Abstract
La presente invención se refiere a procedimiento para la producción del polideoxirribonucleótido Defibrotide, a partir de una solución de ácido nucléicos de partida, cuya solución se encuentra al menos substancialmente libre de polisacáridos y proteínas, caracterizado porque comprende las etapas de: 1) formar polideoxirribonucleótidos altamente polimerizados e interrumpidos por agregación estabilizante de los ácidos nucléicos de partida, ajustar, si es necesario, la solución a una fuerza iónica predeterminada de por lo menos 1 molar y ajustar el pH de la solución hasta alcanzarse la máxima hipercromicidad reversible; 2) despolimerizar los polideoxirribonucleótidos hasta la hipercromicidad reversible de la solución se encuentra en un valor de h igual a 15 + (-) 5, medido como un porcentaje en DNA natural, por calentamiento de la solución resultante a una temperatura de despolimerización para dicho polideoxirribonucleótido y manteniendo la solución a una temperatura de despolimerización hasta conseguirse dicho valor de hipercromicidad reversible; 3) detener entonces la reacción de despolimerización y separar los enlaces hidrógeno de cualquier fragmento de doble filamento en la solución despolimerizada, para formar fragmentos polideoxirribonucleotídicos de un solo filamento; y 4) estabilizar posteriormente los fragmentos de polideoxirribonucleótidos de un solo filamento, resultantes, por calentamiento de la solución resultante a una temperatura y a un pH mayores que los correspondientes parámetros utilizados en la reación de despolimerización, para evitar la reformación de los enlaces hidrógeno.
