overskudd av
utfellingsmiddel, fraskiller bunnfallet,
spalter ved oppløsning i salter eller salt-oppøsninger og befrir oppløsningen for
saltene etter i og for seg kjente metoder.
Egnede salter av polyfosforsyrene for gjen-nomføring av fremgangsmåten ifølge oppfinnelsen er fremfor alt alkali- og ammo-niumsalter.
Spaltningene av den tungtoppløselige
forbindelse kan foregå ved oppløsning i
natronlut eller i saltoppløsninger, eksem-pelvis vandige oppløsninger av NaCl eller
NH,C1 i konsentrasjoner på mere enn 1 pst.
Fjerningen av polyfosforsyrene resp.
deres salter foregår etter i og for seg kjente
metoder, f. eks. ved behandling av oppløs-ningene med utvekslere, dialyse osv.
Fremgangsmåten ifølge oppfinnelsen
kan anvendes på oppløsninger som inneholder kallikrein-inaktivatoren og som ble
fremstillet fra inaktivatorholdige dyriske
organer, som parotis, lunge, lever, pankre-as, milt, lymfekjertler osv.
Ved fremgangsmåten ifølge oppfinnelsen fås kallikrein-inaktivatoren i utbytte
på ca. 80 pst. i form av omtrent rene opp-løsninger. Lyofiliserer man oppløsningene så får man et lett vannoppløselig stoff, hvorav 0,16 til 0,19 y har den biologiske aktivitet av en kallikrein-inaktivator-en-het.
Eksempel 1.
1,080 g av en kallikrein-inaktivator med renhetsgrad 0,54 y/KIE oppløses i 40 ems aqua redestillata. og utfelles med
5 cm<:>>> av en nyfremstillet 10 pst.-ig poly-fosforsyre. Etter en halvtime sentrifugeres utfellingen, vaskes med 10 ems destillert vann og sentrifugeres igjen. Residuet sus-penderer man i 100 ems aqua redestillata. Deretter oppløøses det under omrøring med
n
5 cm/' — natronlut. Denne oppløsning lar 10
man strømme gjennom en utvekslersøyle som var fylt med 15 cm^ kationutveksler.
(«Levatit» S 100 eller «Amberlit» IR 120)
og 45 cm-"' anaonutveksler «Amberlite» IRA 410.
Den oppløsning som strømmer ut fra søylen inneholder 1,6 millioner KIE med en renhetsgrad 0,2 y/KIE.
Eksempel 2.
2 millioner KIE med renhetsgrad 0,39 y/KIE oppløses i 49 ems aqua redestillata og utfelles med 45 ems av en 1 pst.-ig nyfremstillet oppløsning av natriumpolyfos-fat som er innstilt på en pH på 11,8. Deretter sentrifugeres det, bunnfallet vaskes med 10 cma aqua redestillata og suspen-deres etter gjentatt sentrifugering i 100 cm:i aqua redestillata. Deretter oppløses
n. det under omrøring med 5 ems — natron-10
Lut. Denne oppløsning lar man strømme gjennom en utvekslersøyle som var fylt med 15 cm3 kationutveksler («Levatit» S 100 eller «Amberlite» IR 120) og 45 ems anionutveksler «Amberlite» IRA 410.
Den oppløsning som strømmer ut av søylen inneholder 1,5 millioner KIE med en renhetsgrad på 0,17 y/KIE.

- Fremgangsmåte til fremstilling av rene preparater av kallikrein-inaktivatoren, karakterisert ved at man utfeller kallikrein-inaktivatoren fra dets oppløsning med polyfosforsyrer eller deres vannoppløselige salter under unngåelse av et overskudd av utfellingsmiddel, fraskiller utfellingen, spalter ved oppløsning i lut eller saltoppløsninger og befrir oppløsnin-gen for saltene etter i og for seg kjente metoder.
