Acylami.no mitosanen? werkwijze voor het bereiden daarvan.
. De onderhavige uitvinding heeft betrekking op mitomycine analoga die een of meer acylaminogroepen bevatten (klasse 260^ subklasse 326.24). Deze verbindingen zijn mitomycine C-derivaten, waarin de 7-aminogroep t a of het N -stikstofatoom of beide zijn opgenomen in een acylamino-5 substituent. Deze verbindingen zijn actieve antitumorstoffen die een in vivo tumor inhibitor activiteit vertonen tegen tumoren bij proefdieren.
Nomenclatuur - de systematische Chemical Abstracts naam voor mitomycine C is: [laR- (laa, 8β ,8aa,8ba) ] -6-amino-8- [ (aminocarbonyl) oxy) methyl] 10 l,la^2,8, 8a ,8b-hexahydro-8a-methoxy-5-methy 1-azirino£21,3* ,3,4]-pyrrolo[l,2-a lindool-4,7-dion, volgens welke naam het azirinopyrroloindool ringsysteem genummerd is volgens formule 1 van het formuleblad.
Een triviaal nomenclatuur systeem dat in de mitomycine literatuur 15 veel gebruikt wordt, identificeert het eerder genoemde ringsysteem met inbegrip van verschillende van de karakteristieke substituenten van de mitomycinen als mitosaan. Dit mitosaan en de nummering daarvan worden getoond in formule 2 van het formuleblad.
Hoewel dit systeem geschikt en gepast is voor een aantal een-20 voudige derivaten zoals die welke substituenten aan het azirinoring stikstofatoom of op de 7- of 9a-posities dragen, leidt. het. aan bepaalde dubbelzinnigheden en tekortkomingen voor algemeen gebruik. Met betrekking tot de verbindingen volgens de onderhavige uitvinding, die mitomycine C derivaten zijn met substituenten aan zowel het azirinoring stikstof-25 atoom als aan het aromatische ring amino stikstofatoom, is in deze beschrijving gekozen voor een verwijzing naar het azirino stikstofatoom als N^a en naar het aromatische ring amino stikstofatoom als gebruikmakend van het mitosaan nomenclatuursysteem. Wat de stereoche-mische configuratie van de prpdukten volgens de uitvinding betreft, 30 is de bedoeling dat ingeval van een aanduiding door de stamnaam "mitosaan" of door een structuurformule de stereochemische configuratie daarvan als gelijk aan die van mitomycine C wordt geïdentificeerd.
Mitomycine C is een antibioticum dat bereid wordt door fermentatie en momenteel onder goedkeuring van de Food and Drug Administration 35 in de handel is in de therapie voor uitgezaaide adenocarcinoma van de 8403852 -2- ί ? maag of pancreas in bewezen combinaties met andere goedgekeurde chemo- therapeutische middelen en als palliatieve behandeling wanneer andere (R) modaliteiten gefaald hebben (Mutamycin. Bristol Laboratories, Syracuse, New York 13201, Physicians' Desk Reference 35th Edition,1981, blz.717 en 5 718. Mitomycine C en zijn bereiding door fermentatie vormen het onder werp van het Amerikaanse octrooischrift 3.660.578 dat op 2 mei 1972 is verleend en voorrang inroept van eerdere aanvragen waaronder een in Japan op 6 april 1957 ingediende aanvrage.
De structuren van mitomycines A, B, C en van porfiromycine werden 10 voor het eerst gepubliceerd door J.S. Webb et al. of Lederle Laboratories Division American Cyanamid Company, J. Amer. Chem. Soc. 8£, 3185-3187 (1962). Een van de chemische transformaties die in dit structuuronderzoek werden gebruikt om mitomycine A en mitomycine C met elkaar in verband te brengen, was de omzetting van eerstgenoemde 7,9a-dimethoxy-15 mitosaan door reactie met ammoniak in het laatstgenoemde, 7-amino-9a-Tno'h’hrtxymi tngaan, Verdringing van de 7-methoxygroep van mitomycine A is een reactie van aanzienlijk belang gebleken in de bereiding van antitumor actieve derivaten van mitomycine C. De volgende artikelen en octrooien hebben elk betrekking op de omzetting van mitomycine A in een 7-gesub-20 stitueerd amino mitomycine C derivaat met antitumor activiteit. De stand van de techniek geeft echter geen voorbeelden van een 7-acylamino-mitomycine C analogon van het door de onderhavige uitvinding gepresenteerde type.

