Abstract

PURPOSE: Homogenized milt of fish is made to a specific acidic value in pH to permit agitation without dilution with water and prevent the deterioration caused by deamination or others and followed by extraction with alkali, thus isolating DNA, an intermediate for drugs or food.
CONSTITUTION: After homogenized, milt of fish is adjusted to 4.5W6.5 in pH by inorganic or organic acid and stirred at 80W100°C for 0.5W2 hr. Then, an alkali aqueous solution is added to adjust the pH to over 8 and the stirring is continued, the solid part is separated and an acid is added to the filtrate to isolate deoxyribonucleic acid (DNA). The adjustment of the homogenized milt of fish to a specific pH value permits the increase in quantity to be treated per batch and the deterioration caused by deamination or others on heating can be prevented.
