Claims (13)

  1. 수불용성 입자에 공유결합된 비면역반응성 단백질이나 탄수화물을 포함하는 수불용성 시약에 있어서, 비면역반응성 단백질이나 탄수화물이 6 또는 그 미만의 pI를 갖고 아실화제, 아킬화제 또는 설포닐화제로 화학적 개질되었으며 입자 표면에 기타 단백질이나 탄수화물이 실질적으로 없는 것으로 특징되는 수불용성 시약.
  2. 수불용성 입자에 결합된 올리고누클레오타이트로 구성된 수불용성 헥산 프로브에 있어서, 결합이 단백질이나 탄수화물인 결합 그룹을 통해 이루어지며 단백질이나 탄수화물이 6 또는 그 미만의 pI를 갖고 올리고누클레오 타이드와의 공유결합 반응에 대한 그룹들을 제공하기 위해 아실화제, 아킬화제 또는 설포닐화제로 화학적 개질되며 기타 단백질이나 탄수화물이 실질적으로 없는 입자 표면에 공유결합되는 것으로 특징되는 프로브.
  3. 제2항에 있어서, 올리고누클레오타이드가 HIV-I DNA 사슬과 상보적인 핵산 서열인 프로브.
  4. 제2항 또는 제3항에 있어서, 결합되어있는 검출 가능한 라벨을 갖는 프로브.
  5. 제1항 내지 제4항중 어느 한 항에 있어서, 수불용성 입자가 0.1-10㎛의 평균 직경을 갖는 합성 중합체 입자인 수불용성 시약 및 프로브.
  6. 제1항 내지 제5항중 어느 한 항에 있어서, 중합체 입자가 카복시, 활성인 할로겐 원자, 활성화된 2-치환 에틸설포닐, 비닐설포닐 및 비닐설포닐알킬렌 그룹으로 구성된 그룹으로 부터 선택된 반응 그룹을 갖는 단량체로 부터 제조되는 수불용성 시약 및 프로브.
  7. 제1항 내지 제6항중 어느 한 항에 있어서, 화학적으로 개질된 단백질이나 탄수화물이 숙시닐화된 카제인, 글루타릴화된 카제인, 숙시닐화된 클라겐, 숙시닐화된 소의 혈청 알부민, 카복시메틸 셀룰로스 또는 카복시에틸 셀룰로스인 수용성 시약 및 프로브.
  8. A. 타겟 핵산을 함유하고 있는 테스트 샘플을 제2항 내지 제7항중 어느 한 항에 수불용성 핵산 프로브와 접촉시키는데, 이러한 프로브의 올리고누클레오타이드는 실질적으로 타겟 핵산에 상보적이며 상기 접촉시키는 것을 타겟 핵산과 올리고누클레오타이드의 잡종형성이 이루어져 수불용성의 잡종형성 생성물이 만들어지도록 하는 잡종형성 조건하에서 행하고, B. 잡종형성된 생성물을 나머지 테스트 샘플로 부터 분리하는 것으로 구성되는 타겟 핵산을 포획 및 분리하는 방법.
  9. A. 타겟 핵산을 함유하고 있는 테스트 샘플을 제2항 내지 제7항중 어느 한 항의 수불용성 핵산 프로브와 접촉시키는데, 이러한 프로브의 올리고누클레오타이드는 실질적으로 타겟 핵산에 상부족이며 상기 접촉시키는 것을 타겟 핵산과 올리고누클레오타이드의 잡종형성이 이루어져 수불용성의 잡종형성 생성물이 만들어지도록 하는 잡종형성 조건하에서 행하고, B. 테스트 샘플내 타겟 헥산의 존재 표시로서 잡종형성된 생성물을 검출하는 것으로 구성되는 타겟 핵산의 검출방법.
  10. 제9항에 있어서, 수불용성 핵산 프로브가 라벨링되지 않은 포획 프로브이고 타겟 핵산이 또한 라벨링된 수용성 프로브와 잡종분자를 형성하는 샌드위치 잡종형성 분석법으로 행해지는 방법.
  11. 제9항 또는 제10항에 있어서, 타겟 핵산이 단계 A의 접촉 단계전에 중합효소 연결반응을 이용해 증폭되는 방법.
  12. (a) 제2항 내지 제7항중 어느 한 항에 수불용성 핵산 프로브, 및(b) 수불용성 핵산 프로브의 올리고누클레오타이드에 실질적으로 상보적인 타겟 핵산에 상보적인 수용성 프라이머 또는 프로브를 포함하는 진단 테스트키트.
  13. 제12항에 있어서, 타겟 핵산의 중합효소 연결반응에 필요한 시약을 더 포함하는 진단 테스트 키트.
