Dialkoxyfosfinoylmethylbenzamideverbindingen en farmaceutische preparaten, die deze bevatten
De uitvinding heeft betrekking op dialkoxyfosfinoylmethylbenzamideverbindingen en op farmaceutische preparaten voor de behandeling van hyperlipidemie, die als actieve ingrediënt één of meer der genoemde S dialkoxyfosfinoylmethylbenzamideverbindingen bevatten.
Dialkoxyfosfinoylmethylbenzamideverbindingen zijn bekend uit JP-A-61/151199. De in dit document beschreven verbindingen vertonen een calcium-antagonistische werking en hebben een effect tegen ontstekingen en profylactische of genezende werking tegen ischemische ziekten zoals angina pectoris, myocardiaal infarct of aritmie en hypertensie.
10 De veibindingen zoals gedefinieerd volgens de onderhavige uitvinding worden niet in JP-A-61/151199 beschreven. Evenmin wordt van de bekende verbindingen vermeld dat ze een werking tegen hyperlipidemie zouden hebben.
De uitvinding heeft ten doel het verschaffen van nieuwe dialkoxyfosfinoylmethylbenzamideverbindirtgen.
Een verder doel van de uitvinding is het verschaffen van een farmaceutisch preparaat voor het behande- 15 len en/of voorkomen van hyperlipidemie, die naast een drager als actief ingrediënt een of meer der genoemde carbonamideverbindingen bevat.
Volgens de uitvinding, worden de nieuwe dialkoxyfosfinoylmethylbenzamidevetbindingen voorgesteld door de algemene formule (I),

45 In de bovengenoemde algemene formule (I), worden de substituenten, gedefinieerd door de symbolen R1, R2 en R3 als volgt toegelicht
Als Ct_e alkyigroep kan een groep zoals de methyl, ethyl, propyl, isopropyl, butyl, tert-butyl, pentyl en hexylgroep als voorbeeld dienen.
Als alkoxygroep, kan een groep zoals de methoxy, ethoxy, propoxy, butoxy, t-butoxy, pentyloxy en 50 hexyloxygroep ais voorbeeld dienen.
Als C1-e alkanoylgroep kan een groep zoals de acetyl, propionyl, butyryl, isobutyryl, valeryl, isovaleryl, pivaloyi en hexanoylgroep als voorbeeld dienen.
Als halogeenatoom kunnen het fluoratoom, chlooratoom, broomatoom en joodatoom als voorbeeld dienen.
55 Als N.N-ditC^ alkyl)aminogroep kan een groep zoals de Ν,Ν-dimethylamino, N,N-diëthytamino, N,N-dipropylamino, Ν,Ν-dihexylamino, N-methyl-N-ethylamino, N-methyl-N-butylamino en N-ethyl-N-hexylaminogroep als voorbeeld dienen.

Als naftylgroep kunnen α-naftyl en β-naftyl als voorbeeld dienen.
Als pyrimidinylgroep, worden 2-pyrimidinyl, 4-pyrimidinyl, 5-pyrimidinyl en 6-pyrimidinylgroep genoemd.
Als N-ftalazinylaminogroep, worden N-(1-ftalazinyl)amino, N-(5-ftalazinyl)amino en de N-(6-ftalazinyl)aminogroep genoemd.

Dialkoxyfosfinoylmethylbenzamideverbindingen weergegeven door de algemene formule (I) bezitten een uitstekende werkzaamheid bij het verlagen van de lipidespiegel, en zijn derhalve bruikbaar als middelen voor het behandelen van hyperlipidemia, en effectief bij het behandelen en voorkomen van verscheidene ziektes (hyperlipidemia) zoals hypercholesterolemia, hypertriglyceridemia, hyperfosfolipidemte, hyperiipacide-30 mie, en dergeiijke.
De werkwijzen ter bereiding van de verbindingen volgens de uitvinding zijn voor analoge verbindingen bekende werkwijzen, zie bijvoorbeeld JP-A-61/151199.

