Description

Tripeptiden nuttig als immunostimulantia en voorts bij de voorkoming van metastasen.
Het is bekend dat de wisselwerking tussen tumorcellen en de extracellulaire matrix, veroorzaakt of begunstigd door fibronectine, wordt belemmerd door de intraveneuze toediening van het synthetisch pentapeptide 5 Gly-Arg-Gly-Asp-Ser, waarvan de volgorde dezelfde is als die van de celbindende plaats van fibronectine (Humphries M. J. e.a., Science 233:467, 1986).
Ruoslahti en Pierschbacher (cell 44:517,1986) hebben de hypothese voorgesteld, dat de biologische 10 activiteit van dit pentapeptide zou kunnen worden toegeschreven aan de tripeptidevolgorde Arg-Gly-Asp die daarin is opgenomen, zonder echter de activiteit van deze laatste te onderzoeken.
Lam e.a. (J. Biol. Chem. 262:947,1987) hebben 15 in plaats daarvan bewezen dat Arg-Gly-Asp in staat is om enige bloedplaatjes-oppervlak-glycoproteïnen te binden, zodat een eventuele biologische werking van dit tripeptide als een belemmeraar van bloedplaatjesaanhechtingsreacties wordt gesuggereerd.
20 Uitgaande van de wetenschap die voortvloeit uit de bovengenoemde stand van de techniek heeft aanvraagster enige tripeptiden gesynthetiseerd die behoren tot de algemene formule X-Gly-Y, waarin X L-Arg is of D-Arg is en Y L-Asp of D-Asp is, waarbij dan de doeltreffendheid · 25 daarvan wordt beoordeeld in experimentele onderzoeken van antimetastatische activiteit.
De resultaten hebben hun doeltreffendheid als antimetastatische agentia bevestigd, maar ook verrassenderwijs hun opmerkelijke immunostimulerende activiteit aan-30 getoond, die niet voorspelbaar is op basis van de wetenschap van het ogenblik.
Dit immunostimulerende effekt werd in vitro in experimenteel murine en voorts mensmodellen waargenomen, en bestaat zowel uit een rijping van onrijpe T-lymfocyten .8801932 tt - 2 - als uit een versterking van T-celfunktïe.
Een soortgelijk immunostimulerend gedrag is beschreven voor de tripeptiden Arg-Lys-Glu {Amerikaanse octrooiaanvrage Nr. 035.045 van 6 april 1987) , Arg-Ala-Arg 5 (Amerikaanse octrooiaanvrage Nr. 035.044W van 6 april 1987) en Arg-Lys-Asp (Europese octrooiaanvrage Nr. 67 425 van 22 december 1982).
Bovendien hebben de tripeptiden die het voorwerp van de onderhavige uitvinding zijn stabiliteit betoond 10 bij gesimuleerd maagsap: op basis van wat was waargenomen in het geval van de peptiden Arg-Lys-Glu en Arg-Ala-Arg, waarvan de stabiliteit in gesimuleerde maagomgeving was gekoppeld aan activiteit na orale toediening aan dieren, is het mogelijk aan de tripeptiden, die het voorwerp van 15 de onderhavige uitvinding zijn, een immunostimulerende activiteit na orale, naast parentale, toediening toe te schrijven.
Dit betekent een opmerkelijk voordeel in de therapeutische toepassing, waarbij in het bijzonder wordt 20 verwezen naar kinderen en andere patiënten, die de parenterale toediening niet verdragen, en ook met het oog op een beter voldoen voor de patiënt.
Op basis van wat hierboven is vermeld, blijken deze produkten zeer nuttig te zijn bij de voorkoming van 25 metastatisering bij neoplastische patiënten na chirurgische verwijdering van de tumor, en voorts bij de gelijktijdige stimulering van het immune systeem, onderdrukt door zowel de radiochemotherapeutische behandelingen als de operatie zelf: deze beide faktoren zijn in feite notoir-immuno-30 depressant.