5 Mitomycine C derivaten met een gesubstitueerde amino substituent op de 7-positie zijn ook bereid door directe biosynthese, d.w.z. door fermentatievloeistoffen aan te vullen met een reeks primaire amines, en de conventionele mitomycine fermentatie uit te voeren (C.A.Claridge et al. Abst. of the Annual Metting of Amer.Soc. for Microbiology 1982 10 Abs. 028).
De uitvinding heeft betrekking op weer een andere groep van 7 N -gesubstitueerde mitomycine C derivaten waarin de aminogroep op de 7-positie gesubstitueerd is door een organische acylgroep. Carboxamides, thiocarboxamides, ureas, thioureas, urethanen, thiofos for amides, fos-15 foramides, en sulfonamides worden geïllustreerd. Deze verbindingen beantwoorden aan de formule 3 van het formuleblad, waarin 1 7 R gekozen wordt uit H, C, - alkyl, en R , 7 - R gekozen wordt uit de groepen met de formules 4 t/m 15, waarin 20 Ra gekozen wordt uit H, Cj_g alkyl, A-gesubstitueerde C^g alkyl,

Deze verbindingen zijn inhibitors van proeftumoren in dieren.
Ze zijn vergelijkbaar net mitomycine C met betrekking tot de typen ^ tumoren waarvan ze inhibitie geven, maar bezitten in vele gevallen een 8403852 y $ -4- hoge activiteit in de zin dat ze een grótere mate van inhibitie uitoefenen dan met mitomycine C kan worden bereikt. Hun toxiciteit is in het algemeen kleiner dan die van mitomycine C, hetgeen in het bijzonder tot uiting komt in hun lage myelosuppressieve werking.Voor antitumor doeleinden : 5 worden ze aan een zoogdier dat een tumor heeft, toegediend in een in essentie niet-toxische antitumor werkzame dosis.
Ze worden primair toegediend door injectie op overwegend dezelfde wijze als mitomycine C.. In het algemeen'kunnen ze in hogere dosis worden gebruikt dan mitomycine C in verband met hun geringere toxiciteit en 10 grotere tumorinhibitiewerking bij de hogere doses. Ze worden gemakkelijk verspreid in de vorm van droge farmaceutische preparaten die verdun-ningsmiddelen, buffers, stabilisatoren, oplosbaarmakende middelen, en aan de farmaceutische elegantie bijdragende ingrediënten bevatten.
Deze preparaten kunnen vlak voor het gebruik voor de vuistweg met 15 behulp van een injecteerbare vloeistof worden verwerkt. Geschikte injectievloeistoffen omvatten water, isotonische zoutoplossing, enz.
De verbindingen met formule 3 worden uit mitomycine C bereid door eerst een deprotonering uit te voeren om de anionogene vorm van mitomycine C te verkrijgen, gevolgd door een reactie van het anion met 20 een organisch isocyanaat of isothiocyanaat om een mitosaan te verkrijgen met een gesubstitueerde ureido of thioureidogroep op de 7-positie of door een reactie van de anionogene vorm van mitomycine C met een acylerende vorm van een organisch zuur zoals een acylhalogenide, een reactieve ester van een carbonzuur of thiocarbonzuur zoals een fenyl, p-nitrofenyl, 25 p-nitrobenzyl, enz. ester of met een ester vein een carbonzuur met een N-hydroxyamine zoals N-hydroxysuccinimide, N-hydroxybenzotriazol, N-hydroxyftaalimide, N-hydroxypiperidine, enz., een organisch carbonaat, of een organisch halogeen formiaat om een mitosaan te verkrijgen met een amidogroep, of carbamoylgroep opÖe 7-positie. De acylanhydride kunnen 30 eveneens als acyleringsmiddelen worden gebruikt.
