Titel: 7-oxo-k-thia-l-aza-bicyclo/3,2,O/heptaanderivaten en hun bereiding.
De uitvinding "betreft 7-oxo-U-thia-l-aza-bicyclo/3 ,2,Ojhe-ü-taanderivaten, him bereiding uit esters van penicillanzuur-l-oxide en de bij dit procédé gevormde tussenprodukten.
In het bijzondér betreft de uitvinding nieuwe verbindingen 5 met de formule 1 van het formuleblad, waarin R waterstof, alkyl met 1-5 koolstofatomen, trichloorethyl, benzyl, p-nitrobenzyl, difenylmethyl, acetoxymethyl, pivalqylcocymethyl, ftalidyl, trinet hylsilyl of een groep met de formule -CHOCOOCHgCH^ ; r"* CHg 10 -CHgOCOR2, -CH20R3, -C00R2, -CEO, -CEgSH, -CE^SR^, -CEgMg of CEgHHCOR2, waarbij R2 alkyl met 1-5 koolstof atomen, aryl, aralkyl, of een heterocyclische 5- of é-ring met een of meer heteroatcmen,

R alkyl met 1-5 koolstof at omen, benzyl, trityl of trialkylsilyl, en R^ een heterocyclische 5- of β-ring met een of meer hetero- t atomen, benzyl, trityl of trialkylsilyl voorstellen, alsmede hun synthese.
Verbindingen met de formule 1, (E+Z-isomeren), die met ciavulanzuur verwant zijn («T. Chem. Soc. Chem. Comm. 266, 197^) vertonen een antibacteriële activiteit en zijn als therapeutics 20 bij het behandelen van infectieziekten van belang. Voor dit doel * kunnen ze parenteraal of oraal als zuren, farmaceutisch geschikte zouten of esters worden toegediend.· farmaceutisch geschikte zouten van verbindingen met de formule 1 vallen dus eveneens binnen het kader van de uitvinding.
25 De verbindingen met de formule 1 kunnen volgens het schema van het formuleblad worden bereid.
In de in dit schema getékende verbindingen hebben de süb-stituenten de volgende betekenis: Eg is alkyl, trichloorethyl, benzyl, p-nitrobenzyl- difenylmethyl, acetoxymethyl, pivaloyloxy-30 methyl, ftalidyl, trimethylsilyl of een groep met de formule 790 84 34 * * i * „ ’ -2- 2 3 -CHiCH^OCOOCgïï^; is een groep met de formule CHgOCOR , CHgOR , COOR^, CHpRlCOR^ of CHpSR^, waarbij R^ alkyl, aryl, aralkyl of * 3 een heterocyclische groep voorstelt, R : alkyl, benzyll tnalkyl- k , . .
silyl of tntyl en R een heterocyclische 5- of o-nng met een of 5 meer heteroatcmen, danwel henzyl, trityl of trialkylsilyl, R,- is alkyl, aryl, aralkyl of een heterocyclische groep en X een elec- 2 tronen-aantrekkende groep, h.v. een met de fomule COOR , CN of CORHg, danwel waterstof, waarbij R de hoven weergegeven'bètekenis bezit. De reactie A cmvat de omzetting van de verbinding met 10 formule 2 met een alleenverbinding met de formule CHX=C=CHR^ of > met een acetyleenverbinding met de formule CX^C+CHgR.j, waarin X en R.j de boven weergegeven betékenis bezitten, door verhitting op 50-120°C in een inert oplosmiddel. Geschikte inerte oplosmiddelen zijn benzeen, tolueen en dimethylformamide.
15 De reactie B anvat een isomerisatie, die in een inert op losmiddel bij kamertemperatuur plaatsvindt. Dichloormethaan is een geschikt inert oplosmiddel.
De reactie C omvat drie op elkaar volgende omzettingen: • reductiec-van. de sulfoxidebinding van de verbinding··^, ozonolyse van 20 beide dubbele bindingen en hydrolyse.
De reactie D omvat de condensatie van de verbinding 8 met een glyoxylzuurester met de formule CH0-C00Rg, waarbij Rg de bevenweergegeven betekenis bezit, en wel bij een temperatuur van 10-100°C De verbinding 10 wordt als mengsel van zijn diastereoisemeren ver-25 kregen.
De reactie E is een chlorering. Geschikte voorwaarden hiervoor zijn de toepassing van thionylchloride als chloreringsmiddel bij aanwezigheid van een base, zoals pyridine, bij -20 tot +2Ó°C.
De verbinding 12 wordt als een mengsel van zijn diastereoisomeren 30 verkregen.
De reactie F is een condensatiereactie tussen de verbinding 12 en trifenylfosfine, die bij een temperatuur van ten hoogste 50°C en bij aanwezigheid van een base wordt uitgevoerd. Gewoonlijk wordt deze reactie bij kamertemperatuur uitgevoerd; pyridine en 35 2,6-lutidine zijn voorkeursbasen.

