Abstract
La presente invención se refiere a una composición que comprende (a) una sonda que se une a la bilirrubina pero no se une significantemente al ácido graso que comprende una muteína de proteína enlazadora de lípidos intracelulares (iLBP) etiquetada con un primer fluoróforo, en donde la muteína de iLBP corresponde a SEQ ID NO: 3 comprende una o más sustituciones de aminoácidos en posiciones seleccionadas del grupo que consiste en las posiciones 14, 18, 23, 25, 27, 31, 36, 38, 55, 60, 72, 73, 74, 78, 102, 104, 106, 115 y 117, y en donde el primer fluoróforo está unido a un residuo de lisina, el grupo amino de término N de la iLBP, o una substitución de cisteína, y (b) un segundo fluoróforo, en donde el segundo fluoróforo está libre en la solución o unido a una proteína que no se une a la bilurrubina, y en donde el primer fluoróforo y el segundo fluoróforo son capaces de la excitación en la misma longitud de onda, y en donde la longitud de onda de emisión del primer fluoróforo y el segundo fluoróforo es diferente, y en donde el segundo fluróforo no cambia su emisión en respuesta a la bilurribuna que se une a la muteína de iLBP.