De acyleringsmiddelen voor reactie met anionogeen mitomycine C worden gekenmerkt door de formules 16-27.
In deze formules is X een groep die in de stand van de techniek bekend is geworden als een vertrekkende groep. Op het gebied van de orga-35 nische synthese zijn veel van deze groepen ontwikkeld en deze omvatten de halogeniden en versschillende andere reactieve esters zoals die welke 8 4 0?«??.
» t ê -5- bovenstaand vermeld zijn alsook andere acylgroepen in welk geval het resulterende acyleringsmiddel een anhydride is. Gemengde anhydriden kunnen worden g^ruikt waarin een deel van het anhydride de acyleringsfunctie wordt en het andere deel de vertrekkende groep wordt.
5 Be deprotonering van mitomycine C om de anionogene vorm daarvan met nucleofiele eigenschappen te verkrijgen, werd voor het eerst beschreven in de samenhangende Amerikaanse octrooiaanvrage 492.903, die op 9 mei 1983 is ingediend. In deze vorm reageert mitomycine gemakkelijk met electrofiele reagentia, in dit geval acyleringsmiddelen.
10 De omstandigheden voor de deprotonering van mitomycine C omvatten een behandeling daarvan in dimethylformamide oplossing met ongeveer 1,5 mol hoeveelheden natriumhydride bij kamertemperatuur of lager.
De reactie van het anion met een acyleringsmiddel, tenminste een chemisch equivalent daarvan en bij voorkeur 1,5-2 chemisch equivalenten daarvan 15 ten opzichte van de anionogene vorm van mitomycine C, wordt uitgevoerd onder watervrije omstandigheden bij een temperatuur van ongeveer kamertemperatuur tot ongeveer -60°C. Temperaturen van -20 tot -30°C worden gemakkelijk gerealiseerd onder zowel labo-ratoriumomstandigheden als prodüktieomstandigheden en zijn voor het 20 uitvoeren van de onderhavige werkwijze zeer bevredigend.
Aprotische polaire organische oplosmiddelen zoals pyridine, dimethylformamide, hexamethylfosforamide, of dimethylsulfoxyde worden gebruikt. De werkwijze is echter niet beperkt tot de vorming van anionogeen mitomycine op de bovenstaand beschreven wijze, omdat aan de 25 deskundigen modificaties voor ogen-zullen staan.
Vöorkeursverbindingen van de onderhavige uitvinding vanwege hun hoge antitumor activiteit en betrekkelijk lage toxiciteit in vergelijking tot mitomycine C,zijn 7-(formyl) amino~9a-methoxymitosaan (voorbeeld I), 7- (acetyl) amino-9a-methoxymitosaan (voorbeeld III) en 7-(cyclopropaan-30 carbonyl) amino-9a-methoxymitosaan (voorbeeld XIII) .Deze verbindingen zijn 7 representatief voor de subgroep met formule 3, waarin R alkanoyl is met 1—7 koolstof atomen of cycloalkylcarbonyl is met 4-9 koolstofatomen, welke groepen de voorkeurstoffen volgens de onderhavige uitvinding omvatten.
Activiteit tegen P-388 muizenleukemie 35 Tabel A bevat resultaten van laboratoriumproeven met CDF^ vrouwtjesmuizen, waarin intraperitoneaal een tumorent was geïmplanteerd 8403852 4 » -6- c van 10 ascites cellen van P-388 muizenleukemie en die met verschillende doses behandeld waren van hetzij een testverbinding volgens de uitvinding, hetzij mitomycine C. De verbindingen werden door intraperitoneale injectie toegediend . Groepen van zes muizen werden voor elke doseringshoe-5 veelheid gebruikt en deze werden op de dag van inenting met dén enkele doses van de verbinding behandeld. Een groep van 10 met zoutoplossing behandelde controlemuizen werd in elke reeks proeven opgenomen.
