Abstract
MÉTODO PARA CULTIVAR UMA CÉLULA HOSPEDEIRA DE BACILLUS, E, CULTURA DE CÉLULAS HOSPEDEIRAS DE BACILLUS. A presente invenção refere-se ao campo da fermentação industrial. Em particular, refere-se ao método para cultivar uma célula hospedeira de Bacillus compreendendo as etapas de (a) inocular um meio de fermentação com uma célula hospedeira de Bacillus compreendendo um construto de expressão para um gene que codifica uma proteína de interesse, cultivar, para uma primeira fase de cultivo, a célula hospedeira de Bacillus no dito meio de fermentação em condições favoráveis para o crescimento da célula hospedeira de Bacillus e da expressão da proteína de interesse, em que o cultivo da célula hospedeira de Bacillus compreende a adição de pelo menos uma solução de alimentação, e em que a pelo menos uma alimentação solução provê uma fonte de carbono em taxas crescentes, e (c) cultivar, para uma segunda fase de cultivo, a cultura de células hospedeiras de Bacillus obtida na etapa (b) em condições favoráveis para o crescimento da célula hospedeira de Bacillus e da expressão da proteína de interesse, em que o cultivo compreende a adição de pelo menos uma solução de alimentação, e em que a pelo menos uma solução de alimentação provê uma fonte de carbono em uma taxa constante, em taxas decrescentes ou em taxas que aumentam menos que as taxas na etapa (b), em que a dita taxa constante ou a taxa inicial das ditas taxas decrescentes ou a taxa inicial das ditas taxas que aumentam menos que as taxas na etapa (b) está abaixo da taxa máxima da primeira fase de cultivo. É adicionalmente contemplada uma cultura de células hospedeiras de Bacillus que pode ser obtida pelo dito método.
