Titel: Difenylbutyl-piperazinecarboxamiden.
De uitvinding heeft betrekking op een nieuwe groep van U',ΙΓ1,2,3,^,556-gesubstitueerde 1-piperazinecarboxami-den en carbothioamiden, zuuradditiezouten daarvan, farmaceutische preparaten die deze verbindingen bevatten alsmede werkwijzen 5 ter bereiding en toepassing daarvan.
De·meeste geneesmiddelen die momenteel worden toegepast voor de behandeling van mentale stoornissen behoren* tot de groep van antidepressiva, anxiolytiea en de belangrijkste tranquillizers, d.w.z. neuroleptica. Al deze genoemde geneesmid-10 delen blijken geschikt te zijn voor de behandeling van mentale storingen, maar zij hebben tevens aanmerkelijke nadelen. Aldus is de doelmatigheid van de antidepressiva slechts ongeveer 60% en veroorzaken hun anticholinergische werking alsmede hun cardiovasculaire neveneffecten belangrijke nadelen. Onder de anxiolytiea 15 werden niet-giftige verbindingen aangetroffen maar die blijken allen een verslavende werking te bezitten. De neuroleptica zijn geschikt ter onderdrukking van symptomen van schizofrenie maar het grootste nadeel daarvan is dat zij verschillende extrapyrami-dale neveneffecten induceren, waarvan sommigen omkeerbaar zijn, 20 d.w.z. tardieve dyskinesie. 2ij induceren tevens mentale neveneffecten, zoals verminderde emotionele gevoelens, in het bijzonder bij niet-psychotische patiënten. Er moet aan herinnerd worden dat alle neuroleptica werken volgens een niet-specifieke interferentie met de. dopaminergische overdracht in de hersenen, hoewel nog 25 nooit is bewezen dat de anti-psychotische werking daarvan afkomstig is uit de antidopaminergische, d.w.z. de dopaminereceptor-biokkerende activiteit. Dopaminergische overdracht in de hersenen is betrokken bij motorische functies, maar tevens bij gedrags-functies, alsmede endocrine en autonome functies. Derhalve kan 79077 41 é * 2 de tamelijk niet-specifieke antidopaminergische activiteit vele soorten neveneffecten veroorzaken. Er is aldus veel "bewijsmateriaal aanwezig dat er op wijst dat de extra pyramidale neveneffecten als- bovenvermeld afkomstig zijn van deze blokkering, 5 De meeste neuroleptica veroorzaken tevens een sterke kalmering. In vele gevallen is het derhalve moeilijk te beslissen of bij voorbeeld de anti-aggressieve eigenschappen van neuroleptica veroorzaakt worden door gebrek aan coördinatie van de spieractiviteit (ataxie, immobiliteit of catalepsie) of door 10 een werkelijke anti-aggressieve eigenschap, die inherent is aan het geneesmiddel.
De nieuwe 1-piperazine-carboxamiden en carbo-thioamiden volgens de uitvinding kunnen structuursgewijze worden voorgesteld door de algemene formule 1 van het formuleblad, 15 waarin de R^- en Rg-groepen onafhankelijk worden gekozen uit waterstof, rechte of vertakte alkylgroepen met 1 - 3 koolstofatomen, cycloalkylgroepen met 3-8 koolstofatomen, aralkylgroepen met 7-9 koolstofatomen of alkenylgroepen met 2-10 koolstofatomen, fenyl al of niet gesubstitueerd met 1 - 3 substituent en geko-20 zen uit halogeen, met inbegrip van F, Cl en Br, lagere alkyl met 1 - 5 koolstof at omen, lagere alkoxy met 1 - 5 koolstof at omen, amine al of niet gesubstitueerd met 1 of 2 lagere alkylgroepen met 1 - 5 koolstofat omen, -CF^ en -CH" groepen, de R^-, R^-, R^- en Rg-groepen onafhankelijk worden gekozen uit waterstof, lagere 25 alkylgroepen met 1 - 3 koolstof at omen en fenyl, een groep is gekozen uit waterstof, halogeen met inbegrip van F, Cl en Br, lagere alkoxygroepen met 1 - 3 koolstofatomen en -CF^ groepen, terwijl X 0 of S is, alsmede de farmaceutisch aanvaardbare zuur-additiezouten daarvan.