De met mitomycine C behandelde groepen werden als positieve controle opgenomen. Er werd een 30 dagen durend protocol toegepast waarbij 10 de gemiddelde overlevingstijd in dagen voor elke groep muizen werd bepaald en het aantal overlevenden op het einde van de 30-daagse periode werd genoteerd.De muizen werden voor de behandeling en opnieuw op de zesde dag gewogen. De verandering in gewicht werd als een maat voor de toxiciteit van het geneesmiddel genomen. Er werden muizen gebruikt 15 die elk 20 g wogen en een gewichtsverlies tot aan ongeveer 2 g werd niet buitensporig geacht. De resultaten werden bepaald in termen van % T/C, hetwelk de verhouding van de gemiddelde overlevingstijd van de behandelde groep tot de gemiddelde overlevingstijd van de met zoutoplos-* sing behandelde controlegroep -maal 100 is. De met zoutoplossing be- 20 handelde controledieren stierven gewoonlijk binnen negen dagen. Het "’maximum effect" in de volgende tabel is uitgedrukt als % T/C en de dosis die het effect geeft, wordt vermeld. De waarden tussen haakjes zijn de waarden die verkregen werden met mitomycine C als positieve controle in hetzelfde experiment. Aldus kan een maat van de relatieve activiteit 25 van de stoffen volgens de uitvinding ten opzichte van mitomycine C worden geschat. Een minimum effect in termen van % T/C werd 125 geacht. De minimum effectieve dosis, die in de volgende tabel wordt vermeld, is de dosis die een % T/C van ongeveer 125 heeft. De twee waarden die steeds in de kolom "gemiddelde gewichtsverandering" worden gegeven, stellen respec-30 tievelijk de gemiddelde gewichtsverandering per muis bij de maximum effectieve dosis en bij de minimum effectieve dosis voor. · 8403852 -7-

Tabel B bevat resultaten van antitumortests waarbij het in muizen gegroeide B16 melanoma wordt gebruikt. BDFj muizen werden gebruikt en subcutaan ingfceht met het tumorimplantaat. Er werd gebruik gemaakt van een '60 dagen omvattend protocol. Groepen van tien.muizen werden 5 voor elke geteste · doseringshoeveelheid gebruikt en de gemiddelde overlevingstijd voor elke groep werd vastgesteld. Controledieren, die op dezelfde wijze waren ingeënt als de proefdieren en met de injectiedrager waren behandeld zonder geneesmiddel, vertoonden een gemiddelde over- t levingstijd van 24,5 dagen. De overlevingstijd in verhouding tot die 10 van de controles (% T/C) werd genomen als maat voor de effectiviteit, en voor elke proefverbinding werden de maximale effectieve dosis en de minimale effectieve dosis bepaald. De minimale effectieve dosis werd gedefinieerd als de dosis die een % T/C waarde van 125 vertoonde.
Voor elk doseringsniveau, werden de proefdieren met de proefverbinding 15 behandeld op de dagen 1,5 en 9 langs intraveneuze route.

Tabel C bevat verdere resultaten van antitumorproeven waarbij gebruik gemaakt werd van het B16 melanoma, gegroeid in BDF. muizen, 3 welke ditmaal intraperitoneaal waren ingeënt met de tumor (0,5 cm , 30 10% brei). Men gebruikt een protocol van 60 dagen. Groepen van tien muizen werden voor elke onderzochte doseringshoeveelheid gebruikt en de gemiddelde overlevingstijd voor elke groep werd vastgesteld.

Tabel D verwijst naar de meting van myelosuppressieve werking van de verbindingen van voorbeelden I, III en XIII wanneer ze intraveneus aan muizen worden toegediend, vergeleken met mitomycine C.
Aan de muizen werd een enkele dosis van de proefverbinding toegediend 25 op dag 0 en op de dagen 0, 4 en 7 werden het totaal aantal witte bloedcellen geteld (WBC). De waarden voor de dagen 4 en 7 worden in tabel D vermeld als procentuele wijziging (Δ%). Differentiële tellingen werden uitgevoerd op de dagen 0 en 4 en aangegeven als procentuele verandering (A%) op dag 4. Aan verschillende groepen van tien dieren 30 werden verschillende doses van de proef verbindingen, zoals getoond in de tabel, toegediend. De waarden voor Δ%, vermeld tussen haakjes, zijn vergelijkingswaarden voor mitomycine C, die op dezelfde wijze zijn gemeten. De verbindingen van de voorbeelden I, III en XIII werden minder myeosuppressief bevonden dan mitömycine C. Bij doses met 35 equivalente lethaliteit, hebben de onderhavige verbindingen aanzienlijk minder effect op de neutrofieltellingen dan mitomycine C.