De reactie G is een ringsluitingsreactie, die eenvoudig o door verhitting van de verbinding 3A op 20-1^0 C in een inert oplosmiddel, zeals tolueen, benzeen of ethylacetaat, en bij aanwezigheid van een katalytische hoeveelheid van een organische base, 5 liefst pyridine, wordt uitgevoerd.
De reactie H is een ozonolyse, die bij -20 tot -T8°C in een oplosmiddel, zoals dichloormethaan, ethylacetaat of tetrahydrofuran wordt uitgevoerd.
De reactie I is een reductie, die met fosfortribremide, 10 liefst tussen 0 en -kO°C en bij voorkeur in dime thy If ormami de wordt uitgevoerd.
De reactie J is een hydrolyse, die gewöonlijk in methanol bij aanwezigheid van silicagel of onder milde basische voorwaarden plaatsvindt.
15 De reactie K cmvat de condensatie van de verbinding 7 met een glyoxylzuurester met de formule CHQ. CQORg, waarin E^ de bovenweergegeven betekenis bezit en wel bij i|Q-lQ0°C. De verbinding 9 wordt daarbij als een mengsel van zijn diastereoisameren verkregen.
De reactie L is een chlorering. Geschikte voorwaarden zijn 20 een toepassing van thionylchloride als chloreringsmiddel bij aanwezigheid van een base, zoals pyridine, en wel bij -20 tot +20°C.
De verbinding 11 wordt daarbij als een mengsel van zijn diastereo-isaneren verkregen.
De reactie M wordt onder de bij reactie F beschreven voor-25 waarden uitgevoerd, waarbij evenwel de temperatuur UC-80°C bedraagt.
De reactie N is een ozonolyse, waarbij de koolstof-fosfor-dubbele binding door een protonisatie met een sterk zuur, zoals trifluorazijnzuur, wordt beschermd, waarna de deprotonisatie aan het 30 eind van de ozonbehandeling onder toepassing van natriumbicarbonaat plaatsvindt. De reactietemperatuur bedraagt liefst -20 tot -78°C.
De reacties 0, P en Q zijn ozonolysereacties, die onder de bij reactie E weergegeven voorwaarden worden uitgevoerd.
Verbindingen met de formule 1, waarin E waterstof en E^ 35 -CHgOH, -CHgSE, of -CH0 voorstellen, worden eenvoudig door 790 S434 9 .. - s*r* - -U 2 3 verwijdering van de verschillende "beschermende groepen R , R en k R , door reductie van de groep -COORg of door een hydrolyse van de groep COORg verkregen.

5 Voorbeeld I

... . . 3 1,5 g methylpenicillinaat-l-oxide werd m 30 cm tolueen op- gelost. Aan deze oplossing werd 2,2 g glutinezuurdimethylester toegevoegd en de verkregen oplossing b uren onder terugvloeikoelen gekookt. Hèt hoofdprodukt kon door kolamchromatografie onder toepassing van benzeen-ethylaeetaat worden gezuiverd, waarbij 1,8 g van het titelprodukt werd verkregen.

k^-vinylthi o- (l-carbcmethoxymethyl-2-methoxycarbonyl) -1- (1-methoxycarbonyl-2-methyl-l-propenyl )-aeetidin-2-on-S-oxide (zie schema 2).
2,8 g U^-vinylthi o-(l-carbamethoxymethyl-2-methoxy carbonyl)- 30 1- (1-methoxyc arbonyl-2-methyl-2-propenyl) -azetidin-2-on-S-oxide werd 3 3 in |0 cm dichloormethaan opgelost en 0,5 cm triëthylamine hieraan toegevoegd. Deze oplossing werd een nacht bij kamertemperatuur bewaard en daarna in vacuo tweemaal uit tetrachloorkoolstof ingedampt. Het residu bestond uit het zuivere titelprodukt 35 en wel in kwantitatieve opbrengst.

