Paracetamolderivaat, de bereiding en toepassing daarvan.
Paracetamol of ïï-acetyl p-aminofenal neemt onder tegenwoordig gebruikte analgetische en antipyretische geneesmiddelen een belangrijke plaats in.
Paracetamol, een moeilijk in water oplossend pro-5 dukt (circa 1,3 g/100 ml) wordt gewoonlijk oraal ingenomen enwel in de vorm van tabletten of suspensies, die oplossende middelen nodig hebben, zoals glycerol, glycolen in verschillende mengverhoudingen en andere adjuvants, die bepaalde bezwaren kunnen opleveren wat betreft de smaak of viscosi-10 teit van het produkt.
De uitvinding betreft een nieuw derivaat van paracetamol met een zeer goede oplosbaarheid in water, zodat daarvan sterk geconcentreerde oplossingen kunnen worden ver-. kregen, die parenteraal, intramusculair of intraveneus kun- 15 nen worden toegediend, evenals oraal, bijvoorbeeld in zakjes, waarvan de inhoud in water dient te worden opgelost.
De· verbinding van de uitvinding vertoont volgens farmacologische proeven bij dieren analgetische en antipyretische werkingen, die geheel te vergelijken zijn met die • 20 van paracetamol, waardoor het gebruikt kan worden voor de behandeling van pijnen van verschillende oorsprong bij de mens, al of niet gepaardgaande met een verhoogde lichaamstemperatuur. Het kan oraal of parenteraal in een dosis van 0,5-6 g per etmaal toegediend worden, als naar de wijze van 25 toediening en de gevoeligheid van de patient.
Het gaat hier- om de fosforzure ester van paracetamol of p-aceetaminofenylfosfaat van formule 1 van het formuleblad, dat wordt gebruikt in de vorm van zouten met anorganische of organische basen zoals het dinatriumzout, 30 zouten met aminozuren, zoals glycine, lycine en adenosine.
De uitvinding verschaft tevens een werkwijze voor de bereiding van p-aceetaminofenylfosfaat uit de fosforzure • . ester van p-nitrofenol’in de vorm van het dinatriumzout, dat in aanwezigheid van 5%tig palladium/koolstof katalytisch 7908988 -2“ t %- wordt gereduceerd tot p-aminofenylfosfaat, dat dan met azijn-zuuranhydride wordt geacetyleerd. De bereiding zal worden verduidelijkt aan de hand van de volgende niet beperkende voorbeelden.
5 Voorbeeld I
Eerst stap; -Bereiding van het dinatriumzout van p-aninofenyl-fosfaat.
25 g dinatrium p-nitrofenylmonofosfaathexahydraat wordt 2 liter 50%tig ethanol bij kamertemperatuur en onder 10 atmosferische druk gehydrogeneerd in aanwezigheid van 4 g 5%tig palladium/koolstof, waarna de katalysator wordt afgefiltreerd en het filtraat wordt örooggedampt.
Het residu wordt met watervrij ethanol behandeld, afgezogen en 4 uren bij 60°C gedroogd ter verschaffing van 15 circa 15 g van het watervrije beoogde tussenprodukt.
Tweede stap: Bereiding van het dinatriumzout van p-aceetamino-fenylfosfaat."
In een rondkolf wordt aan 200 ml azijnzuuranhydri-de portiegewijs 15 g van de beide eerste stap verkregen 20 verbinding toegevoegd. Nadat het mengsel 30 min is geroerd wordt daaraan 500 ml ether toegevoegd, waarna het geheel weer 30 min wordt geroerd en daaraan 250 ml aceton en 10 ml water worden toegevoegd, gevolgd door 15 min roeren. Het neerslag wordt afgefiltreerd, tweemaal met telkens 450 ml 25 aceton uitgewassen en met 150 ml ethanol behandeld, waarna men het geheel 12 uren bij kamertemperatuur laat staan.
Het neerslag wordt afgefiltreerd, tweemaal met telkens 15 ml ethanol uitgewassen en bij 60°C gedroogd ter verschaffing van circa 13 g ongezuiverd produkt, aan een 30 oplossing waarvan in 60 ml water' 1,5 g actieve koolstof wordt toegevoegd. Het mengsel wordt gefiltreerd en het filtraat wordt met 500 ml aceton verdund, waarna het geheel wordt geroerd. Het kristallijne neerslag wordt afgefiltreerd enige malen met aceton uitgewassen en 2 uren bij 60°C en dan 35 1 nacht bij 105°C gedroogd ter verschaffing van circa 7,5 g van de beoogde verbinding in de vorm van een kleurloze kristallijne stof met een smeltpunt van meer dan 265°C.
Het produkt is zeer gemakkelijk en wateroplosbaar, met name 7908988 * -3- 50 g/100 ml bij kamertemperatuur. De oplossing is stabiel en vertoont een neutrale pH.
Voorbeeld II
p-aceetamino£osfaat wordt verkregen door het 5 dinatriumzout daarvan met zuur te behandelen ter verschaffing van een neerslag van het vrije zuur in de vorm van kleurloze kristallen met 1 molecuul water per mol./ welk monohydraat onder ontleding bij 135°C smelt, terwijl het watervrije produkt een smeltpunt van 205°C vertoont.
10 Het glycinezout van de verbinding van formule 1 wordt verkregen door equimolaire hoeveelheden glycine en het vrije zuur in water bij 25°C op te lossen, waarna aan de oplossing aceton wordt toegevoegd ter verschaffing van een kristallijn neerslag, dat uit 85%tig ethanol wordt herge-15 kristalliseerd ter verschaffing van het beoogde glycinezout in de vorm van kleurloze kristallen met een smeltpunt van 195°C.
Op dezelfde wijze-wordt het lycinezout verkregen, · doch voor het neerslaan wordt aceton. in plaats van ethanol 20 gebruikt ter verschaffing van het gewenste zout in de vorm van kleurloze kristallen met een smeltpunt van 179—181°C (Büchi-capillair).
Het onder dezelfde omstandigheden als in vorenstaande gevormde adenosinezout slaag spontaan uit het reaktie-25 milieu neer in de vorm van het kleurloze monohydraat met een smeltpunt van 163°C (Büchi-capillair).
De analgetische en antipyertische werkingen van de zouten van de verbindingen van de uitvinding zijn dezelfde als die van paracetamol, als uit het volgende blijkt.
30 Het dinatriumzout.
1) Acute toxiciteit.
De bij orale toediening aan muizen bedraagt circa 1200 mg/kg, hetgeen in mol uitgedrukt evenveel is als die van paracetamol.
35 2) Analgetische werking.
Deze wordt bepaald volgens R. Koster c.s. (Ped. Proc., _18, 412 (1959)), waarbij muizen hét dinatriumzout oraal krijgen toegediend en een uur later intraperitoneaal 7908988 -4- % 0,5 ml 0,5%tig azijnzuur, waarna het aantal kronkelingen in de volgende 10 min wordt bepaald. De analgetische werking wordt gevonden als het percentagevermindering van het aantal kronkelingen ten opzichte van de controlegroep. De resulta- 5 ten zijn in tabel A te vinden.

