Abstract
La presente invención se refiere a un procedimiento para recuperar y purificar hormona del crecimiento monomerica, biológicamente activa, de cuerpos de inclusión insolubles producidos por expresión de un gene heterólogo en un microorganismo, caracterizado porque comprende: a) solubilizar y desnaturalizar la hormona del crecimiento extrayendo los cuerpos de inclusión en una solución de una sal de guanidina; b) renaturalizar cuando menos una porción de la hormona del crecimiento en la solución, reemplazando la solución de sal de guanidina por una solución reguladora libre de desnaturalizante, sin el uso de un desnaturalizante intermedio; c) separar los contaminantes precipitados y agregados de la solución reguladora libre de desnaturalizante; y d) purificar la hormona del crecimiento monomerica en la solución, mediante cromatografía de cambio de iones.