Bovendien stelt de beschikbaarheid van geneesmiddelen, die kunnen worden toegediend langs orale weg, voor de therapie van deze patiënten een ander ontwijfelbaar voordeel voor.
35 De onderhavige uitvinding beschrijft de synthese en de chemische en voorts de biologische kenmerken van een nieuwe klasse van tripeptiden, gedefinieerd door de algemene formule X-Gly-Y, waarbij X L-Arg of D-Arg is en Y L-Asp of D-Asp is.
40 Deze produkten worden gekenmerkt doordat zij zijn 8801952 - 3 - begiftigd met antimetastatische eigenschappen, waargenomen bij het onderzoek van murinemelanoma experimentele metasta-sen, en voorts immunostimulerende activiteit, aangetoond in vitro in onderzoeken van murinemiltlymfocyt - rijping 5 (Thy. 1.2 membraan merkstofinductie) en in proeven die betrekking hebben op de activering van rijpe lymfocyt-funktie van de mens (groeifaktorproduktie, DNA en RNA synthese na mitogene stimulus, toeneming van mitosen).
Bovendien hebben in vitro onderzoeken, die zijn 10 uitgevoerd in gesimuleerde maagomgeving, de stabiliteit van deze peptiden aangetoond, zodat het mogelijk is hun activiteit ook na orale toediening te voorzien.
Zo kunnen de produkten die het voorwerp van de onderhavige uitvinding zijn, worden gebruikt dankzij hun 15 unieke combinatie van antimetastatische en immunostimulerende activiteiten als geneesmiddelen bij patiënten die de chirurgische tumorverwijdering ondergaan, teneinde de metastasevorming te voorkomen, terwijl tegelijkertijd wordt geholpen de immune status te verbeteren.
20 Fig. 1 en 2 stellen respectievelijk voor de HPLC profielen van de tripeptiden Arg-Gly-Asp en Arg-Gly-D-Asp, die verkregen zijn in overeenstemming met de werkwijzen beschreven in het voorbeeld II.
De uitvinding zal beter worden begrepen aan de 25 hand van de volgende voorbeelden waarin de volgende afkortingen worden gebruikt:

35 Synthese van L-arginyl-glycyl-L-aspartinezuur 1. t-Butyloxycarbonyl-glycyl-L-aspartinezire -dibenzylester.
t-Butyloxycarbonyl-glycine (7,1 g) werd opgelost in ethylacetaat (50 ml), gekoeld op -10 tot -15°C onder roeren en behandeld met N-methylmorfoline (5,6 ml), gevolgd .8801932 * - 4 - door isobutylchloorformiaat (5,18 ml). Het mengsel werd 10 min. lang bij -15°C geroerd. Ondertussen werd L-aspartinezuurdibenzylester p-tosylaatzout (2°,1 g) opgelost in dimethylformamide (80 ml), afgekoeld tot -10°C 5 en geneutraliseerd door toevoeging van N-methylmorfoline (5,6 ml). Deze oplossing werd toegevoegd aan de bovenstaande oplossing van het voorafgevormd gemengd anhydride en het reactiemengsel liet men over een periode van 3 uur onder roeren langzaam opwarmen tot kamertemperatuur.
10 Het reactiemengsel werd daarna verdund met ethylacetaat (300 ml) en de oplossing werd gewassen met pekel (tweemaal) gevolgd door 4 % waterig natriumbicarbonaat, water, 5 % waterig citroenzuur en ten slotte met water tot neutraal. De organische fase werd gedroogd 15 (magnesiumsulfaat) en daarna ingedampt tot een olie.
Opbrengst: 20 g. Het produkt was homogeen door TLC (systeem: chloroform:methanol:azijnzuur, 360:32:8; = 0,8).
2. Tribenzyloxycarbonyl-L-arginyl-glycyl-L-aspartinezure dibenzylester.
20 Het produkt uit de voorafgaande stap (20 g) werd behandeld met 50 % trifluorazijnzuur in methyleenchloride (125 ml) gedurende 25 min. bij kamertemperatuur en de oplosmiddelen werden onder verminderde druk afgedampt.