De volgende voorbeelden vormen gedetailleerde procedures voor de bereiding van verschillende specifieke uitvoeringsvormen van de onderhavige uitvinding. De verbindingen werden in het algemeen gekarakteriseerd door hun kernmagnetische resonantie, infrarood absorptie, 5 en ultraviolet absorptiespectra. De spectra worden in de volgende voorbeelden beschreven in conventionele bewoordingen die voor dit type-gegevens in de stand van de techniek aanvaard zijn. In de meeste gevallen worden elementair analyses verstrekt, die beantwoorden aan de getoonde structuurformules hetgeen een verdere bevestiging van de identiteit 10 van deze stoffen geeft. 7- (Formyl) amino-9a-methoxymitosaan (BL-6859) met formule 28.
Hen voegde 6 ml dimethylformamide toe aan een mengsel van 549 mg, 1,78 mmol mitomycine c en 214 mg van een 50% oliedispersie van 4,45 mmol NaH, die in een argonatmosfeer werd gehouden. Na gedurende 25 10 minuten te hebben geroerd bij kamertemperatuur, werd het mengsel tot -20°C afgekoeld en werd fenylformiaat toegevoegd (0,60 ml 65%-ig zuiver materiaal, 3,50 mmol, H.L. Yale, J. Org. Chem. 36_, 3234 (1971)).
Het reactiemengsel werd gedurende 30 minuten bij -20°C geroerd en daarna in een periode van 1 uur opgewarmd op kamertemperatuur. Na af schrikken 30 van het reactiemengsel door de toevoeging van vast CC^, werd het reactiemengsel verdund met EtOAc. Het verkregen precipitaat werd af gefiltreerd en het oplosmiddel werd onder verminderde druk verwijderd.
Het residu werd chromatografisch behandeld over silicagel (5% MeOH-CHjC^) 8403852 -12- waarblj 306 mg, 47% van de titelverbinding werd verkregen. Men gebruikte als acy 1 eringsmidde1 432 mg, 1,80 mmol p-nitrofenyl trifluoroace-20 taat. Een silicagel dunnelaagchromatografie (10% MeOH-C^C^) gaf de titelverbinding als een roodachtigpaarse amorfe vaste stof: ; f91 mg, 24%): smp 93-95°C; NMR (pyridine-dg, ί) 2,02(s, 3H), 2,74(m, 1H), * 3,11 (d, 1H, J=4 Hz), 3,20(s, 3H), 3,52(dd, 1H, J=12, 1 Hz), 4,00.. Hen voegde 6 ml dimethylformamide toe aan een mengsel van 668 mg, 2 mmol mitomycine C en 4 mmol NaH onder een argonatmosfeer. Na 20 min. roeren bij kamertemperatuur werd het reactiemengsel afgekoeld tot -25°C ^ en werd 454 mg, 2 mmol, p-nitrofenylmethaansulfonaat toegevoegd (Kametani et al. Yakugaku Zasshi, 84,237 (1963)). Het reactiemengsel werd gedurende 4 uur op -25°C gehouden. Na afschrikken van het reactiemengsel met vast C02, werd het reactiemengsel verdund met EtOAc en met een zoutoplossing gewassen. 7- (Methoxycarbonyl)amino-9a-methoxymitosaan (BMY-25069) met formule 34.
