Titel: N-methyl-D-glucaminezouty oplossing daarvan.
De onderhavige uitvinding heeft betrekking op het zuuradditiezout van 3,4-dihydro-5-methy1-6-(2-methylpropyl)-4-oxothieno[2,3-d]pyrimidine-2-carbonzuur met N-methyl-D-glucamine dat een. base/zuur molecuulverhou-dlng van 1.:1 heeft. De verbinding volgens de uitvinding wordt in zijn 5 algemeenheid geclassificeerd als een geneesmiddel, een type verbinding voor het beinvloeden van het leven en het behandelen van het lichaam.
De zuurcomponent van het zout volgens de uitvinding wordt hier aange-„ duid met het codegetal MJ 12175 en heeft formule X van het formuleblad.
Door D.L. Temple, Jr., wordt in de Amerikaanse octrooischriften 10 4.054.656 en 4.159.377 de bereiding van MJ 12175 heschreven, alsmede zijn gebruik als anti-allergisch middel, en het kaliumzout daarvan. Laatstgenoemde is voordelig vahwege zijn betere oplosbaarheid in water, maar oplossingen daarvan zijn nlet gewenst voor. neusdruppeLeno . in ver-band met hun basische pH. Zoals daarin vermeld kan MJ 12175 voor anti-* 15 allergische doeleinden worden toegepast in de vorm van het zuur of de metaalzouten daarvan. Er worden dan ook voorbeelden gegeven die gericht zijn op zouten van de alkalimetalen, natrium en kalium. Teneinde een snellerresponsie met een anti-allergisch materlaal voor therapeutische doeleinden te realiseren, is een daseringsvorm vereist die tppicaal op 20 slijmmembranen kan worden toegediend. Dit vereist in het algemeen dat het geneesmiddel gepresenteerd wordt als een niet-irriterende oplossing of droog poeder, waarbij de oplossing de voorkeur heeft omdat het daar-mede minder lastig is om de dosering te beheersen wanneer deze bijvoor-beeld als neusdruppelpreparaat wordt geleverd.

Het. doel van de onderhavige uitvinding is derhalve om een in water oplosbare, stabiele, therapeutisch aanvaardbare vorm van het anti-allergischemiddel MJ 12175 te verschaffen, die topicaal kan worden toegediend. Een ander doel van de uitvinding is om een oplossing te verschaffen, bij voorkeur. verdunde oplossing, die geschikt is voor 30 neusdruppelen en een vloeibare pharmaceutische drager omvat welke een hoeveelheid van het N_-methyl-D_-glueaminezout_van_.3 ,4-dihydro-5-methyl- 6—(2-methy;%ropyli}-4*-oxo-thieno[ 2,3-d] pyrimidine-2-carbonzuur bevat die· effectief een onmiddellijke hypergevoeligheidsreactie inhibeert.

Zoals eerder vermeld heeft de onderhavige uitvinding betrekking op een aminezout, dat MJ 12175 alsmede N-methyl-D-glucamine omvat, waarin de zuur/base molecuulverhouding een 1:1 verhouding van equiva— lenten van beide omvat..
5 Vele conventionele pharmaceutisch aanvaardbare aminezouten van MJ 12175 zijn niet erg geschikt voor het onderhavige gebruik als. gevolg van een beperkte oplosbaarheid, gebrek aan stabiliteit,en pH. gebieden. die » r in voldoende mate van de fysiologische pH (7,4) afwijken om ervoor te zorgen dat weefselirritatie een probleem wordt.