Het residu werd opnieuw uit tolueen ingedampt 25 en daarna in vacuo gedroogd. Het ontstane trifluoracetaat-zout van het dipeptide werd opgelost in dimethylformamide (75 ml), gekoeld tot -10°C en geneutraliseerd door behandeling met N-methylmorfoline (5,6 ml). Ondertussen werd tribenzyloxycarbonyl-L-arginine (23,1 g) omgezet in het 30 gemengd anhydride door oplossing in tetrahydrofuran (100 ml), afkoeling tot -10 tot -15°C en behandeling met N-methyl-morfoline (5,6 ml) gevolgd door isobutylchloorformiaat (5,18 ml) als boven beschreven. De vooraf gekoelde oplossing van het geneutraliseerd dipeptide werd toegevoegd 35 aan de oplossing van het gemengd anhydride en het mengsel werd 3 uur lang geroerd, waarbij langzaam tot kamertemperatuur werd opgewarmd. Tijdens de reactie stolde het mengsel en dimethylformamide (50 ml) werd toegevoegd teneinde roeren gemakkelijker te maken. Het tetrahydro-40 furan werd afgedampt onder verminderde druk en het residu .8801932 - 5 - verdund met ethylacetaat (500 ml).
Deze oplossing werd met verzadigd waterig natrium-chloride gewassen, gedurende welke bewerking een witte vaste stof neersloeg. Deze vaste stof werd afgefiltreerd, 5 gewassen met ethylacetaat en gedroogd ter verkrijging van het beschermd tripeptide. Opbrengst: 25 g. Dunne-laag chromatografie (systeem: chloroform:methanol:azijnzuur, 85:10:5) vertoonde een kleine verontreiniging door niet-gereageerd hebbend tribenzyloxycarbonylarginine. Het 10 produkt (14 g) werd daartoe gezuiverd door oplossing in chloroform:methanol (98:2) en aanbrenging op een kolom van silicagel (600 g). De kolom werd geëlueerd met geleidelijk toenemende hoeveelheden methanol in chloroform ter verkrijging van het zuiver beschermd tripeptide. OpbrengS:: 15 10 g.
3. L-arginyl-glycyl-L-aspartinezuur
Het beschermd tripeptide uit de voorafgaande stap (10 g) werd bij 50 psi (350 kPa) gehydrogeneerd in methanol-azijnzuur-water (60:20:20, 300 ml) gedurende 72 20 uur boven palladium op koolstof (5 %, 3 g), toen de reactie volledig was door TLC. De katalysator werd afge-filtreerd, de methanol afgedampt onder verminderde druk en het residu gelyofiliseerd ter verkrijging van het tripeptide als een wit poeder. Opbrengst: 3 g. Het produkt 25 was homogeen door dunne-laag chromatografie (systemen: A, isopropanol:ammoniak, 1:1; B, n-butanol-azijnzuur: water:pyridine, 60:12:40:48).
Synthese van L-arginyl-glycyl-D-aspartinezuur 1. Bereiding van Boc-g-benzyl-D-aspartinezuurharsen.
30 Chloor-gemethyleerd polystyreen (1 % verknoopt; 200-400 mesh (0,074-0,037 mm)) werd gebracht in een rond-bodemkolf en men deed het zwellen in dimethylformamide (bij benadering 8-10 ml per gram hars), het werd daarna behandeld met het Boc-6-benzyl-D-aspartinezuur (1 mmol 35 per gram hars), gevolgd door kaliumfluoride (2 mmol per gram hars). De kolf werd uitgerust met een mechanische roerder en condensor en onder vacuum verhit totdat een kleine hoeveelheid (5-10 ml) van het oplosmiddel was gedestilleerd. De vacuumbron werd verwijderd en het meng-40 sel 16-18 uur lang verhit tot 80-100°C. Bij afkoeling .8801932 % - 6 - werd de hars gefiltreerd, gewassen met dimethylformamide, dimethylformamide:water (1:1), water, ethanol, dichloor-methaan en methanol, en daarna onder vacuum gedroogd.