Aan een mengsel van 334 mg, 1 mmol mitomycine C en 96 mg 2 mmol NaH in 50%-ige oliedispersie werd onder stikstofatmosfeer 5ml droog DMP 30 toegevoegd. Het mengsel werd gedurende 10 minuten bij kamertemperatuur geroerd en vervolgens tot -30°C gekoeld. Men voegde 482 mg, 2,3 mmol methyl £-nitrobenzylcarbonaat toe als vaste stof en zette het roeren gedurende 1 uur voort bij ~30°C. Het reactiemengsel werd afgeschrikt door toevoeging van een kleine hoeveelheid droogijs, en het reactie-35 mengsel werd verdeeld over EtOAc en water. De organische laag werd . -Λ 8403852 -15- _ gewassen met zoutoplossing en gedroogd boven ^280^. Aan een mengsel van 668 mg, 2 mmol mitomycine C, en 192 mg, 5 4 mmol NaH in 50%-igeC^ispersie werd onder stikstof 8 ml droog DMP toe gevoegd. De verkregen oplossing werd gedurende 20 minuten bij kamertemperatuur geroerd en vervolgens tot -25°C gekoeld; Men voegde 340 mg 4 mmol isopropylisocyanaat toe en zette het roeren gedurende 1 uur bij “25°C voort. Het reactiemengsel werd afgeschrikt door toevoeging van een 10 kleine hoeveelheid droogijs, en het reactiemengsel werd verdeeld over EtOAc en water. De organische laag werd gewassen met zoutoplossing en gedroogd boven Na^SO^. te gébruiken, verkrijgt men analoge produkten als die van de voor- beelden en beantwoorden aan formule 3, waarin R staat voor lage alkyl.
De N*a-gesubstitueerde mitomycine C uitgangsmaterialen kunnen worden 20 verkregen als beschreven door Matsui et al., J. Antibiotics, 21, MO. 3 189-198(1968). De verbindingen met formule 3 waarin R* een groep 7 als gedefinieerd voor R is} zijn bis-gesubstitueerde re.actie-produkten, die uit de eerder vermelde voorbeelden als beschreven worden geïsoleerd. Onder gepast gekozen reactiecondities, kan de gevormde 25 hoeveelheid bis -produkt worden vergroot. Mitomycine C uitgangsmaterialen waarin de a-substituent een acylgroep is, kunnen worden bereid als beschreven door Matsui et al. in het Amerikaanse octrooischrift 3,450,705 van 17 juni 1969.

17. Verbinding volgens conclusie 1, te weten N^a- (benzyloxycarbonyl) -7-benzyloxycarbonylamino-9a-methoxymitosaan.
18. Verbinding volgens conclusie 1, waarin EÜ· waterstof voorstelt en cycloalkylcarbonyl met 4-9 koolstofatomen voorstelt.
19. Verbinding volgens conclusie 1, te weten 7-(Cyclopropaancarbonyl)-amino-9a-methoxymitosaan.

23. Verbinding volgens conclusie 1, te weten 7-[((2-chloroethyl) amino-carbonyll amino-9a-methoxymitosaan.

Werkwijze voor het bereiden van een verbinding volgens conclusie 1, waarbij men anionogeen mitomycine C laat reageren mé: 1-2 mol hoeveelheden van een acyleringsmiddel met een van de formule 16-27waarin 30. een conventionele vertrekkende groep is en R en ΈΓ dezelfde betekenis hebben als in conclusie 1, in een aprotisch polair organisch oplosmiddel, bij een temperatuur in het bereik van -60 tot +25°C.
25. Werkwijze voor het bereiden van een verbinding volgens :· conclusie 1, waarbij men mitomycine C opgelost in dimethylformamide 35 (of een ander verenigbaar oplosmiddel) laat reageren met 1,0 tot 1,5 mol hoeveelheden natriumhydride, waarbij anionogeen mitomycine C wordt 8403852 :* v J -24- gevormd, en daarna het aniogene mitomycine C laat reageren met 1-2 mol hoeveelheden van een acyleringsmiddel met een van de formules 16-27, cl b waarin X een conventionele vertrekkende groep is en R en R dezelfde betekenis hebben als in conclusie 1, in een aprotisch polair organisch-5 oplosmiddel bij een temperatuur in het bereik van -60° tot +25°C.
26. Werkwijze volgens conclusie 24 of 25, waarin het aprotische polaire organische oplosmiddel dimethylformamide of pyridine is.
27.
