Abstract
La presente invención se refiere a la inmunoterapia convencional del cáncer resulta deficiente para expandir eficientemente los linfocitos T que se dirigen específicamente a células cancerosas en números suficientes para reducir significativamente el tamaño del tumor o el número de células cancerosas in vivo. Para superar esta limitación, se proporcionan en la presente nanopartículas recubiertas con moléculas MHC Clase I y/o Clase II que presentan antígenos tumor-específicos y moléculas co-estimuladoras y su uso para expandir linfocitos T antígeno-específicos anti-tumorigénicos a niveles no alcanzados en las técnicas inmunoterapéuticas actuales. Estos linfocitos T antígeno-específicos y anti-tumorigénicos incluyen linfocitos T citotóxicos, linfocitos T efectores, linfocitos T de memoria, y linfocitos T auxiliares que son necesario para iniciar y mantener una respuesta inmunitaria sustancial contra células metastáticas o no metastáticas cancerosas, pre-cancerosas, o neoplásicas in vivo. La presente invención describe un procedimiento sistémico para dirigirse a células cancerosas o pre-cancerosas que son células en circulación, como en linfomas, células metastáticas migratorias, y tumores sólidos.