Substituëring (als berekend door gewichts-5 toeneming) = 0,6 mmol per gram.
2. Syntheseprotocol
Een hoeveelheid van 17,6 gram Βοσ-β-benzyl-D-aspartinezuurhars werd gebracht in een glazen reactievat uitgerust met een mechanische roerder, een gesinterd 10 glasvoetstuk en een vacuumbron voor filtratie. De hars werd achtereenvolgens bij omgevingstemperatuur (20-25 °C) met de volgende oplosmiddelen of reagentia behandeld: a. methyleenchloride b. 50 % trifluorazijnzuur:methyleenchloride (v/v) 15 c. 50 % trifluorazijnzuur:methyleenchloride gedurende 25 min.
d. methyleenchloride (driemaal) e. isopropanol f. 10 % triethylaminemethyleenchloride (v/v) (tweemaal) g. methyleenchloride 20 h. methanol (tweemaal) i. methyleenchloride (tweemaal).
Een contacttijd van 3-5 min. werd voor elke behandeling toegelaten. Bij benadering 10-15 ml oplosmiddel of reagens-oplosmiddelmengsel per gram hars werd in elke 25 stap gebruikt.
j. De hars werd geroerd met een oplossing van Boc-glycine (3 equivalenten) in methyleenchloride, en hieraan werd toegevoegd dicyclohexylcarbodixmide (3 equivalenten) in methyleenchloride. De reactietijd voor koppeling was 30 minimaal 2-4 uur maar kon een nacht lang duren (16-18 uur).
De peptide-hars werd gefiltreerd en gewassen met methyleenchloride, methanol en methyleenchloride en gecontroleerd op volledigheid van koppeling door de ninhydrinereactie.
Als de koppeling onvolledig was, werd hetzelfde aminezuur 35 opnieuw gekoppeld onder toepassing van de helft van de hoeveelheid reagentia.
De cyclus werd herhaald voor Boc-N^-tosyl-L-arginine (in dimethylformamide). Na verwijdering van de N-eind-Boc-groep werd het harspeptide grondig gewassen 40 en onder vacuum gedroogd.

Opbrengst van harspeptide: 21,0 g.
Het peptide werd afgesplitst van de hars en van zijn bescherming ontdaan gepaard gaande aan behandeling met watervrij vloeibaar waterstoffluoride (bij benadering 5 10 ml per gram harspeptide) bevattende anisool (10 % v/v) gedurende 1 uur per 0°C. Na afdamping van het waterstoffluoride onder verminderde druk, werd het ruwe peptide geëxtraheerd door de hars te wassen met verdund, waterig azijnzuur en het produkt geïsoleerd door lyofilisering.
10 Opbrengst van ruw peptide: 3,9 g.
3. Zuivering van het ruwe peptide
Het ruwe peptide zou kunnen worden gezuiverd door preparatieve, omgekeerde-fase HPLC onder toepassing van C18-afgeleide silica, onder toepassing van bijv. een 15 Waters Prep 500 instrument. Onder toepassing van een 5 X 30 cm kolom, in evenwicht gebracht met de geschikte waterige buffer, zoals 0,1 % waterig trifluorazijnzuur, werd het ruwe peptide (bij benadering 2 g) aangebracht op de kolom en geëlueerd met een gradiënt bevattende 20 toenemende hoeveelheden acetonitril. Frakties werden gevolgd door analytische HPLC en die welke het produkt bevatte op het gewenste niveau van zuiverheid ( > 97 %) werden gecombineerd en gelyofiliseerd. Ten slotte werd het gezuiverd produkt omgezet in het gewenste zout ervan 25 door behandeling met de gewenste zoutvorm van een ionen-wisselaarhars.
Opbrengst van gezuiverd peptide als acetaatzout: 2,5 g.