30 De verbindingen volgens formule 1 hebben een nieuw farmacologisch profiel dat niet in een andere bekende verbinding eerder is aangetroffen. Bij toediening aan mannetjes muizen, die aggressief zijn gemaakt door isolering, veroorzaken zij een krachtige en langdurende remming van deze aggressie zonder enige 790 7 7 41 *· 4 3 ataxie of catalepsie te veroorzaken. Verder hebben de verbindingen volgens formule 1 bij muizen een goed verminderend effect op opwinding en slapeloosheid gedurende anaesthesie.
In tegenstelling tot de neuroleptica remmen de 5 nieuwe verbindingen volgens formule 1 de door apomorfine of amfetamine geïnduceerde stereotypie of hypermotiliteit niet, hetgeen betekent dat zij geen krachtige dopaminereceptorantago-nisten zijn. Bij hogere doses verlagen de nieuwe verbindingen het explöratieve gedrag en hebben invloed op de ziekte vermijdende 10 respons. Deze en andere bevindingen suggereren dat de verbindingen antipsychotische eigenschappen bezitten. In kleine doses induceren de verbindingen gedragseffecten in apen die soortgelijk zijn aan die welke worden waargenomen na antidepressiva. Dit betekent dat de verbindingen geschikt zullen zijn voor de behandeling van 15 depressies. De nieuwe verbindingen tonen verder waardevolle anal-getische eigenschappen, die door naloxon niet worden omgekeerd.
In tegenstelling tot morfine blijken de verbindingen bij chronische toediening geen fysische afhankelijkheid te veroorzaken.
De anti-onrust en de beschermende invloed tegen 20 opgewekte stress getoond door de verbindingen volgens formule 1 is van waarde voor de behandeling van psychomatische storingen en van ulcus bij mensen. Verder onderstrepen het anti-ontstekings-effect van de verbindingen, het effect op het immunologische systeem alsmede de psychotropische effecten hun toepassing voor oude-25 re en depressieve patiënten.
Een uniek bindingsprofiel van de verbindingen aan specifieke bindingsplaatsen in de membranen van verschillend weefsel kan worden benut voor het moduleren of van ionen-afhanke-lijke processen, secretie, metabolisme en aggregatie van cellen 30 alsmede de vascularisatie van verschillende weefsels. Sommige van deze invloeden komen tot uiting in een hypothermia che respons.
Hierop gebaseerd is geconcludeerd dat de verbindingen volgens formule 1 geschikt zullen zijn voor de behandeling van mentale storingen bij mensen alsmede bij dieren, hoewel 79077 41 „ * ï k hun farmacologische activiteiten verschillen van die van de antidepressiva, anxiolytica en de neuroleptica die nu hij klinische behandelingen worden toegepast. Het werkingsprofiel van de verbindingen volgens formule 1 suggereert invloed op de limbische, 5 hypothalamische en hypofyse gebieden van de hersenen.
De nieuwe verbindingen blijken geschikt te zijn voor de behandeling van aggressief gedrag bij dieren, in het bijzonder varkens, voor.het bevorderen zonder uitbarsting van aggres-sie van de ontwikkeling van een natuurlijke hierarchy in een 10 groep dieren en in het kalmeren van onrustige en gespannen dieren.
Volgens de uitvinding kunnen de nieuwe verbindingen met de algemene formule 1 worden bereid door de volgende'reacties.

Door reactie tussen een isocyanaat - UCO en 10 een 1-benzylpiperazine volgens formule 5 in ether, en dergelijke ter vorming van een verbinding volgens formule 6 van het formuleblad.