In tegenwoordigheid van een sterke base, kan het proton aan het ringstikstof van MJ 12175 ook door een kation zijn vervangen.
2. Zout no.· 11 kon niet worden gezuiverd en de oplosbaarheid van het gelsoleerde materiaal werd niet nauwkeurig vastgesteld; het isolaat 25 gaf echter geen volledige oplosbaarheid in water.
De aminezouten van MJ 12175, getoond in tabel A, werden eerst in situ bereid door het amine toe^te voegen aan een suspensie van MJ 12175 in een hoeveelheid water die voldoende was onr een 4% us. waterige op-lossing van het resulterende zout te verkrijgen.Wanneer zich bij het 02 0 3 2 8 0 ' - 4 - toevoegen van amine een precipitaat vormde (hetwelk een relatieve op-losbaarheid aangaf), zoals in de gevallen van de zouten 4 tot 8 in tabel. Hoewel oplo.ssingen werden verkregen . met de andere in situ gevormde zouten lijkt o'ververzadiging van deze 5 oplossingen waarschijnlijk omdat de vier_zouten (nrs. 2, 3, 9 en li) die geisoleerd werden, minder dan 4% oplosbaar waren bij opnieuw oplos— sen» Drie van deze gelsoleerde zouten neigden stabiele kristallijne materialen te zijn, terwijl het vierder.zout (no. 11) tijdens zuiverings-pogingen een niet-kristallijne teer bleef. De drie kristallijne zouten 10 waren, de enige voorbeelden waarbij uit de- in situ oplossingen bij staan vaste: stof neersloeg. Van de drie kristallijne geisoleerde aminezouten van. MJ 12175, voldoet het glucaminezout het best aan de criteria voor de wateroplosbaarheid en pH. Verdere ·onderzoekingen met het glucaminezout hebben de stabiliteit van zijn waterige oplossingen bij langdurig 15 bewaren aangetoond. Zoals onderzoek van tabel A laat zien, kan het geschikt’zijn van de verschillende aminezouten niet op basis van de wateroplosbaarheid en de pH worden voorspeld.
• De bereidiing van het glucaminezout van MJ 12175 omvat het suspenderen van MJ 12175 in alcohol en het behandelen van de suspensie 20 met een hete alcoholische oplossing van de base, N^nethyl-D-glucamine. Door het roeren voort te zetten en te filtreren wordt het zuivere zout produkt verkregen dat een zuur/zout verhouding van 1:1 heeft.
Behalve de bovenstaande vermelde gewenste oplosbaarheid, fysische geschiktheid en pH eigenschappen die door het onderhavige zout worden 25 vertoond, zijn zijn andere fysische eigenschappen zodanig dat geen compatibiliteitsproblemen bestaan bij het formuleren van het zout in gebufferde oplossingen die bestemd zijn voor topicale toediening.
Een geprefereerde oplossing voor topicale toediening is een geconserveerde waterige oplossing, gebufferd in het pH gebied vein 30. 5,0 tot 8,0, enneen MJ 12175 glucamlnezoutconcentratie van ongeveer 0.,2% tot ongeveer 1,2% (w/v) bevattend. Deze oplossing kan ook conser— veringsmiddelen bevatten zoals een methylparaben, propylparaben, fenylalcohol en benzylalcohol,* buffers zoals citraat, fosfaat, en acetaati chelaatvoxmende middelen zoals ethyleendiamine, tetraazlj nzuur-35. zouten, bis-(2-hydroxyethyl) glycine en wxjnsteenzuur? en:.antioxydatie- “T203280 : : ........ ' - 5 - .
middelen zoals natriumbisulfiet, ascorbinezuur en cysteinehydrochloride. Deze pharmaceutische additieven worden frequent in produkten van dit type gebruikt*
Het N-methyl-D-glucaminezout van MI 12175 kan ook ondar toepassing 5 van andere doseringsvormen worden toegediend. Het zout kan worden opge-lost of gesuspendeerd in een halogeen koolstof of koolwaterstof drijf-middelsysteem enrverpaktiworden in een aerosolhouder met dosisregeling, Nevenoplosmiddelen, oppervlakactieve stoffen en suspendeermiddelen kunnen ook in deze doseringsvorm worden opgenomen. Het zout kan ook 10 worden toegediend als een ihblaaspoeder, bestaande uit een mengsel van inerte ingredienten, gemengd met een geschikte hoeveelheid van het glucaminezout met gepaste deeltjesafmetingen, toegediend door een poe-derinblaasapparaat.
De uitvinding wordt aan de. hand van de volgende,. niet beperkende 15 voorbeelden toegelicht. .
VOORBEELD I
Bereiding van MJ 12175 carbonzuur 3,4-dihydro- 5-methyl-6- (2-me thy-lpropyi 1-4-oxothieno [ 2,3 -d ] pyrimidine-2-carbonzuur.
20 Een oplossing van 37,6'g (0,01 mol) dikalium. 3,4-dihydro-5- methyl-6- (2-methy Ipropy 1) -4-oxothieno [ 2,3 -d] pyrimidine-2-carboxy laat-hydraat in 1,1 liter water werd bij 40°C behandeld met 5 g geactiveerde houtskool en werd gefilteerd. Het filtraat werd onder roeren gedurende een periode van 3 uur toegevoegd aan een oplossing van 21 ml (0,25 moll 4 25 37%-ig zoutzuur in 0,5 liter water, gehouden op een temperatuur van 35 - 38°C. Het mengsel werd. gedurende nog. eens· twee uur geroerd, en daaraa werd. de vaste stof op een filter verzameld,. gespoeld met water en aan de lucht gedroogd, waarbij 26,5 g van een gebrokea witrpradukt met een smeltpunt van 14Q,5-141,5°C (ontleding) werd. verkregen. Het 30 produkt werd met 250 ml methanol tot een brij. gemaakt teneinde een lichtgele? kleur te verwijderen en daarna gefiltreerd en aan de lucht gedroogd. De IR en NMR spectra waren consistent voor de aan MI 12175 _______ _ - - 6 - toegekende structuur.
Anal, berekend voor C^2H14N2°3S*S20: C: Ht 5»67; N: 9,85,* H^Or. 6,34. Gevonden: Cr 50,87; H: 5,65; N: 10,00; H^Or S>19*
Het dikaliumzout van MJ 12175, gebruikt in het bovenstaande voor-5 beeld, kan worden verkregen met de procedure die in de eerdergenoemde octrooischriften van Temple is beschreven, in het bijzonder procedvire - 97 van het Amerikaanse octrooischrift 4.159.377, dat hier door ver-wijzing opgenomen moet worden geacht.
VOORBEELD II
10 Bereiding van MJ? 12175. glucaminezout 3,4 dihydro-5-methyl-6- (2-meth.ylpropyl} -4-oxothieno [ 2,3-d ] pyrimidine-2=carbonzuur-N-methyl-D-glucaminezout 48,2 g (0,18 mol) MJ 12175, het produkt van voorbeeld I, werd bij 25°C geroerd in. 1,5 liter methanol. Een oplossing van 35,3 g (0,18 mol) 15 N^methyl-D-glucamine in 300 ml kokende methanol werd in een. keer onder hevig roeren toegevoegd.. Bijna alle vast® stof.:iaste op voordat zich een dik. precipitant begon te vormen. Dit mengsel werd gedurende 2 uur geroerd en daarna tot 10°C afgekoeld. De vaste stof werd verzameld op een filter, gespoeld met extra methanol en aan de lucht gedroogd. De 20 witvaste stof woog 75,5 g (90,4% van de theorie), en had een smeltpunt van 180,5-182°C (met bellenvorming).
Anal, berekend voor ci2314N203S’C7H17N05: C: 49,45; H: 6,77; N: 9,10; S: 6,95. Gevonden: C: 49,47; Hr 6,84; N: 9,06; Sr 6,98.
NMR (DMSO-d,.): Chemische verschuiving (aantal protonen, multipliciteit) 6 25 - 0,94 (6,dl; 1,85 (l,m); 2,40 (3,s); 2,60 (3,s); 2,64 -(2,d)? 3,10 (2,m); 3,52 (4,m); 3,71 (l,m); 3,97 (l,m); 6,00 (8,bs).
IR (KBr) t 1080, 1370, 1470, 1570, 1635, 1670, 2900, 2930, 2960, 3260, -1 3370 en 3460 cm. .