VOORBEELD III: BIOLOGISCHE KENMERKEN Stabiliteit in gesimuleerd maaqmilieu in vitro
Het tripeptide Arg-Gly-Asp bleek stabiel te 5 zijn bij 37°C 3 uur lang in gesimuleerd maagmilieu in vitro onder toepassing van de oplossing van gesimuleerd maagsap ÜSP XXI (HC1 + pepsine).
Antimetastatische activiteit '
De capaciteit van het tripeptide Arg-Gly-Asp 10 om te longkolonisering te belemmeren van B16-BL6 murine-melanomacellen gelnoculeerd in wijfjes C57BL/6 muizen met een gemiddeld lichaamsgewicht van 20 g (10 dieren per groep) werd beoordeeld.
Arg-Gly-Asp werd toegediend met een dosis van 5 15 3 mg/muis door intraveneuze inspuiting samen met 2x10 melanomacellen.
Veertien dagen later werden de dieren gedood, de longen werden er uit gehaald en na formalinefixatie onderzocht op de aanwezigheid van oppervlaktemelanoma-20 kolonies. Een groep van vergelijkingsdieren werd alleen behandeld met de tumorcelsuspensie.
Terwijl bij de vergelijkingsleren het gemiddeld aantal metastasen boven 500/muis was, was deze waarde bij behandelde dieren verminderd, waar zij 173/muis was.
4 25 Onder toediening van 7x10 tumorcellen was het gemiddeld aantal metastasen bij de controledieren 21,6, nam af tot 6,8 bij dieren die waren behandeld met 3 mg/muis intraveneus van Arg-Gly-Asp (-69 %).
Inductie van Thy 1,2 antigeen in miltcellen van normale 30 muizen.
Het vermogen van het tripeptide Arg-Gly-Asp tot het induceren in vitro van de differentiatie van T-celvoorlopers in lymfocyten die T-celmerkstoffen uitdrukken, werd getest door het aantonen van de inductie 35 van Thy 1,2 membraanantigeen.
CELBEREIDING:
Miltcellen van normale muizen werden gebruikt als bron van "nul"-cellen.
Muizen werden gedood door halsdislokatie.

Milten werden aseptisch verwijderd en fijngehakt met forceps in Hank's in evenwicht gebrachte zoutoplossing (HBSS) (Gibco Ltd., Paisley, Schotland). Een suspensie van enkelvoudige cellen werd verkregen door filtrering 5 op een draadzeef met fijne maaswijdte. Mononucleaire cellen (MNC) werden geïsoleerd door differentiële centrifugering op Lymphoprep (Nyegaard, Oslo, Noorwegen) met continue dichtheidsgradiënt gedurende 20 min. bij 450 g.
Na drie wassingen in HBSS, werd MNC gesuspendeerd 6 10 bij een concentratie vna 5 xlO cellen/ml in 199 medium (Gibco Ltd.) aangevuld met 1 % BSA (Cohn Fr. V. Sigma,
St. Louis), L-glutamine 2 mM en 10 mcg/ml van gentamycine sulfaat (Schering, Klworth, NJ, Ver. Staten) (TC 199-BSA). De cellen werden gebruikt toen hun levensvatbaarheid 15 boven 95 % was bij de Trypan Blue uitsluitingstest.
THY 1,2 ANTIGEENINDÜCTIEBIOPROEF:
Tweehonderd microliter van de milt MNC suspensie werd gemengd met hetzelfde volume tripeptide geschikt verdund met TC 199-BSA medium. De vergelijkingscellen 20 waren slechts behandeld met het medium TC 199-BSA. Alle cellen waren 18 uur lang bij 37°C geïncubeerd in vochtig-gemaakte atmosfeer-bevattende 5 % CC^, daarna tweemaal gewassen met HBSS-bevattende 5 % onder invloed van warmte inactief gemaakt serum van een pasgeboren kalf (Gibco) 25 (HBSSS-CS) en ten slotte opnieuw gesuspendeerd in hetzelfde medium bij een concentratie van 10 cellen/ml.
