FIELD OF TECHNOLOGY The disclosure relates to a method for promoting breeding of larimichthys polyactis by artificial induction of labor, which be- longs to field of agricultural seawater seed breeding.
BACKGROUND Pseudosciaena polyactis Bleeker, also known as small fresh (Xiao Xian), yellow croaker, belongs to Perciformes, Sciaenidae. Pseudosciaena polyactis Bleeker is widely distributed in the East China Sea, the Yellow Sea and the Bohai Sea, and the offshore wa- ters on the west bank of the Korean Peninsula. It is a warm- temperature near-bottom fish. It has plays an important role in China's traditional marine fishery. Pseudosciaena polyactis Bleek- er, Larimichthys crocea, cuttlefish (Sepiella maindroni de Rochebrune), and Trichiurus haumela were once called “Four Sea- food“. However, since the 1970s, the production of Pseudosciaena polyactis Bleeker has been decreasing year by year due to factors such as overfishing and environmental degradation, forcing the price of adult Pseudosciaena polyactis Bleeker to soar. At pre- sent, the market price of chilled Pseudosciaena polyactis Bleeker is more than 80 yuan/kg, and it is in short supply. At the same time, Pseudosciaena polyactis Bleeker features growing fast, espe- cially suitable for artificial breeding. Therefore, artificial in- duction of labor for Pseudosciaena polyactis Bleeker is the prima- ry problem in the development of artificial breeding and cultiva- tion.
SUMMARY In order to solve above technical problems in the prior art, the disclosure provides a method for promoting breeding of larim- ichthys polyactis by artificial induction of labor includes forti- fied culturing of broodstock and injection of exogenous medicines for induction of labor, so as to promote the further development of the gonads of the broodstock, and cause larimichthys polyactis to naturally lay eggs in the indoor cement pit, thereby obtaining high-quality fertilized eggs. The disclosure provides technical support for the implementation of artificial breeding of larimich- thys polyactis.
In order to achieve above purpose, the present invention em- ploys the following technical solution: The method for promoting breeding of larimichthys polyactis by artificial induction of labor includes steps of: step one, indoor fortified culturing of broodstock: selecting one-year-old larimichthys polyactis as a broodstock, with a ratio of male to female of 2:1; step two: artificial induction of labor: a medicine for in- duction of labor is LHRH-A2, which is dissolved and diluted with a medical 6% glucose injection, with an injection dose of 1.25ug/kg for the females and half of the injection dose for the males; starting the injection at 5 to 6 in the evening by one-time injec- tion with an injection dose for induction of labor of 0.3-0.5mL per tail, then covering the entire pit with a black shade cloth to keep the surroundings quiet, so that the broodstock may lay eggs in the cement pit after 24 to 26 hours.
The method for promoting breeding of larimichthys polyactis by artificial induction of labor, wherein in the step one, a start time of the fortified culturing is February to March per year when an indoor water temperature reaches 10 °C or above.
The method for promoting breeding of larimichthys polyactis by artificial induction of labor, wherein conditions for the for- tified culturing are as follows: water temperature of 10-13 ° C, illumination of 1200LX, salinity of 30-35, dissolved oxygen of 5- 8mg/L, daily water exchange of 40%; a fodder made from oyster meat and squid pieces in a ratio of 1:1, which is fed twice a day with a feeding amount accounting for 3% of the weight of the brood- stock.
The method for promoting breeding of larimichthys polyactis by artificial induction of labor, wherein in the step two, a tim- ing for induction of labor is when the water temperature is 14 to
16 °C, the abdomen of the male may be squeezed to shed white se- men, and the female gonad has developed to stage IV as determined from anatomical observation; before induction of labor, the brood- stock is anesthetized with MS-222 of 15-20 ppm.
With above technical solution in the disclosure, the larim- ichthys polyactis can successfully lay eggs indoors, solving the problem of inability of laying eggs naturally during spawning pe- riod for one-year-old broodstock cultured indoors, wherein the yield for induction of labor can reach 75%, and the hatching rate of fertilization rate reaches 80%, which may meet the needs of ar- tificial breeding for the larimichthys polyactis.
DESCRIPTICN OF THE EMBODIMENTS The present invention will be further elaborated hereafter. In the disclosure, the method for promoting breeding of larimichthys polyactis by artificial induction of labor includes steps of:
1. Indoor fortified culturing of broodstock a start time of the fortified culturing is February to March per year when an indoor water temperature reaches 10 °C or above. One-year-old larimichthys polyactis is selected as a broodstock, with a ratio of male to female of 2:1. Method for managing the fortified culturing: water temperature of 10-13 ° C, illumination of 1200LX, salinity of 30-35, dissolved oxygen of 5-8mg/L, daily water exchange of 40%; a fodder made from oyster meat and squid pieces in a ratio of 1:1, which is fed twice a day with a feeding amount accounting for 3% of the weight of the broodstock.
2. Artificial induction of labor Timing for induction of labor: when the water temperature is 14 to 16 °C, the abdomen of the male may be squeezed to shed white semen, and the female gonad has developed to stage IV as deter- mined from anatomical observation; before induction of labor, the broodstock is anesthetized with MS-222 of 15-20 ppm.
Preparation and injection of medicine for induction of labor: a medicine for induction of labor is LHRH-A2, which is dissolved and diluted with a medical 6% glucose injection, with an injection dose of 1.25ug/kg for the females and half of the injection dose for the males; starting the injection at 5 to 6 in the evening by one-time injection with an injection dose for induction of labor of 0.3-0.5mL per tail and a ratio of male to female of 2:1, then covering the entire pit with a black shade cloth to keep the sur- roundings quiet, so that the broodstock may lay eggs in the cement pit after 24 to 26 hours. Embodiments From the beginning of March 2015 to the end of April 2015, a fortified culturing and artificial induction of labor for the broodstock were carried out in a 20 square indoor cement pit in a test site of Xishan Island of Zhejiang Mariculture Research Insti- tute, in which there were 200 broodstock each weighing 100-2009, and a total of 800 mL of high-quality fertilized eggs of larimich- thys polyactis were obtained. The test is specifically carried out as follows:
1. Indoor fortified culturing of broodstock Early in March, an indoor water temperature reaches 10 °C or above. One-year-old larimichthys polyactis is selected for forti- fied culturing, with a ratio of male to female of 2:1. Method for managing the fortified culturing: water temperature of 10-13 ° C, illumination of 1200LX, salinity of 30-35, dissolved oxygen of 5- 8mg/L, daily water exchange of 40%; a fodder made from oyster meat and squid pieces in a ratio of 1:1, which is fed twice a day with a feeding amount accounting for 33 of the weight of the brood- stock.
2. Artificial induction of labor In mid-April, the water temperature reaches 14 °C, the abdo- men of the male may be squeezed to shed white semen, and then the three females are dissected and observed to find that the gonads have developed to stage IV, so the induction of labor may be per-
formed.
Before induction of labor, the broodstock is anesthetized with MS-222 of 15-20 ppm.
Preparation and injection of medicine for induction of labor:
a medicine for induction of labor is LHRH-A2, which is dissolved
5 and diluted with a medical 6% glucose injection, with an injection dose of 1.25ug/kg for the females and half of the injection dose for the males; starting the injection at 5 to 6 in the evening at the origin of pectoral fin by one-time injection with an injection dose for induction of labor of 0.3-0.5mL per tail and a ratio of male to female of 2:1, then covering the entire pit with a black shade cloth to keep the surroundings quiet, so that the broodstock may lay eggs in the cement pit after 24 to 26 hours, finding that the yield for induction of labor is 75% and the fertilization rate is 80%.
