Abstract
La presente invención se refiere al campo de la fermentación industrial. En particular, se refiere a un proceso para cultivar una célula huésped de Bacillus que comprende las etapas de (a) inocular un medio de fermentación con una célula huésped de Bacillus que comprende un constructo de expresión para un gen que codifica una proteína de interés, cultivar durante una primera fase de cultivo la célula huésped de Bacillus en dicho medio de fermentación en condiciones propicias para el crecimiento de la célula huésped de Bacillus y la expresión de la proteína de interés, en donde el cultivo de la célula huésped de Bacillus comprende la adición de al menos una solución de alimentación y en donde la al menos una solución de alimentación proporciona una fuente de carbono a velocidades en aumento, y (c) cultivar durante una segunda fase de cultivo el cultivo de la célula huésped de Bacillus obtenido en la etapa (b) en condiciones propicias para el crecimiento de la célula huésped de Bacillus y la expresión de la proteína de interés, en donde el cultivo comprende la adición de al menos una solución de alimentación y en donde la al menos una solución de alimentación proporciona una fuente de carbono a una velocidad constante, a velocidades disminuidas o a velocidades que aumentan menos que las velocidades en la etapa (b), en donde dicha velocidad constante o la velocidad inicial de dichas velocidades o la velocidad inicial de dichas velocidades que aumenta menos que las velocidades en la etapa (b) está por debajo de la velocidad máxima de la primera fase de cultivo. Se contempla además un cultivo de células huésped de Bacillus que se puede obtener mediante dicho proceso.