DïREKTE IMMUNOFLUORESCENTIE (IF):
De uitdrukking van Thy 1,2 antigeen werd geanalyseerd onder toepassing van IF en anti-Thy 1,2 mono-30 clonaal antilichaam geconjugeerd met fluoresceïne (Bio-
Yeda, verschaft door Technogenetics, S. Mauro Torinese, 6
Italië) met de concentratie van 2 mcg/10 cellen. De cellen werden geïncubeerd met het antilichaam gedurende 30 min. bij 4°C.
35 Na drie wassingen in HBSS werden de cellen opnieuw gesuspendeerd in hetzelfde medium en waargenomen met een Leitz Orthomat microscoop die was uitgerust met epi-verlichting.
Ten minste 300 cellen werden in elke beoordeling 40 geteld.

Zoals weergegeven in de tabel was het tripeptide Arg-Gly-Asp actief in vitro bij de inductie van Thy 1,2 inductie bij concentraties die liggen in het traject van 5 1 en 200 ^ig/ml, waarbij de optimale concentratie 10 mcg/ml is.

15 Stimulering van lymfokinebereiding in vitro
Mens-T-lymfocyten werden geïncubeerd met PHA en met of zonder de peptiden in onderzoek gedurende 24 uur (in het geval van IL-2) of 72 uur (in het geval van BCGF).
De bovendrijvende materialen werden verzameld 20 gefiltreerd (0,2 ^im) en beproefd op de aanwezigheid van IL-2 of BCGF activiteit, door hen toe te voegen bij verschillende concentraties aan verse T-lymfocyten of aan lange-termijn gekweekte B-cellen. De proliferatie-activiteit van deze cellen, afhankelijk van de aanwezigheid 25 van de respectievelijke groeifaktor, werd beoordeeld door 3H-thymidineopneming. „
De resultaten toonden een opmerkelijk effekt van stimulering aan van de groeifaktorbereiding door Arg-Gly-Asp, iets lager dan dat van ARg-Lys-Glu, maar hoger 30 dan datgene wat was verkregen met ARg-Lys-Asp.

Stimulering van lymfokine in vitro door zuivere mens- ? T-lymfocyten zonder macrofagen.
De lymfokinebereiding door mens-T-lymfocyten gezuiverd door massage op nylonwolkolom, werd beoordeeld 5 met dezelfde werkwijzen als in het voorafgaande experiment.
T-celzuiverheid was hierboven 95 %, terwijl macro-fagen/monocyten en voorts B-cellen onder 1 % waren.
Teneinde het gebrek aan macrofagen op te heffen, werd 10 recombinant IL-1 (Genzyme) toegevoegd met de concentratie van 25 eenheden/ml.
De verkregen resultaten, zowel voor IL-2 als BCGF, geven te zien dat het effekt van Arg-Gly-Asp vergelijkbaar is met dat van de andere referentieverbindingen, 15 waaronder ook thymosine fraktie 5, een gedeeltelijk gezuiverd thymisch derivaat, werd gebruikt.

Het tripeptide Arg-Gly-Asp vertoont een LD5Q hoger dan 1000 mg/kg i.p. in muizen.
5 VOORBEELD V: KLINISCHE TOEPASSING
De voorafgaande voorbeelden III en IV hebben aangetoond, dat de tripeptiden die het doel van de onderhavige uitvinding zijn, in vitro actief zijn als immuno-stimulerende agentia in experimentele modellen van zowel 10 dier als mens, en in vivo als antimetastatische produkten in laboratoriumdieren, behalve dat zij niet toxisch zijn.
Zo kan met grote redelijkheid worden voorspeld, dat zij klinisch nuttig zullen zijn bij het voorkomen van metastatisering bij patiënten die chirurgische 15 tumorverwijdering ondergaan, terwijl tegelijkertijd de immune afweren van het organisme worden verbeterd dankzij hun immunostimulerende eigenschappen.