5 1 f-vinylthio-(l-carbamethoxymethyl-2-inethaxycarbonyl)-l-methoxy- oxaloyl-azetidin-2-on-S-oxyde (zie schema 3).
1,8 g l£-vinylthio-(l-carbomethoxymethyl-2-methaxycarbonyl) -1-(1-methoxy earbonyl-2-methyl-l-propenyl)-azetidin-2-on-S=oxide verd in 100 em^ methyleenchloride opgelost en op -78°C gekoeld.
10 Een strocm ozon in zuurstof verd door deze oplossing geblazen tot een blauvkleuring optrad. Het verkregen ozonide verd door schudden met een waterige NagS^O^-oplossing geschud; de organische fase verd met natriumsulfaat gedroogd en in vacuo ingedampt, waarbij 1,1 g titelprodukt verd verkregen.

Een oplossing van 1,5 g lf-virylthio-(1-carbcmethoxymethyl- 2-me thoxy carbonyl )-l-methoxyoxaloyl-azetidin-2-on-S-oxide in 10 cm~ vatervrij dimethylformamide verd op -20°C gekoeld en

0,6 cm fosfortribrcmide onder roeren toegevoegd. Na 10 minuten 25 verd het mengsel in ethylacetaat uitgegoten en tweemaal met water gewassen. De organische laag verd met natriumsulfaat gedroogd en in vacuo ingedampt, waarbij 1,1 g titelprodukt verd verkregen.

Een oplossing van 1,6 g 1/Winy Ithi o-(l-carbame thoxyme thyl- 2 35 2-methoxycarbonyl)-l-methoxyoxaloyl-azetidin-2-on in 50 cm 700 84 34 -6- methanol werd een nacht onder goed roeren tezamen met 5 g silicagel "bewaard. Ha af filtreren van het onoplosbare materiaal werd de methanoloplossing ingedampt en het residu over silicagel gechro-matografeerd, waarbij met benzeen-ethylacetaat werd geëlueerd; 5 daarbij werd 0,8 g titelprodukt verkregen.

Een oplossing van 0,250 g tf-vinylthio-( 1-carbcmethoxy-

methyl-2-methaxycarbanyl)-azetidin-2-on in 30 cm dichloormethaan werd op -J8°C gekoeld en een stroom ozon in zuurstof door deze oplossing geblazen tot een blauvkleuring optrad. Het mengsel werd 15 met een waterige NagSgO^-oplossing geschud en met natriumsulfaat gedroogd. Aldus werd 0,150 g titelprodukt verkregen.

1,8 cur methylglyoxylaat (vers bereid door een ozonolyse *3 van dimetbylfumaraat) werd in 50 cm benzeen opgelost en deze oplossing 30 minuten met een lëan-Stark-apparatuur gekookt.
25 Na afkoelen op 50°C werd 0,8 g k -vinylthio-(l-carbamethoxy- methyl-2-methoxycarbonyl)-azetidin-2-on toegevoegd en de verkregen oplossing wederom 3 uur gekookt. Na kort chromatograferen over silicagel ter zuivering van het produkt van overmaat methylglyoxylaat werd 1,7 g titelprodukt in de vom van een mengsel 30 van de diastereoiscmeren verkregen.

Een oplossing van 0,^50 g vers bereid methylglyoxylaat in 30 ar' benzeen werd met een Dean-Stark-apparatuur 30 minuten ge-5 kookt. 0,200 g ^-methoxycarbonylacetyltbio-azetidin-2-on werd toegevoegd en het mengsel 3 uren qp de kooktemperatuur gehouden.
Na een korte kolcmchrcmatografie ter zuivering van overmaat methylglyoxylaat werd 0,110 g titelprodukt verkregen.

l-trifenylfosforanylideenmethyl)-azetidin-2-on werd in 3 cm/ tolueen opgelost en 30 minuten op 120°C verhit. Het tot een ring 15 gesloten produkt methyl- (23,5R)-E-3-methoxycarhonylmethyleen-7-oxo-H-thia-l-azahicyclop, 2p7heptaani2-carbGxylaat werd van trifenylfosfinoxyde door een korte kolcmchrcmatografie gezuiverd. Aldus werd 0,036 g van de eindverbinding als een mengsel van de E+Z-isomeren verkregen. De zuivering van de hoofdecmponent (E-20 iscmeer) vond dunnelaagchramatografisch plaats·.