Alkylering van het anion gevormd uit het carboxamide van formule 6 door eerst behandelen met een geschikte 15 sterke base, b.v. lithiumdiisopropylamiöe, in tetrahydrofuran en daarna met een alkylhalogenide R0-X, waarin Z wordt gekozen uit Br en I leidt tot de ΙΓ ,ΙΓ-digealkyleerde verbindingen volgens formule 7. De verbindingen volgens formule 6 en 7 worden gehydro-geneerd over een edele metaalkatalysator waarbij de verbindingen 20 volgens formule 2 wordt verkregen.
Reactie B (b)
Door reactie tussen een earbamoylchloride, R^-StEgi-COCl en een 1-benzylpiperazine volgens formule 5 in chloroform en dergelijke wordt een verbinding volgens formule 7 25 van het formuleblad verkregen. De benzylering over een edele metaalkatalysator geeft de verbinding volgens formule 2.
In de reactie A (a) wordt de verbinding volgens formule 2 in reactie gebracht met de verbinding volgens formule 3 (gesynthetiseerd volgens het Franse octrooischrift M 3&95) in een 30 geschikt oplosmiddel, b.v. lagere alkanol, zoals methanol, ethanol, n-butanol en dergelijke, in aanwezigheid van een zuuraccep-tor, d.w.z. een passende base, b.v. een alkalimetaalcarbonaat of bicarbonaat, die benut kan werden voor het binden van het zuur dat gedurende het verloop van de reactie vrij wordt gemaakt, 79077 41 - k * 6 waarbij de verbinding volgens formule 1 wordt verkregen. Verhoogde temperaturen kunnen worden toegepast voor het bevorderen van de reactiesnelheid.
In de reactie A (b) wordt de verbinding volgens 5 formule k (gesynthetiseerd volgens de Nederlandse octrooiaanvrage 6507312) in reactie gebracht met een isocyanaat (of een isothiocyanaat) - NCX of een carbamoylchloride (of een thio- carbamoylchloride), R^ - Ν^)0Χ Cl, in een geschikt oplosmiddel, b.v. diëthylether, chloroform, tolueen en dergelijke, waarbij 10 de verbinding volgens formule 1 wordt verkregen. Het mengsel wordt over een ruim traject van temperaturen van ongeveer 10 tot 110°C in reactie gebracht, hoewel het mogelijk is temperaturen boven en beneden dit traject toe te passen.
In de reactie A (c) wordt de verbinding volgens 15 formule 1+ in reactie gebracht met een alkalimetaalcyanaat in een waterig medium dat azijnzuur bevat onder vorming van de verbinding volgens formule 1.
In de reactie A (d) wordt de verbinding van formule h in reactie gebracht met een fenylcarbamaat met de for-20 mule Ph-0-C0NHR^ in een geschikt oplosmiddel, bij voorbeeld een aromatische koolwaterstof, d.w.z. benzeen, tolueen, xyleen en dergelijke in aanwezigheid van een geschikte base, bij voorbeeld alkalimetaalcarbonaat ter vorming van de verbinding volgens formule 1. Er wordt een verhoogde temperatuur toegepast om de reac-25 tiesnelheid te bevorderen.
Gebruikelijke neuroleptica zijn krachtige remmers van door amfetamine geïnduceerde gedragsstereotypie. Men bepaalt in Sprague Dawley wijfjes ratten de geschiktheid van sub-cutane toediening van de verbindingen voor het remmen van de ge-30 dwongen knaag- en kauwresponsies op een subcutane dosis van 10 mg/kg amfetamine dat · een half uur na de te beproeven verbinding werd toegediend. De nieuwe verbindingen volgens formule 1 zijn in deze proef zwakke antagonisten.
Mannetjesmuizen onderworpen aan langdurige iso- 790 77 41 A -* 7 lering ontwikkelen aggressief gedrag ten opzichte wan elkaar wanneer zij hij elkaar worden gezet. De proef op het aggressie-ve gedrag opgewekt door isolering werd toegepast om de kalmerende activiteit van de verbindingen volgens tabel A te bepalen (S.