ZOUTEN VAN DE TRIPEPTIDEN
De bovenvermelde voorbeelden verwijzen naar het 20 acetaatzout van de tripeptiden. Het is echter mogelijk analoge resultaten te verkrijgen met andere zouten van organische en anorganische zuren, zoals trifluorace-taat, hydrochloride, sulfaat.

Claims (18)
1. Tripeptiden, gekenmerkt door de algemene formule H - Gly - Y waarbij X = L-Arg of D-Arg en Y = L-Asp of D-Asp, en 5 zouten daarvan van organische en anorganische zuren, zoals acetaat, trifluoracetaat, hydrochloride, sulfaat.
2. Tripeptiden volgens conclusie 1, gekenmerkt door de volgende volgorde: Arg-Gly-Asp 10 D-Arg-Gly-Asp Arg-Gly-D-Asp D-Arg-Gly-D-Asp.
3. Tripeptiden volgens conclusie 1, gekenmerkt door in het bijzonder Arg-Gly-Asp en Arg-Gly-D-Asp.
4. Werkwijze voor het bereiden van antimetastatische agentia, met het kenmerk, dat men daarbij tripeptiden volgens conclusie 1 gebruikt.
5. Werkwijze voor het bereiden van immunostimulerende agentia, met het kenmerk, dat men daarbij 20 tripeptiden volgens conclusie 1 gebruikt.
6. Tripeptiden volgens conclusie 1, gekenmerkt doordat zij antimetastatische en immunostimulerende activiteit hebben.
7. Werkwijze voor het bereiden van geneesmiddelen 25 met antimetastatische activiteit na parenterale toediening, met het kenmerk, dat men daarbij tripeptiden volgens conclusie 1 en tripeptiden, verkregen onder toepassing van de werkwijze volgens conclusie 4 gebruikt.
8. Werkwijze voor het bereiden van geneesmiddelen met antimetastatische activiteit na orale toediening, 30 met het kenmerk, dat men daarbij tripeptiden .8801932 4 - 22 - volgens conclusie 1 en tripeptiden, verkregen onder toepassing van de werkwijze volgens conclusie 4 gebruikt.
9. Farmaceutische preparaten verkregen onder toepas sing van de werkwijze volgens conclusie 5, voor orale toediening.
10. Farmaceutische preparaten verkregen onder toepas sing van de werkwijze volgens conclusie 5, voor parenterale toediening.
11. Werkwijze voor het bereiden van geneesmiddelen met immunostimulerende activiteit, met het ken- 10 merk, dat men daarbij tripeptiden volgens conclusie 1 en tripeptiden verkregen onder toepassing van de werkwijze volgens conclusie 5 gebruikt.
12. Therapeutische toepassing van farmaceutisch aanvaardbare zouten of de tripeptiden volgens conclusie 1 en 4 - 10 als geneesmiddelen bij patiënten die chirurgische 15 tumorverwijdering ondergaan, teneinde de vorming te voorkomen van metastasen, terwijl tegelijkertijd wordt bijgedragen aan de verbetering van immune omstandigheden.
13. Toepassing van tripeptiden volgens conclusie 12 in de vorm van acetaatzouten.
14. Toepassing van tripeptiden volgens conclusie 12, als trifluoracetaatzouten.
15. Toepassing van tripeptiden volgens conclusie 12, als hydrochloridezouten.
16. Toepassing van tripeptiden volgens conclusie 12, 25 als sulfaatzouten.
17. Werkwijze voor het bereiden van tripeptiden volgens conclusie 1, gekenmerkt door de volgende fasen: - bereiding van det-butyloxycarbonyl-Gly-Asp-dibenzyl- 30 ester; - bereiding uit de aldus verkregen verbinding van de .8801932 % - 23 - tribenzyloxycarbonyl-Arg-Gly-Asp-dibenzylester; daarop volgende katalytische hydrogenering ter verkrijging van het tripeptide Arg-Gly-Asp.
18. Werkwijze voor het bereiden van de tripeptiden volgens conclusie 1, gekenmerkt door een vaste-5 fase synthese volgens het onderhavige voorbeeld I. .8801932