7· Werkwijze voor het bereiden van verbindingen met de formule 1, waarin de substituenten de in conclusie 1 weergegeven betekenis bezitten, met het kenmerk, dat men een verbinding met de formule 2, waarin de substituenten de in conclusie 6 weergegeven betekenissen bezitten, met een verbinding met de formule CHX = C = CHR^ ^5 0f een verbinding met de formule CX = C - CH^R^, waarin X en R^ de in conclusie 6 weergegeven betekenis bezitten,, bij 50-120°C in een inert oplosmiddel, liefst benzeen, tolueen of dimethyl-formamide, omzet; de daarbij verkregen verbinding met de formule 3, waarin de substituenten de in conclusie 6 weergegeven betekenis bezitten, m een inert oplosmiddel, liefst dichloormethaan, tot een verbinding met de formule k, waarin de substituenten de weergegeven betekenissen bezitten, isomeriseert’ de verbinding, met de formule 6 bij -20 tot -78°C in een geschikt oplosmiddel, liefst dichloormethaan, ethylacetaat of tetrahydrofuran, ozoniseert; 25 de daarbij verkregen verbinding met de formule 5, waarin de substituenten de in conclusie 6 weergegeven betékenissen bezitten, met PBr^ bij 0 tot -kO°C in een geschikt oplosmiddel, liefst dimethylformamide reduceert; de daarbij verkregen verbinding met de formule 6, waarin de substituenten de reeds genoemde betekenissen bezitten, onder milde basische voorwaarden of bij aanwezigheid van silicagel tot een verbinding met de formule 7, waarin de substituenten de in conclusie 6 weergegeven betekenis bezitten, hydrolyseert; deze verbinding met de formule 7 met een verbinding met de formule CHO.COORg, waarin Rg de boven weergegeven betékenis 35 bezit, bij k0-100°C tot een verbinding met de formule 9, waarin 79 0 8 4 S4 -15- de substituent en de Doren weergegeven "betékenis "bezitten, condenseert; de verbinding met de formule 9 met een geschikt chlorerings-middel, liefst thionylchioride, "bij aanwezigheid van een "base, liefst pyridine, "bij -20 tot +20°C tot een verbinding met de formule :5 11» waarin de substituenten de "boven weergegeven "betekenis "bezitten, chloreert; deze verbinding met de formule 11 bij -20 tot -j8°C tot een verbinding met de formule 12, waarin en Eg de boven weergegeven betekenis bezitten, ozoniseert; deze verbinding met de formule 12 met trifenylfosfin bij een temperatuur van ten hoog-10 ste 50°C, liefst bij kamertemperatuur, bij aanwezigheid van een base, liefst pyridine of 2,6-lutidine, condenseert; de daarbij verkregen verbinding met de formule lh, waarin de substituenten de boven weergegeven betekenis bezitten, bij 20-1^0°C bij aanwezigheid van een organische base, liefst pyridine, tot een ring 15 sluit en, indien nodig, de schermgroepen verwijdert en de verkregen verbinding tot een verbinding met de formule 1 hydrolyseert.
8. Werkwijze voor het bereiden van verbindingen met de formule 1, waarin de substituenten de in conclusie 1 weergegeven betekenis bezitten, met het kenmerk, dat men een verbinding met de formule 2, 20 waarin de substituenten de boven weergegeven betekenis bezitten, met een verbinding met de algemene formule CHX = C = CER^ of een verbinding met de formule CX = C - CHgR^, in welke formules X en R.J de boven weergegeven betékenis bezitten, big een temperatuur van 50-120°C in een inert oplosmiddel, liefst benzeen, tolueen 2p of dimethylformamide, cmzet; de daarbij verkregen verbinding met de formule 3 in een inert oplosmiddel, liefst dichloormethaan, tot een verbinding met de formule U, waarin de substituenten de boven weergegeven betekenis bezitten, isomeriseert; deze verbinding met de formule ^ bij -20 tot -T8°C in een geschikt oplos-30 middel, liefst dichloormethaan, ethylacetaat of tetrahydrofuran, ozoniseert; de verkregen verbinding met de formule 5 met PBr^ bij 0 tot -iOC in een geschikt oplosmiddel, liefst dimethylformamide reduceert; de daarbij verkregen verbinding met de formule 6 onder milde basische voorwaarden of bij aanwezigheid van silica-35 gel tot een verbinding met de formule T hydrolyseert; deze ver- 799 84 54 -l6- "binding met de formule T met een verbinding met de formule CHO.COORg, bij b0-100°C tot een verbinding met de formule 9 condenseert! deze verbinding met de formule 9 "bij -20 tot -78°C tot een verbinding met de formule 10 ozoniseert; de verbinding met de formule 10 met 5 een geschikt chloreringsmiddel, liefst thionylchloride, bij aanwezigheid van een base, liefst pyridine, bij -20 tot +20°C bot een verbinding met de formule 12, waarin de substituenten de in conclusie 7 weergegeven betekenis bezitten, chloreert; deze verbinding met de formule 12 met trifenylfosfin bij een temperatuur 10 van ten hoogste 50°C, liefst bij kamertemperatuur, bij aanwezigheid van een base, liefst pyridine of 2,6-lutidine, condenseert; de daarbij verkregen verbinding met de formule 1^ bij 20-1^0°C bij aanwezigheid van een organische base, liefst pyridine, tot een ring sluit en indien nodig de scheurgroepen verwijdert en 15 de verkregen verbinding tot een verbinding met" de formule 1 hy-drolyseert.