5 Garattini en Ξ.Β. Sigg, Aggressive Behaviour, 1969)· Proeven werden 60 min na een subcutane toediening van het geneesmiddel uitgevoerd. De nieuwe verbindingen van formule 1 zijn in deze proef krachtige middelen.
Een ander model dat voor de aggressie werd toe-10 gepast, d.w.z. het doden van muizen (muricide) door ratten, is gebaseerd op de bestaande aggressie tussen de soorten (Karli, P.,
Behaviour 10, 81 (1956). Bij Long Evans mannetjesratten is het doden van muizen nog steeds een spontaan gedrag. In de proeven werden Long Evans mannetjesratten geplaatst in individuele kooien, 15 waarbij door voorafgaande proeven was vastgesteld dat deze binnen 5 min na confrontatie muizen doden. De proeven werden 60 min na de subcutane toediening uitgevoerd. De nieuwe verbindingen bleken zeer krachtig te zijn in het blokkeren van dit type aggressie.
20 Eet effect van de geneesmiddelen op de motorieke coördinatie werd onderzocht door de prestatie van muizen op de "rotarod". De roterende staafproef is een bekende proef voor ataxie in muizen. De verbindingen gerangschikt in tabel A werden subcutaan toegediend. De ED^-waarde is 50% van de doses die 25 bij de muis een uur na de toediening van het geneesmiddel ataxie opwekt. De nieuwe verbindingen volgens formule 1 produceren geen ataxie bij doses gelijk of kleiner dan 20 mg/kg.
De verbindingen volgens de uitvinding blijken krachtige analgetische eigenschappen te bezitten. Het analgeti-30 sche effect van de verbindingen werd geëvalueerd met het stuip-trekkingssyndroom dat door analgetica kan worden beëindigd. De ED-Q-waarde is de subcutane dosis waardoor 50% van de muizen bevrijd wordt van de stuiptrekkingssyndroom-symptomen.
De EDj-Q-waarden uitgedrukt in milligram per 79077 41 8 kilogram lichaamsgewicht in de anti-amfetamine-proef, in aggres-sie remmingsstof, in de spontane muisdodingsproef, in de roterende staafproef en in de stuiptrekkingsproef vorden voorgesteld in tabel A.

De bases volgens formule 1 zijn omzetbaar in therapeutisch actieve niet-giftige zuuradditiezouten door behandeling met een passend zuur, b.v. een anorganisch zuur, zoals halogeenvaterstofzuur, in het bijzonder chloorwaterstofzuur 5 en broomvaterstofzuur, of zwavelzuur, salpeterzuur, fosforzuur en dergelijke, of een organisch zuur zoals azijnzuur, propion-zuur, glycolzuur, melkzuur, malonzuur, barnsteenzuur, fumaar-zuur, wijnsteenzuur, citroenzuur en dergelijke. Omgekeerd kunnen de zouten door behandeling met alkali in de vrije basevorm wor-10 den omgezet.
Effectieve hoeveelheden van elk van de voornoemde farmacologisch actieve verbindingen volgens formule 1 kunnen voor therapeutische doeleinden aan een mens of dier worden toegediend volgens de gebruikelijke toedieningsmogelijkheden en 15 in gebruikelijke vormen, zoals oraal in oplossingen, emulsies, suspensies, pillen, tabletten en capsules, in farmaceutisch aanvaardbare dragers en parenteraal in de vorm van steriele oplossingen. Voor de parenterale toediening van de actieve stof kan de drager of het excipient een steriele, parenteraal 20 aanvaardbare vloeistof zijn, b.v. water of een parenteraal aanvaardbare olie, b.v. aardnootolie.
Hoewel zeer kleine hoeveelheden van de actieve materialen van de uitvinding effectief zijn bij een lichte therapie of in gevallen van toediening aan subjecten die een be-25 trekkelijk laag lichaamsgewicht hebben, zijn de eenheidsdose-ringen gewoonlijk 2 mg tot bij voorkeur 25 — 50 of 100 mg of zelfs hoger, afhankelijk van de toestand, de leeftijd en het gewicht van de patiënt alsmede de respons op de toediening van het medicijn.
