제1도 식물세포의 단면, 제2도 Aspergillus niger DS16813(CBS 323.90)로 부터 얻은 배양 여액의 HPLC 용출 곡선, 이 균주는 나중에 종 Aspergillus tubigensis에 더 속하는것 같아 재분류되었다, 제3도 Aspergillus tubigensis XYL A단백질(식1)의 N-말단 아미노산 서열로부터 설계된 올리고 뉴클레오티드 프로브 AB801-AB806.
Claims (25)
- 크실라나아제 활성을 가진 폴리펩티드를 코드하는 정제 및 단리된 DNA서열에 있어서, DNA서열이, (a)크실라나아제 활성을 가진 폴리펩티드를 코드하는 곰팡이원의 DNA서열, (b)상기 (a)의 서열의 유전자 변종, (c)상기 (a) 및 (b)의 어느 한 서열로 혼성화될수 있는 DNA서열로 구성된 군으로부터 선택되는 특징으로 정제 및 단리된 DNA 서열.
- 제1항에 있어서, DNA서열이 Aspergillus, Disporotrichum, Penicillium, Neurospora, Fusarium 및 Trichoderma로 구성된 속으로 부터 선택된 곰팡이에서 유래하는 것을 특징으로 하는 정제 및 단리된 DNA서열.
- 제2항에 있어서, DNA서열이 Aspergillus tubigensis, Aspergillus niger, Aspergillus awamori, Aspergillus aculeatus, Disporotrichum dimorphosporum 및 Trichoderma reesei로 구성된 군으로 부터 선택된 곰팡이 종에서 유래하는 것을 특징으로 하는 정제 및 단리된 DNA 서열.
- 제1항에 있어서, DNA서열이, a)제8도에 기재된 DNA서열이, (b)상기 a)의 서열의 유전자 변종, c)상기 a) 및 b)의 어느한 서열로 혼성화될수 있는 DNA서열로 구성된 군으로부터 선택되는 특징으로 정제 및 단리된 DNA서열.
- 크실라나아제 활성을 가진 폴리펩티드의 적당한 발현숙주내 과발현을 지시할수 있는 조절영역에 작동적으로 연결되어 제1항의 DNA 서열을 포함하고 있는 특징으로 하는 DNA 구성체.
- 제5항에 있어서, 크실라나아제를 코드하는 유전자가 Aspergillus, Disporotrichum, Penicillium, Neurospora, Fusarium 및 Trichoderma로 구성된 속으로부터 선택된 곰팡이에서 유래하는 것을 특징으로 하는 DNA 구성체.
- 제5항에 있어서, 곰팡이 크실라나아제를 코드하는 유전자가 Aspregillus tubigensis, Aspergillus niger, Aspergillus awamori, Aspergillus aculeatus, Disporotrichum dimorphosporum 및 Trichoderma reesei로 구성된 군으로 부터 선택된 곰팡이종에서 유래하는 것을 특징으로 하는 DNA 구성체.
- 제5항에 있어서, 조절영역은 아밀로글루코시다아제 유전자에서 유래된 프로모터 및 크실라나아제 유전자에 천연적인 프로모터로 구성된 군으로부터 선택된 프로모터를 포함하는 것을 특징으로 하는 DNA구성체.
- 제5항에 있어서, 조절영역이 아밀로글루코시다아제 유전자에서 유래된 분비리이더 서열 및 크실라나아제 유전자에 천연적인 분비 리이더 서열로 구성된 군으로부터 선택된 분비 리이더 서열을 포함하는 것을 특징으로 하는 DNA구성체.
- 곰팡이 크실라나아제를 과발현할수 있는 형질전화된 미생물 숙주에 있어서, 그 미생물 숙주가 제5항내지 제9항중의 어느 한 항에 의한 발현 구성체를 내포하고 있는 것을 특징으로 하는 형질전환된 미생물 숙주.
- 제10항에 있어서, 미생물 숙주가 Aspergillus, Kluyveromyces, Trichoderma, Saccharomyces 및 Bacillus구성된 속으로 부터 선택되는 것을 특징으로 하는 형질전환된 미생물 숙주.
- 제10항에 있어서, 미생물 숙주가 Aspergillus tubigensis, Aspergillus niger, Aspergillus awamori, Aspergillus aculeatus, Aspergillus, Oryzae Trichoderma reesei, Bacillus subtilis, Bacillus licheniformis, Kluyveromyces latis 및 Saccharomyces cerevisiae에 구성된 속으로부터 선택되는 것을 특징으로 하는 형질전환된 미생물 숙주.
- 크실라나아제를 활성을 가진 폴리펩티드를 과발현하는 방법에 있어서, a)크실라나아제 활성을 가진 풀립펩티드를 코드하는 유전자를 발현시키기에 적당한 조건하에서 제10항에 의한 미생물 숙주를 배향하는 단계, 및b)크실라나아제 활성을 가진 폴리펩티드를 회수하는 단계로 되어 있는 것을 특징으로 하는 방법.
- 폴리펩티드가 제13항의 방법에 의해 생산되는 것을 특징으로 하는 크실라나아제 활성을 가진 폴리펩티드.
- 크실란을 함유하는 기질의 분해에 제14항에 의한 크실라나아제 활성을 가진 폴리펩티드의 이용.
- 동물사료 조제에 제14항에의한 크실라나아제 활성을 가진 폴리펩티드의 이용.
- 제14항에 의한 크실라나아제 활성을 가진 폴리펩티드를 함유하는 것을 특징으로 하는 동물사료조성물.
- 빵 제조용 반죽에 제14항에 의한 크실라나아제 활성을 가진 폴리펩티드의 이용.