Oplossing voor topical® nasale toediening-
Het MJ 12175 glucaminezout volgens de'onderhavige. uitvinding wordt bij voorkeur toegediend als een geconserveerde gebufferde waterige 5 oplossing. De samenstelling van enkele typerende werkbare oplossingen· f ‘ van MJ 12175 glucaminezout worden in onderstaande tabel B getoond.

Claims (7)
1. Het N-methyl-D-glucaminezout van 3,4-dihydro-5-methyl-6-(2- methylpropy1)-4-oxothienyl[2,3-d]pyrimidine-2-carbonzuur.
2. Het zout volgens conclusie 1, waarin de molecuulverhouding van N-methyl-D_-glucamine tot genoemd zuusr 1:1 bedraagt.
3. Zout volgens conclusie 1 in een oplossing, bij voorkeur een ver- dunde oplossing, geschikt voor neusdruppelen en omvattend een vloeibare pharmaceutische drager, welke een hoeveelheid van genoemd zout bevat, die effectief een onmiddellijke hypergevoeligheidsreactie inhibeert.
4. Gebufferde waterage oplossing voor nasale toediening, welke in 10 hoge mate niet irriterend voor slijmvliezen is, en ongeveer 0,2% tot 1,2% (w/v) van het zout volgens conclusie 1 bevat.
5. Werkwijze voor het bereiden van een N-methyl-D-glucaminezout van 3,4-dihydro-5-methyl-6--(2-methylpropyl ).-4.-roxothienyl [ 2,3,fd ] pyrimidine-2-carbonzuur, met het kenmerk, dat 3,4-dihydro-5-methyl-6-(2-methylproyl)-15 4-oxothieno[2,3-d]pyrimidine-2-carbonzuur wordt gesuspendeerd in een geschikt oplosmiddel, by voorkeur alkohol, daarna in reactie wordt gebracht met N-methyl-D-glucamine, bij voorkeur met een hefce alkoho-lische oplossing van N-methyl-D-glucamine, waarbij de hoeveelheden van de reactiecomponenten zodanig zijn, dat het verkregen zout een 20 zuur/base molverhouding van 1:1 heeft.
6. Werkwijze voor het bereiden van een topicale oplossing, geschikt voor neusdruppelen, met het kenmerk, dat een effectieve hoeveelheid om een onmiddellijke hypergevoeligheidsreactie te inhiheren van het zout volgens conclusie 1 wordt opgelost in een farmaceutisch aanvaard- 25 bare drager.
7. Werkwijze volgens conclusie 6, waarin de oplossing een waterige, gebufferde oplossing is, die ongeveer 0,2 - 1,2% (w/v) van het zout volgens conclusie 1 bevat. 82 0128 0
