Abstract
본 발명은 내독소(ET)의 활성을 저해하기 위하여 ET와 결합하는 성질을 가진 펩티드 유도체 또는 단백질 및 하나 이상의 계면활성제를 ET를 함유하는 시료와 접촉시킴으로서 ET의 활성을 저해시킬 수 있으며, 심지어 ET활성을 가지는 ET 유사물질이 처리한 시료중에 존재해 있을 때에도 이 물질을 ET의 효과없이도 측정할 수 있는 내독소 저해 방법에 관한 것이다.

제 1 도는 실시예 1 에서 수득하였고, 혈장내 내독소(이하 "ET"라 약칭함)(대장균 055:B5 LPS) 측정에 대한 계면활성제 농도의 효과를 보이는 그래프이다' 제2도는 실시예 1 에서 수득하였고, 혈장내 내독소(대장균 055:B5 LPS) 측정에 대한 폴리믹신B(이하 "Px B"라 약칭함) 농도의 효과를 보이는 그래프이다.
Claims (17)
- 내독소의 활성을 저해시키기 위하여 내독소에 결합하는 성질이 있으며, 펩티드 유도체와 단백질로부터 선택된 내독소를 저해하는 펩티드 및 하나 이상의 계면활성제를 내독소 함유 시료와 접촉시키는 것을 포함하는 내독소 활성의 저해 방법.
- 제 1 항에 있어서, 열처리를 동시에 수행하는 방법.
- 제 1 항에 있어서, 내독소를 저해하는 펩티드 및 계면활성제가 시료와 접촉한 이후에 시료를 열처리하는 방법.
- 제 1 항에 있어서, 계면활성제의 농도는 0.005~5.0w/v%이고, 내독소를 저해하는 펩티드의 농도는 0.00001~0.1w/v%인 방법.
- 제 2 항에 있어서, 60~220℃에서 3~60분동안 가열시키는 열처리를 수행하는 방법.
- 제 1 항에 있어서, 내독소를 저해하는 펩티드가 폴리믹신인 방법.
- 제 1 항에 있어서, 계면활성제가 비이온성 계면활성제 또는 양성(兩性) 계면활성제인 방법.
- 상기 제 1 항의 방법으로 처리한 시료를 참게의 혈구 용해물과 반응시키고, 상기 반응에 의해 일어난 효소 활성화 반응으로 활성화된 효소의 활성 또는 상기 반응에 의해 일어난 젤화 반응에 기인하는 반응 용액의 탁도 변화나 젤화 정도를 측정기구를 사용하거나 나안으로 측정하는 것을 포함하며, 시료중에 존재하고, 참게의 혈구 용해물과 반응하여 효소 활성화 반응 또는 젤화 반응을 일으키는 성질을 가진 하나 이상의 내독소 유사물질을 측정하는 방법.
- 제 8 항에 있어서, 내독소 유사물질이 (1→3)-β-D-글루칸 및/또는 그 유도체인 방법.
- 참게의 혈구 용해물과 반응하지 못하고, 참게의 혈구 용해물과 내독소 유사물질과의 반응을 저해 및 촉진시킬 수도 없는 용액으로, 내독소를 저해하는 펩티드 및 하나 이상의 계면활성제를 함유하는, 내독소의 활성을 저해하는 전처리 수용액.
- 제10항에 있어서, 계면활성제 농도는 0.01~10.0w/v%이고 내독소를 저해하는 펩티드의 농도는 0.00002~0.2w/v%인 전처리 수용액.
- 제10항에 있어서, 내독소를 저해하는 펩티드가 폴리믹신인 전처리 수용액.
- 제10항에 있어서, 계면활성제가 비이온성 계면활성제 또는 양성 계면활성제인 전처리 수용액.
- 내독소를 저해하는 펩티드, 하나 이상의 계면활성제 및 AL 용액을 함유하는 하나 이상의 내독소 유사물질을 측정하는 시약.
- 제14항에 있어서, 내독소를 저해하는 펩티드가 폴리믹신인 시약.
- 제14항에 있어서, 계면활성제가 비이온성 계면활성제 또는 양성 계면활성제인 시약.
- 제14항에 있어서, 내독소 유사물질이 (1→3)-β-D-글루칸 및/또는 그 유도체인 시약.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
