안넥신의 정제방법

제1도는 페닐-20 세파로즈 급속 유동 크로마토그래피의 용출 프로파일(←→=VAC-풀(fool))이다. 제2도는 Q-세파로즈 급속 유동 크로마토그래피의 용출 프로파일(←→=VAC-풀)이다.

Claims (15)

  1. a) 균질화된 세포물질의 pH값을 8.0 내지 10.0으로 조절하고, b) Ca2+, Cd2+, Zn2+, Mn2+및 Co2+중에서 선택된 하나 이상의 이가 양이온을 가하며, c) 인지질을 가하고, d) 불용성 세포 잔사를 세척하여 용해성 조성물을 제거하며, c) 킬레이트화제를 사용하여 안넥신을 세포잔사로부터 추출하는 단계를 특징를 하여, 안넥신을 정제하는 방법.
  2. 제1항에 있어서, 이온성 그룹을 가진 수지를 단계 (e) 이후 가하고 형성된 침전물을 분리하는 방법.
  3. 제2항에 있어서, 수지를 강한 양이온성 그룹으로 작용화시키는 방법.
  4. 제1항 또는 2항에 있어서, 단계(e)에서 수득한 안넥신을 이전에 공지된 방법으로 단백질 침전 및 연속 단백질 정제 단계에 의해 추가로 정제하는 방법.
  5. 제1항 또는 2항에 있어서, 단계(a)에서 pH를 9.0으로 조절시킨 방법.
  6. 제1항 내지 3항중 어느 한 항에 있어서, ca2+을 단계(b)에서 이가 양이온으로서 가하는 방법.
  7. 제6항에 있어서, Ca2+및 Zn2+를 단계(b)에서 이가 양이온으로서 가하는 방법.
  8. 제1항 내지 7항중 어느 한 항에 있어서, 레시틴을 단계(c)에서 인지질로서 가하는 방법.
  9. 제1항 내지 8항중 어느 한 항에 있어서, 단계(b) 및 (c)를 실질적으로 동시에 수행하는 방법.
  10. 제1항 내지 9항중 어느 한 항에 있어서, EDTA를 단계(e)에서 킬레이트화제로서 사용하는 방법.
  11. 제1항 내지 10항중 어느 한 항에 있어서, 세포를 균질화 및 단계(a) 이전에 비활성시키는 방법.
  12. 제11항에 있어서, 세포를 벤젠, 톨루엔, 페놀, 크실렌 및 크레졸 중에서 선택된 제제에 의해 비활성시키는 방법.
  13. 제12항에 있어서, m-크레졸을 비활성화제로서 사용하는 방법.
  14. 제1항 내지 13항중 어느 한 항에 있어서, 정제할 안넥신이 VAC 또는 이의 유사체인 방법.
  15. 제1항 내지 14항중 어느 한 항에 따른 방법으로 수득한 안넥신.