50 In het bovengenoemde reactieschema, bezitten R1, R2 en R3 de bovenvermelde betekenissen.
Volgens het reactieschema, kan een carbonamideverbinding (la) volgens de uitvinding bereid worden door reactie van een carbochloridederivaat (IV) met een amine (lil).
De reactie wordt in het algemeen uitgevoerd in een geschikt oplosmiddel, in aanwezigheid van een ontzuringsmiddel. Met betrekking tot het ontzuringsmiddel, kan elk bekend ontzuringsmiddel, dat geen 55 nadelig effect op de reactie geeft, gebruikt worden en de voorbeelden die de voorkeur genieten omvatten triëthylamine, diëthylaniline, N-methylmorfoline, pyridine en 4-dimethylaminopyridine. Met betrekking tot het oplosmiddel kunnen benzeen, tolueen, xyleen, petroleumether, diêthylether, dimethoxyethaan, tetrahydrofu- 3 194239 ran (THF), 1,4-dioxaan, aceton, methylethylketon, acetofenon, dichtoormethaan, chloroform, tetrachloor* koolstof en 1,2-dichloorethaan worden toegepast
In de bovengenoemde reactie, wordt de verhouding van de hoeveelheid carbonylchloride (IV) ten opzichte van de hoeveelheid amine (III) niet specifiek beperkt en kan er gekozen worden uit een breed 5 traject, en kan er in het algemeen, een equimolaire tot overmatige hoeveelheid van de eerste gebruikt worden ten opzichte van de laatste. Verder kan in het algemeen bij voorkeur een equimolaire hoeveelheid tot geringe overmaat van het bovengenoemde ontzuringsmiddel ten opzichte van het carbonylchloride-derivaat (IV) gebruikt worden. De reactie kan uitgevoerd worden hetzij bij kamertemperatuur hetzij onder koelen bij verwarmen en bij voorkeur bij een temperatuur tussen kamertemperatuur en de refiuxtemperatuur 10 van het oplosmiddel; in het algemeen wordt de reactie in ongeveer 0,5 tot 10 uur voltooid.
De verbindingen, weergegeven door de algemene formule (I) volgens de uitvinding, kunnen gebruikt worden als de actieve ingrediënt in farmaceutische preparaten voor het behandelen en voorkomen van verscheidene ziektes, veroorzaakt door hypertipidemie, zoals hypercholesterolemia, hypertriglycridemie, hyperfosfolipidemie en hypertipacedimie, en wel op grond van hun uitstekende farmacologische werkzaam-15 heid, in het bijzonder het verlagen van de (ipideconcentratie in bloed, zoals het verlagen van de concentraties aan cholesterol, triglyceriden, fosfolipiden en vetzuren in bloed. Verder zijn de verbindingen, weergegeven door de algemene formule (I) volgens de uitvinding, ook bijzonder werkzaam bij het voorkomen en behandelen van arteriosclerose, geïnduceerd door de bovengenoemde ziekten die veroorzaakt worden door hypertipidemie.
20 Naast het bovenstaande worden de verbindingen, weergegeven door de algemene formule (I) volgens de uitvinding gekenmerkt doordat hun farmacologische werkzaamheid op grond van hun lagere toxiciteit voor een bepaalde tijdsperiode verlengd kan worden. De verbindingen volgens de uitvinding zijn derhalve erg geschikt als de actieve ingrediënten voor het behandelen en voorkomen van de verscheidene bovengenoemde ziektes, die door hypertipidemie veroorzaakt worden.
