Description

Fremgangsmåte for fremstilling av et tropasyreamid.
Det er kjent at tropasyre-N-(|3-pikolyl) -amid virker kraftig spasmolytisk på over-levende kanintarm såvel som på kanintarm in situ.
Det er nu funnet at tropasyre-N-(y
-pikolyl)-amid på overraskende måte viser
en ennu kraftigere spasmolytisk virkning
enn tropasyre-N-((3-picolyl)-amid ved lik
toksisitet.
Gjenstand for foreliggende oppfinnelse
er en fremgangsmåte for fremstilling av
tropasyre-N-(y-pikolyl)- amid, som består
i at man omsetter et eventuelt 0-acylert
tropasyre-halogenid med 4-aminometyl-pyridin og eventuelt spalter av O-acylgruppen.
De forbindelser som fremstilles i hen-hold til oppfinnelsen kjennetegnes ved den
etterfølgende generelle formel:
i hvilken Z betyr enten vannstoff eller et
acylradikal.
Eksempel.
Til en blanding av 31 vektsdeler 4-amino-metyl-pyridin og 23 vektsdeler tørr
pyridin i 100 vektsdeler tørr kloroform tilsettes etterhvert rått, av 50 vektsdeler tropasyre på vanlig måte erholdt acetyltropa-
syreklorid under omrøring og isvanns-kjøling. Til slutt røres det ytterligere en time ved 20°.
Derpå fortynnes kloroformoppløsnin-gen med 150 vektsdeler eter og rystes med 3 n-saltsyre. Den kongosure oppløsning oppvarmes 1 time på dampbad, hvorved acetylgruppen av reaksjonsprodukter spal-tes av. Det filtreres over kull og tilsettes overskudd av konsentrert ammoniakk. Det utfelte kondensasjonsprodukt tas opp i kloroform, kloroformen tørkes og destille-res av.
Etter gjenoppløsning av destillasjons-resten i etanol-lavtkokende petroleter får man tropasyre-N-(y-pikolyl)-amid i form av farveløse krystaller med smeltepunkt 143—144°.

Claims (2)

1. Fremgangsmåte til fremstilling av et tropasyreamid, karakterisert ved at man omsetter eventuelt O-acylert tropasyreklo-rid med 4-amino-metyl-pyridin og eventuelt spalter av O-acylgruppen.
2. Fremgangsmåte etter påstand 1, karakterisert ved at man omsetter acetyl-tropasyreklorid med 4-amino-metyl-pyridin og avacetylerer kondensasjonsproduktet til tropasyre-N-(Y-picolyl)-amid.
