Oppfinnelsen vedrorer en. fremgangsmåte for fremstilling av forbindelser med den generelle formel hvor B betyr en alkylengruppe med 2-4 karbonatomer,
R^ hydrogen eller halogen og
R 3 og R. hydrogen, halogen, lavere alkyl, lavere alkoksy eller trifluormetyl, ifolge norsk patent nr. 124.111
hvilken fremgangsmåte karakteriseres ved behandling av en forbindelse med den generelle formel
hvor , R^ og R^ har foran angitte betydning og
X er halogen, som klor, brom og jod, med et diamin med den generelle formel
hvor B har foran angitte betydning.
Kondensasjonen av forbindelser med formel II med diaminer med formel III kan lett utfores ved å blande komponentene eller ved å la dem reagere i nærvær av et inert organisk opplosningsmiddel, f.eks. etere, hydrokarboner, som benzen, toluen; alkoholer, som lavere .alkanolér og lignende. Kondensasjonen utfores hensikts-messig ved værelsetemperatur eller over, fortrinnsvis ved en temperatur mellom 20 og 15Q°C. Alternativt kan diaminreaksjons- f' komponentene med formel III anvendes som et salt derav, i hvilket tilfelle reaksjonen kan utfores ved oppvarmning av blandingen av reaksjonskomponentene til en smelte. Forbindel-
ser med formel II kan fremstilles ved behandling av en forbindelse med den generelle formel
med et halogeneringsmiddel, f.eks. halogen, som klor, brom eller jod, N-bromsucciriimid, N-klorsuccinimid og lignende.
Forbindelser med formel IV -er Xjente forbindelser og kan fremstilles analogt kjente forbindelser.
Forbindelser med formel I er verdifulle som psykostimulerende, antiinflammatoriske og antipyretiske midler.
Det redegjores nærmere for oppfinnelsen i folgende eksempel. Alle temperaturer er angitt i Celsius<g>rader. :Spaltningssmel.te-punktene tas i åpne kapillarer. De kan variere fra -10° avhengig av oppvarmningshastigheten.
EKSEMPEL
Til en opplosning av 4 g o-cyano-difenyl-metan i 50 ml karbon-
tetraklorid tilsettes 3,6 g N-brom-succinimid, og blandingen tilbakelopbehandles i 6 timer. Etter avkjoling, filtreres suspen-
sjonen, og filtratet konsentreres til en orange olje som opp-
loses i 40 ral benzen og 13,5 ml etylendiamin. Opplosningen tilbakelopbehandles i 2 timer, helles på isavkjolt vandig natriumhydroksyd og ekstraheres med benzen. Benzenopplosningen ekstraheres med 2-n saltsyre, og dette ekstrakt gjores basisk.
Ekstraksjon med metylenklorid gir en orange olje som opploses
i etanol og ansyres med svovelsyre. Fortynning med etylacetat gir et krystallinsk bunnfall som etter omkrystallisasjon fra en blanding av etanol i etylacetat gir hvite prismer av 2,3-dihydro-5-fenyl-5H-imidazo[2,l-a]isoindol-sulfat, som smelter ved 224-227° (spaltning).

Claims (2)

1. Analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser med den generelle formel
hvor B betyr en alkylengruppa med 2-4 karbonatomer, hydrogen eller halogen og R3 og R4 hydrogen, halogen, lavere alkyl, lavere alkoksy eller trifluormetyl, ifolge norsk patent nr. 124.111 karakterisert ved behandling av en forbindelse med den generelle formel hvor R^, R^ og R^ har foran angitte betydning og X er halogen, som klor, brom og jod, med et diamin med den generelle formel hvor B har foran angitte betydning.
2. Fremgangsmåte etter krav 1, karakterisert ved at kondensasjonen med et diamin utfores ved en temperatur i området fra 20 til 150°C.