25 Het farmaceutische preparaat voor het behandelen en voorkomen van hypertipidemie volgens de uitvinding omvat, als de essentiële factor ten minste één van de dialkoxyfosfinoylmethylbenza-mideverbindingen met de algemene formule (I). In het algemeen wordt het genoemde farmaceutische preparaat bereid in verschillende farmaceutische toepassingsvormen, afhankelijk van de manier van toediening, door de verbinding te mengen met atoxische farmaceutisch acceptabele dragers die gewoonlijk 30 in farmaceutische preparaten gebruikt worden; elk farmaceutisch preparaat, aldus bereid, wordt toegediend aan een patiënt met hypertipidemie en/of een patiënt met arteriosclerose als behandelingsmiddel, of wordt toegediend ter voorkoming van deze ziekten.
Toedieningseenheidsvormen van farmaceutische preparaten volgens de uitvinding zijn niet specifiek beperkt en kunnen op ruime schaal geselecteerd worden, afhankelijk van de verschillende therapeutische 35 doeleinden. Orale toediening geniet in het bijzonder de voorkeur.
De farmaceutische preparaten kunnen bereid worden via de gebruikelijke werkwijzen.
Injectiepreparaten, en andere vloeistofpreparaten zoals emulsies, suspensies en sirooppreparaten, kunnen op conventionele wijze bereid worden, met voordeel onder toepassing van geschikte oplosmiddelen.
Bij het bereiden van zetpillen, kunnen op conventionele werkwijze excipiênten gebruikt worden zoals 40 polyethyleenglycolen, lanoline en kokosolie.
Doseringen van het gewenste farmaceutische preparaat voor het behandelen en het voorkomen van hypertipidemie volgens de uitvinding kunnen op geschikte wijze geselecteerd worden afhankelijk van de toedieningswijze, de preparaatvorm, leeftijd van de patiënt, lichaamsgewicht van de patiënt, gevoeligheden van de patiënt, omstandigheden van de ziekte, en andere factoren. In het algemeen, kan de hoeveelheid 45 actief ingrediënt, dat elk van deze farmaceutische preparaten bevat, liggen tussen 0,05 tot 80 mg/kg, bij voorkeur tussen 0,1 tot 50 mg/kg lichaamsgewicht per dag.
De onderhavige uitvinding zal nader worden toegelicht aan de hand van de volgende voorbeelden, farmaceutische preparaten en farmacologische testresultaten. De uitvinding is echter hiertoe niet beperkt.
50 Voorbeeld I
1,36 gram (5 mM) 4-diëthoxyfosfinoylmethylbenzoëzuur en 0,409 (5 mM) 3-amino-5-methylisoxazool werden opgelost in 15 ml droge Ν,Ν-dimethylformamide (DMF). Aan deze oplossing werd druppelsgewijs 2 ml droge DMF-oplossing toegevoegd met 1,00 g (5,5 mM) diêthylfosforocyanidaat; vervolgens werd druppelsgewijs 55 3 ml droge DMF-oplossing met 0,56 g (5,5 mM) triëthylamine toegevoegd gedurende 5 minuten bij 0eC. Het resulterende reactiemengsel werd gedurende 30 minuten bij 0°C geroerd en werd gedurende nog 1 uur bij kamertemperatuur geroerd. Aan dit reactiemengsel werd 30 ml water toegevoegd en werd geëxtraheerd met 194239 4 • ethylacetaat.
De ethylacetaatoplossing werd met water gewassen, gedroogd op watervrij natriumsulfaat en geconcentreerd. De rest werd onderworpen aan silicagelkolomchromatografie (geëlueerd met chloroform:ethytacetaat = 1:1), vervolgens werden de ruwe kristallen geherkristalliseerd uit benzeen-n-hexaan ter levering van 0,53 5 g 4-diëthoxyfosfinoylmethyl-N-(5-methyl-3-isoxazolyl)benzamide als kleurloze naalden.