Deze wordt bepaald volgens Winder c.s. (J. Pharm. exp. Therap., 133, 117 (1961)), waarbij hei: temperatuur-verloop van de ratten van de controle- en behandelde groep 10 4 uren wordt gevolgd, na een onderhuidse toediening van een koortverwekkend middel (op basis van biergist).
De werkingen worden nagegaan bij orale, intraperi-toneale en intraveneuse toediening in de in tabel B vermelde doses, in welke tabel ook de resultaten zijn verzameld.

Bij orale toediening bedraagt de DA_g (dosis voor een werking van 50%. 520 mg/lichaamsgewicht.
5 2) Antipyretische werking (bij ratten).
De toegediende dosis bedraagt 1 mmol, alszijnde 306 mg/kg. De gemeten lichaamstemperaturen zijn in de tabellen C en D opgenomen.

Claims (7)
- 2. Verbinding volgens conclusie 1, met het kenmerk, 5 dat het bestaat uit het dinatrium-, glycine-, lycine- of adenosine-zout.
- 3. Werkwijze voor de bereiding van geneeskrachtige verbindingen, met het kenmerk, dat men de verbinding van formule 1 van het formuleblad op een voor analoge verbindin- 10 gen bekende wijze bereid.
- 4. Werkwijze volgens conclusie 3, met het kenmerk, dat het dinatriumzout wordt bereid door de fosforzure ester van p-nitrofenol in aanwezigheid van palladium/koolstof katalytisch wordt gereduceerd tot het p-aminoderivaat, waar- 15 van de aminogroep dan met azijnzuur anhydride wordt geacety-leerd.

Werkwijze volgens conclusie 3, met het kenmerk, dat het glycine-, lycine- of adenosine-zout wordt verkregen door het dinatriumzout met zuur te behandelen ter verschhfi- 20 fing van het vrije zuur, dat dan aan een reaktie wordt onderworpen met een equimolaire hoeveelheid resp. glycine, lycine of adenosine.
- 6. Geneesmiddel met analgetische en antipyretische werking, met het kenmerk, dat het een werkzame hoeveelheid 25 van de verbinding van conclusies 1 of 2 bevat.
- 7. Middel' volgens conclusie 6, in de vorm van j tabletten of in zakjes, oplosbaar in water, voor een orale toediening of in ampullen voor intramusculaire of intra— veneuse toediening.