30 De eenheidsdosis kan 0,1 - 200 mg vormen, maar is bij voorkeur 10 - 1.5 mg. Dagelijkse doseringen dienen bij voorkeur te liggen in het gebied van 10 tot 200 mg. De exacte individuele doseringen alsmede de dagelijkse doseringen zullen uiteraard worden bepaald volgens standaard medicinale principes 790 77 41 11 onder het toezicht van een dokter of dierenarts.

Voorbeeld I
N *-Ethyl-4-benzyl-1-piperazine-carboxamide 5 Aan een oplossing van 13»5 g (0,19 mol) ethyl- isocyanaat in 300 ml ether werd druppelsgewijze bij 25°C gedurende een periode van 30 min 37>0 g (0,21 mol) 1-benzylpiperazine toegevoegd. Het mengsel werd gedurende 2 uur onder terugvloei-koeling gekookt. 250 ml Petroleumether werd toegevoegd en het 10 mengsel tot 50°C gekoeld. De vaste stof die zich uit de oplossing afscheidde werd door filtratie verzameld en gerekristalli-seerd uit tolueen-petroleumether 1:1, waarna men U3 g ΪΓ-ethyl-U-benzyl-1-piperazine-carboxamide als produkt verkreeg. Smeltpunt 100 - 102°C.
15 Voorbeeld II
Nf -Ethyl-H* -methyl-^·-benzyl-1 -piperazinecarboxamide hydrochloride Aan 0,068 mol lithi'umdiïsopropylamide, bereid uit n-butyllithium en diisopropylamine in tetrahydrofuran, werd bij -40°C lks9 g (Ο,ΟβΟ mol) N'-ethyl-Wbenzyl-1 -piperazinecar-20 boxamide in 100 ml tetrahydrofuran gedurende 30 min toegevoegd. Het mengsel werd gedurende 30 min tot 0°C verwarmd. Ha koeling tot -30°C werd 11,6 g (0,082 mol) methyljodide gedurende 20 min toegevoegd. Men liet het mengsel bij kamertemperatuur gedurende de nacht staan. Het mengsel werd op ijs geschonken en geëxtra-25 heerd met ether. De gecombineerde extracten werden gedroogd met natriumsulfaat en de ether door destillatie verwijderd. De rest-olie werd opgelost in ether en het hydrochloride met ethanolisch HC1 neergeslagen. De vaste stof werd verzameld door filtratie en gaf na rekristallisatie uit isopropanol 1^-,0 g N’-ethyl-H’-me-30 thyl-lj—benzyl-1 -piperazine-carboxamide-hydrochloride.
Voorbeeld III
N1,H'-Diëthyl-^-benzyl-l-piperazinecarboxamide
Sen oplossing van 26,h g (0,15'mol) 1-benzyl-piperazine in 70 ml chloroform werd verhit tot de terugloop be- 790 77 41 13 gon, waarna een oplossing van 20,¼ g (0,15 mol) diëthylearbamide-chloride in 80 ml chloroform werd toegevoegd. Het mengsel werd gedurende 2 uur onder terugloop behandeld. Het gekoelde mengsel werd basisch gemaakt met natriumhydroxyde (0,25 mol, 10,0 g in 5 6o ml water) en geëxtraheerd met chloroform. De chloroformextrac-ten werden gedroogd met natriumsulfaat, het oplosmiddel verwijderd en het residu gedestilleerd, waarbij men ^5,¼ g ff' ,fff-di-ethyl-^-benzyl-1 -piperazinecarboxamide verkreeg, kookpunt 1U0 -150°C bij 0,12 mm Hg.