Voorbeeld VII
5 1,80 gram (15 mM) indoline, 1,82 g (18 mM) triëthylamine en 0,37 g (3 mM) 4-dimethylaminopyridine werden opgelost in 30 ml droge dichloorethaan; aan deze oplossing werd langzaam druppelsgewijs 30 ml droge dichloormethaanoplossing met 5,21 g (15 mM) 4-diëthoxyfosfinoylmethylbenzoylchloride onder roeren bij 0°C toegevoegd. Nadat het roeren gedurende 10 uur bij kamertemperatuur voortgezet was, werd 50 ml water aan het reactiemengsel toegevoegd en met chloroform geëxtraheerd. De extracten werden op 10 watervrij natriumsulfaat gedroogd en geconcentreerd. De aldus verkregen rest werd volgens silicagelko-lomchromatografie gezuiverd (geëlueerd met chloroform:ethylacetaat = 1:1).
De ruwe kristallen werden geherkristalliseerd uit benzeen-n-hexaan ter levering van 3,90 g 1-(4-diëthoxyfosfinoylmethylbenzoyl)indoline als kleurloze kristallen.
Smeltpunt: 93,0-94,5°C.

Voorbeeld XXVII
45 0,77 gram (2,0 mM) 1 -(4-diëthoxyfosfinoylmethylbenzoyl)-1,2,3,4-tetrahydrochïnoline en 1,0 g (4,6 mM) fosforpentasulfide werden gesuspendeerd in een gemengd oplosmiddel bestaande uit 20 ml watervrij benzeen en 5 ml watervrije pyridine. Deze suspensie werd verhit en gedurende 7 uur gerefluxt. Na afkoeling van het reactiemengsel tot kamertemperatuur werd het mengsel in 50 ml ijswater gegoten, en werd de waterige fase aangezuurd met 4 N zoutzuur en vervolgens geëxtraheerd met chloroform. De chtoroform-50 oplossing werd gedroogd op watervrij natriumsulfaat en geconcentreerd.
De aldus verkregen rest werd gezuiverd volgens silicagelkolomchromatografie (geëlueerd met chloroform:ethylacetaat = 1:1). De ruwe kristallen werden geherkristalliseerd uit benzeen-n-hexaan ter levering van 0,23 g 1 -(4-diëthoxyfosfinoylmethylfenyl)thiocarbonyl]-1,2,3,4-tetrahydrochinoline als geelachtige kristallen.

Farmacologische proeven
De mate van toename (%) van de plasma hoge dichtheid lipoproteïne cholesterol (HDLC)-concentratie in het serum, en de mate van afname (%) van de plasma triglyceride (TG)-concentratie in het serum, welke teweeggebracht werden door een carboxamideverbinding met de algemene formule (I) volgens de uitvinding 25 werden als volgt uitgevoerd.
Proefmethoden
Er werden zeven weken oude mannelijke Wistar ratten gebruikt.
De proefverblnding werd oraal toegediend gedurende 2 dagen in een dagelijkse dosis van 300 mg/kg lichaamsgewicht aan elk van de zes (6) ratten in de testgroep. De doseringshulpstof was 0,5% 30 natriumcarboxymethylcellulose-oplossing en het doseringsvolume was 5 ml/kg lichaamsgewicht.
Overeenkomstig de methoden, toegepast bij de ratten in de testgroep werd aan elk van de zes (6) ratten in de controlegroep alleen 0,5% natriumcarboxymethylcellulose-oplossing toegediend.
Op dag 2, nadat gedurende 20 uur gevast was, werd uit de "jagular" sinus met een gehepariniseerde spuit bloed afgetapt. Plasma werd verkregen d.m.v. centrifugeren.
35 Plasma HDCL werd bepaald na precipitatie van de andere lipidefractie met heparine en Ca2+ [HDL-C Kit-N, (gefabriceerd door Nlhon Shoji Kabushild Kaisha)].
Plasma TG werd bepaald volgens de werkwijze Van Handel [Clin. Chem., 7, 241 (1961)], [Triglyceride G-Test Wako (gefabriceerd door Wako Pure Chemicals Co., Ltd.)].