Werkwijze voor het bereiden van verbindingen met de 15 foimule 1, waarin de substituent en de in conclusie 1 weergegeven betekenissen bezitten, met het kenmerk, dat men een verbinding met de algemene formule 2, waarin de substituenten de in conclusie 6 weergegeven betékenis bezitten, met een verbinding met de formule CSX = C = CEF^ of een verbinding met de formule 20 CX = C - CHg^i > waarin en X de in conclusie 6 weergegeven betekenis bezitten,bij 50-120°C in een inert oplosmiddel, liefst benzeen, tolueen of dimethylformamide amzet; de daarbij verkregen verbinding met de formule 3 in een inert oplosmiddel, liefst dichloormethaan, tot een verbinding met de formule ^ isomeriseert; 25 de verbinding met de formule U met PEr^ "bij 0 tot -k0°C in een geschikt oplosmiddel, liefst dimethylformamide, reduceert; de daarbij verkregen verbinding bij -20 tot -78°C in een geschikt oplosmiddel, liefst dichloormethaan, ethylacetaat of tetrahydro-furan ozoniseert; de daarbij verkregen verbinding onder milde 30 basische voorwaarden of bij aanwezigheid van silicagel tot een verbinding met de formule 8 hydrolyseert; deze verbinding met de formule 8 met een verbinding met de formule CHO.COORg bij U0-100°C tot een verbinding met de formule 10 condenseert; de verbinding met de formule 10 met een geschikt chloreringsmiddel, 35 liefst thionylehloride, bij aanwezigheid van een base, liefst 790 14 34 -18- pyridine, tij -20 tot +20°C tot een vertinding met de formule 12 chloreert; de verbinding met de formule 12 met trifenylfosfin hij een temperatuur van ten hoogste 50°C, liefst hij kamertemperatuur, hij aanwezigheid van een hase, liefst pyridine of 2,6-5 lutidine, condenseert; de daarbij verkregen verbinding met de formule 1¾ bij 20-1^0°C bij aanwezigheid van een organische base, liefst pyridine, tot een ring sluit en, indien nodig, de scherm-groepen verwijdert en de verkregen verbinding tot een verbinding met de formule 1 hydrolyseert.
11. Geneesmiddelpreparaat, met het kenmerk, dat dit als actieve stof een verbinding met de formule 1 of een farmaceutisch geschikt zout hiervan al of niet gemengd met een farmaceutisch geschikt verdunningsmiddel of drager bevat.
12.