10 Voorbeeld IV
ff*-Ethyl-1-piperazinecarboxamide 22,3 g (0,09 mol) ff*-Ethyl-^-benzyl-1-piperazinecarboxamide opgelost in 370 ml ethanol en aangezuurd met ethanoliseh. HC1 werd behandeld met waterstof over een palladium-15 katalysator in een Parr hydrogenator bij 2,U - 2,8 bar. De katalysator werd door filtratie verwijderd en het oplosmiddel onder verlaagde druk verwijderd. Het residu werd opgelost in 25 ml water en werd basisch gemaakt met 10 g (0,25 mol) natriumhydroxyde in 50 ml water. Het mengsel werd geëxtraheerd met chloroform.
20 De gecombineerde extracten werden gedroogd met natriumsulfaat en geconcentreerd. Het witte produkt werd gerekristalliseerd uit isobutylacetaat en gaf 10,0 g ff1-ethyl-1-piperazinecarboxamide, smeltpunt 81 - 83°C.

Een geroerd mengsel van b3J g (0,03 mol) ff1-ethyl-1-piperazinecarboxamide, 10,1 g (0,036 mol) chloor-1,1-(di-p-fluorfenyl)butaan, 5,0 g natriumbicarbonaat en 10 ml etha-30 nol werd gedurende 60 uur onder terugvloeikoeling verhit. Er werd 50 ml water toegevoegd. Het mengsel werd tweemaal met ether geëxtraheerd. De gecombineerde extracten werden met natriumsulfaat gedroogd en geconcentreerd. Het residu werd opgelost in ethanol-ether en het hydrochloride werd neergeslagen met ethano- 7907741 lit lisch HC1. De vaste stof werd verzameld door filtratie en gere-kristalliseerd uit 2-butanon-isopropanol it : 1 en gaf 6, it gN’-ethyl-it- (it ,it- [ di-p-fluorfenyl) butyl ]-1-piper azinecarboxamide hydrochloride. Smeltpunt 17T - 1T8°C. Infrarood-speet mm nr. 1.
5 Voorbeeld VI
IT * -Methyl-it- [ it, it- (di-p-fluorfenyl)butyl ] -1 -piperazinecarboxa-Tni a s Aan een oplossing van 20,0 g (0,06 mol) l-[lt,it-(di-p-fluorfenyl)butyl]-piperazine in 150 ml ether -werd druppelsgewijze gedurende een periode van 30 min it,5 g (0,079 mol) 10 methylisocyanaat in 10 ml ether toegevoegd. Men liet het mengsel bij kamertemperatuur staan. Na kort staan verscheen een wit kristallijn neerslag dat door filtratie werd verzameld en gere-kristalliseerd uit ethanol-ether, waarna 21,0 g N’-methyl-it-[ it, it-(di-p-fluorfenyl)butyl]-1-piperazine-carboxamide werd verkregen.

N * -Fenyl-it- [ it, U- (di-p-f luorf enyl )butyl ] -1 -piper azinec arboxami de-hydrochloride
Aan een oplossing van 5,0 g (0,015 mol) 1—[it,it— 20 (di-p-fluorfenyl)butyl]-piperazine in 30 ml ether werd druppelsgewijze 2,0 g (0,017 mol) fenylisocyanaat in 2 ml ether toege-voegd. Het mengsel werd gedurende een uur onder terugloop behandeld. Na gedurende de nacht staan werd het mengsel aangezuurd met ethanolisch HC1. De vaste neerslag werd door filtratie ver-25 zameld en gerekristalliseerd uit ethanol waarna men 5,6 g van de titelverbinding verkreeg. Smeltpunt 202 - 20it°C.

Trans-2,5-dimethyl-N * -methyl-it- [it, it- (di-p-fluorfenyl )butyl ]—1 -piperazinecarboxamide-oxalaat 30 Aan een oplossing van 5,0 g (0,01^ mol) trans- 2,5-dimethyl-1 - [ it, it- (di-p-fluorfenyl )butyl ] -piperazine in 25 ml ether werd druppelsgewijze een oplossing van 0,9 g (0,016 mol) methylisocyanaat in 5 ml ether toegevoegd. Na gedurende 30 min roeren werd het mengsel gedurende 2 uur onder terugloop behandeld.

