Abstract
본 발명은 유전자 재조합된 대장균에서 발현된 닭 성장 호르몬을 정제하는데 있어서, (가) 닭 성장 호르몬이 발현된 대장균을 분쇄하여 불용성의 닭 성장호르몬 굴절체를 분리하고, (나)분리된 불용성 굴절체를 계면활성제 및 에틸렌디아민테프라아세트산이 함유된 용액으로 세척한 다음 다시 100 내지 500mM 염화나트륨 또는 증류수로 세척하고, (다) 불용성 굴절체를 수산화나트륨을 사용하여 pH11.5 내지 13.0에서 용해시킨 후, (라) 원심분리 및 한 외여과법에 의해 용해되지 않은 침전물을 제거하고, (마) 음이온 교환수지 컬럼 크로마토그래피하여서 닭성장 호르몬을 정제함으로써, 닭 성장 호르몬을 저렴한 비용으로 고순도 및 고수율로 대량 정제하는 방법을 제공하는 것이다.

제1도는 닭 성장 호르몬의 아미노산 서열을 나타낸 것이고, 제2도는 닭 성장 호르몬이 발현된 대장균 추출액을 소듐 도데실 설페이트 폴리아크릴 아마이드 젤 전기영동(SDS-PAGE)한 결과를 나타낸 것이며, 제3도는 정제된 닭 성장 호르몬을 고성능 액체 크로마토그래피(HPLC)로 분석한 결과이다.
Claims (4)
- 유전자 재조합된 대장균에서 발현된 닭 성장호르몬을 정제하는 방법에 있어서, (가)닭 성장 호르몬이 발현된 대장균을 분쇄하여 불용성의 닭 성장 호르몬 굴절체를 분리하고, (나)분리된 불용성 굴절체를 계면활성제 및 에틸렌디아테트라아세트산이 함유된 용액으로 세척한 다음 농도 100 내지 500mM의 염화나트륨 또는 증류수로 세척하고, (다)불용성 굴절체에 수산화나트륨을 첨가하여 pH11.5에서 용해시킨 후, (라) 원심분리 및 한외여과법에 의해 용해되지 않은 침전물을 제거하고, (마)음이온 교환수지 컬럼 크로마토그래피로 분리하는 단계를 포함하는 방법.
- 제1항에 있어서, 상기 (나)단계의 계면활성체가 0.01 내지 1%농도의 트리톤 엑스-100(Triton X-100)인 방법.
- 제1항에 있어서, 상기 (라)단계의 한회여과는 분자량 30만 이하의 물질만을 통과시키는 한회여과막을 사용하는 것인 방법.
- 제1항에 있어서, 상기 (마)단계의 음이온 교환 수지 컬럼 크로마토그래피는 pH8.5 내지 pH10.5에서 실시하며, 음이온 교환 수지가 디에틸아미노에틸을 작용하기로 하고, 용출제로서 70 내지 150mM의 염화나트륨 용액을 사용하는 것인 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