De ether -werd onder verlaagde druk verwijderd. Een analytisch monster werd van het olie-aehtige residu geïsoleerd door prepara-tieve-dunnelaag-chromatografie (tic). Preparatieve tic werd uitgevoerd met platen die bedekt waren met een 2,0 mm dikke laag 5 silicagel (Merck eluerri: vas chloroform-diëthylamine- methanol (1J : 2 : 1). De zone van silicagel die het produkt bevatte werd gedetecteerd door UV-licht, afgeschrapt en geëlueerd met ether. R^-waarde 0,67. Infrarood-spectrum nummer 3.
Het olie-achtige residu werd opgelost in 10 ml 10 2-butanon en 2,5 g (0,028 mol) oxaalzuur in 30 ml 2-butanon werd toegevoegd. De neergeslagen vaste stof werd door filtratie verzameld en gerekristalliseerd uit isopropanol en gaf 4,0 g van een kristallijn complex tussen trans-2,5-dimethyl-U'-methyl-4-[4,4-(di-p-fluorfenyl)butyl] -1 -piperazine-carboxami.de en oxaalzuur,
15 dat ongeveer 1,5 mol oxaalzuur per mol vrije base bevatte, alsmede een bepaalde hoeveelheid isopropanol, die alleen door extreme middelen kon worden verwijderd. Smeltpunt 153 - 155°C. Voorbeeld IX
4-[4,4- (di-p-Fluorfenyl)butyl]-1-niperazineearboxamide-hydro-20 ehloride
Aan een oplossing van 5,0 g (0,015 mol) 1—[4,4— (di-p-fluorfenyl)butyl]-piperazine in 25 ml ijsazijnzuur werd druppelsgewijze 1,6 g (0,02 mol) kaliumcyanaat in 10 ml water toegevoegd. Men liet het mengsel gedurende de nacht bij kamer-25 temperatuur staan. Het gekoelde mengsel werd verdund met 75 ml water en basisch gemaakt met 5H natriumhydroxyde. De vaste stof die neersloeg werd verzameld door filtratie, gewassen met water en gedroogd. De verkregen vaste stof werd opgelost in ethanol en de oplossing behandeld met een geringe overmaat ethanolisch HC1.

ff' -Methyl-**-Γ ** Λ-(di-p-fluorfenyl)butyl1-1-piperazinecarbothio-amide-hydrochloride
Aan een oplossing van 3,3 g (0,01 mol) 1-[**,*+-,5 (di-p-fluorfenyl)butyl]-piperazine in 25 ml ether -werd druppels gewijze een oplossing van 0,75 g (0,01 mol) methylisothiocyanaat in 5 ml ether toegevoegd. Men liet het mengsel onder roeren gedurende de nacht staan. De ether werd onder verlaagde druk verwijderd. Het olieachtige residu werd opgelost in ethanol en om-10 gezet in het zout door toevoeging van ethanolisch HC1. Ether werd toegevoegd en de vaste stof die neersloeg werd verzameld door filtratie. Het zout werd gerekristalliseerd uit methanol en gaf 3,5 g van de titelverbinding.
Smeltpunt 21U - 216°C. Infrarood spectrum nummer b.

Een geroerd mengsel van 9,9 g (0,03 mol) 1-[^,^-(di-p-5 fluorfenyl)butyl]piperazine en 5,0 g (0,03 mol) fenyl-N-ethyl- carbamaat, 6,6 g kaliumcarbonaat en 100 ml tolueen werd onder terugyloeikoeling gedurende U5 min gekookt. Het mengsel werd afgefiltreerd en het oplosmiddel verwijderd. De restolie werd opgelost in ethanol-ether en het hydrochloride werd neergeslagen 10 met ethanolisch ÏÏC1. De vaste stof werd door filtratie verzameld en gerekristalliseerd uit 2-furanon-isopropanol h : 1, waarbij 6,8 g van de titelverbinding werd verkregen. Smeltpunt 177 - 178°C.
